Synergic activity of oligostyrylbenzenes with amphotericin B against Candida tropicalis biofilms.

Antimicrobial drug resistance is a serious challenge in clinical settings worldwide, with biofilm formation having been associated with this problem. In the present study, the synergism of oligostyrylbenzene (OSB) compounds in combination with amphotericin B (AmB) against Candida tropicalis biofilms was investigated. In addition, the toxicity in human blood cells was determined. Synergistic combinations of OSBs and AmB were evaluated to consider future effects of OSBs in vivo. The checkerboard microdilution method was used to study the interactions of one anionic (1) and two cationic (2 and 3)

Candida tropicalis biofilm's matrix--involvement on its resistance to amphotericin B.

Candida tropicalis has emerged as one of the most prevalent fungal pathogens, and its ability to form biofilms has been considered one of the most important virulence factors, since they represent high tolerance to antifungal agents. However, the mechanisms of biofilm resistance to antifungal agents remain poorly understood. Thus, the main goal of this study was to infer about the ability of amphotericin B (AMB) to control and combat C. tropicalis biofilms. Additionally, it was also intended to determine the influence of matrix components in biofilm resistance. AMB was unable to totally preven

EQUAL Candida score, an effective tool for predicting the outcomes of Candida tropicalis candidaemia: A retrospective cohort study.

BACKGROUND: Candida tropicalis is the most common non-albicans Candida species found in Asia-Pacific countries, including Thailand. The pathogen is known for its great virulence, which causes a high case-fatality rate. Associations between case fatality and patient characteristics, infectious disease unit consultation and EQUAL Candida score were investigated. METHODS: This retrospective cohort study was conducted with 160 cases of C. tropicalis bloodstream infection between 2015 and 2019 at a single, large, tertiary centre in Thailand. Clinical characteristics, clinical presentations, patien

Biofilm formation directly correlates with cell viability in Candida tropicalis on polypropylene.

Candida tropicalis, the most prevalent non-Candida albicans Candida species, is an emerging pathogen forming robust biofilms on medical devices, contributing to biofouling, virulence, and antifungal resistance. In this study, growth conditions for six C. tropicalis clinical isolates (C4, U873, U951, U1179, U1309, U1360) and a standard strain (MTCC-184) were optimized on polypropylene using central composite design-based response surface methodology. The parameters tested included temperature, pH, shaker speed, inoculum size, and incubation time, with biofilm formation quantified by crystal vio

Research progress on the drug resistance mechanisms of Candida tropicalis and future solutions.

Candida tropicalis is an important member of the non-Candida albicans species. It is closely associated with candidemia, especially common in neutropenic and critically ill patients. Drug-resistant C. tropicalis isolates have been found not only in clinical patients but also in animals, fruits, and the environment. In recent years, the detection rate of azole-resistant C. tropicalis isolates has increased. Drug-resistant C. tropicalis is related to persistent, recurrent, and breakthrough infections. Therefore, understanding its drug resistance is crucial for clinical treatment. The review expl

Elbow osteomyelitis by Candida tropicalis in acute lymphoblastic leukemia. A case report.

INTRODUCTION: musculoskeletal involvement of the extremities is common in pediatric patients with acute lymphoblastic leukemia. The insidious and oligosymptomatic presentation of several musculoskeletal conditions in these patients often leads to different diagnostic suspicions, which may delay appropriate management of the pathology. Non-albicans Candida osteoarticular infections are a rare entity that may affect immunodeficient patients. We describe a case of osteoarticular infection caused by an opportunistic fungus in a pediatric patient with acute lymphoblastic leukemia during the second

Candida tropicalis-Associated Osteomyelitis in an Intravenous Drug User: A Case Report.

Candida species represent the most frequent fungal cause of osteomyelitis in patients with a history of drug use, with Candida albicans historically being the predominant pathogen. However, non-albicans Candida species, particularly Candida tropicalis, have emerged as increasingly important pathogens associated with more complex clinical presentations and treatment challenges. Candida tropicalis (C. tropicalis) demonstrates enhanced virulence factors, including superior adherence capabilities and biofilm formation, contributing to its pathogenicity in immunocompromised hosts and those with ind

Metabolic Engineering of Candida tropicalis for the De Novo Synthesis of β-Ionone.

β-ionone, a norisoprenoid, is a natural aromatic compound derived from plants, which displays various biological activities including anticancer, antioxidant and deworming properties. Due to its large biomass and strong environmental tolerance, the nonconventional oleaginous yeast Candida tropicalis was selected to efficiently synthesize β-ionone. We initially investigated the capacity of the cytoplasm and subcellular compartments to synthesize β-ionone independently. Subsequently, through adaptive screening of enzymes, functional identification of subcellular localization signal peptides and

Systematic metabolic engineering for improved synthesis of perillic acid in Candida tropicalis.

Perillic acid has been studied as an anticancer and antimicrobial drug. Production of perillic acid has attracted considerable attention. Meanwhile, Candida tropicalis is an unconventional diploid yeast, most significantly characterized by its ability to metabolize alkanes or fatty acids for growth and proliferation. Therefore, perillic acid's precursor (L-limonene) in C. tropicalis was firstly synthesized by expressing a Mentha spicata L-limonene synthase gene, LS_Ms in this work. Expression of a gene which encoded for a truncated version of tLS_Ms increased the production of L-limonene with

Candida tropicalis distribution and drug resistance is correlated with ERG11 and UPC2 expression.

BACKGROUND: Candida tropicalis (C. tropicalis) is an important opportunistic pathogenic Candida species that can cause nosocomial infection. In this study, we analyzed the distribution and drug susceptibility of C. tropicalis and the relationship between ERG11 and UPC2 expression and resistance to azole antifungal agents. METHODS: C. tropicalis was cultured and identified by Sabouraud Agar Medium, CHROM Agar Candida and ATB tests (Bio-Mérieux, France). Total RNA was extracted from the collected strains, and the ERG11 and UPC2 mRNA expression levels were analyzed by quantitative real-time PCR.

Fungus ball and emphysematous cystitis secondary to Candida tropicalis: A case report.

Fungus ball and fungal emphysematous cystitis are two rare complications of fungal urinary tract infection. A 53-year-old male patient presented with these complications caused by Candida tropicalis simultaneously. The predisposing factors were diabetes mellitus and usage of broad-spectrum antibiotics. The fungus ball, measuring 3.5 × 2.0 cm on the left wall of the urinary bladder, shrank significantly to 1.6 × 0.8 cm after 5 days of intermittent irrigation with saline before surgery. With transurethral removal of the fungus ball and antifungal treatment with fluconazole, the patient fully rec

The Gastrointestinal Tract Is Pinpointed as a Reservoir of Candida albicans, Candida parapsilosis, and Candida tropicalis Genotypes Found in Blood and Intra-Abdominal Samples.

BACKGROUND: Candida spp., as part of the microbiota, can colonise the gastrointestinal tract. We hypothesised that genotyping Candida spp. isolates from the gastrointestinal tract could help spot genotypes able to cause invasive infections. MATERIALS/METHODS: A total of 816 isolates of C. albicans (n = 595), C. parapsilosis (n = 118), and C. tropicalis (n = 103) from rectal swabs (n = 754 patients) were studied. Genotyping was conducted using species-specific microsatellite markers. Rectal swab genotypes were compared with previously studied blood (n = 814) and intra-abdominal (n = 202) genot

Moonlighting proteins are variably exposed at the cell surfaces of Candida glabrata, Candida parapsilosis and Candida tropicalis under certain growth conditions.

BACKGROUND: Adaptability to different environmental conditions is an essential characteristic of pathogenic microorganisms as it facilitates their invasion of host organisms. The most external component of pathogenic yeast-like fungi from the Candida genus is the multilayered cell wall. This structure is composed mainly of complex polysaccharides and proteins that can undergo dynamic changes to adapt to the environmental conditions of colonized niches. RESULTS: We utilized cell surface shaving with trypsin and a shotgun proteomic approach to reveal the surface-exposed proteins of three import
