A novel approach to resilience and its links with education and Alzheimer's disease genetics.

INTRODUCTION: Cognitive resilience refers to maintaining cognitive function despite Alzheimer's disease (AD) pathophysiology. METHODS: We analyzed amyloid-positive individuals across clinical stages of AD in two cohorts: the Amsterdam Dementia Cohort (ADC, N = 1036) and Alzheimer's Disease Neuroimaging Initiative (ADNI, N = 685). Cognitive resilience was conceptualized from a canonical correlation analysis of magnetic resonance imaging and neuropsychological data in each cohort separately. Model validation involved education as a resilience proxy and key genetic factors (apolipoprotein E [APO

A data-driven investigation of relationships between bipolar psychotic symptoms and schizophrenia genome-wide significant genetic loci.

The etiologies of bipolar disorder (BD) and schizophrenia include a large number of common risk alleles, many of which are shared across the disorders. BD is clinically heterogeneous and it has been postulated that the pattern of symptoms is in part determined by the particular risk alleles carried, and in particular, that risk alleles also confer liability to schizophrenia influence psychotic symptoms in those with BD. To investigate links between psychotic symptoms in BD and schizophrenia risk alleles we employed a data-driven approach in a genotyped and deeply phenotyped sample of subjects

Relationship of differentiation of self to adult clients' perceptions of the alliance in brief family therapy.

The authors examined differentiation of self, a multidimensional construct in Murray Bowen's (1976, 1978) family systems theory, as a predictor of clients' perceptions of the therapeutic alliance. Adult clients (N=38) in brief family therapy completed the Differentiation of Self Inventory-Revised (Skowron & Schmitt, 2003) during prescreening and the self-report version of the System for Observing Family Therapy Alliances (Friedlander & Escudero, 2002) after Session 3. More differentiated adults reported significantly more positive overall alliances. In a canonical correlation analysis, four ro

Disentangling white matter correlates of symptom severity and general impairment in early psychosis.

White matter (WM) alterations are well documented in individuals at clinical high-risk for psychosis (CHR-P) and with recent-onset psychosis (ROP), yet it remains unclear whether they reflect vulnerability, psychosis-related symptom severity, or general impairments shared with other disorders such as depression. To disentangle these correlates, we analyzed diffusion MRI data from 882 individuals (457 females) of the multisite Personalized Prognostic Tools for Early Psychosis Management (PRONIA) study, including 183 CHR-P, 206 ROP, 298 healthy controls, and 195 individuals with depression who w

Integration of Transcriptomics and Metabolomics Reveals Organ-Specific Biosynthesis and Accumulation of Pharmacologically Active Flavonoids in Rhododendron yedoense var. poukhanense.

Rhododendron yedoense var. poukhanense is an important medicinal plant, but still little is known about how its bioactive flavonoids are made in different organs. Comprehensive metabolomic profiling was performed using ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS), coupled with reference-based RNA sequencing (RNA-seq), to analyze root, stem, and leaf tissues from three independent biological replicates. Subsequently, we performed two-way orthogonal partial least squares (O2PLS) regression, canonical correlation analysis (CCA), and Pearson correlation analyses. A

Concomitant Dysregulation of Cerebral Vasoreactivity and Arterial Blood Pressure Is Closely Related in Patients with Carotid Stenosis.

Background: In patients with severe atherosclerotic internal carotid artery stenosis (ICAS), the capacity of cerebral vasoreactivity (CVR)-an independent risk factor for cerebral ischemia-is reduced, and dysregulation of arterial blood pressure (ABP) may also be present. Thus, this study assessed the relationship between changes in cerebral blood flow velocity (BFV) in response to vasoactive stimuli (as measured by transcranial Doppler (TCD)), characterizing CVR and cardiovascular autonomic nervous system (CANS) function. Methods: Common carotid artery compression (CCC n = 26), hyperventilatio

Clinical data mining on network of symptom and index and correlation of tongue-pulse data in fatigue population.

BACKGROUND: Fatigue is a kind of non-specific symptom, which occurs widely in sub-health and various diseases. It is closely related to people's physical and mental health. Due to the lack of objective diagnostic criteria, it is often neglected in clinical diagnosis, especially in the early stage of disease. Many clinical practices and researches have shown that tongue and pulse conditions reflect the body's overall state. Establishing an objective evaluation method for diagnosing disease fatigue and non-disease fatigue by combining clinical symptom, index, and tongue and pulse data is of grea

Brain signatures of childhood trauma and polygenic scores for mental health drive clinical subtypes in depression: a UK Biobank study.

Gene-environment correlations (rGE) may drive the clinical heterogeneity of major depressive disorder (MDD) through their effects on brain structure. However, previous literature focused on isolated components of these interplays. Here, we jointly investigate how rGE shape neurobiological profiles in MDD, and whether rGE-driven brain signatures can disentangle depression subtypes. In 5951 MDD patients with genetic, trauma-related and neuroimaging data from the UK Biobank, cross-validated sparse canonical correlation analysis was employed to assess multivariate associations between polygenic sc

Relating the shape of protein binding sites to binding affinity profiles: is there an association?

BACKGROUND: Various pattern-based methods exist that use in vitro or in silico affinity profiles for classification and functional examination of proteins. Nevertheless, the connection between the protein affinity profiles and the structural characteristics of the binding sites is still unclear. Our aim was to investigate the association between virtual drug screening results (calculated binding free energy values) and the geometry of protein binding sites. Molecular Affinity Fingerprints (MAFs) were determined for 154 proteins based on their molecular docking energy results for 1,255 FDA-appr

A powerful score-based test statistic for detecting gene-gene co-association.

BACKGROUND: The genetic variants identified by Genome-wide association study (GWAS) can only account for a small proportion of the total heritability for complex disease. The existence of gene-gene joint effects which contains the main effects and their co-association is one of the possible explanations for the "missing heritability" problems. Gene-gene co-association refers to the extent to which the joint effects of two genes differ from the main effects, not only due to the traditional interaction under nearly independent condition but the correlation between genes. Generally, genes tend to
