The clinical application of cantharidin (CTD) is seriously limited due to its nephrotoxicity. Therefore, this study aims to investigate sensitive biomarkers for the evaluation and prediction of nephrotoxicity induced by CTD in rat. A total of 80 rats were randomly divided into four groups: control group and three doses of CTD groups. After 0, 1, 5, 15, and 28 days of intragastric administration, rat serum and urine were collected for biochemical indexes, then serum was used for metabolomic analyses, and rat kidney was collected for pathological and ultrastructural observation. The levels of se

Natural product cantharidin can inhibit multiple myeloma cell growth in vitro, while serious adverse effects limited its clinical application. Therefore, the structural modification of cantharidin is needed. Herein, inspired by the structural similarity of the aliphatic endocyclic moiety in cantharidin and TRIP13 inhibitor DCZ0415, we designed and synthesized DCZ5418 and its nineteen derivatives. The molecular docking study indicated that DCZ5418 had a similar binding mode to TRIP13 protein as DCZ0415 while with a stronger docking score. Moreover, the bioassay studies of the MM-cells viability

Condyloma Acuminatum is a sexually transmitted viral disease caused by the human papilloma virus (HPV). It is the most common viral sexually transmitted disease. In this randomized controlled trial, cantharidin was found to be more effective and better tolerated than trichloroacetic acid for the treatment of these lesions. Patients treated with cantharidin healed with less scarring than those treated with TCA (P<0.034), had less pain during treatment (P<0.01), and required fewer treatments to eradicate warts (P<0.01) when compared to Trichloroacetic acid.

BACKGROUND: Cantharidin (CTD), a potent anti-tumor compound from traditional medicine, has its clinical application restricted by severe hepatotoxicity. Astragalus polysaccharides (APS), the primary active components of the food-medicine homologous herb Astragalus membranaceus, are recognized for their hepatoprotective properties.
PURPOSE: This study aimed to elucidate the molecular mechanism by which APS protects against CTD-induced liver injury.
METHODS: In vivo and in vitro models of CTD-induced liver injury were established. The protective effects of APS were evaluated by serum biochemis

Although cantharidin (CTD) exhibits significant clinical efficacy against various cancers, its clinical utility is severely hampered by profound systemic toxicity. Thus, structural modification of CTD to obtain analogues with potent anticancer activity and lower toxicity is essential for advancing CTD-based therapeutics. Building on our previous synthesis of a series of C-5 alkynyl-substituted CTD analogues with potent anti-inflammatory properties, this study investigates the anticancer potential of a new analogue NCTD-01 demonstrated a favorable safety profile in mice, with the maximum tolera

As a protein kinase inhibitor, cantharidin (CTD) exhibits antitumor activities. However, CTD is highly toxic, thereby limiting clinical applications. Moreover, relatively few studies have investigated CTD-induced reproductive toxicity, thus the underlying mechanism remains unclear. In this study, the toxic effects of CTD on mouse testis were confirmed in vivo and the potential mechanism was predicted by network toxicology (NT) and molecular docking technology. Proteins involved in the signaling pathways and core targets were verified. The results showed that different concentrations of CTD ind

Cantharidin (CTD) has been traditionally used in Traditional Chinese Medicine (TCM) and exhibits pronounced antitumor activity, particularly in hepatocellular carcinoma (HCC). However, its preclinical translation has been substantially constrained by dose-limiting toxicity. Inspired by traditional Mylabris and Ginseng combinations, we conducted an investigation into the synergistic efficacy of CTD and Ginsenoside-Rg3 (Rg3), one of the most prevalent and bioactive ginsenosides, in combating HCC. We find that CTD/Rg3 co-treatment synergistically suppressed HCC cell proliferation, migration, and

The hepatotoxicity mechanism of cantharidin (CTD), a major active component of Mylabris was explored based on liver lipidome alterations and spatial distributions in female and male rats using lipidomics and matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI). After oral CTD exposure, the livers of female rats were screened for 104 differential lipids including lysophosphatidylethanolamine(LysoPE)(20:2/0:0) and diacylglycerol(DG)(18:2/22:4), whereas the livers of male rats were screened for 76 differential lipids including fatty acid(FA)(24:6) and DG(18:0/22:4). BACKGROUND: In China, sodium cantharidinate/vitamin B6 (SC/VB6) injection has been approved since 2002 for the treatment of lung cancer and primary liver cancer. In addition to these authorized indications, clinical application of SC/VB6 is also common in various other types of cancer. However, there is a lack of comprehensive understanding on this topic. Thus, this systematic review and meta-analysis aims to consolidate evidence regarding the efficacy and safety of off-label use of SC/VB6 in oncology.
METHODS: International databases including PubMed, Embase, Cochrane Library, Web of Science

The present study was designed to investigate the effect of cantharidin on cell proliferation, ability of selfrenewal, cell cycle arrest and induction of apoptosis in HepG2 hepatocellular carcinoma stem cells (HCSCs). It was observed that cantharidin treatment exhibited dose- and time-dependent inhibitory effect on the viability of HCSCs. The inhibition of cell viability by cantharidin in HepG2 CD133+ and parental cells was significant at the concentration 5 and 15 µM, respectively after 48 h. Cantharidin treatment inhibited the self-renewal ability of the HCSCs and the expression of β-catenin

BACKGROUND: Ginsenosides, such as ginsenoside RG3, demonstrate antitumor potential in hepatocellular carcinoma (HCC) and are often combined with cantharidin (CTD) in traditional Chinese medicine to achieve synergistic effects while mitigating CTD’s toxicity. However, the precise molecular mechanisms underlying this synergy remain elusive. METHODS: The progression of HCC was assessed using a series of in vitro assays, including CCK-8 for cell viability, EdU staining for proliferation, wound healing for migration, and transwell assay for invasion. The antitumor efficacy and hepatotoxicity were a

Triple-negative breast cancer (TNBC) is a subtype of breast cancer that poses a great threat to women's health. Herein, we report that cantharidin (CTD), a main ingredient of Mylabris, can effectively inhibit the proliferation of TNBC cells, making it a potential therapeutic agent for this kind of tumor. However, the poor water solubility, short half-life, and systemic toxicity of CTD greatly limit its clinical application. To address these issues, we developed a cancer cell membrane and iRGD peptide co-modified CTD liposomes (CTD@LipsMR). The results of in vitro and in vivo experiments showed

Cantharidin (C(10)H(12)O(4)) is a monoterpene defensive toxin in insects involved in chemical defence as well as in courtship and mating behaviours. It is relatively well known in the medical literature because of its high anticancer activity and as an effective therapy for molluscum contagiosum. However, little is known about its biosynthesis pathway in vivo, and no enzyme involved in cantharidin biosynthesis has been identified. The purpose of this study was to identify the crucial enzyme that is involved in the biosynthesis of cantharidin. Using the homology cloning method, a 3-hydroxy-3-me

Topical cantharidin is a commonly used treatment for molluscum contagiosum (MC). However, studies validating its safety and efficacy are limited. We conducted a 6-week, randomized, double-blind, placebo-controlled trial with subsequent open-label extension to assess the safety and effectiveness of cantharidin in treating pediatric MC. Ninety-four participants with MC were randomized to receive cantharidin or placebo, with or without occlusion. The primary outcome was complete lesion clearance. Secondary outcomes included post-treatment lesion count, adverse events, and side effects. No signifi
