Atom-Efficient Synthesis of Trimethine Cyanines Using Formaldehyde as a Single-Carbon Source.

Herein, we present an innovative and atom-efficient synthesis of trimethine cyanines (Cy3) using formaldehyde (FA) as a single-carbon reagent. The widespread application of Cy3 dyes in bioimaging and genomics/proteomics is often limited by synthetic routes plagued by low atom economy and substantial side-product formation. Through systematic investigation, we have developed a practical and efficient synthetic pathway for both symmetrical and unsymmetrical Cy3 derivatives, significantly minimizing resource utilization. Notably, this approach yields water as the by-product, in alignment with sus

Simultaneous detection of lysosomal SO2 and viscosity using a hemicyanine-based fluorescent probe.

The changes in sulfur dioxide and viscosity of lysosomes are significant indicators in physiological processes and the cell microenvironment. This study aimed to synthesize a hemicyanine-based probe for simultaneous detection of SO2 and viscosity. The probe could not only rationally detect sulfur dioxide in a semi-aqueous solution with high sensitivity (limit of detection = 0.78 μM) and fast response (within 30 s) but also monitor viscosity via fluorescence emission enhancement at 580 nm. Further, the dual-response probe was successfully used to image SO2 and viscosity in the lysosomes of livi

Preparation of a cyanine-based fluorescent probe for highly selective detection of glutathione and its use in living cells and tissues of mice.

Glutathione (GSH) is a major endogenous antioxidant that has a central role in cellular defense against toxins and free radicals. This protocol describes the preparation of CPDSA, a cyanine-based near-infrared (NIR) fluorescent probe for the detection of GSH in cells and in vivo. CPDSA is prepared with high yield through a simple two-step process. The first step is to react commercially available IR-780 iodide with excess anhydrous piperazine in anhydrous N,N-dimethyl formamide at 85 °C to form cyanine-piperazine (CP). The second step is the sulfonylation of CP with dansyl chloride in anhydrou

Granulocyte stimulating factor attenuates hypoxic-ischemic brain injury by inhibiting apoptosis in neonatal rats.

PURPOSE: This study was undertaken to determine the neuroprotective effect of granulocyte stimulating factor (G-CSF) on neonatal hypoxic-ischemic brain injury. MATERIALS AND METHODS: Seven-day-old male newborn rat pups were subjected to 110 minutes of 8% oxygen following a unilateral carotid artery ligation. Apoptosis was identified by performing terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL) staining and flow cytometry with a combination of fluorescinated annexin V and propidium iodide (PI) and JC-1 (5,5',6,6'-tetrachloro-1,1',3,3'- tetraethylbenzimidazolyl-car

Early development of descending supraspinal pathways: a tracing study in fixed and isolated rat embryos.

Early brainstem-spinal cord projections were studied in the rat using the carbocyanine dye DiI in fixed embryos and biotinylated dextran amine (BDA) in an isolated embryonic brain-spinal cord preparation. A system of staging embryos was used that allows direct comparison with data in other mammals. With both techniques it was shown that in embryos of at least 12 days of age (E12), i.e., at the time of closure of the posterior neuropore, already a variety of brainstem centers innervate the spinal cord. In the interstitial nucleus of the fasciculus longitudinalis medialis and various parts of th

Specific binding of human low-density lipoprotein to the surface of schistosomula of Schistosoma mansoni and ingestion by the parasite.

Low-density lipoproteins (LDL) may be important in human schistosomiasis because LDL bound to the surface of the parasite inhibits the binding of anti-schistosomal antibodies. Low-density lipoproteins also may serve as a source of lipids for the parasite membrane synthesis. Here LDL fluorescently labeled with carbocyanine dye (DiI-LDL) was used to measure the specificity of binding of LDL to the surface of schistosomula of Schistosoma mansoni and to examine the distribution of the LDL particles over time. DiI-LDL binding was saturable and specific, with strong inhibition by unlabeled LDL and a

Hyperbolic Surface Exciton Polaritons and Surface Waves in Near-Zero Permittivity Regions Supported by Artificial Organic Hyperbolic Metamaterials.

Hyperbolic metamaterials enable extreme light confinement and control of photonic states, but their realization has been restricted to inorganic architectures. Here, a fully organic route to fabricate artificial hyperbolic metamaterials based on multilayered thin films of J-aggregate carbocyanine dyes alternated with polyelectrolytes is introduced. These structures exhibit strong optical anisotropy and experimentally support hyperbolic surface exciton polaritons and, for selected dyes, additional surface waves in near-zero permittivity regimes. Spectroscopic ellipsometry confirms a uniaxial di

Canine adipose tissue-derived mesenchymal stem cells ameliorate severe acute pancreatitis by regulating T cells in rats.

Severe acute pancreatitis (SAP) is associated with systemic complications and high mortality rate in dogs. Mesenchymal stem cells (MSCs) have been investigated for their therapeutic potential in several inflammation models. In the present study, the effects of canine adipose tissue-derived (cAT)-MSCs in a rat model of SAP induced by retrograde injection of 3% sodium taurocholate solution into the pancreatic duct were investigated. cAT-MSCs labeled with dioctadecyl-3,3,3'-tetramethylindo-carbocyanine perchlorate (1 × 10⁷ cells/kg) were systemically administered to rats and pancreatic tissue was

Connectivity of ectopic neurons in the molecular layer of the somatosensory cortex in autoimmune mice.

Approximately 50% of New Zealand Black mice (NZB/BINJ) and 80% of NXSM-D/EiJ mice prenatally develop neocortical layer I ectopias, mostly in somatosensory cortices. These cortical anomalies are similar to those seen in the brains of individuals with dyslexia. Neurofilament staining revealed a radial column of tightly packed fiber bundles in the layers underlying ectopias. This suggested that the connectivity of the ectopic neurons was aberrant. The present study used the tracers 1,1'-dioctadecyl- 3,3,3',3'-tetramethylindo- carbocyanine perchlorate (DiI) and biotinylated dextran amine (BDA) to

Anatomical relationship between vagal afferent fibers and CCK-immunoreactive entero-endocrine cells in the rat small intestinal mucosa.

There is evidence for a pathway involving small intestinal CCK-producing entero-endocrine cells and visceral afferent nerve fibers in signaling the effect of luminal nutrients on gastrointestinal and food intake regulation. In order to investigate the type of anatomical apposition that exists between CCK cells and vagal afferents, CCK immunocytochemistry was performed on tissue from rats whose vagal afferent fibers to the abdomen had previously been labeled in vivo by injecting the fluorescent carbocyanine dye DiI into the nodose ganglia. CCK immunoreactive (CCK-IR) cells were more abundant th

Visceral afferent and efferent columns in the spinal cord of the teleost, Ictalurus punctatus.

In tetrapod vertebrates, neural circuitries subserving visceral and somatic reflexes are each represented in distinct columns of cells within the gray area of the spinal cord. To determine the location of visceral elements of the spinal cord of a teleost fish, crystals of the carbocyanine dye 1,1'dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine (DiI), were placed on either the abdominal sympathetic (mesenteric) nerves, the coeliac ganglia, or on the rostral three somatic spinal nerves, in fixed specimens of the channel catfish, Ictalurus punctatus. Spinal and facial innervation of the skin in the gadid fish Ciliata mustela (Teleostei).

The pattern of innervation of the skin of the rockling Ciliata mustela was investigated to sort out spinal from facial nerve innervation of cutaneous chemosensory and mechanosensory systems. This fish has a variety of appendages with different functional sensory specializations, i.e., the chin barbel, pelvic fin, anterior dorsal fin, and dorsal trunk skin. The carbocyanine dye, diI, was applied to nerve stumps in dissected aldehyde-fixed tissue. In the case of the chin barbel, the dye was applied to both the trigeminal and facial nerve components. In the other cases, the dye was applied either

Smooth-muscle endoplasmic reticulum contains a cardiac-like form of calsequestrin.

It is proposed that smooth-muscle endoplasmic reticulum contains calsequestrin and that this protein in smooth muscle resembles cardiac calsequestrin more than the skeletal-muscle form. This proposal is based on seven similarities between the smooth-muscle protein and cardiac calsequestrin. Proteins with an Mr of 55,000 can be extracted from the membranes of smooth muscle and of cardiac muscle using 100 mM Na2CO3. The protein from smooth muscle binds to phenyl-Sepharose in the absence of Ca2+ and is released by 10 mM CaCl2, as has been observed for cardiac calsequestrin. The protein from smoot

The state of energization of the membrane of Escherichia coli as affected by physiological conditions and colicin K.

The bacterial protein colicin K, when added to sensitive Escherichia coli in the presence of 3,3'-dihexyloxacarbocyanine, cuases a doubling in fluorescence of the probe. Glucose and oxygen cause a decreased fluorescence while anoxia and cyanide cause a rise in fluorescence. These results in conjunction with the work of other laboratories suggest that colicin K causes a depolarization of the transmembrane electrical potential. Fluorescence in the absence of colicin K was relatively independent of KCl, NaCl, and MgCl2 concentrations below 0.1 M. Although colicin K caused rapid efflux of the K+ a

Enhanced Single-Molecule Imaging via Intersystem Crossing Suppression.

The red-emitting cyanine dye Cy5 is widely used in single-molecule imaging, but its utility is limited by triplet-state accumulation. This state facilitates energy transfer to molecular oxygen, producing reactive oxygen species (ROS) that lead to photobleaching. The triplet state also causes the dye's fluorescence intermittency and introduces temporal noise. Furthermore, Cy5 in this state is susceptible to nucleophilic attacks in cellular environments, resulting in an inactive off-state. To address these challenges, we engineered a series of phenyl-substituted meso-Cy5 derivatives to suppress

Equivalent Response Strategy for Sensing Total Biothiols in Human Serums and Living Cells Using a Hemicyanine-Based Self-Immolative Probe.

Biothiols including cysteine (Cys), homocysteine (Hcy), and glutathione (GSH) are crucial in maintaining the redox balance in the body, and the metabolism and transportation of biothiols rely on the coreaction of diverse proteins and enzymes. The abnormal concentrations and metabolism of biothiols are closely associated with many diseases. However, due to the same active reaction site of the sulfydryl group in biothiols, it is inevitable to bear a confused signal of mutual influence on both nonselective detection and discriminate detection, which presents a serious challenge of accurately sens

Synergistic anti-cancer and attenuation effects of thymosin on chemotherapeutic drug vinorelbine in tumor-bearing zebrafish.

Vinorelbine, the standard chemotherapy drug on advanced lung cancer, causes adverse events such as immunosuppression and bone marrow suppression. Thus, it is necessary to find drugs that could improve immune function and synergistically enhance the anti-tumor effect of vinorelbine. Thymosin is reported to inhibit tumor growth as an immunomodulator. Herein, to study the synergistic anti-cancer and attenuation effects of thymosin on vinorelbine, human lung cancer A549 cells that were labeled with CM-DiI were transplanted into zebrafish to establish the lung cancer xenotransplanted model. A matrix metalloproteinase activation probe for painting human tumours.

A probe that allows specific 'painting' of human tumours is described. Probe activation was mediated by specific matrix metalloproteinases, resulting not only in disruption of a FRET pair, but in the generation of a fragment that "fluorescently paints" human tumours. This probe demonstrated rapid and effective human tumour labelling with the potential to allow margin detection during surgical resection.

Size-matched alkyne-conjugated cyanine fluorophores to identify differences in protein glycosylation.

Currently, there are few methods to detect differences in posttranslational modifications (PTMs) in a specific manner from complex mixtures. Thus, we developed an approach that combines the sensitivity and specificity of click chemistry with the resolution capabilities of 2D-DIGE. In "Click-DIGE", posttranslationally modified proteins are metabolically labeled with azido-substrate analogs, then size- and charge-matched alkyne-Cy3 or alkyne-Cy5 dyes are covalently attached to the azide of the PTM by click chemistry. The fluorescently-tagged protein samples are then multiplexed for 2DE analysis.

Design, synthesis and biological evaluation of the positional isomers of the galactose conjugates able to target hepatocellular carcinoma cells via ASGPR-mediated cellular uptake and cytotoxicity.

Galactose as a recognizing motif for asialoglycoprotein receptor (ASGPR) is a widely accepted vector to deliver cytotoxic agents in the therapy of hepatocellular carcinoma (HCC), however, the individual hydroxyl group of galactose (Gal) contributed to recognizing ASGPR is obscure and remains largely unanswered in the design of glycoconjugates. Herein, we designed and synthesized five positional isomers of Gal-anthocyanin Cy5.0 conjugates and three Gal-doxorubicin (Dox) isomers, respectively. The fluorescence intensity of Gal-Cy5.0 conjugates accumulated in cancer cells hinted the optimal modif
