Pharmacological and Toxicological Profile of Harmane-β-Carboline Alkaloid: Friend or Foe.

BACKGROUND: The plant secondary metabolites have an outstanding therapeutic potential and success over the years. In fact, it is the foundation of numerous clinically used drugs. Similarly, these is a general perception that these products are inherent safety. However, such products might have toxic/unwanted lethal effects therefore, along with biological relevance, toxicological evaluation is equally important for clinical applications. Therefore, harmane- β-carboline alkaloid was investigated for both therapeutic and toxicological potential. METHODS: The literature related to the therapeuti

In vitro evaluation of β-carboline alkaloids as potential anti-Toxoplasma agents.

BACKGROUND: Toxoplasmosis is a worldwide infection caused by the protozoan parasite Toxoplasma gondii, which causes chorioretinitis and neurological defects in congenitally infected newborns or immunodeficient patients. The efficacy of the current treatment is limited, primarily by serious host toxicity. In recent years, research has focused on the development of new drugs against T. gondii. β-Carbolines (βCs), such as harmane, norharmane and harmine, are a group of naturally occurring alkaloids that show microbicidal activity. In this work, harmane, norharmane and harmine were tested against

Review on in-vitro anti-malarial activity of natural β-carboline alkaloids.

Malaria is one of the major health problems in developing countries. It kills 1-2 million people every year and also it affects financial status of many countries. Developed resistance to aminoquinoline (chloroquine), quinoline methanols (Quinine, Mefloquine) created troubles in malarial chemotherapy and signs of appearance of resistance to artemisinin based combination therapy (ACT) created emergency to develop novel antimalarial agents with high efficacy before spreading of resistance to ACT. From the ages natural products played an important role in antimalarial therapy, identification of n

Riluzole antagonizes the anxiogenic properties of the beta-carboline FG 7142 in rats.

The possible anxiolytic activity of riluzole, a drug which interferes with glutamic acid neurotransmission, was studied in rats using operant conflict procedures. In both "anxiolytic" and "anxiogenic" procedures, riluzole alone did not possess any anticonflict or proconflict effect at doses of 2 and 4 mg/kg PO. Riluzole over the same dose-range was able to antagonize the well known proconflict effect of the beta-carboline derivative FG 7142, an inverse agonist at the GABA-benzodiazepine-chloride ionophore receptor complex. This effect could be related to the possible interaction of riluzole wi

[Effect of the beta-carboline derivative FG 7142 on inhibition in hippocampal sections].

The action of anxiogenetic derivative of beta-carboline FG7142 on evoked activity of neurons in hippocampal sections was investigated using extra- and intracellular recordings. The activity in CAI area was registered upon stimulation of Schaffer's collaterals (SC). Excitatory effect of FG7142 (5 microM) on population spike (PS) was blocked by simultaneous diazepam (5 microM) or RO15-1788 (5 microM) application. This suggests that FG7142 action is mediated by benzodiazepine receptors. To evaluate the action of FG7142 on hippocampal inhibitory processes paired-pulse stimulation of SC was used. I

Neuropharmacological antagonism of the beta-carboline-induced "anxiety" response in rhesus monkeys.

A behavioral and physiological syndrome of stress-related responses was reported in primates following treatment with the benzodiazepine receptor antagonist, beta-carboline-3-carboxylic acid ethyl ester (beta-CCE). The behavioral and physiological effects of beta-CCE are similar to those observed during stressful or "anxiety"-related conditions characterized in rhesus monkeys under natural conditions. Pharmacological agents which are known to antagonize anxiety responses in other paradigms were tested for their ability to antagonize the actions of beta-CCE. Diazepam (1 mg/kg) completely blocke

The effects of the beta-carboline FG 7142, on intracranial self-stimulation in the rat.

The effects of FG 7142 were examined, alone and in combination with chlordiazepoxide, on self-stimulation of the mid-lateral hypothalamus. Rewarding stimuli were delivered according to a 10-sec variable-interval schedule of reinforcement. FG 7142 (1-20 mg/kg) produced a dose-related depression in responding, and chlordiazepoxide (5 mg/kg) enhanced it. When these two drugs were given together, response rates did not differ significantly from control rates.

Rational Design of β-Carboline Photosensitizers for Combating Multidrug-Resistant Bacterial Infections.

The misuse of antibiotics has accelerated the emergence of multidrug-resistant bacteria such as methicillin-resistant Staphylococcus aureus (MRSA), creating an urgent need for novel antimicrobial strategies. Antimicrobial photodynamic therapy, which employs light-activated photosensitizers to generate reactive oxygen species (ROS), is promising since it kills both sensitive and resistant pathogens without inducing resistance. β-Carbolines, a class of natural alkaloids with notable antimicrobial potential, have mostly been studied for cancer therapy and rarely adapted as antibacterial photosens

β-Carboline (FG-7142) modulates fear but not anxiety-like behaviour in zebrafish.

The β-Carboline FG-7142 is a partial inverse agonist at the benzodiazepine allosteric site on the GABA-A receptor that induces anxiogenic, proconvulsant, and appetite-reducing effects in many species, including humans. Seizure-kindling effects have been well studied, however anxiogenic properties are relatively unexplored. This study aimed to investigate concentration-dependent effects of FG-7142 on anxiety-like behaviour and fear responses in zebrafish (Danio rerio) using the open-field test (OF) and novel object approach test (NOA). A U-shaped distribution was found with maximal responses in

β-Carboline Alkaloids Resist the Aggregation and Cytotoxicity of Human Islet Amyloid Polypeptide.

β-Carboline alkaloids have a variety of pharmacological activities, such as antitumor, antibiosis and antidiabetes. Harmine and harmol are two structurally similar β-carbolines that occur in many medicinal plants. In this work, we chose harmine and harmol to impede the amyloid fibril formation of human islet amyloid polypeptide (hIAPP) associated with type 2 diabetes mellitus (T2DM), by a series of physicochemical and biochemical methods. The results indicate that harmine and harmol effectively prevent peptide fibril formation and alleviate toxic oligomer species. In addition, both small molec

In vitro digestion modifies Lepidium meyenii extract, altering its major glucosinolates, lepidilines, β-carbolines, and gut bacterial growth.

The tubers of Lepidium meyenii (commonly known as maca) have been used in the Andean cuisine for ages. The study aims to determine the behavior of the extract from the red phenotype of maca in the in vitro gastrointestinal tract model and also to define the overall impact of the extract on the microbiota. Glucosinolates, lepidilines, and a derivative of β-carboline alkaloid, i.e. 1-methyltetrahydro-β-5,6-hydride-carboline-3-carboxylic acid, as representatives of one of the main groups of metabolites were selected for the quantitative analysis after in vitro digestion at every stage of the dige

The endogeneous formation of highly chlorinated tetrahydro-beta-carbolines as a possible causative mechanism in idiopathic Parkinson's disease.

The causative interrelationship between long-term, low level exposure to chlorinated volatile organic solvents (VOSs) and neurodegenerative diseases (polyneuropathy, encephalopathy) are still an issue of controversial debate. Endogeneously formed chlorinated tetrahydro-beta-carbolines found by Bringmann 1995 (TaClo hypothesis) may contribute, in particular, to the development of (idiopathic) Parkinson's disease (PD) in the presence of the sufficient amount of trichloroacetaldehyde, an intermediate in metabolism of trichloroethylene (TRI). Long-term storage of specific VOSs over years, evident

Inhibition of histamine-N-methyltransferase (HNMT) by fragments of 9-amino-1,2,3,4-tetrahydroacridine (tacrine) and by beta-carbolines.

Histamine-N-methyltransferase (HNMT), the major enzyme for the metabolism of histamine in rat brain, is potently inhibited by 9-amino-1,2,3,4-tetrahydroacridine (tacrine). Structural fragments of tacrine were less potent inhibitors of rat brain HNMT than was tacrine itself. Harmaline and a number of other beta-carbolines inhibited HNMT with IC50 values in the range of 1-10 microM. HNMT inhibition by harmaline was competitive with respect to both substrates, S-adenosylmethionine and histamine (Ki = 1.4 microM). These findings are discussed in the context of mechanisms for HNMT inhibition.

Hyperphagic and anorectic effects of beta-carbolines in a palatable food consumption test: comparisons with triazolam and quazepam.

The triazolobenzodiazepine triazolam (0.1-1.0 mg/kg i.p.) and quazepam (0.3-30.0 mg/kg i.p.) were administered to non-food-deprived rats which had been partially-satiated on a palatable diet. In a subsequent 30 min feeding test, both compounds produced a significant increase in the level of food consumption. While triazolam had a dose-related effect and produced a 151.5% increase in the level of food intake, quazepam exerted only a partial effect, achieving a 73.9% increase in food intake at 3.0 mg/kg but no additional increase in food intake at higher doses. The two beta-carbolines, ZK 93423

Stress and beta-carbolines decrease the density of low affinity GABA binding sites; an effect reversed by diazepam.

Cerebral cortex membranes from rats habituated to manipulations preceding decapitation (habituated rats) had 40% higher GABA binding than membranes from naive animals. Diazepam (5 X 10(-6) M), added to membranes from naive rats, increased GABA binding to the level of habituated rats, but failed to induce any further increase in membranes from the latter animals. Vice versa, beta-carbolines (FG 7142, beta-CCE, DMCM) added to membranes from habituated rats lowered GABA binding to the level of naive animals, but caused no further decrease in the membranes from this last group. Diazepam removed th

Advances in Structural Diversity, Pharmacological Activities, and Drug Development of β-Carboline Alkaloids.

β-Carboline alkaloids are natural products built around an indole-pyridine tricyclic core. Their relatively simple structure and ease of modification have drawn lasting interest from medicinal chemists. This review organizes their pharmacological activities according to structural types: simple β-carbolines, those with substitutions at the C-1, N-2/N-9 or C-3 positions, cyclized derivatives, and bis-β-carboline dimers. For each type, we discuss reported mechanisms, such as inhibition of CDK4, HDAC, and topoisomerases. The available data point to promising applications in cancer, inflammatory d
