The cellular effects of carbon monoxide in the airway.

The notion of inhaled carbon monoxide (CO) being a toxic chemical under all circumstances is currently being challenged, as recent research has suggested that low concentrations of CO may have therapeutic value, especially in the airway. This review evaluates CO's effects on cellular functions that may result in beneficial outcomes in the settings of airway disease, inflammation, and injury. CO can modulate the stress response system of the cell by decreasing levels of reactive intermediates over time, produced by mitochondrial iNOS and NADPH oxidase. Intracellular stress-induced response fact

[Carbon monoxide as an external cause of fatality].

INTRODUCTION: Carbon monoxide intoxications mainly occur as a result of fires in houses and failures of heating systems. Case of fatal accident involving a charcoal factory worker is described in this paper. MATERIAL AND METHODS: Authors based on a medico-legal autopsy protocol and material collected in prosecutor files. Medico-legal autopsy of a 46 year old male was performed in the forensic medicine department. Interlocutory proceedings indicated that the immediate cause of death was a head-crush caused by a hydraulic cover. Previously self-ignition in the factory was noted. RESULTS: Autop

Mechanism of the ventilatory response to carbon monoxide.

The effects of carbon monoxide on ventilation were studied in unanesthetized goats. Responses to single breaths of 10-25% CO in O2, which rapidly raised carboxyhemoglobin (COHb) from 5 to 60%, were considered to reflect peripheral chemoreceptor-mediated reflexes whereas responses to continuous inhalation of 1% CO in O2, which slowly raised COHb from 0 to 60%, were considered to reflect both peripheral chemoreceptor and nonperipheral chemoreceptor mechanisms. In each of six goats, single breaths of CO failed to elicit any immediate ventilatory response. However, slow buildup of carboxyhemoglobi

Edible Fungi Are a Hidden Source of Carbon Monoxide and Carbon Dioxide.

This study provides the first clear evidence that edible mushrooms, such as Lentinula edodes (shiitake), Pleurotus ostreatus and Pleurotus eryngii, can generate carbon monoxide (CO) as part of their metabolic activity-independent of bacteria, illumination or oxygen limitation. Systematic measurements of CO and CO2 emissions were performed over 60 days using multiple fungal species, substrates and growth conditions. Microscopy observations (light, scanning and transmission electron microscopy) confirmed no extracellular and intracellular bacterial endosymbionts involved, supporting a fungal gen

Continuous extracorporeal hyperoxygenation therapy reduces carbon monoxide half-life time in a carbon monoxide-poisoned pig model: a feasibility study.

Carbon monoxide (CO) poisoning is a leading cause of toxic exposure-related morbidity and mortality worldwide. Although hyperbaric oxygen therapy increases CO elimination, its clinical use is limited by accessibility, side effects, and inconclusive long-term efficacy. We developed a cEHT system - a device for continuous extracorporeal hyperoxygenation therapy that enhances CO removal from blood without exposing patients to systemic hyperbaric conditions. The system was tested in vitro and in vivo in a porcine CO poisoning model. Carboxyhemoglobin (COHb) half-life, hemodynamics, hemolysis, and

Astrocyte-produced carbon monoxide and the carbon monoxide donor CORM-A1 protect against cerebrovascular dysfunction caused by prolonged neonatal asphyxia.

Neonatal asphyxia leads to cerebrovascular disease and neurological complications via a mechanism that may involve oxidative stress. Carbon monoxide (CO) is an antioxidant messenger produced via a heme oxygenase (HO)-catalyzed reaction. Cortical astrocytes are the major cells in the brain that express constitutive HO-2 isoform. We tested the hypothesis that CO, produced by astrocytes, has cerebroprotective properties during neonatal asphyxia. We developed a survival model of prolonged asphyxia in newborn pigs that combines insults of severe hypoxia, hypercapnia, and acidosis while avoiding ext

Immobilized Neuroglobin Scavenges Carbon Monoxide From Circulating Carboxyhemoglobin.

Carbon monoxide (CO) poisoning is responsible for around 50,000 emergency department visits per year in the United States alone. Currently, there is no available antidote for CO poisoning. We have previously developed an antidotal therapy based on an engineered hemeprotein, human neuroglobin (Ngb-H64Q-CCC). Intravenous infusion of Ngb-H64Q-CCC removes CO from circulating red blood cells and improves survival in a lethal CO poisoning mouse model. However, the infusion of heme-containing proteins has inherent heme toxicity risks that may limit the dose that can be used safely without liver or ki

Heme oxygenase/carbon monoxide in thermoregulation and neuroenergetics.

This review explores the heme oxygenase/carbon monoxide (HO/CO) pathway as a central integrator of thermoregulation and neuroenergetics during inflammatory and oxidative stress. Thermoregulation is framed not merely as a homeostatic process, but as a continuous problem of energy allocation where body temperature (Tb) serves as an indicator of an organism's bioenergetic capacity. The core mechanistic thesis identifies the HO/CO system as a state-dependent "coupling pathway" that links inflammatory and redox tone to mitochondrial performance and, through central autonomic hierarchies, to the sel

Immobilized neuroglobin scavenges carbon monoxide from circulating carboxyhemoglobin.

Carbon monoxide (CO) poisoning is responsible for around 50,000 emergency department visits per year in the U.S. alone. With the present standard of care, persistent neurological sequelae occur in ~30-40% of severe CO poisoning cases. Currently, there is no available targeted molecular antidote for CO poisoning. In previous work, we have developed an antidotal therapy for CO poisoning based on an engineered hemeprotein, human neuroglobin (Ngb-H64Q-CCC). Emerging role of carbon monoxide in intestinal transplantation.

Intestinal transplantation has become an established therapeutic option that provides improved quality of life to patients with end-stage intestinal failure when total parenteral nutrition fails. Whereas this challenging life-saving intervention has shown exceptional growth over the past decade, illustrating the evolution of this complex and technical procedure from its preclinical origin in the mid-20th century to become a routine clinical practice today with several recent innovations, its success is hampered by multiple hurdles including technical challenges such as surgical manipulation du

Emerging cellular-based therapies in carbon monoxide poisoning.

Carbon monoxide (CO) is an odorless and colorless gas with multiple sources that include engine exhaust, faulty furnaces, and other sources of incomplete combustion of carbon compounds such as house fires. The most serious complications for survivors of consequential CO exposure are persistent neurological sequelae occurring in up to 50% of patients. CO inhibits mitochondrial respiration by specifically binding to the heme a3 in the active site of CIV-like hydrogen sulfide, cyanide, and phosphides. Although hyperbaric oxygen remains the cornerstone for treatment, it has variable efficacy requi

Patterns of cardiac dysfunction after carbon monoxide poisoning.

OBJECTIVE: To describe the structural sequelae of carbon monoxide (CO) poisoning on the heart assessed using stress cardiac MRI (CMR). CO poisoning is common. While acute cardiac injury is frequent among survivors, the mid- and long-term effects of CO on the myocardium are unclear. METHODS: CMR studies performed between the years 2005 and 2014 for a primary diagnosis of CO poisoning at a tertiary care center were reviewed by an experienced cardiologist. Variables of interest were compared between patients with normal and abnormal studies to identify factors associated with cardiac dysfunction

Carbon monoxide-releasing molecule attenuates allograft airway rejection.

Acute rejection after lung transplantation is the main risk factor for the development of bronchiolitis obliterans (BO). Carbon monoxide (CO) can provide anti-inflammatory effects and may serve to limit tissue injury in airway transplant. Here, we tested the ability of carbon monoxide releasing molecule-2 (CORM-2) to prevent airway rejection. Tracheal grafts from BALB/c or C57BL/6 were transplanted to C57BL/6 recipients. Experimental groups were treated with multiple doses of CORM-2. Histopathological evaluation of luminal obliteration was blindly reviewed. Immunohistochemistry and real-time R

Increased carbon monoxide clearance during exercise in humans.

PURPOSE: Hyperventilation increases the clearance of carbon monoxide (CO) from blood; thus, we hypothesized that CO elimination would be enhanced with exercise. Accordingly, this study examined the effect of exercise on the half-life of carboxyhemolobin elimination. METHODS: Six healthy subjects (three males and three females) with mean ± SD ages of 23 ± 4 yr were exposed to CO sufficient to raise blood carboxyhemolobin concentration to 10-14% on five separate days. The half-life for CO elimination was measured breathing room air at rest and during exercise at three intensities. RESULTS: Com
