Carboxylic Acid Transporters in Candida Pathogenesis.

Opportunistic pathogens such as Candida species can use carboxylic acids, like acetate and lactate, to survive and successfully thrive in different environmental niches. These nonfermentable substrates are frequently the major carbon sources present in certain human body sites, and their efficient uptake by regulated plasma membrane transporters plays a critical role in such nutrient-limited conditions. Here, we cover the physiology and regulation of these proteins and their potential role in Candida virulence. This review also presents an evolutionary analysis of orthologues of the Saccharomy

General Cascade Esterification-Pentafluorosulfanyloxylation of Carboxylic Acids with Cyclic Ethers using Silver Pentafluorooxosulfate.

As the analog of the trifluoromethoxy (OCF3) group, the pentafluorosulfanyloxy group (OSF5) exhibits comparable electronic properties and unique geometry. Previous research has primarily focused on pentafluorosulfanyloxylation of aryl-based substrates; however, a similar transformation in alkyl substrates is exceedingly rare. In this work, we report the first cascade esterification of carboxylic acids with cyclic ethers to achieve the selective synthesis of alkyl-OSF5 products. In this transformation, AgOSF5 serves as the pentafluorosulfanyloxide source, cyclic ethers act as the coupling partn

CO2 Fixation into Useful Aromatic Carboxylic Acids via C (sp2)-X Bonds Functionalization.

Carbon dioxide (CO2) is an abundant and readily available carbon source. Its transformation into high-added-value chemicals is a beneficial strategy, which mitigates greenhouse gas emissions and provides new raw material sources for the chemical industry. Among these chemicals, the aromatic carboxylic acids and derivatives have broad applications in medicine, pesticides, and materials science. Therefore, the carboxylation of C(sp2)-X (X = metal, halide, H, O, or S) bonds with CO2 to efficiently construct aromatic carboxylic acids and their derivatives is a synthetic strategy of significance. Ultrafast Solid-Phase Oxidation of Aldehydes to Carboxylic Acids by Atmosphseric Plasma Treatment.

Although atmospheric plasma treatment is an industrially widespread, scalable, and environmentally friendly method, it has been generally used for surface modification, decontamination, or sterilization. In this paper, a novel, sustainable, green, and ultrafast oxidation method is described for aldehydes on a preparative thin-layer chromatographic plate as a solid support. The plasma treatment has proven to be suitable for producing the corresponding carboxylic acids by using only air as a reactant source under mild reaction conditions, while the isolation of the products is also directly inte

Peptide Conjugates of Benzene Carboxylic Acids as Agonists and Antagonists of Amylin Aggregation.

Human islet amyloid polypeptide (hIAPP), also known as amylin, is a 37 residue peptide hormone that is stored and co-secreted with insulin. hIAPP plays a pivotal role in type 2 diabetes and is the major component of amyloid deposits found in the pancreas of patients afflicted with the disease. The self-assembly of hIAPP and the formation of amyloid is linked to the death of insulin producing β-cells. Recent findings suggest that soluble hIAPP oligomers are the cytotoxic species responsible for β-cell loss whereas amyloid fibrils themselves may indeed be innocuous. Potential avenues of therapeu

Selective 19 F-Labeling of Functionalized Carboxylic Acids with Difluoromethyl Diazomethane (CF2 HCHN2 ).

The incorporation of fluorinated moieties into organic structures is a particularly powerful approach for generating compounds with useful biological applications in industry, pharmacology, and agriculture. Here, the recently developed reagent-CF2 HCHN2 -selectively esterifies carboxyl groups on organic molecules in the presence of other polar functional groups. The reaction is remarkably simple, proceeding at room temperature in chloroform without addition of a catalyst. The procedure allows "last-step" incorporation of a fluorinated ester moiety into organic structures, including bioorganic

Carbonic anhydrase inhibitory activity of sulfonamides and carboxylic acids incorporating cyclic imide scaffolds.

A series of sulfonamides incorporating cyclic imide moieties were investigated as inhibitors of several human α-carbonic anhydrase (hCA, EC 4.2.1.1) isoforms. Several carboxylic acids possessing the same scaffolds as the sulfonamides were also included in the study, since the sulfonamidate and the carboxylate are among the frequently used zinc-binding groups (ZBGs) for obtaining zinc enzymes inhibitors. The cytosolic isoform hCA I was moderately inhibited by most of the 30 investigated derivatives; many low nanomolar hCA II inhibitors were detected, whereas some of these compounds were low nan

Acyloxyamines as prodrugs of anti-inflammatory carboxylic acids for improved delivery through skin.

An N,N-dialkylhydroxylamine derivative of indomethacin has been synthesized. It has been shown to improve the delivery of indomethacin through mouse skin (compared to indomethacin itself) by a factor of two, to be more effective than indomethacin in inhibiting thermal inflammation (two to three times) in animal models, but to be only as effective as indomethacin in inhibiting UV-B radiation erythema in human volunteers.

Decarboxylative selenylation of carboxylic acids: a reliable strategy for the synthesis of selenoethers.

Decarboxylative cross-coupling reactions have emerged as powerful and highly significant transformations for the construction of carbon-carbon and carbon-heteroatom bonds, benefiting from the use of stable, abundant, and cost-effective carboxylic acids as readily accessible coupling partners. These processes offer attractive alternatives to conventional cross-coupling or addition reactions that rely on organometallic reagents. Building on this strategy, a variety of methods have been developed for the synthesis of synthetically and biologically relevant selenoethers through decarboxylative sel

Enantioselective Radical Addition of Carboxylic Acids to Imines through Cooperative Copper/Acridine Catalysis.

Amino acids and their derivatives are indispensable as both synthetic building blocks and functional entities in the chemical industry and life sciences. Herein, we report a modular strategy for the asymmetric synthesis of chiral α-amino acids and amines through the decarboxylative radical addition to imines. This protocol leverages readily available, cost-effective carboxylic acids, aldehydes, and anilines as starting materials, enabled by the synergistic catalysis of acridine and copper. The reaction operates under ambient conditions, exhibits excellent functional group compatibility, and de

Single-enzyme redox-neutral oxidation of alcohols to carboxylic acids using alcohol dehydrogenases.

Oxidation of primary alcohols to carboxylic acids is a fundamental reaction in organic chemistry, traditionally dependent on toxic oxidants and often limited by poor selectivity. In this study, we demonstrate the multifunctional capability of some alcohol dehydrogenases (ADHs) to catalyze both alcohol and aldehyde oxidation while regenerating their NAD+ cofactor through concomitant reduction of acetone. Screening of a panel of ADHs revealed that the enzymes from Paracoccus pantotrophus (Pp-ADH) and Aromatoleum aromaticum (Aa-ADH) have strong overoxidation activity to carboxylic acids. The bioc

Formal Reductive Amination of Carboxylic Acids via Iron-Catalyzed Photoredox Decarboxylative α-Aminomethylation.

Here we report an iron-catalyzed photocatalytic protocol utilizing ligand-to-metal charge transfer (LMCT) for efficient decarboxylative reductive amination to synthesize tertiary alkylamines. The method employs readily available carboxylic acids and N,O-acetals under mild conditions (room temperature, 390 nm LED), achieving excellent yields (up to 89%) across diverse alkyl carboxylic acid substrates, including primary, secondary, and tertiary types (>40 examples). Furthermore, the protocol exhibits good tolerance toward sensitive functional groups and enables late-stage functionalization of ph

Deprotective Functionalization: A Direct Conversion of Nms-Amides to Carboxamides Using Carboxylic Acids.

The nature of protecting group chemistry necessitates a deprotection step to restore the initially blocked functionality prior to further transformation. As this aspect of protecting group manipulation inevitably adds to the step count of any synthetic sequence, the development of methods enabling simultaneous deprotection and functionalization ("deprotective functionalization"-distinct from "deprotection followed by functionalization") is appealing, as it has the potential to improve efficiency and streamline synthetic routes. Herein, we report a deprotective functionalization of the newly in

Ligand-enabled ruthenium-catalyzed meta-C-H alkylation of (hetero)aromatic carboxylic acids.

Carboxylates are ideal directing groups because they are widely available, readily cleavable and excellent linchpins for diverse follow-up reactions. However, their use in meta-selective C-H functionalizations remains a substantial unmet catalytic challenge. Herein, we report the ruthenium-catalyzed meta-C-H alkylation of aromatic carboxylic acids with various functionalized alkyl halides. A bidentate N-ligand increases the electron density at the metal center of ortho-benzoate ruthenacycles to the extent that single-electron reductions of alkyl halides can take place. The subsequent addition

Metal-free synthesis of α-acyloxy ketones from carboxylic acids and sulfoxonium ylides.

A straightforward, catalyst- and additive-free approach has been described for synthesizing α-acyloxy ketones from β-ketosulfoxonium ylides and carboxylic acids. Moderate to high yields of α-acyloxy ketones were produced using sulfoxonium ylides and carboxylic acids adorned with various functional groups. Eventually, the applicability of this approach has been shown via a large-scale reaction and transforming the synthesized α-acyloxy ketone derivatives into other valuable compounds.

"One-Pot" Synthesis of γ-Pyrones from Aromatic Ketones/Heteroarenes and Carboxylic Acids.

Despite the various attractive properties of γ-pyrones, there are still some deficiencies in their synthetic approaches such as lower atom economy, multistep processes, and prefunctionalization of the reagents. In this work, an efficient and simple (CF3CO)2O/CF3SO3H-mediated "one-pot" approach was realized to produce γ-pyrones by applying aromatic ketones/heteroarenes and carboxylic acids as the starting materials. The target products were isolated in moderate to excellent yields. The reaction mechanism was studied by density functional theory calculational methods. The results of experimental

Mechanism of Phosphorus-Carbon Bond Formation in the Amidoalkylation of Phosphonous Carboxylic Acids.

An unusual greater reactivity of phosphonous propionic acids was found in comparison with phosphonous propionic esters in carbamate version of Kabachnik-Fields reaction. Compounds of tricoordinated phosphorus generated in situ during the amidoalkylation of hydrophosphorylic compounds in acetyl chloride/acetic anhydride mixture were found by 31P NMR analysis. A hypothesis is proposed about the generation of spirophosphoranes in situ to explain the mechanism for the formation of the phosphorus-carbon bond in the reaction under study.
