Gallate Moiety of Catechin Is Essential for Inhibiting CCL2 Chemokine-Mediated Monocyte Recruitment.

Leukocyte recruitment witnesses an orchestrated complex formation between the chemokines and their molecular partners. CCL2 chemokine that regulates monocyte trafficking is a worthwhile system from the pharmaceutical perspective. In the current study, four major catechins (EC/EGC/ECG/EGCG) were assessed for their inhibitory potential against CCL2-regulated monocyte/macrophage recruitment. Interestingly, catechins with the gallate moiety (ECG/EGCG) could only attenuate the CCL2-induced macrophage migration. These molecules specifically bound to CCL2 on a pocket comprising the N-terminal, β0-she

Trehalose-Conjugated, Catechin-Loaded Polylactide Nanoparticles for Improved Neuroprotection against Intracellular Polyglutamine Aggregates.

Intracellular/extracellular protein aggregation is linked to a variety of neurodegenerative diseases. Current research focuses on identifying antiamyloidogenic small molecules to inhibit such protein aggregation and associated cytotoxicity. We have recently demonstrated that transforming these antiamyloidogenic small molecules into nanoparticle forms can greatly improve their performance, and biocompatible/biodegradable formulation of such nanoparticles is critical for therapeutic applications. Here, we report polylactide (PL)-based biodegradable nanoparticles for improved neuroprotection agai

Synergistic protection of PC12 cells from beta-amyloid toxicity by resveratrol and catechin.

beta-Amyloid peptide (beta-AP) elicits a toxic effect on neurons in vitro and in vivo. Many environmental factors including antioxidants, metal ions and proteoglycans modify beta-AP toxicity. We have investigated on PC12 cells, the protective effect from beta-AP (1-41) of two plant polyphenols, resveratrol and catechin. PC12 cells treated with 10(-6)M beta-AP (1-41) for 16h decrease the cell viability at 24% of the control with an IC(50) value of 1.1+/-0.14 x 10(-8)M. Twenty-five micromolar resveratrol and 50 microM catechin protect PC12 cells from beta-AP (1-41) toxicity with the IC(50) value

Liquid chromatographic determination of methylxanthines and catechins in herbal preparations containing guaraná.

Herbal preparations derived from the dried seeds of guaraná (Paullinia cupana) have become a popular nutritional supplement used for stimulatory purposes. Once considered a drug substance in the United States, guaraná currently is classified as a food additive and dietary supplement. The pharmacological activity of guaraná-containing products is primarily due to methylxanthine alkaloids. For guaraná preparations, methylxanthine levels and, more significantly, the presence of several polyphenol compounds (i.e., catechins) provide phytochemical markers of authenticity. Methylxanthines and polyph

Iron, Catechin, and Ferulic Acid Inhibit Cellular Uptake of β-Carotene by Reducing Micellization.

Green leafy vegetables have low β-carotene bioavailability, which we hypothesized to be, at least in part, due to high contents of fiber, minerals, and phenolics. We investigated the effects of pectin (40-120 μg/mL), iron (50-150 μg/mL), ferulic acid (30-90 μg/mL), and catechin (50-150 μg/mL), in a model system, on β-carotene micellization (in vitro digestion) and intestinal absorption (Caco-2 cell model). Iron, pectin, ferulic acid, and catechin on average reduced ( p < 0.05) β-carotene micellization (1.49 ± 0.05 μmol/L) by 66.9, 59.3, 43.2, and 51.7%, respectively. Iron reduced micellization

Catechin solubilization by spontaneous hydrogen bonding with poly(ethylene glycol) for dry eye therapeutics.

Catechin exhibits various pharmacological effects, yet its poor aqueous solubility limits its clinical use. Here, we investigate a facile catechin solubilization method via spontaneous hydrogen bonding between catechin and poly(ethylene glycol) (PEG). The method is extremely simple in that mixing PEG with catechin followed by lyophilization completely converts insoluble catechin to soluble PEG/catechin nanoscale complexes. This solubilized catechin formulation is useful for preparing eyedrop medicine, and we demonstrate that the solubilized catechin exhibits therapeutic effect upon dry eye dis

Catechin, taxifolin, and proanthocyanidin derivatives from Entada elephantina rhizome and their antimicrobial activity assessment.

Twenty-two compounds, predominantly catechin, taxifolin, and proanthocyanidin derivatives, were isolated and characterized from the methanol extract of Entada elephantina tuberous rhizomes. Five taxifolin-based metabolites, elephantinosides A-E (1-5) and one proanthocyanidin, elephantinoside F (19), were previously undescribed. Their structures were elucidated primarily based on 1D- and 2D-NMR and HRESIMS data. The absolute configuration of isolates was determined via NOESY NMR technique and experimental and calculated ECD analyses. None of the tested compounds 3-6, 8-10, 13-17, and 19 exhibit

The tea catechin epigallocatechin gallate inhibits NF-κB-mediated transcriptional activation by covalent modification.

Potential health benefits of consuming tea are thought to include anti-inflammatory actions of its constituent flavonoids including catechins, which are well-recognized antioxidants. We analyzed and discovered a novel mechanism by which epigallocatechin gallate (EGCG), the most abundant polyphenol in tea and a putative health-promoting constituent, inhibits activation of the nuclear transcription factor NF-κB, which mediates inflammatory responses to cytokines and other agents. We found that EGCG inhibits NF-κB-p65 transcriptional activity, by preventing NF-κB-p65 binding to κBs in normal huma

Catechin supplementation prevents kidney damage in rats repeatedly exposed to cadmium through mitochondrial protection.

Nephrotoxicity is recognized as a serious disorder affected by chronic cadmium exposure. Imbalance between radical generation and elimination is considered a critical factor involved in the initiation and progression of renal injury caused by this heavy metal. The present study investigated the possible protection by catechin, a natural phenolic antioxidant, against cadmium nephrotoxicity and elucidated its potential mechanism. Male Wistar rats were assigned to receive vehicle, cadmium (CdCl2 2 mg/kg, i.p.) and cadmium plus catechin (25, 50, and 100 mg/kg, orally, respectively). After 4 weeks
