Multi-halogenation enhances the antimicrobial and antibiofilm activities of catechols against A. baumannii via iron chelation and ROS-mediated killing.

AIMS: The rise of multidrug-resistant Acinetobacter baumannii, listed by the WHO as a top-priority pathogen, necessitates the urgent development of new antimicrobial agents. Here, we identified and evaluated halogenated catechol derivatives with potent antimicrobial and antibiofilm activity against A. baumannii and other ESKAPE pathogens. MATERIALS AND METHODS: Fifteen halogenated catechol derivatives were screened for antimicrobial activity. The most active compounds, namely tetrachlorocatechol (TCC) and tetrabromocatechol (TBC), were evaluated for their impact on biofilm formation, membrane

Catechol-O-Methyltransferase Loss Drives Cell-Specific Nociceptive Signaling via the Enteric Catechol-O-Methyltransferase/microRNA-155/Tumor Necrosis Factor α Axis.

BACKGROUND & AIMS: The etiology of abdominal pain in postinfectious, diarrhea-predominant irritable bowel syndrome (PI-IBS-D) is unknown, and few treatment options exist. Catechol-O-methyltransferase (COMT), an enzyme that inactivates and degrades biologically active catecholamines, plays an important role in numerous physiologic processes, including modulation of pain perception. Our objective was to determine the mechanism(s) of how decreased colonic COMT in PI-IBS-D patients contributes to the chronic abdominal pain phenotype after enteric infections. METHODS: Colon neurons, epithelial cel

MnFe aminoclay as a novel catalyst: structural characterization and potential for catechol detection.

The development of enzyme-mimicking nanomaterials offers a promising route toward sensitive, stable, and cost-effective detection of phenolic compounds. In this study, MnFe hybrid aminoclay (MnFeAC) was synthesized and systematically characterized by XRD, XPS, SEM, TEM, and elemental mapping to confirm its amorphous structure, homogeneous elemental distribution, and mixed-valence states of Mn and Fe that underpin its catalytic activity. The MnFeAC material exhibited oxidase-like properties, enabling the oxidation of catechol in the presence of 4-aminoantipyrine (4-AAP) with a distinct colorime

First Synthesis of Shimobashiric Acid C With Acetonide as the Catechol Protecting Group.

Shimobashiric acid C (SBA-C) shows excellent hyaluronidase inhibition but its natural isolation yield is only 0.0092 %. We report its first synthesis via a convergent seven-step route from caffeic and rosmarinic acids in 35% overall yield. Direct protection of D-Danshensu (D-DSS) with 2,2-dimethoxypropane gave an isochroman; the desired acetonide-protected D-DSS building block was instead prepared from methyl rosmarinate via bis-acetonide formation followed by methanolysis. Acetonide-protected caffeic acid was synthesized and crystallized in a P-1 lattice, where linear hydrogen-bonded caffeic

Sustainable synthesis of catechol compounds using tyrosinase in ethanol cosolvent system.

Ethanol is environmentally friendly, not hazardous, and can be produced from renewable sources. In this study, ethanol was applied as a cosolvent for the tyrosinase-catalyzed synthesis of catechol compounds from phenolic compounds, which are abundant in fruit pomace. To investigate the cosolvent function of ethanol on tyrosinase, six types of tyrosinase from Priestia megaterium and water-insoluble phenolic substrates (i.e., phloretin and resveratrol) were used. Biocatalytic activity and stability of tyrosinase were maintained in the ethanol-water system up to 80 % (v/v) ethanol with an optimum

Music Aggravates Catechol-Induced Behavioural Abnormality and Redox Imbalance in Zebrafish.

Catechol, also known as pyrocatechol, is a widespread antioxidant carcinogen that has been shown to cause central nervous system damage and metabolic abnormalities, and in zebrafish, it has been shown that exposure to aqueous solutions of catechol results in reduced pigmentation and decreased basal locomotor rate in juvenile zebrafish. However, the effects of catechol on zebrafish redox and behaviour are unknown. The aim of this study was to investigate the effects of catechol on oxidative stress levels in early zebrafish development and on behaviour in later stages of zebrafish development an

Synthesis, Cytotoxicity and Antioxidant Activity Evaluation of Some Thiazolyl-Catechol Compounds.

A series of thiazolyl-catechol compounds with antioxidant and cytotoxic activities were synthesized by a Hantzsch heterocyclization, using diverse thioamides as the thiocarbonyl component and 4-chloroacetyl-catechol as haloketone. These compounds were characterized by MS, IR spectroscopy, and NMR. Their antioxidant potential was evaluated by antiradical, electron transfer, and ferrous ion chelation assays using ascorbic acid, Trolox, and EDTA-Na2 as references. The cytotoxicity of the synthesized compounds was evaluated on two different cell types, normal human foreskin fibroblasts (BJ) and hu

Mucoadhesive chitosan-catechol as an efficient vaccine delivery system for intranasal immunization.

The mucosal barrier and scavenging effect of the mucosal layer are two main obstacles in inducing mucosal immunization. To overcome these obstacles, we synthesized a bio-inspired mucoadhesive material, chitosan-catechol (ChiC), for surface modification of inactive porcine epidemic diarrhea virus (PEDV). Studies have revealed that PEDV particles can be facilely and mildly modified by Chi-C forming Chi-C-PEDV nanoparticles (Chic-Ps) through the covalent and electrostatic bond, which effectively prolongs the retention time of PEDV in the nasal mucosa. The cell co-culture model demonstrated that C

Catechol-mimicking transition-state analogues as non-oxidizable inhibitors of tyrosinases.

Tyrosinases are copper-containing metalloenzymes involved in several processes in both mammals, insects, bacteria, fungi and plants. Their phenol oxidation properties are especially responsible for human melanogenesis, potentially leading to abnormal pigmentation, and for postharvest vegetable tissue browning. Thus, targeting tyrosinases attracts interest for applications both in dermocosmetic and agrofood fields. However, a large part of the literature about tyrosinase inhibitors is dedicated to the report of copper-interacting phenolic compounds, that are more likely alternative substrates l

Design and catalytic studies of structural and functional models of the catechol oxidase enzyme.

The catechol oxidase activity of three copper/bicompartmental salen derivatives has been studied. One mononuclear, [CuL] (1), one homometallic, [Cu2L(NO3)2] (2), and one heterometallic, [CuMnL(NO3)2] (3) complexes were obtained using the ligand H2L = N,N'-bis(3-methoxysalicylidene)-1,3-propanediamine through different synthetic methods (electrochemical, chemical and solid state reaction). The structural data indicate that the metal ion disposition models the active site of type-3 copper enzymes, such as catechol oxidase. In this way, their ability to act as functional models of the enzyme has

Unraveling Substrate Specificity and Catalytic Promiscuity of Aspergillus oryzae Catechol Oxidase.

Catechol oxidases and tyrosinases are coupled binuclear copper enzymes that oxidize various o-diphenolic compounds to corresponding o-quinones. Tyrosinases have an additional monooxygenation ability to hydroxylate monophenol to o-diphenol. It is still not clear what causes the difference in the catalytic activities. We solved a complex structure of Aspergillus oryzae catechol oxidase with resorcinol bound into the active site. Catalytic activity of A. oryzae catechol oxidase was studied, for the first time, by high-resolution FT-ICR mass spectrometry to shed light on the reaction mechanism. Th

Lithium bis-catechol borate as an effective reductive quencher in photoredox catalysis.

The use of lithium bis-catechol borate (LiB(cat)2) as a reductive quencher for the photoredox mediated intermolecular C-H functionalization of various heteroaromatics with bromopyrroloindolines is described. LiB(cat)2 offers a financial benefit over state-of-the-art quenchers currently in use while eliminating the side reactions that typically plague these couplings. The advantage of this methodology is highlighted by the synthesis of C3-C2' (-) gliocladin C. Furthermore, additional examples of reactivity with various bromopyrroloindolines sets the stage for expedient routes towards other phar

Medicinal chemistry of catechol O-methyltransferase (COMT) inhibitors and their therapeutic utility.

Catechol O-methyltransferase (COMT) is the enzyme responsible for the O-methylation of endogenous neurotransmitters and of xenobiotic substances and hormones incorporating catecholic structures. COMT is a druggable biological target for the treatment of various central and peripheral nervous system disorders, including Parkinson's disease, depression, schizophrenia, and other dopamine deficiency-related diseases. The purpose of this perspective is fourfold: (i) to summarize the physiological role of COMT inhibitors in central and peripheral nervous system disorders; (ii) to provide the history

Schistosome and liver fluke derived catechol-estrogens and helminth associated cancers.

Infection with helminth parasites remains a persistent public health problem in developing countries. Three of these pathogens, the liver flukes Clonorchis sinensis, Opisthorchis viverrini and the blood fluke Schistosoma haematobium, are of particular concern due to their classification as Group 1 carcinogens: infection with these worms is carcinogenic. Using liquid chromatography-mass spectrometry (LC-MS/MS) approaches, we identified steroid hormone like (e.g., oxysterol-like, catechol estrogen quinone-like, etc.) metabolites and related DNA-adducts, apparently of parasite origin, in developm

Catechol-O-methyltransferase in complex with substituted 3'-deoxyribose bisubstrate inhibitors.

The biological activity of catechol neurotransmitters such as dopamine in the synapse is modulated by transporters and enzymes. Catechol-O-methyltransferase (COMT; EC 2.1.1.6) inactivates neurotransmitters by catalyzing the transfer of a methyl group from S-adenosylmethionine to catechols in the presence of Mg²⁺. This pathway also inactivates L-DOPA, the standard therapeutic for Parkinson's disease. Depletion of catechol neurotransmitters in the prefrontal cortex has been linked to schizophrenia. The inhibition of COMT therefore promises improvements in the treatment of these diseases. The con

Serotonin-2A receptor and catechol-O-methyltransferase polymorphisms in panic disorder.

Catechol-O-methyltransferase (COMT) and serotonin receptor 2A (5HTR2A) polymorphisms have been investigated for their possible role in panic disorder (PD). The aim of this study was to investigate the genotype distribution of the COMT val158met and 5HTR2A 102T/C polymorphisms in PD. COMT val158met is a polymorphism at codon 158 that results in variations in COMT enzymatic activity with high- (H) and low-activity (L) alleles. The 5HTR2A 102T/C polymorphism comprises a T-to-C mutation at position 102. The effects of symptom severity, gender, and age of onset were also investigated. The participa
