Divalent cation transport by the distal nephron: insights from Bartter's and Gitelman's syndromes.

Elucidation of the gene defects responsible for many disorders of renal fluid and electrolyte homeostasis has provided new insights into normal and abnormal physiology. Identifying the causes of Gitelman's and Bartter's syndromes has greatly enhanced our understanding of ion transport by thick ascending limb and distal convoluted tubule cells. Despite this information, several phenotypic features of these diseases remain confusing, even in the face of molecular insight. Paramount among these are disorders of divalent cation homeostasis. Bartter's syndrome is caused by dysfunction of thick asce

Divalent cation-activated-ATPase and ecto 5'-nucleotidase activities in chronic tonsillitis.

The activities of two ectoenzymes,Mg(2+)-ATPase (ATPase) and 5'-nucleotidase (5'-NT) from tonsillar mononuclear cells (TMC) and peripheral blood mononuclear cells (PBMC) were investigated in idiopathic tonsillar hyperplasia and recurrent tonsillitis. The ATPase activity of TMC was significantly higher in recurrent tonsillitis than in idiopathic tonsillar hyperplasia, whereas no difference was demonstrated in ATPase activity of PBMC. The activity of 5'-NT was similar in both investigated groups. However, ATPase and 5'-NT activities were significantly higher in TMC compared to PBMC. Such results

Trypsinized BHK21 cells aggregate in the presence of metabolic inhibitors and in the absence of divalent cations.

The rapid formation of adhesions in suspension by lightly trypsinized BHK21 cells is not dependent on protein synthesis, and only in part on cellular metabolism, although it is completely inhibited by heat- and aldehyde-fixation of the cells. A requirement for protein synthesis becomes evident only if cells are exposed to high levels of trypsin for long periods. Formation of adhesions does not require addition to the medium of divalent cations, although it is increased by divalent manganese and cobalt ions. It is promoted by cytochalasin B and by cyclic AMP and is not inhibited by p-mercuriphe

Inhibitory effect of divalent metal cations on zinc uptake via mouse Zrt-/Irt-like protein 8 (ZIP8).

AIMS: There is controversy regarding the substrate specificity of ZIP8, a ZIP isoform, involved in regulation of extra- and intracellular zinc levels. Here, we investigated the inhibitory effects of divalent metal cations on zinc uptake via mouse ZIP8 (mZIP8). MAIN METHODS: mZIP8 cDNA was transfected into HEK293T cells by a lipofection method, and its functional expression was evaluated by immunocytochemistry, Western blotting and 65Zn (65ZnCl2) uptake measurement. KEY FINDINGS: Transfection of mZIP8 cDNA into HEK293T cells induced expression of mZIP8 in the cells, and increased zinc uptake.

Supramolecular assemblies of phenyl-pyridyl-triazolopyridine and β-cyclodextrin as sensor of divalent cations in aqueous solution.

The chemosensor 3-phenyl-7-(pyrid-2-yl)-[1,2,3]triazolo[1,5-a]pyridine (PhPTP) used in combination with two different cyclodextrins, enable its solubilization and stabilization in aqueous solution. The behavior of the inclusion complex, and its binding ability in both cyclodextrins were investigated by means of absorption and fluorescence spectroscopy. The best results were obtained for PhPTP-DMβCD assembly, and its orientation in the DMβCD nano cavity was obtained by 2D-NMR. This inclusion geometry was confirmed by docking studies. The binary complex was proved as chemosensor upon the presenc

High affinity binding of long-chain polysialic acid to antibody, and modulation by divalent cations and polyamines.

Long-chain polysialic acid (PSA) is expressed on the vertebrate neural cell adhesion molecule (NCAM) during neuronal plasticity. Its structural similarity to the capsular PSAs of some pathogenic bacteria has hampered the development of polysaccharide vaccines against meningitis. The antibodies formed during immunization require a long epitope for binding, and cross-react with host tissue PSA. The nature of the epitope and possible external effectors involved are still unclear. We have evaluated the interaction of PSA with its antibody mAb735 by surface plasmon resonance. The influences of PSA

Cyclic GMP-activated channels of the chick pineal gland: effects of divalent cations, pH, and cyclic AMP.

Chick pineal cells maintained in dissociated cell culture express an intrinsic photosensitive circadian oscillator, but the mechanisms of phototransduction in avian pinealocytes are not fully understood. In this study, we have used inside-out patches to examine the characteristics of cyclic GMP-activated channels of chick pinealocytes in more detail, concentrating on the effects of factors known to modulate the secretion of melatonin and/or the function of circadian pacemakers. In most patches, the predominant conductance state was 19 pS in symmetrical 145 mM NaCl. But in some patches, a secon

Reduction behaviors of permanganate by microbial cells and concomitant accumulation of divalent cations of Mg2+, Zn2+, and Co2.

Permanganate treatment is widely used for disinfection of bacteria in surface-contaminated water. In this paper, the fate of the dissolved permanganate in aqueous solution after contact with cells of Pseudomonas fluorescens was studied. Concomitant accumulation of divalent cations of Mg2+, Zn2+, and Co2+ during precipitation of Mn oxides was also studied. The time course of the Mn concentration in solution showed an abrupt decrease after contact of Mn(VII) with microbial cells, followed by an increase after ~24 hr. XRD analysis of the precipitated Mn oxides, called biomass Mn oxides, showed th

Effects of divalent cations and sulfhydryl reagents on the p-aminohippurate (PAH) transporter of renal basal-lateral membranes.

A specific system of transport for p-aminohippurate (PAH) is demonstrated in rabbit renal basal-lateral membrane vesicles. The PAH uptake into an intravesicular space is inhibited by probenecid in concentrations above 0.2 mM. The transport is saturable and is also temperature-dependent with an optimum between 37 and 45 degrees C. Divalent cations are able to enhance the uptake 2- to 3-fold. The stimulatory effect of the divalent cations diminishes in the following order: Mg++ = Mn++, Ba++, Ca++ and Sr++. Maximum stimulation occurs between 2.5 and 5 mM Mg++. The divalent cation stimulatory effe

The use of elevated divalent cation solutions to isolate monosynaptic components of sensorimotor connections in Aplysia.

A commonly used method to remove polysynaptic components of test PSPs is to elevate action potential threshold of interneurons with high extracellular concentrations of divalent cations ('Hi-Di'). Extrapolation to normal conditions requires that Hi-Di have negligible effects on synaptic transmission. We examined effects of Hi-Di on EPSPs from sensory neurons (SNs) onto motor neurons (MNs) of Aplysia in the pleural-pedal and abdominal ganglia, and in dissociated cell culture. In ganglia, standard Hi-Di solutions eliminated spontaneous input from interneurons as well as polysynaptic components o

Separation of chloride and sulfate ions in univalent and divalent cation forms from aqueous streams.

The precipitation and separation of chloride and sulfate in several cation forms (sodium, potassium, magnesium, calcium, strontium, and barium) from aqueous streams were studied using isopropylamine (IPA) and ethylamine (EA) as precipitation solvents. The precipitation fractions (P) of the tested chloride salts at 5000 and 10,000 ppm by both IPA and EA over the studied range of solvents volume ratio (V(R)) were relatively identical (18-60%) and their small variations were within their experimental uncertainty. The P of combined sulfate at 1000 ppm (56-99.5%) and chloride at 5000 ppm (28-62%) i

A study of the mechanism by which inhibitors of the plasmamembrane ATPase enhance uptake of divalent cations in yeast.

The enhancement of divalent cation uptake in yeast provoked by the membrane ATPase inhibitors trifluoperazine, miconazole, compound 48/80, ethidium, DIO-9 and calmidazolium should be ascribed to an increase in cation permeability of the yeast rather than to hyperpolarisation of the yeast cell membrane. For trifluoperazine and miconazole it is unequivocally shown that the cells are hyperpolarized though for miconazole only transiently. Whether the other drugs also hyperpolarize the yeast cells is uncertain. The apparent hyperpolarisation caused by trifluoperazine and miconazole may be attribute

Parameterization of the CHARMM force field for divalent cations (Ca2+/Be2+) binding to anionic lipids with isothermal titration calorimetry.

Beryllium toxicity manifests as a rare lung disease, berylliosis, characterized by persistent granulomas. Studies indicate that, in the presence of Be2+, macrophage phagocytosis is markedly reduced, potentially hindering the removal of apoptotic cells. Although the exact mechanism by which Be2+ ions reduce phagocytosis is unclear, one hypothesis suggests that these small ions outcompete Ca2+ in critical ion-lipid and lipid-protein interactions mediating lipid recognition. To explore these interactions at the atomic level using molecular dynamics (MD), we aim to improve existing MD force fields

Effect of Divalent Cations (Cu, Zn, Pb, Cd, and Sr) on Microbially Induced Calcium Carbonate Precipitation and Mineralogical Properties.

Microbially induced calcium carbonate precipitation (MICP) is a bio-geochemical process involving calcium carbonate precipitation and possible co-precipitation of other metals. The study investigated the extent to which a urease-positive bacterium, Sporosarcina pasteurii, can tolerate a range of metals (e.g., Cu, Zn, Pb, Cd, and Sr), and analyzed the role of calcium carbonate bioprecipitation in eliminating these divalent toxicants from aqueous solutions. The experiments using S. pasteurii were performed aerobically in growth media including urea, CaCl2 (30 mM) and different metals such Cu, Zn

The fundamental roles of monovalent and divalent cations with sulfates on molybdenite flotation in the absence of flotation reagents.

Due to regional shortage of freshwater, the use of saline/seawater for Cu-Mo sulfide ore flotation has received considerable attention. However, the effects of various salts, especially the cations present in seawater, on molybdenite flotation and the mechanisms involved remain unclear due to the complexity of the solutions applied. In this work, the influence of some common cations (i.e., Na+, K+, Ca2+ and Mg2+) with sulfate (SO4 2-) anions on molybdenite flotability was investigated in the absence of flotation reagents (i.e., frothers and collectors) at pH 10. Flotation results indicated a g

Effects of zinc gluconate and 2 other divalent cationic compounds on olfactory function in mice.

Intranasal application of zinc gluconate has commonly been used to treat the common cold. The safety of this treatment, however, has come into question recently. In addition to a United States recall of a homeopathic product that contains zinc gluconate, abundant literature reports cytotoxic effects of zinc on the olfactory epithelium. Additional research suggests that divalent cations (such as zinc) can block ion channels that facilitate the transduction of odors into electrical signals on the olfactory epithelium. The purpose of the current study was 2-fold: to confirm whether zinc gluconate

Localisation of ecto-5'-nucleotidase and divalent cation-activated ecto-ATPase in chronic tonsillitis.

The histochemical localisation of two ecto-enzymes, 5'-nucleotidase (5'-NT) and Mg(2+)-ATPase, was investigated in hyperplastic and recurrent tonsillitis. Detection of enzymes was performed on frozen sections using the classical lead nitrate method. Activity of 5'-NT was demonstrated particularly in the cells of lymphoid follicles and in the basal layer of the surface tonsillar epithelium. There was no difference in localisation of 5'-NT between hyperplastic and recurrent tonsillitis, whereas a stronger reaction in follicular mantle zones was observed in recurrent tonsillitis compared to hyper

Mechanisms of cation permeation across apical cell membrane of Necturus gallbladder: effects of luminal pH and divalent cations on K+ and Na+ permeability.

Conventional microelectrode techniques were combined with unilateral mucosal ionic substitutions to determine the effects of luminal pH and luminal alkali-earth cation concentrations on apical membrane cation permeability in Necturus gallbladder epithelium. Acidification of the mucosal solution caused reversible depolarization of both cell membranes and increase of transepithelial resistance. Low pH media also caused: (a) reduction of the apical membrane depolarization induced by high K, and (b) increase of the apical membrane hyperpolarization produced by Na replacement with Li or N-Methyl-D-
