Objective The purpose of this study was to establish the diffusion-weighted magnetic resonance imaging (DW-MRI) findings of the caudate nucleus and putamen in patients with obsessive-compulsive disorder (OCD) and to obtain new information on the etiopathogenesis of OCD, which is still unclear. Methods The study comprised 20 patients with OCDs and 20 healthy volunteers. In these cases, DW-MRI and diffusion-weighted echo-planar images (DW-EPI) at b600 and b1000 gradient values were taken and the measurements were made using the apparent diffusion coefficient (ADC) maps of each group at b600 and

BACKGROUND AND PURPOSE: Whereas traditional views of language processing in the brain have assumed that the language function is concentrated to a limited number of cortical areas (Broca's and Wernicke's areas), current knowledge points at a much more complex system of language and speech processing involving many brain areas, both cortical and subcortical. The purpose of the current study was to make an unbiased assessment of which cerebral areas are affected in first-ever acute ischaemic stroke patients identified as having language and speech impairments according to the National Institutes

INTRODUCTION: Velocardiofacial syndrome (VCFS) is a neurogenetic disorder caused by a microdeletion on chromosome 22q11. Among other cognitive impairments and learning difficulties, affected individuals show difficulties in estimating time intervals (Debbané et al., 2005). Interestingly, neuroimaging studies have found an increased volume of the basal ganglia of people with VCFS (Eliez et al., 2002; Kates et al., 2004; Campbell et al., 2006). Given that the caudate nucleus represents a central component of the cerebral network underlying temporal perception skills, the present report proposes

This review describes experiments on the effects of caudate nucleus lesions on behavior in monkeys, cats and rats. Early work on monkeys and cats focused on the relationship of the caudate to the cortex in motor control, leading to the idea that the caudate serves to inhibit behaviors initiated by the cortex. However, investigation of this hypothesis with systematic behavioral testing in all three species did not support this idea; rather, these studies provided evidence that caudate lesions affect memory functions. Two main types of memory tasks were affected. One type involved reinforced sti

The extracellular levels of gamma-aminobutyric acid (GABA) in the caudate-putamen and the nucleus accumbens of rats following administration of haloperidol decanoate, fluphenazine decanoate, or vehicle for 8 months were assessed using intracranial microdialysis. Basal levels of extracellular GABA were significantly decreased in the nucleus accumbens of both neuroleptic-treated groups while levels of GABA in the caudate-putamen were not significantly different between groups. These results provide evidence for selective chronic neuroleptic-induced effects on in vivo GABA function in different t

In cats electrostimulation of nucleus caudatus and putamen produced contralateral rotation of the head which was used to simulate the extrapyramidal torsion disorders. Single or repeated injections of cataleptogenic neuroleptic drugs (haloperidol, metoclopramide) caused nonuniform changes of the thresholds of rotation induced by putamen stimulation. On the contrary, atypical neuroleptics (clozapine, sulpiride, tioridazine) facilitated equally both types of striatum-induced responses. The authors infer that acute muscular dystonia can result from disorder in intrastriatal relations between nucl

INTRODUCTION: Both ophthalmic and retinal artery occlusion and caudate nucleus infarction are forms of arterial ischemic stroke, which are rare in adolescents. The causal relationship between these two events is still unclear.
CASE PRESENTATION: A 17-year-old Han Chinese boy presented with vision loss in his left eye for 24 h and was diagnosed with ophthalmic and retinal artery occlusion. The ophthalmologic examination showed no light perception and no pupil light reflex. The fundus examination revealed a cherry-red spot at the macula with white ischemic retina and optic disc. Intra-arterial

Previous studies have suggested a dopaminergic regulation of eye blink rates in human and nonhuman primates. Blockade of either dopamine (DA) D1 or DA D2 receptors or DA depletion induced by the dopaminergic neurotoxin MPTP both decrease spontaneous eye blink rates in monkeys. MPTP-induced decreases in blink rates can be reversed by administration of the full efficacy D1 agonist dihydrexidine, which has also been found to have dramatic antiparkinsonian effects in MPTP-treated animals. Increases in blink rates can also be induced by D1 and D2 agonists in normal animals. In the current study, we

Extracellular gamma amino butyric acid (GABA) levels were measured in the caudate nucleus and the prefrontal cortex of the rhesus monkey brain using in vivo microdialysis under isofluorane gas anesthesia. Evoked GABA release was investigated for voltage sensitivity and calcium (Ca2+) dependency. There was a multifold increase in extracellular GABA levels following local perfusion with: (1) high potassium (50 mM, KCI), (2) veratridine (10 microM), and (3) the GABA releasing agent and uptake blocker, (-) nipecotic acid (1 mM). Release of GABA was significantly reduced when veratridine or (-) nip

This study describes the short-term effects of intracaudate microinjections of carbachol in temporarily isolated and restrained Java monkeys. The monkeys were found to display a series of motor disturbances including blepharoptosis, facial twitches, tongue protrusions, ear flattening, torticollis, and compulsive alternations of rapid flexions and extensions of the extremities. In general, carbachol was found to produce consistent effects as far as it concerns its ability to elicit motor disturbances. Three of the five tested monkeys had previously received another series of carbachol injection

Electrolytic ablation of the ventral parts of the cat caudate nucleus head results in an increase of frequency and disorganization of the pattern of the stereotype head turnings, induced by large doses of amphetamine. Lesion of the dorsal parts, on the other hand, is attended with a decreased number and limited manifestation of stereotype movements. A similar effect appears following a low frequency electrical stimulation of the nucleus ventral part. The disrupting action of neuroleptic haloperidol on the amphetamine-induced stereotype is weakened in animals with ventral lesions and enhanced i

Alcohol use disorder (AUD) induces complex transcriptional and regulatory changes across multiple brain regions including the caudate nucleus, which remains understudied. Using paired single-nucleus RNA-seq and ATAC-seq on caudate samples from 143 human postmortem brains, including 74 with AUD, we identified 17 distinct cell types. We found that a significant portion of the alcohol-induced changes in gene expression occurred through altered chromatin accessibility. Notably, we identified novel transcriptional and chromatin accessibility differences in medium spiny neurons, impacting pathways s

Structural and functional alterations in the brain's reward circuitry are present in cocaine use disorder (CocUD), but their molecular underpinnings remain unclear. To investigate these mechanisms, we performed single-nuclei multiome profiling on postmortem caudate nucleus tissue from six individuals with CocUD and eight controls. We profiled 31,178 nuclei, identifying 13 cell types including D1- and D2-medium spiny neurons (MSNs) and glial cells. We observed 1,383 differentially regulated genes and 10,235 differentially accessible peaks, with alterations in MSNs and astrocytes related to neur

BACKGROUND: The mechanisms underlying neurodegeneration in the striatum of Huntingon's Disease (HD) brain are currently unknown. While the striatum is massively degenerated in symptomatic individuals, which makes cellular characterization difficult, it is largely intact in asymptomatic HD gene positive (HD+) individuals. Unfortunately, as striatal tissue samples from HD+ individuals are exceedingly rare, recent focus has been on the Brodmann Area 9 (BA9), a relatively unaffected region, as a surrogate tissue. In this study, we analyze gene expression in caudate nucleus (CAU) from two HD+ indiv

BACKGROUND: Biotin-thiamine-responsive basal ganglia disease (BTBGD) is a treatable neurometabolic disease caused by variants in SLC19A3. Typical imaging features include symmetrical involvement of the caudate nuclei and putamina.
OBJECTIVE: The study sought to explore classical BTBGD without caudate nucleus involvement, to highlight the importance of recognizing this new pattern early in the disease.
METHODS: Individuals with genetically confirmed BTBGD who harbored the same homozygous variant: NM_025243.4 (SLC19A3): c.1264A > G (p.Thr422Ala) and had atypical neuroimaging were recruited.
