Potential involvement of IL-32 in cell-to-cell communication between macrophages and hepatoblastoma.

PURPOSE: This study investigated the expression of interleukin 32 (IL-32) in hepatoblastoma, the most common primary pediatric liver tumor, and its possible roles in tumorigenesis. METHODS: IL-32 expression was investigated in two hepatoblastoma cell lines (Hep G2 and HuH 6) in the steady state and after co-culture with macrophages by RNA-seq analysis and RT-qPCR, and after stimulation with chemotherapy. Cultured macrophages were stimulated by IL-32 isoforms followed by RT-qPCR and western blot analysis. IL-32 immunohistochemical staining (IHC) was performed using specimens from 21 hepatoblas

Constructing cell-type-specific gene regulatory networks from cell-cell communication and a global gene regulatory network.

Accurately inferring cell-type-specific gene regulatory networks (GRNs) is crucial for understanding cellular heterogeneity, lineage determination, and disease progression mechanisms. Although single-cell RNA sequencing (scRNA-seq) enables high-resolution expression profiling, its inherent sparsity and high noise levels pose challenges to reliable GRN reconstruction. To address these issues, we propose cell type-specific gene regulatory network (CTN), a regulatory inference framework based on bilateral self-representation matrix decomposition, which integrates bulk RNA-seq data with scRNA-seq

A transcriptomic-driven segmentation and cell simulation framework for high-resolution spatial transcriptomics and cell-cell communication.

The Visium HD spatial transcriptomics platform enables transcriptome-wide profiling at near-single-cell resolution. However, accurate segmentation of cells to define spatial boundaries relies heavily on histological images. Previous approaches struggle to define cells when the tissues have high cell density, are inflamed, or are mineralized, leading to transcriptomic bleed-through and inaccurate clustering. To address this, we developed TENGU (Transcript-signal Enrichment and Grouping Unit), a comprehensive end-to-end bioinformatic software package. Unlike existing tools, TENGU employs a trans

Immunothrombotic Cell-Cell Communication Networks in Coronary Atherosclerosis: Critical Insights from Single-Cell and Spatial Systems Biology.

Coronary artery disease (CAD) is increasingly recognized as a thromboinflammatory disorder in which innate immune activation and coagulation are tightly coupled within the plaque microenvironment. Emerging single-cell and spatial technologies have refined this paradigm by demonstrating that these processes are not diffusely distributed but instead concentrated within discrete cellular niches. This narrative review critically evaluates mechanistic and translational studies integrating single-cell RNA sequencing, spatial transcriptomics, and ligand-receptor modeling to characterize cell-cell com

Single-cell RNA sequencing reveals cell-cell communication and potential biomarker in sepsis and septic shock patients.

BACKGROUND: Sepsis is a disease characterized by infection-induced multiorgan dysfunction, which can progress to septic shock if not promptly treated. Early identification of sepsis is crucial for its treatment. However, there are currently limited specific biomarkers for sepsis or septic shock. This study aims to identify potential biomarkers for sepsis and septic shock. METHODS: We analyzed single-cell transcriptomic data of peripheral blood mononuclear cells (PBMCs) from healthy individuals, sepsis and septic shock patients, identified differences in gene expression and cell-cell communica

Prediction of cell-cell communication patterns of dorsal root ganglion cells: single-cell RNA sequencing data analysis.

Dorsal root ganglion neurons transmit peripheral somatic information to the central nervous system, and dorsal root ganglion neuron excitability affects pain perception. Dorsal root ganglion stimulation is a new approach for managing pain sensation. Knowledge of the cell-cell communication among dorsal root ganglion cells may help in the development of new pain and itch management strategies. Here, we used the single-cell RNA-sequencing (scRNA-seq) database to investigate intercellular communication networks among dorsal root ganglion cells. We collected scRNA-seq data from six samples from th

A Novel Cell-Cell Communication Structure: Tanycyte and Cilia Hypothalamic Unifying Glia-cilia Structure (HUGS).

Primary cilia, microtubule-based sensory organelles that mediate cell-cell communication, may facilitate signaling in the brain through direct physical contacts (e.g., synapse-like structures). Similarly, specialized glial cells lining the third ventricle (3V) called tanycytes signal through physical interactions and can dynamically alter their morphology in response to external stimuli and physiological changes. Here, we identify robust cilia-tanycyte contacts; we term HUGS ( H ypothalamic, U nifying G lia-cilia S tructures) and discover that these connections are disrupted in a mouse ciliopa

Lipopolysaccharide alters cell communication at the maternal-fetal interface revealed by single-cell RNA-sequencing.

Embryo implantation is a decisive process in pregnancy that highly relies on effective cell communication at the maternal-fetal interface. Embryo implantation failure is frequently caused by gram-negative bacterial infection, therefore, this study aimed to investigate the effect of Lipopolysaccharides (LPS)-induced inflammation on cellular composition, cell-cell interaction and key signaling pathways at the maternal-fetal interface using Single-cell RNA-Sequence (scRNA-Seq). LPS exposure significantly up-regulated the expression of pro-inflammatory cytokines, CCL-2, TNF-α, and IL-1β in materna

B Cell Orchestration of Anti-tumor Immune Responses: A Matter of Cell Localization and Communication.

The immune system plays a crucial role in cancer development either by fostering tumor growth or destroying tumor cells, which has open new avenues for cancer immunotherapy. It was only over the last decade that the role of B cells in controlling anti-tumor immune responses in the tumor milieu has begun to be appreciated. B and plasma cells can exert anti-tumor effects through antibody-dependent cell cytotoxicity (ADCC) and activation of the complement cascade, even though their effector functions extend beyond the classical humoral immunity. In tumor tissues, B cells can be found in lymphoid

Co-cultures of multiple cell types mimic pulmonary cell communication in response to urban PM10.

The current authors evaluated whether a system of co-cultures of relevant cells (pneumocytes (A549), macrophages (THP-1), mast cells (HMC-1) and endothelial cells (EAHY926)) would mimic the responses to particles with a 50% cut-off aerodynamic diameter of 10 microm (PM(10)) previously reported in vivo. The role of mast cells was considered of special interest. Single cultures, bicultures (A549 + HMC-1 in a 10:1 ratio; THP-1 + HMC-1 in a 2:1 ratio) and tricultures (A549 + THP-1 + HMC-1 in a 10:2:1 ratio) were exposed to urban PM(10) (24 h at 0, 10, 30 or 100 microg x cm(-2)). Additionally, EAHY

Oestriol and oestradiol increase cell to cell communication and connexin43 protein expression in human myometrium.

Oestradiol increases the protein expression of connexin43 (Cx43) gap junctions in myometrium but the effect of oestriol on gap junction expression has not been described previously. Oestriol is the most abundant free oestrogen in pregnant women and there is a marked surge in oestriol concentrations before term and idiopathic preterm labour. In order to determine whether oestriol may have a physiological action on the myometrium, cultured human myometrial cells obtained from non-pregnant hysterectomy specimens were exposed to 10 nmol/l oestradiol or oestriol. Hepatocyte-to-intestinal stem cell remote communication regulates blood glucose homeostasis.

The liver is known to play a pivotal role in modulating blood glucose homeostasis through intrahepatic glucose metabolism. Here, we reveal a unique mechanism by which fatty liver exacerbates hyperglycemia through remote communication from hepatocytes to intestinal stem cells (ISCs), independent of enhanced intrahepatic gluconeogenesis. Mechanistically, hepatocyte-derived alkaline phosphatase (ALP) targets α2δ-1 in ISCs to promote the membrane translocation of Cav1.2. This process triggers increased intracellular calcium levels, which subsequently activates the calcineurin/NFATC2 signaling axis

Gut immune cell trafficking: inter-organ communication and immune-mediated inflammation.

Immune cell trafficking is a complex and tightly regulated process that is indispensable for the body's fight against pathogens. However, it is also increasingly acknowledged that dysregulation of cell trafficking contributes to the pathogenesis of immune-mediated inflammatory diseases (IMIDs) in gastroenterology and hepatology, such as inflammatory bowel disease and primary sclerosing cholangitis. Moreover, altered cell trafficking has also been implicated as a crucial step in the immunopathogenesis of other IMIDs, such as rheumatoid arthritis and multiple sclerosis. Over the past few years,

Side-specific valvular endothelial-interstitial cell mechano-communication via cadherin-11.

Calcific aortic valve disease (CAVD) is a condition causing stiffening of the aortic valve, impeding cardiac function and resulting in significant morbidity worldwide. CAVD is thought to be driven by the persistent activation of the predominant cell type in the valve, aortic valve interstitial cells (AVICs), into myofibroblasts, resulting in subsequent calcification and stenosis of the valve. Although much of the research into CAVD focuses on AVICs, the aortic valve endothelial cells (AVECs) have been shown to regulate AVICs and maintain tissue homeostasis. Exposed to distinct flow patterns du

CellCom2: An Integrated Web Platform for Flexible Prediction and Visualization Analysis of Cell-cell Communication.

Cell-cell communication across diverse cell types and tissues underlies proper organismal function and is primarily mediated by interactions between secreted ligands and cell-surface receptors. Understanding these molecular interactions is essential for elucidating regulatory networks governing physiological and pathological processes and for identifying potential therapeutic targets. Although several computational tools have been developed for inferring intercellular communication, many existing methods require substantial programming expertise, rely on restricted or method-specific biologica

Oxidative stress induced by DMSA-IONPs impairs breast cancer cell migration and paracrine cell communication.

BACKGROUND: The migration of tumor cells requires a complex interplay of these cells with their microenvironment. Significantly, these migrating (metastatic) cells undergo metabolic and oxidative changes that render them more sensitive to shifts in their redox homeostasis. Nanoparticle (NP)-based pro-oxidant therapies are strategies that can potentially influence cell viability and migration. Among these, iron oxide NPs (IONPs) stand out due to their biocompatibility and the fact they induce ROS production through the Fenton Reaction. Here we investigate the possibility of using dimercaptosucc

Predicting Relevant Microglia-Associated Cell-Cell Communication Pathways in Alzheimer's Disease: A Role for SPP1.

Microglia play a key role in the pathophysiology of Alzheimer's Disease (AD) and their increased heterogeneity likely affects disease progression. We previously identified distinct microglial signatures that were enriched in AD donors and associated with amyloid and tau, respectively. Here we generated a snRNAseq dataset from postmortem control and AD cases and analyzed alterations in cell-cell communication pathways that might be relevant to AD pathophysiology. One signaling pathway perturbed in AD cases involved SPP1, and while this pathway was also present in control samples, microglia-micr
