BACKGROUND: Cumulus granulosa cells (CGCs) are essential for oocyte support, metabolic cooperation, and steroidogenesis, yet they deteriorate rapidly in conventional 2D culture, limiting long-term studies of these human ovarian somatic cells. Existing 3D systems rarely focus specifically on CGCs or sustain their function over extended periods. Here we developed a novel hCGC spheroid model encapsulated in PEGylated fibrin hydrogel, a stable, ovarian tissue-like biomaterial, to sustain viability, architecture, and endocrine activity over 30 days, providing a preclinical somatic platform for ovar

In vitro models of differentiated respiratory epithelium that allow high-throughput screening are an important tool to explore new therapeutics for chronic respiratory diseases. In the present study, we developed in vivo-like three-dimensional (3-D) models of bronchial epithelial cell lines that are commonly used to study chronic lung disease (16HBE14o-, CFBE41o- and CFBE41o- 6.2 WT-CFTR). To this end, cells were cultured on porous microcarrier beads in the rotating wall vessel (RWV) bioreactor, an optimized suspension culture method that allows higher throughput experimentation than other phy

To study the roles of bacterial secretome in immune cell polarization and gut mucosal homeostasis, we developed a 3-dimensional co-culture model using commercially available inserts in a back-to-back stacked format. We termed this system 3D Flipwells, and successfully co-cultured bacteria, colon epithelial, and immune cells in stratified layers to recapitulate the gut mucosal microenvironment. This co-culture system enables the seeding of colon epithelial cells on one side of the membrane and undifferentiated non-adherent monocytes on the other, allowing for the synchronous progression of colo

Obesity represents a complex systemic disease resulting in the global metabolic syndrome epidemic, yet our understanding of the underlying mechanisms in adipose tissue driving obesity remains limited. As the primary cellular targets of increased caloric intake, adipocytes undergo dramatic morphological and functional changes that trigger systemic metabolic dysfunction. To study the dysfunctional obese adipocyte phenotype in vitro, revolutionary three-dimensional (3D) adipocyte models have shown to be uniquely capable of recapitulating the lipid-laden, hypertrophic adipocyte characteristic of o

Focused Ultrasound (FUS) provides a non-contact, directional acoustic field extensively employed in diverse biomedical applications. However, conventional concave transducers are physically constrained to a single fixed focus, lacking the flexibility for dynamic field modulation necessary to meet increasingly complex functional requirements. In this work, we report a focused ultrasound-based acoustic holography (FUS-AH) approach that enables concave transducers to achieve sophisticated spatial control and generate intricate multi-focal patterns. By systematically investigating the propagation

Haematological malignancies comprise a diverse group of life-threatening systemic diseases, including leukaemia, lymphoma, and multiple myeloma. Currently available therapies, including chemotherapy, immunotherapy, and CAR-T cells, are often associated with important side effects and with the development of drug resistance and, consequently, disease relapse. In the last decades, it was largely demonstrated that the tumor microenvironment significantly affects cancer cell proliferation and tumor response to treatment. The development of biomimetic, in vitro models may promote the investigation

This study investigates the use of industrial-grade polydimethylsiloxane (PDMS) for mass production of microfluidic chips through Liquid Silicone Rubber Injection Molding (LSR-IM) technologies. Injection molding of PDMS is ubiquitous in the automotive and aerospace industries. Yet, it is not currently applied to large-scale production of microfluidic devices. Here, we manufactured PDMS microfluidics devices using industrial IM systems and measured the values of several physicochemical properties of PDMS important in microfluidics. For the two grades of injection-moldable PDMS studied, properti

Fibrocyte degeneration in the cochlear lateral wall is one possible pathology of age-related metabolic hearing loss (presbycusis). Within the lateral wall fibrocytes play a role in potassium recycling and maintenance of the endocochlear potential. It has been proposed that cell replacement therapy could prevent fibrocyte degeneration in the CD/1 mouse model of hearing loss. For this to work, the replacement fibrocytes would need to take over the structural and physiological role of those lost. We have grown lateral wall fibrocytes from neonatal CD/1 mice in a 3D-collagen gel culture with the a

BACKGROUND: Human pluripotent stem cells (hPSCs) offer a promising source of hepatocytes for disease modeling and drug screening. However, hepatocytes derived in conventional two-dimensional (2D) cultures often exhibit incomplete maturation, limiting their physiological relevance. The molecular mechanisms underlying the improved differentiation observed in three-dimensional (3D) culture systems remain poorly understood.
RESULTS: Induced PSCs (iPSCs) were differentiated into hepatic progenitors under 2D monolayer culture. At the hepatic progenitor stage, cells were either continued in 2D or re

Osteoarthritis (OA) is the most prevalent degenerative joint disease worldwide, imposing a substantial global disease burden. However, its pathogenesis remains incompletely understood, and effective treatment strategies are still lacking. Organoid technology, in which stem cells or progenitor cells self-organise into miniature tissue structures under three-dimensional (3D) culture conditions, provides a promising in vitro platform for simulating the pathological microenvironment of OA. This approach can be employed to investigate disease mechanisms, carry out high-throughput drug screening and

Psoriasis is a chronic inflammatory skin disease involving several cell types, including T cells, via the IL-23/IL-17 axis. IL-17A acts on the surrounding epithelial cells thus resulting in an inflammatory feedback loop. The development of immunocompetent models that correctly recapitulate the complex phenotype of psoriasis remains challenging, which also includes both the T cell isolation and activation methods. The purpose of this work was to develop an advanced in vitro 3D psoriatic skin model that enables the study of the impact of T cells on psoriatic epithelial cells. To reach that aim,

High-content imaging (HCI) is a powerful method for quantifying biological effects in vitro. Historically, HCI has been applied to adherent cells growing in monolayers. With the advent of confocal versions of HCI devices, researchers now have the option of performing analyses on 3D cell cultures. However, some obstacles remain in integrating the third dimension, such as limited light penetration and less sophisticated image analysis. Here, we report the development of an HCI technique for imaging human bronchial 3D organotypic air-liquid interface (ALI) cultures (hBR-ALI). In this method, we m

Research insights into uterine function and the mechanisms of labour have been hindered by the lack of suitable animal and cellular models. The use of traditional culturing methods limits the exploration of complex uterine functions, such as cell interactions, connectivity and contractile behaviour, as it fails to mimic the three-dimensional (3D) nature of uterine cell interactions in vivo. Animal models are an option, however, use of these models is constrained by ethical considerations as well as translational limitations to humans. Evidence indicates that these limitations can be overcome b

For the past decades, many advances were made in the field of immunotherapies, most notably the development of chimeric antigen receptor (CAR) T cells, that allowed development of potent and targeted treatment options against cancers and other diseases. To determine the preclinical cytotoxic efficacy of T cell therapies, such as CAR T cell therapy, bioluminescence-based assays have emerged as a common technique, thanks to their high sensitivity, reproducibility and ease of use. Because the extracellular matrix (ECM) plays a key role in tumor development, numerous 3D cancer models have been gen

Three-dimensional (3D) in vitro models provide a physiologically relevant platform that more accurately recapitulates tissue architecture, cell-cell and cell-extracellular matrix interactions, and the physical microenvironment than traditional two-dimensional (2D) cultures. In particular, these models reproduce metabolic gradients, including oxygen and nutrient availability, enabling more precise investigations of cellular metabolism. This chapter describes two methods to grow Ewing sarcoma cell lines as 3D spheroids using physiological media, as well as the collection of spheroids in bulk or

This systematic review aimed to evaluate whether the use of 3D substrates improved cell behaviour compared to 2D substrates for the study of exposure between human dental pulp cells (HDPCs) and bacteria/lipopolysaccharides (LPS). Articles studying the interaction between HDPCs and bacteria/LPS in 3D substrates between January 2000 and May 2025 were included from 4 databases (MEDLINE, Scopus, Web of Science and Embase) following a registered protocol (Prospective Register of Systematic Reviews CRD42024512080) and as per Preferred Reporting Items for Systematic Review and Meta-Analysis guideline

Diabetic keratopathy (DK) is a degenerative corneal disease occurring in more than 50 % of diabetic patients. DK is correlated with the hyperglycemic state causing morphological and functional changes in corneal layers. Currently, most studies on the cornea are performed on two-dimensional (2D) cultures in vitro or animal models. Although 2D culture models can provide large amounts of data at low cost, they poorly represent the complex pathophysiology of the human cornea and hardly predict in vivo responses that can be achieved with animal model studies. However, the use of the latter presents

In vitro cell culture is a well-established technique present in numerous laboratories in diverse areas. In reproduction, gametes, embryos, and reproductive tissues, such as the ovary and endometrium, can be cultured. These cultures are essential for embryo development studies, understanding signaling pathways, developing drugs for reproductive diseases, and in vitro embryo production (IVP). Although many culture systems are successful, they still have limitations to overcome. Three-dimensional (3D) culture systems can be close to physiological conditions, allowing greater interaction between

Decellularization techniques support the creation of biocompatible extracellular matrix hydrogels, providing tissue-specific environments for both in vitro cell culture and in vivo tissue regeneration. We obtained endometrium derived from porcine decellularized uteri to create endometrial extracellular matrix (EndoECM) hydrogels. After decellularization and detergent removal, we investigated the physicochemical features of the EndoECM, including gelation kinetics, ultrastructure, and proteomic profile. The matrisome showed conservation of structural and tissue-specific components with low amou

3D printing has become widespread for the manufacture of parts in various industries and enabled radically new designs. This trend has not spread to bioprocess development yet, due to a lack of material suitable for the current workflow, including sterilization by autoclaving. This work demonstrates that commercially available heat temperature stable poly-lactic acid (PLA) can be used to easily manufacture novel bioreactor vessels with included features like harvest tubes and 3D printed spargers. Temperature responsiveness was tested for PLA, temperature stable PLA (PLA-HP) and glass for tempe
