A membranous basal cell adenoma arose from the minor salivary glands in the hypopharynx of a 62-year-old man. Features distinguishing the membranous variant from other forms of basal cell adenoma include the following: (1) abundant reduplicated basement membrane material, (2) the presence of sialomucin-containing cells, and (3) the lack of distinct encapsulation. To our knowledge, this is the first report of a basal cell adenoma of the hypopharynx, and the first report of the membranous variant in a minor salivary gland.

Polarized monolayers of cultured epithelial cells, such as the kidney-derived MDCK cell line, when infected with enveloped viruses, provide a convenient model system for study of the intracellular routes followed by newly synthesized glycoproteins to reach specific domains of the plasma membrane. The polarized nature of the monolayers is reflected in the asymmetric assembly of enveloped viruses, some of which, such as influenza and simian virus 5 (SV5), bud from the apical surfaces of the cells, while others, such as vesicular stomatitis virus (VSV), emerge from the basolateral surfaces. MDCK

Various cell-cell interactions are triggered by the binding of ligands to receptors on the cell membrane. This binding occurs effectively within a biological membrane crowded with other types of membrane molecules. Understanding these initial binding events provides fundamental knowledge of cellular responses and insights into interface design for various biomedical applications, such as drug delivery systems, cell-based therapies, and the formation of multicellular aggregates. A detailed analysis requires an experimental model that can quantitatively track ligand-receptor binding at the membr

Membrane vesicles (MVs) released from bacteria into the extracellular environment play various roles in bacterial and bacterial-host interactions. In Gram-positive bacteria, cell wall degradation or damage is crucial for cytoplasmic MV (CMV) release because the cytoplasmic membrane must pass through the thick cell wall to the extracellular space. Although Streptococcus mutans, a major causative agent of dental caries, is known to release CMVs that are involved in pathogenicity and biofilm formation, the mechanism by which CMV production is triggered is unknown. In this study, we demonstrate th

The physical and physiological behavior of sickle cell trait carriers (AS) is somewhat equivocal under strenuous conditions, although this genetic abnormality is generally considered to be a benign disorder. The occurrence of incidents and severe injuries in AS during exercise might be explained, in part, by the lactic acidosis due to a greater lactate influx into AS red blood cells (RBCs). In the present study, the RBC lactate transport activity via the different pathways was compared between AS and individuals with normal hemoglobin (AA). Sixteen Caribbean students, nine AS and seven AA, per

The corneal endothelium, comprising densely packed corneal endothelial cells (CECs) adhering to Descemet's membrane (DM), plays a critical role in maintaining corneal transparency by regulating water and ion movement. CECs have limited regenerative capacity within the body, and globally, there is a shortage of donor corneas to replace damaged corneal endothelia. The development of a carrier for cultured CECs may address this worldwide clinical need. In this study we successfully manufactured a gelatin nanofiber membrane (gelNF membrane) using electrospinning, followed by crosslinking with glut

Cell membrane repair is a critical process used to maintain cell integrity and survival from potentially lethal chemical, and mechanical membrane injury. Rapid increases in local calcium levels due to a membrane rupture have been widely accepted as a trigger for multiple membrane-resealing models that utilize exocytosis, endocytosis, patching, and shedding mechanisms. Calcium-sensor proteins, such as synaptotagmins (Syt), dysferlin, S100 proteins, and annexins, have all been identified to regulate, or participate in, multiple modes of membrane repair. Dysfunction of membrane repair from ineffi

In order to maintain persistent infections, microbes that cause chronic disease have to evade detection by the human immune system. To do so, many modulate the expression of plasma membrane receptors that trigger cell signalling pathways and immune responses. Using microscopy and cell sorting techniques, Businger et al. map the morphological changes in the plasma membranes of macrophages infected by human cytomegalovirus or human immunodeficiency virus and find novel differentially expressed receptors.

Congenital haemolytic anaemias are inherited disorders caused by red blood cell membrane and cytoskeletal protein defects, deviant hemoglobin synthesis and metabolic enzyme deficiencies. In many cases, although the causing mutation might be known, the pathophysiology and the connection between the particular mutation and the symptoms of the disease are not completely understood. Thus effective treatment is lagging behind. As in many cases abnormal red blood cell cation content and cation leaks go along with the disease, by direct electrophysiological measurements of the general conductance of

Plasma membrane lipid rafts are highly ordered membrane microdomains enriched for glycosphingolipids and cholesterol, which play an important role during T-cell antigen receptor (TCR) signaling. Our previous work has demonstrated that plasma membrane lipid composition is an important determinant of human CD4+ T-cell function and that defects in lipid raft expression contribute to CD4+ dysfunction in patients with autoimmunity. In this chapter we share three flow cytometry-based methods to quantitatively analyze plasma membrane lipid composition in primary human CD4+ T cells. We describe the qu

Fluorescent inorganic quantum dots are highly promising for biomedical applications as sensing and imaging agents. However, the low internalization of the quantum dots, as well as for most of the nanoparticles, by living cells is a critical issue which should be solved for success in translational research. In order to increase the internalization rate of inorganic CdSe/ZnS quantum dots, they were functionalized with a fluorinated organic ligand. The fluorinated quantum dots displayed an enhanced surface activity, leading to a significant cell uptake as demonstrated by in vitro experiments wit

Ecdysteroids secreted by the prothoracic gland (PG) cells of insects control the developmental timing of their immature life stages. These cells have been historically considered as carrying out a single function in insects, namely the biochemical conversion of cholesterol to ecdysteroids and their secretion. A growing body of evidence shows that PG cells receive multiple cues during insect development so we tested the hypothesis that they carry out more than just one function in insects. We characterised the molecular nature and developmental profiles of cell membrane receptors in PG cells of

A 63-year-old male was admitted with complaints of upper gastrointestinal symptoms with fatigue and myalgia. Investigations revealed severe hyponatremia with elevated creatine phosphokinase levels. Following further workup, it was diagnosed as a case of hyponatremia induced rhabdomyolysis. Because of prompt correction of hyponatremia, his renal function was preserved and myoglobinuria induced renal failure was avoided. The importance of early recognition of this potentially dangerous condition is emphasized.

The term bioelectrical modulators specifically applies to anticonvulsant drugs and mood stabilizers; however, it also embodies the larger more generic concept that bioelectrical modulators can mitigate any pathological condition caused by a dysregulation of the mechanisms that control cellular excitability, especially the excitability of neurons. The beneficial effects of these agents occur primarily as a result of modulatory influences on the bioelectrical properties of the cellular plasma membrane. Channels, transporters, and most other membrane proteins directly or indirectly involved in ex

Spindle cell melanoma (SCM) is an uncommon form of melanoma that may be confused histologically with other tumors, including malignant peripheral nerve sheath tumors (MPNST). Tumors with neural differentiation and melanocytic nevi may both show basement membrane immunohistochemically and at the ultrastructural level. However, most ultrastructural studies of melanoma have failed to demonstrate well formed basement membrane around tumor cells. The presence of basement membrane has been used by some authors as evidence favoring MPNST, as opposed to SCM. To evaluate this distinction immunohistoche

The present study explores the role of different ionic conductances in the regulation of membrane potential under resting conditions and after bradykinin (BK) or thapsigargin (TG) stimulation of cultured bovine aortic endothelial cells. Under resting conditions, the cell membrane potential observed was -62+/- 5 mV. The main conductance under these conditions is an inwardly rectifying potassium (IRK) channel. Application of 50 nM BK induced a transient hyperpolarization to -87 +/- 4 mV followed by sustained depolarization to -35 +/- 5 mV. The transient hyperpolarization was eliminated by 1 micr

HPCA is a rare disease among hemolytic anemias. A dominantly transmitted chronic anemia characterized by an increase of erythrocyte membrane phosphatidylcholine (PC) and cholesterol with decrease of phosphatidylethanolamine (PE). On peripheral blood smears stomatocytes and target cells are evident. The MCV may be increased, but the life span of red cells is decreased. The osmotic fragility is normal or increased, but splenectomy is not effective. These two points are different from other hereditary hemolytic anemias. As in other hemolytic anemias, gallstone and biliary tract disease more frequ

The concentrations of total ([T-Mg]), ultrafilterable ([UF-Mg]), and protein-bound or nonfilterable ([NF-Mg]) magnesium were measured in the plasma and in the intracellular compartment of blood from 8 essential hypertensive patients and 9 normotensive subjects. In the former, [T-Mg] was unchanged in the plasma but decreased in whole blood due to decreases of both [UF-Mg] and [NF-MG]; [UF-Mg] was increased in plasma but decreased intracellularly while [NF-Mg] was decreased in plasma and unchanged intracellularly. These concentrations correlated significantly with the average blood pressures. De

Cytoskeletal abnormalities in red cells were studied in 250 patients at our laboratory, especially on hereditary elliptocytosis (HE), hereditary spherocytosis (HS), and band 4.2 anomalies. First of all, on HE, we have found two patients of a dominantly-inherited trait of a new beta-spectrin variant with 216 kDa peptide. The contents of abnormal beta'-spectrin to the total spectrins were 7.6% in propositus, and 10.5% in her mother. As a functional abnormality, abnormal alpha beta'-spectrin dimer could not be converted to tetramer in both patients. Thus, our patients may differ from HE Nice. Sec

Cross bonding and stiffening of the human red cell membrane was studied using treatments with SH, amino, and carboxyl reagents, oxidizing and denaturing treatments and acidification. Membrane cross bonding was initiated when, after red cell treatment, opposite areas of the cytoplasmic face of the red cell membrane were brought into contact by cell shrinking. Membrane cross bonding was detected by light microscopy when this contact persisted upon swelling the cells in a hypotonic medium. Membrane stiffening was recorded as a decrease in elongation of red cells in the shear field of a viscous de
