The Origin and Fate of Chondrocytes: Cell Plasticity in Physiological Setting.

PURPOSE OF REVIEW: Here, we discuss the origin of chondrocytes, their destiny, and their plasticity in relationship to bone growth, articulation, and formation of the trabeculae. We also consider these processes from a biological, clinical, and evolutionary perspective. RECENT FINDINGS: Chondrocytes, which provide the template for the formation of most bones, are responsible for skeletal growth and articulation during postnatal life. In recent years our understanding of the fate of these cells has changed dramatically. Current evidence indicates a paradoxical situation during skeletogenesis,

Sex differences in anatomic plasticity of gut neuronal-mast cell interactions.

The gut wall houses mast cells that are anatomically situated near enteric neuronal fibers. Roles of specific neuropeptides in modulating function of immune components like mast cells in response to challenge with bacterial components are relatively unknown. Investigating such interactions requires models that include diverse cellular elements in native anatomic arrangements. Using an organotypic slice model that maintains gut wall cellular diversity ex vivo, the present study compared responses between tissues derived from male and female mice to examine neural-immune signaling in the gut wal

Gut resistome plasticity in pediatric patients undergoing hematopoietic stem cell transplantation.

The gut microbiome of pediatric patients undergoing allo-hematopoietic stem cell transplantation (HSCT) has recently been considered as a potential reservoir of antimicrobial resistance, with important implications in terms of patient mortality rate. By means of shotgun metagenomics, here we explored the dynamics of the gut resistome - i.e. the pattern of antibiotic resistance genes provided by the gut microbiome - in eight pediatric patients undergoing HSCT, half of whom developed acute Graft-versus-Host Disease (aGvHD). According to our findings, the patients developing aGvHD are characteriz

Commensal-specific T cell plasticity promotes rapid tissue adaptation to injury.

Barrier tissues are primary targets of environmental stressors and are home to the largest number of antigen-experienced lymphocytes in the body, including commensal-specific T cells. We found that skin-resident commensal-specific T cells harbor a paradoxical program characterized by a type 17 program associated with a poised type 2 state. Thus, in the context of injury and exposure to inflammatory mediators such as interleukin-18, these cells rapidly release type 2 cytokines, thereby acquiring contextual functions. Such acquisition of a type 2 effector program promotes tissue repair. Aberrant

Diverting T helper cell trafficking through increased plasticity attenuates autoimmune encephalomyelitis.

Naive T helper cells differentiate into functionally distinct effector subsets that drive specialized immune responses. Recent studies indicate that some of the effector subsets have plasticity. Here, we used an EAE model and found that Th17 cells deficient in the transcription factor BCL11B upregulated the Th2-associated proteins GATA3 and IL-4 without decreasing RAR-related orphan receptor γ (RORγt), IL-17, and GM-CSF levels. Surprisingly, abnormal IL-4 production affected Th17 cell trafficking, diverting migration from the draining lymph nodes/CNS route to the mesenteric lymph nodes/gut rou

Cerebral protection, brain repair, plasticity and cell therapy in ischemic stroke.

Among available treatments in the acute ischemic stroke, only intravenous thrombolysis has been demonstrated to be efficacious. Although the majority of pharmacological neuroprotectants have been efficacious in experimental studies, they have failed in clinical trials. Hence, we need to consider integrated cerebral protection which includes the concept of cerebral repair by supporting cerebral plasticity. We provide a nonsystematic review of the studies published on cerebral protection and repair treatments of cerebral ischemia considering the possibilities of stimulating brain plasticity by t

MAP2-mediated integrin dysregulation reprograms cancer cell plasticity in hepatocellular carcinoma.

Targeting tumor cell plasticity is central to overcome drug resistance and metastasis. Through lineage-tracing and lineage-ablation studies, our team has previously shown hepatocellular carcinoma (HCC) cells marked by CD133 to represent a functional subset displaying dedifferentiated status with stemness traits, yet there are limited therapies effective for eradicating such resilient cell populations. Here, we explore the therapeutic potential of repurposing an existing drug estramustine phosphate (EMP) for targeting HCC plasticity through inhibiting MAP2, a gene uniquely enriched in CD133+ 'H

The shifting balance: innate lymphoid cell plasticity in chronic gut diseases.

Innate lymphoid cells (ILCs), as core components of innate immunity, coordinate mucosal immune defense and homeostasis through their functional plasticity. They exhibit functional diversity in the intestine and display a complex dual role in intestinal diseases, which is closely associated with their phenotypic transformation and functional reprogramming. Current studies have demonstrated that ILCs play critical roles in various chronic intestinal diseases, including autoimmune intestinal disorders, chronic infectious intestinal inflammation, and colorectal cancer. However, the specific functi

Cancer Heterogeneity and Cancer Cell Plasticity: Molecular Mechanisms and Precision Therapy.

Tumor heterogeneity creates selective pressures and ecological niches that enable cancer cell plasticity; conversely, plasticity continuously generates and reshapes heterogeneity. Together, these processes drive tumor progression, therapeutic resistance, recurrence, and divergent clinical outcomes, thereby limiting the durability of precision oncology. In this Review, we synthesize recent advances in our understanding tumor heterogeneity and cancer cell plasticity across spatial, temporal, and molecular scales, and discuss how the tumor microenvironment regulates plasticity through physical, c

Reactive oxygen species stimulate Paneth cell plasticity and diminish antimicrobial function.

The gut epithelium faces constant environmental oxidative stresses. How it responds to pathological reactive oxygen species (ROS) is not fully understood. Using Paneth cell (PC) lineage reporters and genetic tracing, we show that as PC progenitors mature, a decline in their oxidative phosphorylation (OXPHOS) activity coincides with an increased capacity to produce antimicrobial peptides (AMPs). Direct stimulation by elevated extracellular ROS, often found during gut infection and inflammation, can drive a subset of PCs into mitotic cell division. Increasing mitochondrial superoxide production

Regulated cell death plasticity in cancer: thresholds, reversibility, and therapeutic failure.

Therapy resistance and disease recurrence remain major challenges across cancer types despite substantial advances in targeted therapy, chemotherapy, and immunotherapy. In many clinical settings, treatments successfully engage stress and death signaling pathways and produce rapid tumor regression, yet complete and durable eradication is uncommon. A consistent pattern emerges in which most tumor cells are eliminated while a small fraction survives, persists as residual disease, and later drives relapse. These observations suggest that failure is not always due to the absence of death pathways,

Epithelial cell plasticity in metazoans: Evolutionary insights into roles and mechanisms.

Epithelial tissues function as multicellular communities that preserve tissue integrity while adapting to diverse environmental stresses by altering cell behaviors. A striking manifestation of such adaptability is cell plasticity, the ability of differentiated cells to revert to stem-like states or adopt alternative fates. Once considered rare and confined to highly regenerative species, cell plasticity is now recognized across the metazoan tree. In early-branching animals such as sponges and cnidarians, transdifferentiation and dedifferentiation are integral to life-cycle transitions and rege

Smooth Muscle Cell Plasticity as a Central Determinant of Plaque Stability.

PURPOSE OF REVIEW: This review examines how smooth muscle cell (SMC) fate transitions contribute to the central paradox of atherosclerosis: why thin-cap plaques that only mildly narrow the lumen cause most acute events, whereas many severely stenotic lesions remain stable. We propose that interactions between SMC developmental programming and local microenvironments determine whether SMCs build collagen-rich fibrous caps, undergo medial degeneration, or adopt phenotypes that weaken the vessel wall. RECENT FINDINGS: SMCs show marked plasticity, adopting phenotypes that either strengthen or wea

FOXC1 Controls Smooth Muscle Cell Differentiation and Plasticity in Vascular Disease.

Smooth muscle cell-related transcription factors have been associated with coronary artery disease in large genome-wide association studies, increasing their translational value for therapeutic investigations. Our study shows for the first time that FOXC1 is a major transcriptional regulator in vascular injury and atherosclerotic lesions, causally implicated in the disease. Functionally, our data suggest that FOXC1 modulates smooth muscle cell activation vs quiescence by regulating key pathways involved in cell adhesion, cell cycle progression, and actin cytoskeleton organization. Further inve

Basal cell of origin resolves neuroendocrine-tuft lineage plasticity in cancer.

Neuroendocrine and tuft cells are rare chemosensory epithelial lineages defined by the expression of ASCL1 and POU2F3 transcription factors, respectively. Neuroendocrine cancers, including small cell lung cancer (SCLC), frequently display tuft-like subsets, a feature linked to poor patient outcomes1-9. The mechanisms driving neuroendocrine-tuft tumour heterogeneity and the origins of tuft-like cancers are unknown. Using multiple genetically engineered animal models of SCLC, we demonstrate that a basal cell of origin (but not the accepted neuroendocrine origin) generates neuroendocrine-tuft-lik

Adapt to Persist: Glioblastoma Microenvironment and Epigenetic Regulation on Cell Plasticity.

Glioblastoma (GBM) is the most frequent and aggressive brain tumor, characterized by great resistance to treatments, as well as inter- and intra-tumoral heterogeneity. GBM exhibits infiltration, vascularization and hypoxia-associated necrosis, characteristics that shape a unique microenvironment in which diverse cell types are integrated. A subpopulation of cells denominated GBM stem-like cells (GSCs) exhibits multipotency and self-renewal capacity. GSCs are considered the conductors of tumor progression due to their high tumorigenic capacity, enhanced proliferation, invasion and therapeutic r

DNA methylation in cell plasticity and malignant transformation in liver diseases.

The liver possesses extraordinary regenerative capacity mainly attributable to the ability of hepatocytes (HCs) and biliary epithelial cells (BECs) to self-replicate. This ability is left over from their bipotent parent cell, the hepatoblast, during development. When this innate regeneration is compromised due to the absence of proliferative parenchymal cells, such as during cirrhosis, HCs and BEC can transdifferentiate; thus, adding another layer of complexity to the process of liver repair. In addition, dysregulated lineage maintenance in these two cell populations has been shown to promote

Molecular Factors Mediating Neural Cell Plasticity Changes in Dementia Brain Diseases.

Neural plasticity-the ability to alter a neuronal response to environmental stimuli-is an important factor in learning and memory. Short-term synaptic plasticity and long-term synaptic plasticity, including long-term potentiation and long-term depression, are the most-characterized models of learning and memory at the molecular and cellular level. These processes are often disrupted by neurodegeneration-induced dementias. Alzheimer's disease (AD) accounts for 50% of cases of dementia. Vascular dementia (VaD), Parkinson's disease dementia (PDD), dementia with Lewy bodies (DLB), and frontotempor
