The Xanthomonas axonopodis pv. glycines Cellulase EngXCA Governs Dual Roles in Virulence and Innate Immunity in Soybean.

The Xanthomonas axonopodis pv. glycines (Xag) cellulase EngXCA exhibits a dual functionality in the soybean pathosystem. Deletion of engXCA significantly attenuated bacterial growth and symptom severity, a phenotype that was fully restored by genetic complementation. Plants infected with ΔengXCA exhibited attenuated defense responses, including reduced H2O2 accumulation, callose deposition, and PCD, indicating that EngXCA acts as an immunity elicitor. Soybeans infected with ΔengXCAΔhrcC elicited weaker immune responses than those infected with ΔhrcC alone, confirming that EngXCA functions inde

Cellulase-assisted platelet-rich plasma release from nanofibrillated cellulose hydrogel enhances wound healing.

Platelet-rich plasma (PRP) is a source of growth factors, which are implicated in active tissue regeneration. However, after transplantation the efficacy of these bioactive compounds is often diminished due to rapid degradation and untargeted localization. For this reason, we evaluated the potential of nanofibrillated cellulose (NFC) hydrogel as a PRP carrier. NFC hydrogel is an animal-free biomaterial that, when doped with cellulase, can assist the release of PRP in a wound site. In this study, we examined the effects of 0.5% (m/v) NFC hydrogel formulations, including PRP and cellulase, on th

Development of a pH/Cellulase Dual-Responsive Fe-MOFs Delivery System for Targeted Control of Maize Fungal Diseases.

The iron-based metal-organic frameworks (Fe-MOFs) NH2-MIL-101(Fe) (MIL) has gained widespread attention as a drug carrier material in agriculture. In this study, a multifunctional nanocomposite, MIL@A@D@C, was successfully synthesized by loading the fungicide azoxystrobin (AZOX) and immunoinducer d-cellobiose (D) onto MIL, followed by surface modification with carboxymethyl cellulose (CMC). The loading capacity of MIL was optimized by varying solvent conditions and the mass ratio of MIL to AZOX, revealing that ethanol as the reaction solvent and a MIL: AZOX mass ratio of 1:4 resulted in the hi

Mechanisms and strategies for lignin-targeted pretreatment and β-glucosidase molecular engineering in enhancing cellulase hydrolysis efficiency: A review.

Lignocellulosic biomass (LCB), a second-generation feedstock for cellulosic ethanol production, requires efficient pretreatment. This step is critical in biorefinery processes. During enzymatic hydrolysis, lignin limits cellulase accessibility, whereas β-glucosidase (BGL) functions as a rate-limiting enzyme. This review systematically summarises lignin-targeted pretreatment methods, including acidic, organic solvent, deep eutectic solvent (DES), and biological approaches. Particular emphasis is placed on lignin structural modifications and their implications for enzyme engineering. In addition

Insights into Lignin Structural Divergence Induced by Alkali-Ethanol Pretreatment and Its Regulating Effect on Cellulase Nonproductive Adsorption.

Alkali-ethanol pretreatment effectively removes lignin while preserving cellulose; however, the structural transformations of lignin during this process and their influence on the inhibition of enzymatic hydrolysis remain insufficiently understood. Mitigating the nonproductive adsorption of cellulase enzymes by lignin is of significant industrial importance, as it can reduce enzyme-related costs in large-scale biorefining operations. This study examined lignin fractions derived from alkali-ethanol pretreatment of poplar, specifically black liquor lignin (BL) and pretreatment material milled wo

Trichoderma reesei Nsd3 transcription factor: pleiotropic roles in development, stress response, secondary metabolism, and cellulase production.

Trichoderma reesei is known for its ability to secrete high amounts of cellulases, enzymes of fundamental importance in generating products from lignocellulosic biomass. Diverse signaling pathways and transcription factors (TFs) control the cellulolytic repertoire in T. reesei to ensure correct adaptation to the environment. Here, we analyzed RNA-Seq data and identified a new potential regulator of cellulase production in T. reesei: a novel TF named Nsd3, a homolog of NsdC from Aspergilli. Deletion of nsd3 reduced vegetative growth and conidiation on solid medium. Phenotypic characterization o

Heteroxylan hydrolysis by a recombinant cellulase-free GH10 xylanase from the alkaliphilic bacterium Halalkalibacterium halodurans C-125.

The search for affordable enzymes with exceptional characteristics is fundamental to overcoming industrial and environmental constraints. In this study, a recombinant GH10 xylanase (Xyn10-HB) from the extremely alkaliphilic bacterium Halalkalibacterium halodurans C-125 cultivated at pH 10 was cloned and expressed in E. coli BL21(DE3). Removal of the signal peptide improved the expression, and an overall activity of 8 U/mL was obtained in the cell-free supernatant. The molecular weight of purified Xyn10-HB was estimated to be 42.6 kDa by SDS-PAGE. The enzyme was active across a wide pH range (5

Relatively high-substrate consistency hydrolysis of steam-pretreated sweet sorghum bagasse at relatively low cellulase loading.

Sweet sorghum bagasse (SSB) was steam pretreated in the conditions of 190 °C for 5 min to assess its amenability to the pretreatment and enzymatic hydrolysis. Results showed that pretreatment conditions were robust enough to pretreat SSB with maximum of 87% glucan and 72% xylan recovery. Subsequent enzymatic hydrolysis showed that the pretreated SSB at 2% substrate consistency resulted in maximum of 70% glucan-glucose conversion. Increasing substrate consistency from 2% to 16% led to a significant reduction in glucan conversion. However, the decrease ratio of glucan-glucose conversion was the

A resource-efficient agar segment method for qualitative cellulase screening: reducing processing time, waste generation, and environmental impact.

Sustainable and cost-effective screening methods are essential for identifying microorganisms with cellulolytic activity. Traditional plate-based assays require substantial volumes of dye solutions (typically 30-40 mL per plate for flooding protocols) and generate contaminated liquid waste, including residual Congo red and NaCl solutions, as well as spore-contaminated liquid waste when working with sporulating microorganisms. Here, we present an Agar Segment Method (ASM) for the qualitative detection of carboxymethylcellulase (CMCase) activity, involving excision of small agar segments from pr

Influence of Modification of Cysteine Residues in the Carbohydrate-Binding Domain on Cellulase Activity: Achieving Enhanced Performance and pH-Sensitive Recovery.

The conversion of cellulosic biomass into fermentable sugars represents a pivotal stage in the environmentally friendly and sustainable production of clean fuels and chemicals. In this paper, a new strategy to enhance enzyme performance was proposed. This strategy can achieve pH-sensitive recovery of cellulase while improving the catalytic activity and stability of the enzyme. This strategy was achieved by means of a chemical modification of the cysteine residue (-SH) of the carbohydrate-binding domain (CBD) of cellulase with a PEG-based modifier (Mal-PEG-FA). It was shown that chemical modifi

kdgREcc negatively regulates genes for pectinases, cellulase, protease, HarpinEcc, and a global RNA regulator in Erwinia carotovora subsp. carotovora.

Erwinia carotovora subsp. carotovora produces extracellular pectate lyase (Pel), polygalacturonase (Peh), cellulase (Cel), and protease (Prt). The concerted actions of these enzymes largely determine the virulence of this plant-pathogenic bacterium. E. carotovora subsp. carotovora also produces HarpinEcc, the elicitor of the hypersensitive reaction. We document here that KdgREcc (Kdg, 2-keto-3-deoxygluconate; KdgR, general repressor of genes involved in pectin and galacturonate catabolism), a homolog of the E. chrysanthemi repressor, KdgREch and the Escherichia coli repressor, KdgREco, negativ

Integrated cellulase continuous production and downstream processing using a packed-bed bioreactor for solid-state fermentation by thermophilic fungus.

In situ cellulase production is a key strategy to enhance efficiency, sustainability, and robustness of second-generation bioethanol processes. However, limitations related to heat transfer, bioreactor operation, and downstream processing still hinder large-scale implementation of solid-state fermentation (SSF) for enzymes production. This study systematically evaluated the integrated production, extraction, recovery and stabilization of endoglucanase from the cultivation of Myceliophthora thermophila I-1D3b in a multilayer packed-bed bioreactor operated under batch and continuous modes, using
