Studies on lophocercous cercariae from Bithynia siamensis goniomphalus (Prosobranchia: Bithyniidae).

Cercariae from Bithynia siamensis goniomphalus were studied in an area endemic for opisthorchiasis. Snails emitted different types of cercariae when shedding was induced by illumination. In addition to two lophocercous cercariae, a monostome cercaria (probably Notocotylus sp), two different furcocercariae and a xiphidiocercaria were found. Two similar types of lophocercous cercariae were distinguishable and one could be identified as Opisthorchis viverrini. To date the cercaria of O. viverrini has been regarded as the only lophocercous cercaria in the endemic area of opisthorchiasis. Therefore

Laboratory transmission of Schistosoma mansoni in brackish waters; survival and infectivity of cercariae.

Schistosoma mansoni cercariae were exposed to various dilutions of sea water, both in the absence and in the presence of mice. It was found that whereas 100% and 75% sea water were lethal to the parasites, the parasites survived in lower concentrations of sea water. The lower the concentration the longer was the survival time. Similarly, the rate of infection of mice decreased as the concentration of the sea water increased.

Murine immunization by cesium-137 irradiation attenuated Schistosoma mansoni cercariae.

Cesium-137, becoming a more readily available ionizing gamma radiation source for laboratory use, was shown to effectively attenuate Schistosoma mansoni cercariae for vaccine production. In parallel comparison studies with the murine model, cesium-137 attenuated cercariae consistently afforded better (P greater than 0.05) protection than did the cobalt-60 prepared vaccine. Dose-response data indicated that the optimal total irradiation with cesium-137 was between 45 and 50 Krad.

Immunization of mice with ultraviolet-irradiated Schistosoma mansoni cercariae: a re-evaluation.

Mice immunized by percutaneous exposure to ultraviolet-irradiated Schistosoma mansoni cercariae developed levels of resistance to subsequent S. mansoni infection comparable to those induced by gamma-irradiated cercariae (50-70% reduction in adult worm burden). Cercariae treated with ultraviolet doses ranging from one to three times the minimum dose required to prevent long-term survival induced the highest levels of resistance.

Dermatitis caused by cercariae of Orientobilharzia turkestanicum in the Caspian Sea area of Iran.

Cercarial dermatitis is very common among rice farmers in the Caspian Sea area of Iran. The characteristic eggs of Orientobilharzia turkestanicum were found in the feces of a plowing bull. Fifty-six percent of uninfected Lymnaea gedrosiana maintained at 24-25 degrees C produced cercariae, identical to those from natural infections, 21 days after exposure to the miracidia obtained from the feces of the bull. Cercariae of O. turkestanicum from naturally infected L. gedrosiana produced dermatitis on the arm of one of us.

Schistosoma mansoni Heterochromatin Protein 1 (HP1) nuclear interactome in cercariae.

Schistosoma mansoni causes schistosomiasis, which affects 240 million people, and 700 million people are living at risk of infection. Epigenetic mechanisms are important for transcriptional control and are well-known conserved transcriptional co-regulators in evolution, already described in mammal, yeast, protozoa and S. mansoni, responsible for heterochromatization and gene silence mechanisms through the formation of complexes of transcriptional repression in chromatin. Previous results from another group have shown that HP1 (SmCBX) proteins form chromatin complexes with SmMDB2/3 proteins and

Temperature and light effects on Trichobilharzia szidati cercariae with implications for a risk analysis.

BACKGROUND: Cercarial dermatitis (swimmer's itch) caused by bird schistosome cercariae, released from intermediate host snails, is a common disorder also at higher latitudes. Several cases were observed in the artificial Danish freshwater Ringen Lake frequently used by the public for recreational purposes. The lake may serve as a model system when establishing a risk analysis for this zoonotic disease. In order to explain high risk periods we determined infection levels of intermediate host snails from early spring to late summer (March, June and August) and elucidated the effect of temperatur

Molluscicidal and cercaricidal activities of curcumin on Biomphalaria glabrata and Schistosoma mansoni cercariae.

BACKGROUND: Schistosomiasis control in endemic areas depends on several factors, including chemotherapy, snail control and adequate sanitation. In this context, the employment of compounds isolated from plants is an important issue regarding infection and snail control. The aim of this study was therefore to evaluate the effects of curcumin (CUR), a compound isolated from Curcuma longa, against snails and embryos of Biomphalaria glabrata, which is the most important intermediate host of schistosomiasis in the Americas, as well as in cercariae, the infecting larval stage of Schistosoma mansoni.

Experimental infection of Opisthorchis viverrini cercariae to the cyprinid fish, Barbonymus gonionotus.

Opisthorchis viverrini is an important public health problem, a major cause of cholangiocarcinoma in the Greater Mekong subregion including Lao PDR, Cambodia, Vietnam and Thailand. Humans acquire the infection by consumption of raw, fermented or partially cooked freshwater cyprinid fish containing infective metacercariae. This study examined the effect of cercarial infection dosage (25-200 cercariae), age (1-60 day) and size (1-24mm) of Barbonymus gonionotus fish on infection success of O. viverrini cercariae. Additionally, the site of cyst formation of O. viverrini cercariae and subsequent de

Quantitative detection of Schistosoma japonicum cercariae in water by real-time PCR.

In China alone, an estimated 30 million people are at risk of schistosomiasis, caused by the Schistosoma japonicum parasite. Disease has re-emerged in several regions that had previously attained transmission control, reinforcing the need for active surveillance. The environmental stage of the parasite is known to exhibit high spatial and temporal variability, and current detection techniques rely on a sentinel mouse method which has serious limitations in obtaining data in both time and space. Here we describe a real-time PCR assay to quantitatively detect S. japonicum cercariae in laboratory

Effect of aestivation of Biomphalaria pfeifferi on the survival and infectivity of Schistosoma mansoni cercariae.

Schistosoma mansoni cercariae from post-aestivated Biomphalaria pfeifferi remain motile for 20 hours after release. Thereafter, their activity decreases with age. The difference in mortality rate of cercariae from aestivated and non-aestivated B. pfeifferi studied here proved to be statistically significant (P < 0.05) within the first 10 hours of the experimental period. Results of the percentage recovery of worms from different mouse organs infected with cercariae from aestivated and non-aestivated snails varied. The two main organs infected were the liver and intestine. In conclusion, the pe

The infection of human skin by schistosome cercariae: studies using Franz cells.

Franz cells (2-chambered, air/fluid phase static diffusion devices, previously used for the study of drugs across viable human skin) are utilized for the first time to investigate the process of infection of human skin by Schistosoma mansoni cercariae. Skin obtained from cosmetic surgery sources was used in the Franz cells to describe the temporal dynamics of the early interaction of cercariae with skin. At 38 degrees C, about 50% of cercariae applied in water to the epidermal surface of the skin were irreversibly attached within 1 min and after 5 min about 85%, were similarly irrecoverable. T

Field trial of 1% niclosamide as a topical antipenetrant to Schistosoma mansoni cercariae.

A randomized, double-blind, placebo-controlled field trial of a topical antipenetrant lotion, 1% niclosamide, applied daily to the upper and lower limbs of farmers occupationally exposed to Schistosoma mansoni cercarial-infested water, was conducted in the Nile Delta to assess its safety and efficacy in preventing reinfection. Farmers aged 18-40 years were treated to cure their S. mansoni infections three months prior to the onset of the trial. Subjects were randomly assigned to receive niclosamide or placebo lotion that was self-applied daily for five months. A total of 186 subjects met the i

Effect of ultraviolet radiation on survival, infectivity and maturation of Schistosoma mansoni cercariae.

S. mansoni cercariae exposed to ultraviolet radiation for 1, 3, 5, 10 and 20 s as well as non-irradiated cercariae remained actively motile 30 min post-irradiation. Thereafter the activity decreased with increasing dose level of radiation and age of cercariae. There was no significant difference between the rates of attachment of the batches of cercariae. The recovery rates (0-49% of cercariae to which mice were exposed) of adult worms were, however, significantly different from the number of cercariae calculated to have attached to the mice (93.5-100% of cercariae to which mice were exposed).

Intraspecific cross-protection in mice immunized with irradiated Schistosoma mansoni cercariae.

Several laboratory-maintained strains of Schistosoma mansoni were tested for their relative immunogenicity or susceptibility to anti-schistosome immunity in irradiated cercaria-immunized mice. A total of 11 strains and substrains were used; 7 were of Puerto Rican origin, 3 from Brazil, and 1 from Egypt. Mice were immunized by percutaneous exposure to 50-krad-irradiated cercariae. Immunity was assessed following challenge with cercariae of the homologous or a heterologous strain. The results showed that the choice of either the challenge or immunizing strains was not critical in the development

Schistosoma mansoni polypeptides immunogenic in mice vaccinated with radiation-attenuated cercariae.

We compared the humoral immune response of mice protected against Schistosoma mansoni by vaccination with radiation-attenuated cercariae to that of patently infected mice, and we identified antigens that elicit a greater, or unique, immune response in the vaccinated mice. These comparisons were based upon radioimmunoprecipitations and immunodepletion of [35S]methionine-labeled schistosomular and adult worm polypeptides, followed by one- and two-dimensional polyacrylamide gel analyses. The humoral responses of patently infected mice and of mice vaccinated once were remarkably similar and were d

Schistosoma mansoni: decay of resistance induced by gamma irradiated cercariae in the mouse.

The level of resistance induced by a single percutaneous infection of cercariae which had been previously exposed to 20 kr of gamma irradiation was compared in two strains of mice. Challenge cercariae were given at several time intervals after the preliminary immunization either to the same skin site as that used for immunization or to a previously unexposed skin site. No significant difference in resistance to infection was observed when a different skin site was used for the challenge. A higher degree of resistance to challenge was observed in CBA/Ca mice than in BALB/c mice. Immunity in bot

Secretory glands in cercaria of the neuropathogenic schistosome Trichobilharzia regenti - ultrastructural characterization, 3-D modelling, volume and pH estimations.

BACKGROUND: Cercariae of schistosomes employ bioactive molecules for penetration into their hosts. These are released from specialized unicellular glands upon stimuli from host skin. The glands were previously well-described in the human pathogen Schistosoma mansoni. As bird schistosomes can also penetrate human skin and cause cercarial dermatitis, our aim was to characterize the architecture and ultrastructure of glands in the neurotropic bird schistosome Trichobilharzia regenti and compare it with S. mansoni. In the context of different histolytic enzymes used by these two species, we focuse

From cercariae to chronic inflammation: understanding schistosome infection and host immune responses.

Schistosomiasis is a parasitic disease caused by trematodes of the genus Schistosoma, typically found in tropical and subtropical freshwater environments. Recognized by the World Health Organization as a major emerging disease, schistosomiasis is characterized by the parasite's ability to modulate and evade the host immune system, enabling long-term persistence within the human body. This immunomodulation not only supports chronic infection but also drives disease pathology, particularly through granulomatous inflammation surrounding parasite eggs trapped in host tissues. A deeper understandin
