Pyrenoids are algal CO2-fixing organelles that mediate approximately one-third of global carbon fixation. Most pyrenoids are traversed by membranes that are thought to supply them with concentrated CO2. Despite the critical nature of these membranes for pyrenoid function, they are poorly understood, with few protein components known in any species. Here, we identify protein components of the pyrenoid-traversing membranes from the leading model alga, Chlamydomonas reinhardtii, by affinity purification and mass spectrometry of membrane fragments. Our proteome includes previously known proteins a

Autophagy is a central degradative pathway highly conserved among eukaryotes, including microalgae, which remains unexplored in extremophilic organisms. In this study, we described and characterized autophagy in the newly identified extremophilic green microalga Chlamydomonas urium, which was isolated from an acidic environment. The nuclear genome of C. urium was sequenced, assembled and annotated in order to identify autophagy-related genes. Transmission electron microscopy, immunoblotting, metabolomic and photosynthetic analyses were performed to investigate autophagy in this extremophilic m

Traditional predictions of microalgal growth states rely on empirical or easily implementable kinetic models, leading to significant biases and elevated cost. This study proposes a kinetic-assisted machine learning method for predicting the growth curve of microalgal biomass under small sample conditions. Firstly, a microalgae growth kinetic model is constructed based on the logistic model. A two-stage kinetic fitting strategy is specified to account for the light-dark ratio. The Box-Behnken method is employed for experimental design. Then, using Two-stage TrAdaboost.R2 algorithm, the kinetic

As an essential component of protein cofactors, iron is important for all photosynthetic organisms. In Chlamydomonas reinhardtii, iron levels are strictly controlled by proteins such as iron-assimilating protein 1 (FEA1). This periplasmic protein is expressed under conditions of iron deficiency, but its mechanisms of function remain unknown. Because FEA1 has no amino-acid similarity to protein structures in the Protein Data Bank, its crystal structure cannot be solved by molecular replacement. Here, recombinant FEA1 protein lacking the N-terminal signal sequence was successfully purified and c

Autophagy constitutes an essential process triggered by oxidative stress that enables cells to recycle damaged biomolecules and organelles, which is eventually traced by immunodetection with anti-ATG8. In parallel with autophagy induction, carbon metabolism in Chlamydomonas reinhardtii under abiotic stress is diverged toward lipid biosynthesis and lipid droplet accumulation, which can be analyzed by a simple thin-layer chromatography and in vivo staining with the fluorescent probe BODIPY 493/503. We show the responses in Chlamydomonas cells exposed to mercury or cadmium (0-50 μM doses), as exa

Under high light, oxygenic photosynthetic organisms avoid photodamage by thermally dissipating absorbed energy, which is called nonphotochemical quenching. In green algae, a chlorophyll and carotenoid-binding protein, light-harvesting complex stress-related (LHCSR3), detects excess energy via a pH drop and serves as a quenching site. Using a combined in vivo and in vitro approach, we investigated quenching within LHCSR3 from Chlamydomonas reinhardtii. In vitro two distinct quenching processes, individually controlled by pH and zeaxanthin, were identified within LHCSR3. The pH-dependent quenchi

Magnesium (Mg)-deprived Chlamydomonas reinhardtii cells are capable to sustain hydrogen (H2 ) photoproduction at relatively high photosystem II (PSII) activity levels for an extended time period as compared with sulfur (S)-deprived cells. Herein, we present a comparative study of H2 photoproduction induced by Mg and S shortage to unravel the specific rearrangements of the photosynthetic machinery and cell metabolism occurring under the two deprivation protocols. The exhaustive analysis of photosynthetic activity and regulatory pathways, respiration and starch metabolism revealed the specific r

The present study explores the effect of light with different spectral composition on the stability of CuO-nanoparticle (CuO-NP) dispersions and their effects to green alga Chlamydomonas reinhardtii. The results showed that simulated natural light (SNL) and light with enhanced UVB radiation (UVR*) do not affect the dissolution of CuO-NPs as compared to light irradiation conditions typically used in laboratory incubator (INC). Comparable values of ζ-potential and hydrodynamic size during 24h were found under all studied conditions. Concentrations of CuO-NPs below 1mgL(-1) do not attenuate the l

Sulphur is an essential macroelement that plays important roles in living organisms. The thiol rich sulphur compounds, such as cysteine, γ-Glu-Cys, glutathione and phytochelatins participate in the tolerance mechanisms against cadmium toxicity. Plants, algae, yeasts and most prokaryotes cover their demand for reduced sulphur by reduction of inorganic sulphate. The aim of this study was to investigate, using a bifactorial experimental design, the effect of different sulphate concentrations in the nutrient solution on cadmium toxicity in the freshwater microalga Chlamydomonas moewusii. Cell grow

A genetic screen for Chlamydomonas reinhardtii mutants with copper-dependent growth or nonphotosynthetic phenotypes revealed three loci, COPPER RESPONSE REGULATOR 1 (CRR1), COPPER RESPONSE DEFECT 1 (CRD1), and COPPER RESPONSE DEFECT 2 (CRD2), distinguished as regulatory or target genes on the basis of phenotype. CRR1 was shown previously to be required for transcriptional activation of target genes like CYC6, CPX1, and CRD1, encoding, respectively, cytochrome c(6) (which is a heme-containing substitute for copper-containing plastocyanin), coproporphyrinogen III oxidase, and Mg-protoporphyrin I

The recent identification in Chlamydomonas of the intraflagellar transport machinery that assembles cilia and flagella has triggered a renaissance of interest in these organelles that transcends studies on their well-characterized ability to move. New studies on several fronts have revealed that the machinery for flagellar assembly/disassembly is regulated by homologs of mitotic proteins, that cilia play essential roles in sensory transduction, and that mutations in cilia/basal body proteins are responsible for cilia-related human disorders from polycystic kidney disease to a syndrome associat

UNLABELLED: Pyrenoids are CO2-fixing organelles responsible for approximately one-third of global CO2 fixation. The pyrenoids of many algae are surrounded by a starch sheath proposed to perform the critical function of slowing leakage of concentrated CO2 out of the pyrenoid. How the cell shapes starch granules into curved starch plates that encase the pyrenoid to enable efficient CO2 fixation is currently unknown. Here, we elucidate how starch transitions from granules into a fully formed pyrenoid starch sheath using confocal microscopy of the model green alga Chlamydomonas reinhardtii . We ob

Intraflagellar transport (IFT) carries proteins into flagella but how IFT trains interact with the large number of diverse proteins required to assemble flagella remains largely unknown. Here, we show that IFT of radial spokes in Chlamydomonas requires ARMC2/PF27, a conserved armadillo repeat protein associated with male infertility and reduced lung function. Chlamydomonas ARMC2 was highly enriched in growing flagella and tagged ARMC2 and the spoke protein RSP3 co-migrated on anterograde trains. In contrast, a cargo and an adapter of inner and outer dynein arms moved independently of ARMC2, in

Contamination by mercury (Hg) is a worldwide concern because of Hg toxicity and biomagnification in aquatic food webs. Nevertheless, bioavailability and cellular toxicity pathways of inorganic (IHg) and methyl-Hg (MeHg) remain poorly understood. We analyzed the uptake, transcriptomic, and physiological responses in the microalga Chlamydomonas reinhardtii exposed to IHg or MeHg. Bioavailability of MeHg was up to 27× higher than for IHg. Genes involved in cell processes, energy metabolism and transport were dysregulated by both Hg species. Physiological analysis revealed an impact on photosynthe

Algae and vascular plants are cysteine (Cys) prototrophs. They are able to import, reduce, and assimilate sulfate into Cys, methionine, and other organic sulfur-containing compounds. Characterization of genes encoding the enzymes required for Cys biosynthesis from the unicellular green alga Chlamydomonas reinhardtii reveals that transcriptional and posttranscriptional mechanisms regulate the pathway. The derived amino acid sequences of the C. reinhardtii genes encoding 5'-adenylylsulfate (APS) reductase and serine (Ser) acetyltransferase are orthologous to sequences from vascular plants. The C

Gametic mating by Chlamydomonas reinhardi is inhibited in a dose-dependent and reversible manner by the calmodulin antagonists trifluoperazine (TFP) and W-7, but not by W-5, an analog of W-7 having lower affinity for calmodulin. Quantitation of the sequential steps of mating showed that TFP and W-7 both allow normal levels of flagellar agglutination but prevent all subsequent steps. Gametes agglutinate aberrantly and do not form mating pairs. Further, both of these drugs prevent the translocation of latex beads along the flagellar surface. Our observations suggest that calmodulin may play an i

BACKGROUND: The cilium is a microtubule-based organelle essential for cellular signaling, whose assembly depends on intraflagellar transport (IFT). IFT20, a unique IFT-B component, localizes to both the Golgi apparatus and cilium/flagellum in mammalian systems and plays a role in ciliary membrane protein trafficking. Here, we analyzed IFT20-mediated ciliary trafficking of channelrhodopsin-1 (ChR1) in Chlamydomonas reinhardtii.
METHODS AND RESULTS: Co-immunocytochemistry showed that IFT20 and ChR1 co-localized throughout the flagella of wild-type cells. Using fla8 (kinesin-2) and dhc1b-3 (dyne

The model green alga Chlamydomonas reinhardtii has a pyrenoid within chloroplast for photosynthetic CO2 fixation under limiting CO2 conditions and its chloroplast is an ideal chassis for engineering photosynthetic modules. In contrast, carboxysomes are bacterial microcompartments that encapsulate Rubisco for CO2 fixation. The aim of the present study was to determine if a bacterial carboxysome or its components can replace pyrenoid and function to allow Chlamydomonas growth powered by photosynthesis. We replaced the Chlamydomonas endogenous RbcL gene with cbbL and cbbS from the chemoautotrophi

Microalgae are considered as attractive expression systems for the production of biologics. As photosynthetic unicellular organisms, they do not require costly and complex media for growing and are able to secrete proteins and perform protein glycosylation. Some biologics have been successfully produced in the green microalgae Chlamydomonas reinhardtii. However, post-translational modifications like glycosylation of these Chlamydomonas-made biologics have poorly been investigated so far. Therefore, in this study, we report on the first structural investigation of glycans linked to human erythr

C-reactive protein (CRP) participates in human inflammatory responses and is an important indicator in clinical diagnoses. At present, the use of monoclonal antibodies to detect CRP in the human body is high, but they are unstable and expensive. Understanding the CRP expression pathway is of great significance for developing CRP tests and reagents. Chlamydomonas reinhardtii is a model organism that has great potential as a foreign protein expression system. This study is the first attempt to express human CRP in C. reinhardtii. We selected the endogenous constitutive Rbcs2 promoter and termina
