A chloramine-based antiseptic polymer improves Candida auris skin decolonization compared to chlorhexidine in a murine model.

Candida auris frequently demonstrates resistance to multiple antifungal classes and has been classified as a critical priority pathogen by the World Health Organization and the first fungal pathogen designated an urgent threat by the U.S. Centers for Disease Control and Prevention. Patients with skin colonization face a 7%-20% risk of invasive infection, with 30-day mortality of 30%-72%. While chlorhexidine gluconate (CHG) is widely used for general skin decolonization, it fails to reliably clear C. auris from patient skin, and no proven decolonization protocol exists. We evaluated a novel chl

Reverse Osmosis Shifts Chloramine Speciation Causing Re-Formation of NDMA during Potable Reuse of Wastewater.

UV-based advanced oxidation processes (AOPs) effectively degrade N-nitrosodimethylamine (NDMA) passing through reverse osmosis (RO) units within advanced treatment trains for the potable reuse of municipal wastewater. However, certain utilities have observed the re-formation of NDMA after the AOP from reactions between residual chloramines and NDMA precursors in the AOP product water. Using kinetic modeling and bench-scale RO experiments, we demonstrate that the low pH in the RO permeate (∼5.5) coupled with the effective rejection of NH4+ promotes conversion of the residual monochloramine (NH2

[Biochemical activity of opportunistic antibiotic and chloramine-sensitive and resistant microorganisms isolated from healthy and sick people].

Activity of oxidation-reduction enzymes such as succinate dehydrogenase, peroxidase and catalase was studied in staphylococci isolated from healthy persons and patients as well as from the air and implements of medical institutions. The isolates were resistant either to antibiotics or to chloramine B or to the both. The results showed that development of resistance to antibiotics and chloramine B in the staphylococci was accompanied by a decrease in the activity of succinate dehydrogenase, peroxidase and catalase. In the strains resistant only to chloramine B the activity of the enzymes was pr

Control of aromatic halogenated disinfection byproducts in chlorinated and chloraminated drinking water through pre-oxidation: Effects and mechanisms.

Aromatic halogenated disinfection byproducts (DBPs) are of great concern in disinfected drinking water due to their high toxicity. Pre-oxidation is commonly employed in drinking water treatment to remove emerging contaminants and also to control DBPs formation. However, the control of aromatic halogenated DBPs in drinking water through pre-oxidation has not been clarified. In this study, the effects of four common pre-oxidants, including ozone (O₃), chlorine dioxide (ClO₂), permanganate (Mn(VII)), and ferrate (Fe(VI)), on the formation of aromatic halogenated DBPs in chlorinated and chloramina

Sustainable chloramine-functionalized iron hydroxide nanofiber membrane for arsenic(Ⅲ) removal via oxidation-adsorption mechanism.

Arsenic-contaminated groundwater is an intractable environmental problem worldwide, particularly As(III), which is not only highly toxic but also resistant to removal. In this study, sustainable chloramine-functionalized iron hydroxide cellulose nanofibrous membrane (Fe-CNFM-Cl) was prepared by electrostatic spinning followed by chemical grafting for As(III) decontamination. In situ engineered iron hydroxides were uniformly dispersed in cellulose nanofibers for As adsorption. The oxidative chlorine (+1) in the grafted chloramine could oxidize As(III) to readily removable As(V). Benefiting from

Chloramine-T application for Trichodina sp. in Arapaima gigas juveniles: Acute toxicity, histopathology, efficacy, and physiological effects.

Trichodinids are problematic ectoparasites for intensive arapaima fish farming, which can make their breeding costly and unprofitable. The search for effective and safe therapeutic molecules is constant, and chloramine-T has stood out as an effective biocide to treat various pathogens in aquaculture. Here we investigated the acute toxicity (LC50-4h) of chloramine-T and its behavioural, morphological, histological, physiological, and antiparasitic effects on the control of trichodinids in arapaima juveniles. Initially arapaima were exposed to chloramine-T (0, 10, 20, 40, 50, 60, 80, 100 mg L-1)

Mechanistic Study on the Formation of Cl-/Br-/I-Trihalomethanes during Chlorination/Chloramination Combined with a Theoretical Cytotoxicity Evaluation.

Chlorination followed by chloramination can be used to mitigate the formation of potentially toxic iodinated disinfection byproducts (I-DBPs) while controlling the formation of regulated chloro-bromo-DBPs (Cl-/Br-DBPs). Water samples containing dissolved organic matter (DOM) isolates were subjected to 3 disinfection scenarios: NH2Cl, prechlorination followed by ammonia addition, and HOCl alone. A theoretical cytotoxicity evaluation was carried out based on the trihalomethanes (THMs) formed. This study demonstrates that the presence of bromide not only enhances the yield and rate of iodate form

Some properties of sodium channels in neuroblastoma cells modified with scorpion toxin and chloramine-T. Single channel measurements.

Currents through sodium channels of neuroblastoma cells were measured using patch technique in outside-out configuration. Scorpion toxin (ScTX) produced 3 to 4 fold prolongation of mean open time and increased number of reopenings. The mean open times showed slow fluctuations around some average values. The distribution of channel open times for ScTX-modified channels required more than one exponential to be fitted. Chloramine-T (ChT) produced qualitatively similar, though weaker, prolongation of open times.

Use-dependent blockade of sodium channels by local anaesthetics and antiarrhythmic drugs. Effects of chloramine-T and calcium ions.

Voltage clamp studies were carried out of the effects of chloramine-T(CT) and external Ca++ on the blocking interactions of local anaesthetics (LAs) and antiarrhythmic drugs (lidocaine, tetracaine, N-propyl ajmaline, compound KC 3791) with Na+ channels in frog Ranvier nodes. The results obtained provided direct evidence for the notion that: LAs interact preferentially with inactivated Na+ channels and stabilize their inactivated conformation ("drug-induced slow inactivation": SI); and SI underlies the cumulative inhibition of INa during repetitive membrane stimulation. Normal inactivation is n

Impact of chlorine exposure time on disinfection byproduct formation in the presence of iopamidol and natural organic matter during chloramination.

Chloramines, in practice, are formed onsite by adding ammonia to chlorinated drinking water to achieve the required disinfection. While regulated disinfection byproducts (DBPs) are reduced during chloramine disinfection, other DBPs such as iodinated (iodo-) DBPs, that elicit greater toxicity are formed. The objective of this study was to investigate the impact of prechlorination time on the formation of both halogen-specific total organic halogen (TOX) and iodo/chlorinated (chloro-) DBPs during prechlorination/chloramination in source waters (SWs) containing iopamidol, an X-ray contrast medium

Mechanism of inactivation of single sodium channels after modification by chloramine-T, sea anemone toxin and scorpion toxin.

Single sodium-channel currents were measured in neuroblastoma cells after inhibition of inactivation by chloramine-T (CHL-T), sea anemone toxin II (ATX-II) and scorpion toxin (SCT). The decaying phase of the averaged single-channel currents recorded with 90-msec pulses in cell-attached patches was clearly slower than that of the ummodified channels, suggesting inhibition of macroscopic inactivation. Each substance caused repetitive openings and a moderate increase in the channel open time. At Vm = RP + 20 mV and T = 12 degrees C, the mean channel open times were 1.4, 1.6 and 1.8 msec for CHL-T

Chloramine-T-Enabled Mass Spectrometric Analysis of C═C Isomers of Unsaturated Fatty Acids and Phosphatidylcholines in Human Thyroids.

Specific locations of carbon-carbon double bonds (C═C) in lipids often play an essential role in biological processes, and there has been a booming development in C═C composition analysis by mass spectrometry. However, a universal derivatization and fragmentation pattern for the annotation of C═C positions in lipids is still challenging and attractive. To expand this field in lipidomics, a flexible and convenient N-tosylaziridination method was developed, with high derivatization efficiency, sensitivity, and specificity. The derivatization was very fast (15 s), and C═C numbers as well as locat

Enhanced formation of dichloroacetamide and dichloroacetonitrile during chloramination of drinking water and model organic matters in the presence of copper corrosion products.

The formation of nitrogenous disinfection byproducts (N-DBPs) occurs in chloraminated water in drinking water distribution systems and may be affected by metal pipe materials and their corrosion products. The effect of copper corrosion products, including Cu2+, CuO, and Cu2O, on the formation of dichloroacetonitrile (DCAN) and dichloroacetamide (DCAcAm) was investigated during chloramination of natural organic matter (NOM), model precursors (carboxylic acids and amino acids), and real water samples. Copper corrosion products enhanced DCAN and DCAcAm formation during chloramination of NOM by 33

THMs, HAAs and NAs production from culturable microorganisms in pipeline network by ozonation, chlorination, chloramination and joint disinfection strategies.

Disinfection is an indispensable process to inactivate pathogens, while unexpected disinfection by-products (DBPs) would also be formed between the reaction of residual disinfectants and microorganisms in the water distribution system (WDS). However, there are few studies referring to the formation of DBPs and DBPs-associated toxicity under various disinfection methods based on microorganisms in the real WDS. In addition, the main contributors of bacterial communities or components that generate DBPs are unclear. In this study, the formation of trihalomethanes (THMs), halo-acetic acids (HAAs),

Stable Isotopic Labeling and Automated Reactivity-Directed Approach Accelerating Discovery of Toxic Disinfection Byproducts in Chlorinated and Chloraminated Waters.

Reactivity-directed analysis facilitates prioritization of toxic disinfection byproducts (DBPs), yet its data processing remains laborious and inefficient. Herein, we developed an automated approach integrating stable isotopic labeling with reactivity-directed principles. Normal and deuterated glutathione (GSH) probes were used to generate paired isotopic signatures enabling precise localization of reactive DBPs. Customized bioinformatics tools were employed to efficiently extract paired GSH-DBP adducts. The workflow allowed rapid automated processing of large data sets within minutes while ma

Factors affecting the formation of disinfection by-products during chlorination and chloramination of secondary effluent for the production of high quality recycled water.

During the production of high quality recycled water by reverse osmosis membrane filtration secondary effluent must be disinfected to limit biofouling on the membrane surface. Advanced Water Treatment Plants in South East Queensland, Australia use disinfectant contact times ranging from 30 min up to 24 h. Disinfectants such as chlorine and chloramines react with effluent organic matter to generate disinfection by-products (DBPs) which could be potentially hazardous to human health if the water is destined for supplementing public water supplies. In this context, secondary effluents are of conc

Toward Robust ARB Inactivation in Wastewater: Comparing Performic Acid, Peracetic Acid, and Chloramination Using ICT, Resistance, and Reactivation Metrics.

Antimicrobial resistance (AMR), particularly vancomycin-resistant enterococci (VRE), poses a critical global health threat. Although wastewater disinfection limits pathogen dissemination, conventional chlorine-based disinfection can be ineffective against antibiotic-resistant bacteria (ARB) and may induce viable but nonculturable (VBNC) states. This study compared performic acid (PFA), peracetic acid (PAA), and sodium hypochlorite (NaOCl) for inactivation of VRE and total enterococci (TE) in high-ammonia secondary effluent. Batch disinfection experiments were conducted using an integrated conc

Flushing Temporarily Improves Microbiological Water Quality for Buildings Supplied with Chloraminated Surface Water but Has Little Effect for Groundwater Supplies.

Microbial communities in premise plumbing systems were investigated after more than 2 months of long-term stagnation, during a subsequent flushing event, and during post-flush stagnation. Water samples were collected from showers in buildings supplied with chlorinated groundwater, untreated groundwater, and chloraminated surface water. The building supplied with chlorinated groundwater generally had the lowest bacterial concentrations across all sites (ranging from below quantification limit to 5.2 log copies/L). For buildings supplied with untreated groundwater, bacterial concentrations (5.0
