Imipramine and chlordiazepoxide in depressive and anxiety disorders. II. Efficacy in anxious outpatients.

A double-blind, placebo-controlled comparison at three collaborating university sites included 242 patients diagnosed as having anxiety disorders. A two-week placebo washout period preceded random assignment to eight weeks of imipramine hydrochloride, chlordiazepoxide hydrochloride, or placebo treatment. Antianxiety effects of imipramine were superior to those of the other two agents by the second treatment week; these effects became more clearly significant thereafter and were independent of degree of both baseline depression and anxiety. Excluding patients with possible panic-phobic syndrome

Modulation of the behavioral effects of chlordiazepoxide by methylxanthines and analogs of adenosine in squirrel monkeys.

The effects of chlordiazepoxide (CDAP) on suppressed behavior of squirrel monkeys were determined alone and in combination with selected methylxanthines (caffeine, theophylline, 8-phenyltheophylline, theobromine, 3-isobutyl-1-methylxanthine) and analogs of adenosine (L-PIA, N6-cyclohexyladenosine, NECA). Monkeys responded under a fixed-interval schedule of food presentation in which responding was suppressed by a concurrent fixed-ratio schedule of response-produced electric shock. Dose-effect curves were determined by administering cumulative doses i.v. during timeout periods that preceded seq

The absorption of intramuscular chlordiazepoxide (Librium) in patients with severe alcoholic liver disease.

Intramuscular chlordiazepoxide (CDX) is commonly administered to alcoholic liver disease (ALD) patients requiring prompt management of alcohol agitation, anxiety, and delirium tremors. Sedative action is associated with plasma levels of approximately 2.0 mcg/ml, however, intramuscular CDX has been shown to produce peak concentrations consistently below 2.0 mcg/ml in normal subjects. The present study was designed to define the absorption characteristics of intramuscular 25 mg CDX in males with ALD. Five normal males had mean CDX absorption half-lives of 3.0 h and peaked at 0.8 mcg/ml in 7.2 h,

Sedative effects of PK 9084 and PK 8165, alone and in combination with chlordiazepoxide.

1 The sedative effects in rats of two phenylquinolines, PK 9084 and PK 8165 (5-50 mg/kg), were examined in a holeboard: both when given alone and when given in conjunction with chlordiazepoxide (5 mg/kg). 2 Both phenylquinolines produced significant dose-related decreases in locomotor activity and rearing, with an ED50 about twice that for chlordiazepoxide. 3 When the phenylquinolines were combined with chlordiazepoxide the degree of sedation was equal to that seen with either drug given alone, whichever produced the greater sedation; the sedative effects of the two drugs were never additive.

The effects of chlordiazepoxide HCl administration upon punishment and conditioned suppression in the rat.

Rats were trained to lever press for sucrose on a random interval (RI) 64-s schedule. During a 55 min session there were four 3 min "intrusion periods" signalled by a flashing house-light. In experiment 1 there were two groups matched for baseline response rate. During the intrusion periods one group received response-independent footshock on an independent RI 64 schedule; the other group received response-contingent shock on this schedule. Shock intensity was varied for each rat to match degree of response suppression between the two groups. Chlordiazepoxide HCl (CDP) in doses 0.5-5 mg/kg all

Effects of chlordiazepoxide on maze performance of rats subjected to undernutrition in early life.

The purpose of this research was to study the isolated effects of and interactions between undernutrition and emotional reactivity upon maze performance in rats. This work compares the effects of chlordiazepoxide on learning performance in two groups of adult rats: one undernourished early in life and then rehabilitated, and the other normally nourished. Chlordiazepoxide was used at 5 mg/kg body weight administered i.p. 30 min before each test. The response was evaluated in a Hebb-Williams maze. Water was used as reinforcement. Chlordiazepoxide improved performance in the early undernourished

The inability of CCK to block (or CCK antagonists to substitute for) the stimulus effects of chlordiazepoxide.

To further examine the relationship between cholecystokinin (CCK) and GABA, the present study assessed the ability of the CCK-A antagonist devazepide and the CCK-B antagonist L-365,260 to substitute for the stimulus effects of chlordiazepoxide (CDP), as well as the ability of CCK-8s to block these effects, in female Long-Evans rats within the conditioned taste aversion baseline of drug discrimination learning. Both devazepide and L-365,260 failed to substitute for the discriminative stimulus properties of CDP, and CCK-8s failed to block its stimulus effects. The benzodiazepine diazepam did sub

Isobolographic analysis of chlordiazepoxide and triazolam combinations in squirrel monkeys discriminating triazolam.

RATIONALE: The discriminative stimulus (DS) effects of chlordiazepoxide (CDP) differ from those of other typical benzodiazepine (BZ) agonists in that CDP does not always occasion full substitution for a BZ agonist DS. OBJECTIVES: The present study tested the hypothesis that the unusual DS effects of CDP may result from its relatively low intrinsic efficacy by examining the combinations of CDP and triazolam using isobolographic analysis in squirrel monkeys discriminating triazolam. METHODS AND RESULTS: Squirrel monkeys were previously trained to discriminate triazolam (0.03 mg/kg, i.v.) from

Effects of propranolol, buspirone, pCPA, reserpine, and chlordiazepoxide on open-field behavior.

The study examined the possibility that propranolol, buspirone, pCPA, and reserpine have antianxiety effects by comparing their effects with those of chlordiazepoxide on an open-field test of emotionality in rats. The effects of intraperitoneal injections of d,l, propranolol (5, 10, 20 mg/kg), buspirone (1.25, 2.5, 5 mg/kg), reserpine (0.5 mg/kg), parachlorophenylalanine (pCPA) (100 mg/kg), and chlordiazepoxide (CDP) (2.5, 5, 10 mg/kg) were compared with performance of rats under saline or water using an open-field test on 5 successive days. Significant effects were found on peripheral movemen

Effects of chlordiazepoxide on opioid-induced antinociception and respiratory depression in restrained rats.

This study investigates the influence of possible stress due to housing in Bolman cages on antinociception and on respiratory depression following opioid administration. To evaluate the functional role of this stressor and to modulate it, rats were subcutaneously pretreated with the anxiolytic chlordiazepoxide (CDP; 10 mg/kg) or saline (SAL) before the immobilization in the Bolman cages and before the intravenous administration of small doses of morphine (MOR), sufentanil (SUF), or vehicle (VEH). Antinociception, respiratory impairment and stress were evaluated by means of the tail-flick laten

Inhibition by antimanic drugs of hyperactivity induced by methamphetamine-chlordiazepoxide mixture in mice.

The effects of lithium chloride and other antimanic drugs on locomotor hyperactivity induced by a mixture of methamphetamine (MAMP) and chlordiazepoxide (CDZP) were examined in mice, using an Animex activity meter. CDZP (12.5 mg/kg) given SC in combination with MAMP (1 mg/kg) caused a marked increase in locomotor activity, as compared with that in mice treated with MAMP alone. However, when CDZP (12.5 mg/kg) was administered together with 0.5 or 2.0 mg/kg of MAMP, no significant enhancement was observed. Lithium (2 and 3 mEq/kg, IP) and carbamazepine (4 and 8 mg/kg, IP) inhibited the hyperacti

Imipramine and chlordiazepoxide in depressive and anxiety disorders. I. Efficacy in depressed outpatients.

We randomly assigned 425 outpatients, independently classified as primarily depressed by two trained psychiatrists, to double-blind treatment with Imipramine hydrochloride, chlordiazepoxide hydrochloride, or placebo. Those patients who remained at least moderately depressed (following a two-week placebo washout period) were treated for an additional eight weeks. An endpoint analysis of 387 patients who completed two or more weeks of medication disclosed early therapeutic advantages of chlordiazepoxide. By week 4 of treatment, however, imipramine produced more improvement than did placebo and c

Effects of tifluadom on food consumption compared with chlordiazepoxide and kappa agonists in the rat.

Tifluadom (0.625-10.0 mg kg-1) was administered to non-deprived male rats which had been accustomed to eating a highly palatable diet in a 30 min test period. This compound, an opioid benzodiazepine, produced a significant increase in consumption of food when administered by the subcutaneous route, but not after intraperitoneal injection. Both chlordiazepoxide (1.25-20.0 mg kg-1) and the selective kappa opiate receptor agonist U-50,488 (0.3125-2.5 mg kg-1) also produced significant hyperphagic effects in the same feeding situation. In contrast, the two kappa opiate receptor agonists, ethylketo

[Discrimination reversal learning in rats under the treatment of chlordiazepoxide: effect of overtraining].

A total of 48 rats were run in a black-white discrimination (original) learning task to the learning criterion of 18/20 correct responses. Correct responses were rewarded by food and a non-correction method was used. One group was intraperitoneally injected with 15 mg/kg of chlordiazepoxide (CDP) and the other with physiological saline (SAL), 30 min prior to the beginning of the daily trials. For half of the rats (NOT) of each group, reversal learning began on the next day after reaching the criterion, and for the other half (OT), 100 additional trials(overtraining) were given before the rever

Chlordiazepoxide increases the force of two topographically distinct operant responses in rats.

By using operant conditioning techniques one group of 8 rats was trained to reach through a hole in the wall of an operant chamber, and to exert downward responses on a force-sensing circular disk. Eight other rats learned to reach through the hole and grasp and pull toward them a wire bail attached to a force transducer. Both behaviors were maintained on a fixed ratio 20 schedule of water reinforcement. The effects of chlordiazepoxide (CDP, 2.5, 5.0, 10.0 mg/kg) on response force and rate were assessed for both groups. CDP significantly increased response force in a dose-related manner in bot

Interactions of chlordiazepoxide and anorectic agents on rate and duration parameters of feeding in the rat.

d-Amphetamine (0.5 and 1.0 mg/kg) and dl-fenfluramine (2.0 and 4.0 mg/kg) reduced food intake in a short exposure feeding test, and their effects were counteraated by chloriazepoxide, particularly at 50 mg/kg. Chloriazepoxide reduced latency to eat, extended the durationof feeding and depressed the rate of feeding. Antagonism occurred in combination with d-amphetamine in relation to latency and duration, but in this experiment d-amphetamine did not affect feeding rate. Antagonism also occurred in combination with fenfluramine, with latency and duration measures, but only at the lower chlordiaz

Stimulatory effects of chlordiazepoxide, diazepam and oxazepam on the drug-metabolizing enzymes in microsomes.

5 days' exposure of rats to daily doses of 400 mg/kg body wt. of chlordiazepoxide, diazepam and oxazepam stimulated the microsomal metabolism in the liver, as evidenced by acceleration of both p-hydroxylation of aniline and hydroxylation of benzene. The effect was accompanied by an increased concentration of liver microsomal protein and by the development of tolerance to the drugs. Similar effects were found after exposure of rats to lower doses of the drugs. The metabolism of aniline in vivo in rats treated with chlordiazepoxide was accelerated; this was correlated with development of toleran
