The complete chloroplast genome of Rosa willmottiae Hemsl. (1907).

Rosa willmottiae Hemsl. (1907) is an ornamental plant with certain medicinal value. Its complete chloroplast genome was assembled via high-throughput sequencing. The 157,277 bp circular genome comprises one large single-copy region (LSC; 86,207 bp), one small single-copy region (SSC; 19,264 bp) and two inverted repeat regions (IRs; 25,903 bp each), encoding 130 genes: 86 protein-coding genes, 36 tRNAs and 8 rRNAs. Phylogenetic analyses indicate Rosa willmottiae's closest affinity to Kushui Rose (R. sertata × R. rugosa), and strongly support Rosa willmottiae's role as the putative maternal prog

Evolutionary dynamics of the chloroplast genome in Abutilon (Malvoideae, Malvaceae).

The genus Abutilon Mill. (Malvaceae) comprises approximately 178 species distributed across tropical and subtropical regions, many of which hold significant ornamental, economic, and medicinal value; yet its taxonomic classification remains challenging. In this study, six species were sequenced from herbarium specimens, and the chloroplast (cp.) genomes of ten additional species were assembled de novo from publicly available raw data. Three previously reported cp. genomes were also incorporated to characterise cp. genome structure, identify polymorphic loci, and perform phylogenetic analyses.

Cytokinin availability and chloroplast functional lifetime in senescing leaves.

Leaf senescence is often characterized by visible yellowing, reflecting the degradation of photosynthetic pigments and the functional deterioration of photosynthetic complexes, marking a shift from photosynthetic maintenance to nutrient remobilization. Cytokinins are key modulators in this process. This review examines cytokinin action in mature Arabidopsis leaves from the perspective of chloroplast functional lifetime, focusing on how long-distance supply, local metabolism and candidate transport routes may shape cytokinin availability and action during leaf senescence. Cytokinin signaling is

Why is the chloroplast redox regulatory machinery so complex?

Dithiol-disulfide exchange of cysteine residues has deep effects on protein conformation, hence on enzyme activity, which is the basis of redox regulation. The redox state of cells, including cellular compartments, is dynamically adjusted by reducing, i.e. NAD(P)H, and oxidizing signals, such as reactive oxygen species (ROS), generated during metabolic activity. Among ROS, H2O2 triggers the oxidation of cysteine thiols and the formation of disulfide bridges, thus having a relevant contribution to redox regulation. Disulfide reduction is catalyzed by thioredoxins (Trxs), small polypeptides with

The roles of chloroplast membrane lipids in abiotic stress responses.

Plant chloroplasts have complex membrane systems. Among these, thylakoids serve as the sites for photosynthesis and photosynthesis-related adaptation. In addition to the photosynthetic membrane complexes and associated molecules, lipids in the thylakoid membranes, are predominantly composed of MGDG (monogalactosyldiacylglycerol), DGDG (digalactosyldiacylglycerol), SQDG (sulfoquinovosyldiacylglycerol) and PG (phosphatidylglycerol), play essential roles in shaping the thylakoid architecture, electron transfer, and photoregulation. In this review, we discuss the effect of abiotic stress on chloro

An essential pentatricopeptide repeat protein in the apicomplexan remnant chloroplast.

The malaria parasite Plasmodium and other apicomplexans such as Toxoplasma evolved from photosynthetic organisms and contain an essential, remnant plastid termed the apicoplast. Transcription of the apicoplast genome is polycistronic with extensive RNA processing. Yet little is known about the mechanism of apicoplast RNA processing. In plants, chloroplast RNA processing is controlled by multiple pentatricopeptide repeat (PPR) proteins. Here, we identify the single apicoplast PPR protein, PPR1. We show that the protein is essential and that it binds to RNA motifs corresponding with previously c

Production of biopharmaceuticals and vaccines in plants via the chloroplast genome.

Transgenic plants offer many advantages, including low cost of production (by elimination of fermenters), storage and transportation; heat stability; and absence of human pathogens. When therapeutic proteins are orally delivered, plant cells protect antigens in the stomach through bioencapsulation and eliminate the need for expensive purification and sterile injections, in addition to development of both systemic and mucosal immunity. Chloroplast genetic engineering offers several advantages, including high levels of transgene expression, transgene containment via maternal inheritance and mult

Interactions of lipoidal materials and a pyridazinone inhibitor of chloroplast development.

Formation of chloroplast pigments was inhibited, and free fatty acids accumulated in mustard (Brassica juncea [L.] Coss.) cotyledons and in barley (Hordeum vulgare L.) first leaves developed after treatment with 4-chloro-5- (dimethylamino)-2- (alpha, alpha, alpha-trifluoro-m-tolyl) -3 (2H) -pyridazinone. The inhibitor reduced the amount of fatty acids found in polar lipids (galactolipids) of barley chloroplasts and increased the amount in nonpolar lipids while having little effect on total content of bound fatty acids. The inhibition of chlorophyll formation was circumvented by D-alpha-tocophe

Chloroplast small heat shock protein HSP21 interacts with plastid nucleoid protein pTAC5 and is essential for chloroplast development in Arabidopsis under heat stress.

Compared with small heat shock proteins (sHSPs) in other organisms, those in plants are the most abundant and diverse. However, the molecular mechanisms by which sHSPs are involved in cell protection remain unknown. Here, we characterized the role of HSP21, a plastid nucleoid-localized sHSP, in chloroplast development under heat stress. We show that an Arabidopsis thaliana knockout mutant of HSP21 had an ivory phenotype under heat stress. Quantitative real-time RT-PCR, run-on transcription, RNA gel blot, and polysome association analyses demonstrated that HSP21 is involved in plastid-encoded R

Photosynthetic Electron and Proton Transport in Chloroplasts: EPR Study of ΔpH Generation, an Overview.

This is a brief overview focused on the electron paramagnetic resonance applications to the study of the proton transport processes in chloroplasts. After brief description of structural and functional organization of the chloroplast electron transport chain, our attention is focused on the measurements of trans-thylakoid pH difference (ΔpH) with pH-sensitive spin-probes. The use of spin-probes is based either (i) on measuring the ΔpH-partitioning of spin-probes between the thylakoid lumen and external volume, or (ii) on monitoring changes in the electron paramagnetic resonance spectra of pH-s

Red light promotes sea slug growth while blue light increases longevity of stolen chloroplasts.

In Metazoa, long-term sequestration of photosynthetically active algal chloroplasts - kleptoplasts - is a distinctive physiological trait of sacoglossan sea slugs. In this study, we examined the effects of the light spectrum on the body weight and photosynthetic activity of the sea slug Elysia crispata. Laboratory-reared specimens were exposed to equivalent irradiances of white, red, or blue light during feeding and starvation experiments. Sea slugs fed under red light increased their weight at a faster pace than specimens fed under white or blue light. At the end of the feeding trial, chlorop

Transfer and Expression of Native Human Insulin-Like Growth Factor-1 in Tobacco Chloroplasts.

BACKGROUND: Insulin-like growth factor-1 (IGF-1), in addition to having insulin-like effects, has boosting effects on all cells in human body. Most of the recombinant IGF-1 required for patients suffering from its deficiency is currently produced by bacterial and yeast systems. Plant systems, especially chloroplasts, have many benefits for producing human blood proteins. Production costs are low in these systems, and their side effects are less than other systems. OBJECTIVES: In this study, the transfer and expression of mature IGF-1 protein cDNA in tobacco chloroplasts under the control of s

Subunit movements in single membrane-bound H+-ATP synthases from chloroplasts during ATP synthesis.

Subunit movements within the H(+)-ATP synthase from chloroplasts (CF(0)F(1)) are investigated during ATP synthesis. The gamma-subunit (gammaCys-322) is covalently labeled with a fluorescence donor (ATTO532). A fluorescence acceptor (adenosine 5'-(beta,gamma-imino)triphosphate (AMPPNP)-ATTO665) is noncovalently bound to a noncatalytic site at one alpha-subunit. The labeled CF(0)F(1) is integrated into liposomes, and a transmembrane pH difference is generated by an acid base transition. Single-pair fluorescence resonance energy transfer is measured in freely diffusing proteoliposomes with a conf

Inhibition and uncoupling of photophosphorylation in isolated chloroplasts by organotin, organomercury and diphenyleneiodonium compounds.

1. Trialkyltin, triphenyltin and diphenyleneiodonium compounds inhibited ADP-stimulated O(2) evolution by isolated pea chloroplasts in the presence of phosphate or arsenate. Tributyltin and triphenyltin were the most effective inhibitors, which suggests a highly hydrophobic site of action. Phenylmercuric acetate was a poor inhibitor of photophosphorylation, which suggests that thiol groups are not involved. 2. Triethyltin was a potent uncoupler of photophosphorylation by isolated chloroplasts in media containing Cl(-), but had little uncoupling activity when Cl(-) was replaced by NO(3) (-) or
