The chick embryo chorioallantoic membrane as an in vivo assay to study the skin in melanoma and other skin pathological conditions.

The chick chorioallantoic membrane (CAM) is a well-established model for studying angiogenesis, tumor growth, tissue grafting, and therapeutic responses; however, conventional in ovo approaches are limited by a short experimental window, immune-related constraints, and restricted imaging accessibility. Being naturally immunodeficient, the chick embryo accepts transplantation from various tissues and species. This article analyzes the literature data concerning the use of the CAM model to study melanoma and other skin pathological conditions, due to the lack of literature data summarizing this

Using chorioallantoic membranes for non-lethal assessment of persistent organic pollutant exposure and effect in oviparous wildlife.

David Peakall and co-workers pioneered innovative approaches that utilized extra-embryonic membranes to assess accumulation of organochlorine pesticides in eggs. This technique provided the foundation for an entire line of research to improve non-lethal methods for assessing contaminant exposure in oviparous wildlife. Currently, analysis of chorioallantoic membranes (CAMs) provides predictable estimates of chlorinated contaminant presence in eggs and in maternal tissues. Field studies have been conducted with herons, stilts, alligators, crocodiles, and sea turtles. Controlled dose-response stu

Comparison of titrations on the chorioallantoic membrane of chick embryos with the rabbit scarification technique for the potency assay of smallpox vaccines.

The potency of the U.S. Reference Smallpox Vaccine, Lot 2, the International Reference Preparation of Smallpox Vaccine, and commercial smallpox vaccines was determined by the chorioallantoic membrane (CAM) and rabbit scarification (RS) potency assay methods. The mean titer of the U.S. Reference (based on 107 ampoules) was 10(8.1) pock-forming units (PFU) per ml and that of the International Reference (based on 3 ampoules) was 10(7.8) PFU/ml. A statistical analysis of the CAM data for the U.S. Reference resulted in the establishment of a table of limits of acceptance for smallpox vaccines. Of t

Hydroethanolic root extract of Croton membranaceus suppresses angiogenesis in zinc-testosterone-induced prostate adenocarcinoma and chorioallantoic membrane models.

Angiogenesis plays a pivotal role in the growth, progression, and metastatic spread of cancer cells. However, the clinical utility of current antiangiogenic agents has been limited by the emergence of drug resistance, thereby highlighting the need for novel therapeutic candidates, including natural products. Croton membranaceus has previously been shown to suppress prostate cancer cell growth and may represent a potential antiangiogenic agent. In this study, we investigated the effect of Croton membranaceus hydroethanolic root extract (CMERE) on new blood vessel formation. The zinc-testosteron

Biocompatibility, Tissue Integration, and Angiogenic Potential of Polylactic Acid-Based Scaffolds in the Chicken Embryo Chorioallantoic Membrane Model.

The use of stem cells for the treatment of chronic wounds and burns is a promising trend in regenerative medicine. The efficacy of this approach largely depends on the cell delivery strategy. It is established that the use of polymeric scaffolds as cell carriers can, under certain conditions, facilitate integration of the graft into the host tissues and induce angiogenesis. We demonstrated that porous polylactide scaffolds induce pronounced perifocal vascularization with a minimal inflammatory response and the absence of fibrosis in the chorioallantoic membrane (CAM) model. Immediate Tissue Responses to Multipulse Low Fluence Laser Treatment in a Chorioallantoic Membrane Model of Port Wine Birthmarks.

OBJECTIVES: The standard Port Wine Stains (PWS) treatment involves using single pulse high fluence (SPHF) laser therapy, which often leads to discomfort and requires multiple sessions. According to the Arrhenius theory, employing multiple pulses with low fluence (MPLF) that have lower peak power could potentially reduce pain while still achieving the therapeutic goals. This study aims to explore the potential of MPLF at 530 nm in achieving the desired clinical endpoints in treating PWS. Additionally, it seeks to validate the predictions of the Arrhenius theory regarding thermal denaturation ag

Coordinated development of embryonic long bone on chorioallantoic membrane in ovo prevents perichondrium-derived suppressive signals against cartilage growth.

Perichondrium has been shown to elicit signals to suppress differentiation and proliferation of chondrocytes during endochondral bone formation based on in vitro organ culture. However, these in vitro organ cultures did not allow the growth of bone collar, and thus the effect of perichondrium in a normal environment where development of adjacent embryonic tissues, including bone collar, is taking place has not yet been fully understood. Therefore, we examined the effect of perichondrium on cartilage development using chicken long bone organ cultures on chorioallantoic membrane in ovo, which su

Effects of Two-by-Two Combination Therapy with Valproic Acid, Lithium Chloride, and Celecoxib on the Angiogenesis of the Chicken Chorioallantoic Membrane.

BACKGROUND: The synergistic effects of valproic acid (VPA), lithium (Li), and celecoxib (CX) have been shown in combination therapy against the proliferation and metastasis of numerous cancers. Angiogenesis plays a critical role in the pathogenesis of tumor growth and metastasis. The aim of the present study was to evaluate the antiangiogenic effects of VPA, lithium chloride (LiCl), and CX, alone or in 2-by-2 combinations, using the chicken chorioallantoic membrane (CAM) assay. METHODS: Fertilized chicken eggs were randomly divided into 10 groups: control, VPA (1.8 and 3.6 µmol/CAM), Li (0.15

CD44 and tenascin play critical roles in growth and vascular development of the chick chorioallantoic membrane and are targets of cigarette smoke.

Chorioallantoic membranes (CAMs) were used to determine which extracellular matrix molecules play essential roles in growth and vascular development in vivo and whether expression of critical molecules is affected by cigarette smoke exposure. Treatment of CAMs on day 5 of development with antibodies to CD44 or tenascin, but not to other matrix molecules, inhibited CAM growth and affected various aspects of blood vessel development including normal growth and branching of vessels, migration of vessels, and formation and differentiation of the capillary plexus. DNA synthesis was inhibited by ant

In vivo time-course of the angiogenic response induced by multiple myeloma plasma cells in the chick embryo chorioallantoic membrane.

In this study, we set out to make a fine characterization of the angiogenic response induced by plasma cells obtained from patients with active-multiple myeloma (MM), in comparison with cells obtained from patients with non-active MM and benign lesions such as monoclonal gammopathy of undetermined significance (MGUS), in the chick embryo chorioallantoic membrane (CAM) assay. To achieve this we investigated the time-course of the angiogenic response induced by gelatin sponges soaked in the cell suspensions and implanted on the CAM surface from day 8 to day 12 of incubation by evaluating the num

Implementation of intussusceptive microvascular growth in the chicken chorioallantoic membrane (CAM): 1. pillar formation by folding of the capillary wall.

Intussusceptive microvascular growth is a new mode of capillary network growth originally described in the lungs of rabbits and rats. It constitutes an alternative to endothelial sprouting. The capillary network grows by insertion of new intercapillary meshes with dimensions around 1.5 microns called tissue pillars or posts. In a recent investigation, growth by intussusception was demonstrated in the chicken chorioallantoic membrane (CAM). In the present study the first of several modes of its implementation can now be presented in the CAM by in vivo video microscopy and analyses of light and

Antioxidant Potential of Natural Extract of Ginkgo biloba L. in Relation to Chorioallantoic Membrane (CAM) Vessels of Chicken Embryo.

The chicken chorioallantoic membrane (CAM) model is an embryonic blood capillary system considered a suitable "in vivo" model for studying the irritation effect of plant extracts on the vascular system, including impacts on hemostasis, hyperemia, hemorrhage, and coagulation. The main aim of the present work was to investigate the irritation effects of different concentrated alcohol extracts of Ginkgo biloba L. (GBE) leaves on the blood vessels of the CAM model during early embryogenesis, evaluated using the Luepke scoring system. The antioxidant properties of GBE were assessed using DPPH radic

The chick chorioallantoic membrane (CAM) as a versatile patient-derived xenograft (PDX) platform for precision medicine and preclinical research.

Patient-derived xenografts (PDX) are an increasingly valuable tool in oncology, providing biologically faithful models of many different cancer types, and potential platforms for the development of precision oncology approaches. However, PDX have primarily been established in immunodeficient rodent models, with accompanying cost and efficiency constraints that pose barriers to more widespread adoption. The chicken egg chorioallantoic membrane (CAM) is an alternative in vivo PDX model. We provide here a comprehensive review of studies that grafted primary human tissue, as opposed to cell lines,

Calcium-activated ATPase of the chick embryonic chorioallantoic membrane. Identification, developmental expression, and topographic relationship with calcium-binding protein.

A Ca2+-activated ATPase activity is present in the chick embryonic chorioallantoic membrane (CAM), the placenta-like tissue which translocates eggshell calcium into the embryonic circulation. The enzyme is membrane-bound, ATP-specific, Mg2+-dependent, exhibits dual Km values of 30 microM and 0.3 mM Ca2+, and has a Mr of 170,000. Throughout embryonic development, a single electrophoretic form of the Ca2+-ATPase is found and, furthermore, its specific activity as a function of age follows a bimodal pattern. In particular, from incubation days 14-15 to the end of gestation, a period representing

Real-time Imaging of Vascular Changes and Extravasation in the Chicken Chorioallantoic Membrane (CAM) Model Exposed to Ultrasound and Microbubbles.

OBJECTIVE: The chicken chorioallantoic membrane (CAM) is used to study biological processes, including drug delivery, and provides an in ovo model with vasculature available for intravital microscopy. Ultrasound and microbubbles have been shown to be a promising strategy for improving drug delivery to solid tumors and across the blood-brain barrier. Acoustic cluster therapy (ACT) based on large microbubbles with a volume 50-100 times larger than regular contrast agent microbubbles is of particular interest. The objective of this study was to expand our understanding of the mechanisms underlyin

Chick chorioallantoic membrane as an in vivo model to study vasoreactivity: characterization of development-dependent hyperemia induced by epoxyeicosatrienoic acids (EETs).

Shell-less culture of chick chorioallantoic membrane (CAM) of developing chicken embryos is a useful model to evaluate the effects of vascular agents. We assessed the response of CAM vessels to epoxyeicosatrienoic acids (EETs), derivatives of the essential fatty acid arachidonic acid, that have a number of important biological functions, including dilation of microvessels in the coronary, cerebral, renal, and mesenteric circulations. Three of four regioisomers of EETs, 14,15-, 11,12-, and 8,9-EET, induced a characteristic dose-dependent acute hyperemia within 4 min after application on 10-day-

Investigations concerning the cultivation of myxo- and paramyxoviruses on chorioallantoic membrane fragments. Note I. Data on the multiplication of several myxo- and paramyxoviruses.

Influenza viruses A(H1N1) and A(H3N2) and parainfluenza viruses (Sendai mumps) were cultivated in chorioallantoic membrane (CAM) fragments maintained in media with different formulae, with or without daily medium changes, in roller or stationary tubes. Inoculation was performed either directly on CAM fragments in Petri dishes or by dilution of the virus-containing material in the medium. Infectant titers obtained in CAM fragments were similar to those recorded in embryonated eggs at 48 hours post inoculation (p.i.) in the case of influenza virus A(H1N1) and at 72 hours p.i. in that of Sendai a

Tailored Mesoporous Silica Nanoparticles and the Chick Chorioallantoic Membrane: A Promising Strategy and Model for Efficient Blood-Brain Barrier Crossing.

Crossing the blood-brain barrier (BBB) remains a major challenge for brain-targeted drug delivery. Mesoporous silica nanoparticles (MSNs) with tunable size and surface properties are promising vehicles for crossing the BBB. In this study, we explored the potential applications of the chick chorioallantoic membrane (CAM) model in combination with nanotherapeutics. We synthesized ∼25 nm MSNs and RITC-conjugated MSNs (RMSNs) with short PEG chains and varying amounts of positively charged molecules, specifically tertiary amine (polyethylenimine, PEI) or quaternary amine (trimethylammonium, TA), to
