Induction of chromatid breaks by carbon K-shell ultrasoft X rays.

Chromatid breaks have previously been shown to be induced in G2-phase cells after exposure to ionizing radiation (X and gamma rays) as a linear function of dose, consistent with a single-event mechanism. DNA double-strand breaks (DSBs) are thought to be the initiating lesion, and experiments with a genetically engineered cell line containing a single DSB site also indicate that a single DSB is sufficient to induce a chromatid break. Although the precise mechanism of conversion of an isolated DSB into a chromatid break is not yet understood, it is known that a proportion of chromatid breaks res

Sister-chromatid exchange in highly purified human CD4+ and CD8+ lymphocytes.

Sister-chromatid exchange (SCE) frequencies were determined in human peripheral blood CD4+ and CD8+ T lymphocyte subpopulations which were rapidly and highly purified from pooled T lymphocytes by immunological methods. The purified lymphocytes were stimulated with phytohemagglutinin (PHA) for 4 days. CD4+ lymphocytes showed significantly higher SCE frequencies than autologous CD8+ lymphocytes when measured simultaneously after identical bromodeoxyuridine (BrdU) incubation times. Differences in SCE frequencies between CD4+ and CD8+ lymphocytes were also detected when mitomycin C (MMC) was added

Increase of sister chromatid exchanges in excision repair deficient xeroderma pigmentosum.

The distribution of spontaneous sister chromatid exchanges (SCEs) was studied in PHA-stimulated lymphocytes from 15 patients affected by xeroderma pigmentosum (XP). The study of unscheduled DNA synthesis (UDS) in twelve of these patients showed that seven were deficient and five proficient. The number of SCEs in XP patient cells was higher than in those of 19 controls, and the distributions of SCEs per cell were significantly different. However, the results varied when XP patients were considered in relation to their UDS: the group of XP patients with proficient UDS did not differ, whereas the

Sister-chromatid exchanges induced by ultraviolet light in Bloom's syndrome fibroblasts.

The relationships between the cytotoxic effect of ultraviolet light and the UV-induced sister-chromatid exchanges (SCEs) were compared among fibroblast cell strains from two unrelated Bloom's syndrome (BS) patients, one xeroderma pigmentosum (XP) patient belonging to complementation group A and two unrelated normal controls. The "net" induced SCEs as a function of UV fluence, obtained by subtracting spontaneous SCEs from observed SCEs, were much higher in both BS cells and XP group A cells than in normal cells. The relative efficiency of induced SCE, defined as the "net" induced SCEs as a func

Sister-chromatid exchanges in a special form of xeroderma pigmentosum (form II).

The frequency of sister-chromatid exchanges (SCE) was studied in peripheral blood lymphocytes from a xeroderma pigmentosum (form II, XPII) patient. The cells were irradiated with UV or X-rays. In some experiments novobiocin (NB), inhibitor of topoisomerase II, or caffeine (CA), inhibitor of DNA repair were added to the cultures. The level of spontaneous SCE in the patient's lymphocytes was found to be significantly increased in comparison to that in the cells from normal donors. The inhibitors and UV-light caused a rise in the frequency of SCE in the cells taken from normal donors and except f

Incidence and localization of sister chromatid exchanges induced by nickel and chromium compounds.

Carcinogenic nickel compounds enhanced the incidence of sister chromatid exchanges (SCEs) in a concentration-dependent fashion in intact Chinese hamster ovary cells. There was a preferential induction of these exchanges in the heterochromatic regions of the chromosomes. CaCrO4 also caused a dose-dependent induction of SCEs. However, in contrast to NiCl2, the exchanges induced by CaCrO4 were not localized in any particular chromosomal region. The total incidence of exchanges was higher with CaCrO4 than with NiCl2. CaCrO4, crystalline NiS and NiCl2 enhanced the incidence of SCEs at concentration

Enhanced response to the induction of sister chromatid exchange by gamma radiation in neurofibromatosis.

The study included 8 unrelated patients with neurofibromatosis, and 10 unrelated normal and healthy persons as controls. Whole blood samples were divided into plastic T flasks and exposed at room temperature to gamma rays. The radiation dose was 36 rad/minute, and the doses delivered were 0, 75, 150 and 300 rad. The lymphocytes were cultured in (RPMI) 1640 tissue culture medium and autologous serum (20%). Phytohemagglutinin and bromodeoxyuridine (Brdu) (10 microM) were added at initiation of culture and harvesting was done 64 to 68 hours after culture initiation. Slides were coded, differentia

Further studies on sister-chromatid exchange frequency in users of hormonal contraceptives.

Women using an estrogen-progestogen combination contraceptive exhibited an increased frequency of sister-chromatid exchange (SCE), supporting our previous observation. These elevated SCE frequencies tended to decline 3 months after the discontinuation of the pill. Women using a progestogen injectable contraceptive, on the other hand, had unaltered SCE rates. The analysis, in vitro; of mitomycin-C-induced SCE frequencies in women using either the combination or the progestogen injectable contraceptives showed significantly higher rates of induced SCE.

[Induction of sister chromatide exchange by trichloroethylene and its metabolites (author's transl)].

The mutagenecity of trichloroethylene and its metabolites (trichloroethanol and chloral hydrate) have been tested by using the method of sister chromatide exchanges (SCE). We have found the increasing SCE in workers chronically exposed to trichloroethylene. The increasing of SCE was also observed when the human lymphocytes were exposed to trichloroethanol and chloral hydrate in vitro in certain concentrations. The all results of tests groups are compared with those of the control groups and the difference is statistically significant.

The effects of beta-radiation on sister-chromatid exchanges in cultured human lymphocytes.

The incidence of Sister-Chromatid Exchanges (SCEs) due to beta-radiation was investigated in cultured human lymphocytes using the BrdU/Giemsa technique. Cultures treated continuously with 0.001 and 0.01 microCi of [3H]uridine showed no increase in either chromosome abnormalities or SCEs. Continuous treatment with 0.1 microCi resulted in a significant increase in chromosome aberrations but no increase in SCEs, while treatment with 0.2 microCi gave both an increase in chromosome aberrations and SCEs. Cultures given a 4-h pulse with 1.0 microCi showed a significant increase in both SCEs and chrom

The relationship between sister chromatid exchanges and chromosome aberrations in Bloom's syndrome.

The distribution of the break points of sister chromatid exchanges (SCE) was compared with that of chromosome aberrations in Bloom's syndrome by using differential sister chromatid staining and banding techniques. A comparison was made of the distribution in chromosomes 1, 2, and 3, since the exact identification of other chromosomes is difficult with the differential sister-chromatid staining technique. It was shown that SCE and chromosome breaks do not necessarily correlate as to location. Some chromosome break points, e.g., 1q21, 1p36, 2q31, 3q12, and 3p13, were common with those of SCE, wh

UV-light induced sister chromatid exchanges in xeroderma pigmentosum lymphocytes.

Cultured lymphocytes from 9 patients with clinically different types of xeroderma pigmentosum were exposed to ultraviolet light at 24 h. An increased rate of sister chromatid exchanges were observed in 6 patients (128--148% increase in three, 34--51% in three), but not in three patients with deSanctis-Cacchione syndrome (xeroderma pigmentosum with mental defect), compared to simultaneously cultured controls. A positive result could be useful as preliminary cytogenetic diagnostic test. The results are interpreted as an expression of UV-light induced chromosomal instability due to impaired DNA r

Micronuclei and sister chromatid exchange in lymphocytes from physicians exposed to ionizing radiation.

OBJECTIVE: Occupational exposure to ionizing radiation is a known risk factor for genomic instability. This study evaluated whether physicians occupationally exposed to ionizing radiation exhibit higher frequencies of chromosomal alterations compared to unexposed hospital workers, utilizing micronuclei and sister chromatid exchanges as biomarkers. METHODS: Peripheral blood samples were analyzed from 50 participants: 25 physicians exposed to ionizing radiation (radiologists) and a control group of 25 sex- and age-matched hospital staff with no history of occupational exposure. The cytokinesis-

The alteration of sister chromatid exchange frequencies in Behçet's disease with and without HLA-B51.

BACKGROUND: The analysis of sister chromatid exchange (SCE) is a cytogenetic technique used to show DNA damage as a result of an exchange of DNA fragments between sister chromatids. It is known that there is an increased SCE frequency in Behçet's disease (BD). OBJECTIVE: To investigate whether human leucocyte antigen (HLA)-B51-positive patients with Behçet's disease exhibit higher SCE frequencies than those without HLA-B51. METHODS: Lymphocytes from 75 patients (38 women, 37 men) and from 50 controls (28 women, 22 men) were cultured in darkness for 72 h in the presence of bromodeoxyuridine.

Roles of the sister chromatid cohesion apparatus in gene expression, development, and human syndromes.

The sister chromatid cohesion apparatus mediates physical pairing of duplicated chromosomes. This pairing is essential for appropriate distribution of chromosomes into the daughter cells upon cell division. Recent evidence shows that the cohesion apparatus, which is a significant structural component of chromosomes during interphase, also affects gene expression and development. The Cornelia de Lange (CdLS) and Roberts/SC phocomelia (RBS/SC) genetic syndromes in humans are caused by mutations affecting components of the cohesion apparatus. Studies in Drosophila suggest that effects on gene exp

Sister chromatid exchanges in ring chromosomes following X-irradiation of human lymphocytes.

PURPOSE: To examine whether X-rays induce sister chromatid exchanges (SCE). MATERIALS AND METHODS: Peripheral lymphocytes irradiated in vitro or in vivo were cultured and treated with okadaic acid to generate premature chromosome condensation (PCC). When identical spreads were analysed using conventional Giemsa staining and pan-centromeric fluorescence in situ hybridization painting, ring chromosomes were observed. RESULTS: In PCC preparations, cells in the late G(2) phase and late M phase were observed. In late M phase cells, 17-20% of ring chromosomes lacked one chromatid (single-chromatid

Sister chromatid exchanges in peripheral lymphocytes in newborns treated with phototherapy and vitamin E.

A study was undertaken to determine whether blue fluorescent light might affect the sister chromatid exchange (SCE) frequency of peripheral lymphocytes in icteric newborns undergoing continuous phototherapy treatment (72 h). Also, the potential preventive effect of vitamin E on SCE frequency was studied in a subgroup of 11 preterm and 9 fullterm newborns after daily administration of vitamin E (46.44 mumol/kg/d, im). The results revealed that only the preterm icteric newborns showed an increase in mean SCE frequency of peripheral lymphocytes after phototherapy (9%, p = 0.02), but in no case di

[The relationship between the radiosensitivity of DNA replicative synthesis and the formation of sister chromatid exchanges].

It has been found that irradiation in doses 0.5-2.0 Gy does not enhance the frequency of sister chromatid exchanges in cells of patients with Down's syndrome and ataxia-telangiectasia compared to the normal cells. In the case of ataxia, this phenomenon was accompanied with radioresistant replicative DNA synthesis, whereas in two cases of Down's syndrome the replicative DNA synthesis was found to be as radiosensitive as in the norm. According to these data, the mechanism of sister chromatid exchanges proposed in our previous publication (Pleskach et al., 1988) seems to be rather doubtful.
