Group II-VI magic clusters, featuring precise atomic compositions, well-defined geometric structures, and superior stability, hold great potential in atomic-scale manufacturing. While monodispersed magic clusters exhibit unambiguous characteristics, their response to external stimuli has to be understood. Moreover, strong interaction between clusters and an external field is desirable in order for clusters to assemble or tailor their physicochemical properties instantaneously for device applications. Herein, we explore the effects of an electric field on the absorption and circular dichroism s

Previously we have shown that human red blood cells (RBCs) undergo a sudden change from blocking to passing through a 1.3+/-0.2-microm micropipette when applying an aspiration pressure of 2.3 kPa at a critical transition temperature (Tc = 36.4+/-0.3 degrees C). Low-shear viscosity measurements suggested that changes in the molecular properties of hemoglobin might be responsible for this effect. To evaluate structural changes in hemoglobin at the critical temperature, we have used circular dichroism (CD) spectroscopy. The thermal denaturation curves of human hemoglobin A (HbA) and hemoglobin S

Structural properties, and especially the differential stability, of complexes between carbonic anhydrase (CA) and three sulfonamide inhibitors, acetazolamide, dorzolamide and methazolamide, were investigated by spectroscopic and electrophoretic techniques. These included denaturant gradient gel electrophoresis either across a urea or a steady-state transverse sodium dodecyl sulfate (SDS) gradient. Acetazolamide, the smallest and most hydrophilic of the sulfonamides, forms the most stable complex in the presence of urea, whereas dorzolamide, with a bulky and hydrophobic structure, is most stab

The sequences of 32 phospholipases A2 (EC 3.1.1.4) were analysed by secondary-structure prediction and the results were compared with the available crystallographic data. Good agreement is evident between prediction and experiment, especially for helical structure. Circular dichroic spectra were also determined for six enzymes from Elapid snake venom and these, in association with previously published spectra, confirm the main implication of the predictions, namely that all the homologues have qualitatively similar tertiary structures. Consideration was then given to possible structure/activit

We present a theoretical and conceptual framework for single-shot detection of electronic or vibrational circular dichroism (CD) using vector vortex beams combined with orbital angular momentum (OAM) sorting. In this approach, a linearly polarized Gaussian beam is transformed by a q-plate into a spin-orbit superposition comprising left- and right-circularly polarized components carrying equal and opposite OAM. When this structured beam interacts with a chiral sample, the two spin components experience unequal attenuation, producing an intrinsic intensity imbalance directly proportional to the

Chiral lead halide perovskites (chiral LHPs) have emerged as one of the best candidates for opto-spintronics due to their large spin-orbit coupling (SOC) and unique chirality-induced spin selectivity (CISS) even in the absence of a magnetic field. Here, we report the impact of halide composition on circular dichroism (CD) and magneto-photoluminescence (PL) effects of chiral 2D LHPs (R/S-MBA)2PbBrxI4-x (MBA = C6H5CH2(CH3)NH3). By tuning the mixing ratio of Br/I halide anions, we find that (R/S-MBA)2PbBrxI4-x thin films exhibit tunable and wide wavelength range CD signals. Simultaneously, the ma

Caveolin-1 is a main structural component of caveolae and essential for the invagination of caveolae by forming a hairpin-shaped structure in the membrane-inserting domain (residues, 102-122). In this article, we determined the tertiary structures of the peptides comprising residues 93-126 and 101-126 of caveolin-1 in 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) aqueous solution and sodium dodecyl sulfate (SDS) micelles, respectively, by nuclear magnetic resonance (NMR) study. The self-association of the peptides in SDS and dodecylphosphocholine (DPC) micelles was also studied by circular dichrois

The optical and magnetic properties of the multi-metal rings [NH(2)R(2)][Cr(7)MF(8)(O(2)CCMe(3))(16)], where M = Cd(II), Mn(II) or Ni(II), have been studied using variable-field and variable-temperature magnetic circular dichroism (MCD) in the UV-visible spectra. Spectra of samples were recorded in a frozen organic matrix or cast in a polymethacrylate (PMMA) polymer film between 1.7 and 75 K. The spectra are characteristic of the Cr(III) ion (d(3)) in a rhombic field when M = Cd(II). In the case that M = Ni(II) additional optical transitions arise from the d(8) ion whereas for M = Mn(II) no ad

Aggregation of Abeta peptides is a seminal event in Alzheimer's disease. Detailed understanding of Abeta assembly would facilitate the targeting and design of fibrillogenesis inhibitors. Here comparative conformational and aggregation studies using CD spectroscopy and thioflavine T fluorescence assay are presented. As a model peptide, the 11-28 fragment of Abeta was used. This model peptide is known to contain the core region responsible for Abeta aggregation. The structural and aggregational behaviour of the peptide was compared with the properties of its variants corresponding to natural, cl

Circular dichroism spectroscopy has been used to study how different solvents stabilize secondary structure in the neuropeptide galanin (rat), two N-terminal fragments of galanin, galanin(1-12) and galanin(1-16), and six other differently charged analogs. Among these analogs, the peptide M40, galanin(1-13)-Pro-Pro-Ala-Leu-Ala-Leu-Ala amide, is a high affinity, receptor subtype specific galanin receptor antagonist. The different solvents include sodium dodecyl sulfate (SDS) micelle solutions, 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) and 1,2-dioleoyl-sn-glycero-3-phosphoglycerol (DOPG) ve

Simultaneous binding of two drugs to human serum albumin (HSA) was investigated by difference circular dichroism (delta CD) spectroscopy. Phenylbutazone and diazepam were chosen as specific markers for binding areas I (cumarines) and II (indoles), respectively, and their stereospecific interactions with protein were selectively characterized. Displacers were drugs known to specifically bind to areas I (salicylate) and II (racemic ibuprofen). The results indicate two different interaction mechanisms: a direct competition one (diazepam-ibuprofen and phenylbutazone-salicylate) and an indirect com

S100a is a heterodimeric, acidic calcium-binding protein that interacts with calmodulin antagonists in a Ca2(+)-dependent manner. In order to study the behavior of the hydrophobic domain on S100a when bound to Ca2+, its interaction with trifluoperazine (TFP) was investigated using 16F nuclear magnetic resonance (NMR) and circular dichroism (CD) spectroscopy. The dissociation constant (Kd) values of TFP, as estimated from the chemical shifts of 19F NMR, were 191 and 29 microns in the absence and presence of Ca2+, respectively, and were similar to those previously reported for S100b. However, th

Here, we present the new application of solid-state Vibrational Circular Dichroism (VCD) spectroscopy to differentiate several dutasteride (DS) solvatomorphs - the model active pharmaceutical ingredient (API). Several crystalline DS hydrochloride hydrates solvated with methanol, ethanol, acetonitrile, acetone, and acetic acid were prepared. In contrast to almost identical IR spectra, the VCD ones were very sensitive to changes in the sample composition. We marked significant differences in the shape of VCD spectra of studied DS solvatomorphs, DS hydrates, and DS polymorphic forms. Our findings

The coherent ring current of Mg-phthalocyanine created by a broad band UV-visible pump pulse shows variation with time, where the ring currents at the corner benzene rings, around the Mg cation and on the outer ring oscillate with different time periods and the current density migrates among these regions. The 7 pairs of E u degenerate excited states populated upon photoexcitation, generate 21 distinct coherent ring currents. We further calculate the time-resolved X-ray circular dichroism (TRXCD) spectrum of the coherences contributing to the ring current obtained by an attosecond X-ray probe

Manganese porphyrins are used as catalysts in the oxidation of olefins and nonactivated hydrocarbons. Key to these reactions are high-valent Mn-(di)oxo species, for which [Mn(Porph)(X)] serve as precursors. To elucidate their properties, it is crucial to understand the interaction of the Mn center with the porphyrin ligand. Our study focuses on simple high-spin [MnIII(TPP)X] (X = F, Cl, I, Br) complexes with emphasis on the spectroscopic properties of [MnIII(TPP)Cl], using variable-temperature variable-field magnetic circular dichroism spectroscopy and time-dependent density functional theory

OBJECTIVE: To analyze the protein structure encoded by COL4A5 gene with a missense mutation and to discuss the effect of gene mutation on basic structure and predicted secondary structure of the encoded protein.
METHODS: A fragment of cDNA with a g.3246G T mutation resulting in p.G1015V from an X-linked Alport patient, and that of corresponding cDNA from a control were expressed in E.coli. The recombinant and mutant polypeptide was a fragment of COL4A5, containing 158 amino acid residues with a glycine to valine substitution mutation in it. The secondary structure of the two recombinant prote

Repressor-activator protein 1 (RAP1) has an essential role in the maintenance of yeast telomeres. Yeast telomeric DNA consists of simple repeated G-rich sequences that are bound by RAP1. We have found that RAP1, in addition to its known binding activity for double-stranded DNA, interacts with the G-rich strand containing guanine base (G)-tetrads. We show here using circular dichroism spectroscopy that RAP1 promotes the formation of one particular type of DNA quadruplex, parallel G4-DNA. Furthermore, RAP1 is able to bind to both preformed parallel and antiparallel DNA quadruplexes. These result

Absorption and magnetic curcular dichroism spectra of nonequilibrium states of peroxidase and its complexes with F-, N3-, CN- produced by reduction of oxidased forms of proteins by thermalysed electrons at 77 degrees K were studied. Mixtures of high spin and low spin ferroforms were found in nonequilibrium states of peroxidase and complexes with F- and N3-, the content of the high spin ferroform increasing as follows: N3- complex less than peroxidase less than fluorine complex. Only low spin ferroforms was found after low temperature reduction of the cyanide complex. The existence of the low s

X-ray magnetic circular dichroism (XMCD) and X-ray magnetic linear dichroism (XMLD) are powerful spectroscopic techniques for probing magnetic properties in solids. In this study, we revisit the XMCD and XMLD sum rules within a complete magnetic multipole basis that incorporates both spinless and spinful multipoles. We demonstrate that these multipoles can be clearly distinguished and individually detected through the sum-rule formalism. Within this framework, the anisotropic magnetic dipole term is naturally derived in XMCD, offering a microscopic origin for ferromagnetic-like behavior in ant

Calreticulin of Brugia malayi (BmCRT) play very important role in host-parasite interaction. In previous study it was found that BmCRT is responsible for prevention of host classical complement pathway activation via its interaction with first component C1q of the human host. Therefore, BmCRT is an essential protein for parasite survival and an important drug target to fend filariasis. In the present study, we have carried out a systamatic biophysical characterization of BmCRT protein. Unfolding of BmCRT was found to be non-cooperative two-state process in the presence of both denaturant GdmCl
