Fine-Regional Role of the Claustrum in Anxiety and Higher Sensitivity to Cocaine in Adolescent Cocaine-Exposed Male Mice during Adulthood.

Adolescent cocaine exposure (ACE) induces anxiety and higher sensitivity to substances abuse during adulthood. Here, we show that the claustrum is crucial for controlling these psychiatric problems in male mice. In anxiety-like behavioral tests, the CaMKII-positive neurons in the median portion of the claustrum (MClaustrum) were triggered, and local suppression of these neurons reduced the anxiety-like behavior in ACE mice during adulthood. In contrast, the CaMKII-positive neurons in the anterior portion of the claustrum (AClaustrum) were more activated in response to subthreshold dose of coca

Is REM sleep a paradoxical state?: Different neurons are activated in the cingulate cortices and the claustrum during wakefulness and paradoxical sleep hypersomnia.

Michel Jouvet proposed in 1959 that REM sleep is a paradoxical state since it was characterized by the association of a cortical activation similar to wakefulness (W) with muscle atonia. Recently, we showed using cFos as a marker of activity that cortical activation during paradoxical sleep (PS) was limited to a few limbic cortical structures in contrast to W during which all cortices were strongly activated. However, we were not able to demonstrate whether the same neurons are activated during PS and W and to rule out that the activation observed was not linked with stress induced by the flow

Dopamine receptor 1 on CaMKII-positive neurons within claustrum mediates adolescent cocaine exposure-induced anxiety-like behaviors and electro-acupuncture therapy.

Adolescent cocaine exposure (ACE) increases risk of developing psychiatric problems such as anxiety, which may drive relapse in later life, however, its underlying molecular mechanism remains poorly understood. Methods: ACE male mice model were established by exposing to cocaine during adolescent period. Elevated plus maze (EPM) were used to assess anxiety-like behaviors in mice. Within claustrum, local injection of SCH-23390, a specific antagonist for dopamine receptor 1 (D1R), or D1R knocking-down virus were used to regulate D1R function or expression on CaMKII-positive neurons (D1RCaMKII) i

Febrile infection-related epilepsy syndrome with claustrum sign following a human rhinovirus-positive respiratory illness in an adult: a case report.

BACKGROUND: Human rhinovirus (HRV) is the predominant viral pathogen causing community-acquired upper respiratory tract infections, typically manifesting as mild coryzal symptoms. Severe neurological complications, including febrile infection-related epilepsy syndrome (FIRES), are exceptionally rare. CASE PRESENTATION: We report a case of febrile infection-related epilepsy syndrome (FIRES) with bilateral claustral involvement occurring in temporal association with an HRV-positive febrile respiratory illness, identified at the General Hospital of Ningxia Medical University in April 2025. Connectivity of the Superior Temporal Polysensory Area With the Presubiculum, Parasubiculum, Entorhinal Cortex, and Claustrum-Endopiriform Complex in the Common Marmoset (Callithrix jacchus).

The rodent hippocampal-presubicular-entorhinal circuit is thought to be a key part of the circuitry involved in memory formation. In primates, additional connections are believed to enable more complex and higher-order memory information processing. We investigated whether such additional connections exist in the cortical regions adjacent to the hippocampal formation using a tracer injection method. We discovered that the marmoset parasubiculum (ParS), presubiculum (PreS) (distal portion), and entorhinal cortex (EC) (proximal or medial portion) have strong reciprocal connections with the super

A developmental switch in corticoclaustral signaling.

The anterior cingulate cortex (ACC) is a high-order cortical region involved in different functions including attention,1,2 pain processing,3,4,5 and predictive coding.6,7,8,9 ACC neurons send top-down inputs to several cortical and subcortical brain regions including the claustrum.10,11,12,13,14,15 Synaptic input from the ACC is thought to drive the activation of claustrum neurons, which in turn amplify and distribute ACC-related information back to the cortex to help orchestrate cortical networks during demanding cognitive states.12,16,17,18 However, the circuit mechanisms underlying ACC-cla

Frontoclaustral interrelations in cats and dogs.

In 13 cats and 5 dogs various parts of the frontal lobe were removed. As a result terminal degenerations in the rostral quarter of the insular claustrum were found by Nauta and Fink-Heimer II methods. In cats with motor cortex lesions, the degenerations were localized more dorsally than in cats with removals of the frontoorbital cortex, but there is a large overlap between degenerating areas of the claustrum in both groups of animals. In the dog it even seems possible to differentiate claustral regions in which axons from various gyri of the frontal lobe mainly terminate. It was found under th

Corticoclaustral connections in the cat.

The claustrum is a sheet-like layer of gray matter situated between the external and extreme capsules of the mammalian brain. This structure was first described by the French physician and anatomist Vicq d'Azyr in 1786. The claustrum's phylogeny, ontogeny and functional characteristics have long been the subject of debate and considerable investigative efforts. However, despite such efforts (or perhaps as a result thereof), significant disparities and discrepancies remain, most notably in the context of the claustrum's afferent and efferent connections. For the purpose of this study, we sought

Claustral Input to the Macaque Medial Posterior Parietal Cortex (Superior Parietal Lobule and Adjacent Areas).

The projections from the claustrum to cortical areas within and adjacent to the superior parietal lobule were studied in 10 macaque monkeys, using retrograde tracers, computerized reconstructions, and quantitative methods. In contrast with the classical view that posterior parietal areas receive afferents primarily from the dorsal and posterior regions of the claustrum, we found that these areas receive more extensive projections, including substantial afferents from the anterior and ventral regions of the claustrum. Moreover, our findings uncover a previously unsuspected variability in the pr

Claustral Neurons Projecting to Frontal Cortex Mediate Contextual Association of Reward.

The claustrum is a small nucleus, exhibiting vast reciprocal connectivity with cortical, subcortical, and midbrain regions. Recent studies, including ours, implicate the claustrum in salience detection and attention. In the current study, we develop an iterative functional investigation of the claustrum, guided by quantitative spatial transcriptional analysis. Using this approach, we identify a circuit involving dopamine-receptor expressing claustral neurons projecting to frontal cortex necessary for context association of reward. We describe the recruitment of claustral neurons by cocaine and

Claustral afferents of superior parietal areas PEc and PE in the macaque.

The exposed surface of the primate superior parietal cortex includes two cytoarchitectonically defined areas, the PEc and PE. In the present study we describe the distribution of neurons projecting from the claustrum to these areas. Retrograde neuronal tracers were injected by direct visualization of regions of interest, and the location of injection sites was reconstructed relative to cytoarchitectural borders. For comparison, the patterns of claustral label that resulted from injections involving neighboring cytoarchitectonic areas were analyzed. We found that the claustral territories sendi

Physostigmine enhances blood flow-metabolism ratio in neocortex.

Inhibition of central nervous system cholinesterase with a single pulse of physostigmine induces a pronounced increase of blood flow in the neocortex, cingulate gyrus, claustrum, and amygdala. This phenomenon is not accompanied by an increase in energy metabolism and may help explain the effect of this drug on memory in normal humans and patients with Alzheimer's disease, as well as other conditions. In contrast, a parallel increase of blood flow and metabolism was observed in the superior colliculus, a component of the visual pathways. Prolonged administration of physostigmine lead to persist

Controlling for activity-dependent genes and behavioral states is critical for determining brain relationships within and across species.

The genetic profile of vertebrate pallia has long driven debate on homology across distantly related clades. Based on an expression profile of the orphan nuclear receptor NR4A2 in mouse and chicken brains, Puelles et al. (The Journal of Comparative Neurology, 2016, 524, 665-703) concluded that the avian lateral mesopallium is homologous to the mammalian claustrum, and the medial mesopallium homologous to the insula cortex. They argued that their findings contradict conclusions by Jarvis et al. (The Journal of Comparative Neurology, 2013, 521, 3614-3665) and Chen et al. (The Journal of Comparat

Early memory deficits and extensive brain network disorganization in the AppNL-F/MAPT double knock-in mouse model of familial Alzheimer's disease.

A critical challenge in current research on Alzheimer's disease (AD) is to clarify the relationship between network dysfunction and the emergence of subtle memory deficits in itspreclinical stage. The AppNL-F/MAPT double knock-in (dKI) model with humanized β-amyloid peptide (Aβ) and tau was used to investigate both memory and network dysfunctions at an early stage. Young male dKI mice (2 to 6 months) were tested in three tasks taxing different aspects of recognition memory affected in preclinical AD. An early deficit first appeared in the object-place association task at the age of 4 months, w

Meta-analysis of neural correlates of working memory, reward, and emotion processing in major depressive disorder using ALE.

BACKGROUND: Functional magnetic resonance imaging (fMRI) has revealed inconsistent neural activity patterns in major depressive disorder (MDD) across cognitive and affective domains, and this study used an activation likelihood estimation (ALE) meta-analysis to examine brain function abnormalities in working memory, reward processing, and emotion processing. METHODS: A systematic search was conducted in PubMed, Embase, Web of Science, ScienceDirect, and CNKI for fMRI studies comparing MDD patients with healthy controls (HCs), including data up to 3 December 2024. Claustral neurons projecting to frontal cortex restrict opioid consumption.

The synthetic opioid fentanyl is a major contributor to the current opioid addiction crisis. We report that claustral neurons projecting to the frontal cortex limit oral fentanyl self-administration in mice. We found that fentanyl transcriptionally activates frontal-projecting claustrum neurons. These neurons also exhibit a unique suppression of Ca2+ activity upon initiation of bouts of fentanyl consumption. Optogenetic stimulation of frontal-projecting claustral neurons, intervening in this suppression, decreased bouts of fentanyl consumption. In contrast, constitutive inhibition of frontal-p

An FMRI meta-analysis of interoception in eating disorders.

Eating Disorders (EDs) are associated with disturbed interoception - the sense of the internal condition of the body. Disturbances in interoception across senses have not yet been comprehensively examined in EDs. To do so, we employed an innovative Bayesian author-topic model approach to fMRI meta-analyses that pools together neural deficits across interoceptive senses and task types in participants with and recovered from EDs. Following PRISMA guidelines, our results combine activation patterns from 1,341 initially screened studies and data from 25 manuscripts that met study criteria that com

Functional brain networks associated with the urge for action: Implications for pathological urge.

Tics in Tourette syndrome (TS) are often preceded by sensory urges that drive the motor and vocal symptoms. Many everyday physiological behaviors are associated with sensory phenomena experienced as an urge for action, which may provide insight into the neural correlates of this pathological urge to tic that remains elusive. This study aimed to identify a brain network common to distinct physiological behaviors in healthy individuals, and in turn, examine whether this network converges with a network we previously localized in TS, using novel 'coordinate network mapping' methods. Systematic se

Common and specific neural correlates underlying creative generation and evaluation: An ALE meta-analysis study.

The cognitive process underlying creative thinking is often conceptualized within a dual-state model, dividing it into generation and evaluation sub-processes. The generation process primarily involves the reorganization of existing knowledge to foster novel ideas, which are subsequently assessed for novelty and appropriateness during the evaluation process. Although these processes likely engage distinct cognitive components and neural mechanisms, there is a paucity of neuroimaging studies directly comparing them. This study utilized the activation likelihood estimation technique to pinpoint
