Lineage A Betacoronavirus NS2 Proteins and the Homologous Torovirus Berne pp1a Carboxy-Terminal Domain Are Phosphodiesterases That Antagonize Activation of RNase L.

Viruses in the family Coronaviridae, within the order Nidovirales, are etiologic agents of a range of human and animal diseases, including both mild and severe respiratory diseases in humans. These viruses encode conserved replicase and structural proteins as well as more diverse accessory proteins, encoded in the 3' ends of their genomes, that often act as host cell antagonists. We previously showed that 2',5'-phosphodiesterases (2',5'-PDEs) encoded by the prototypical Betacoronavirus, mouse hepatitis virus (MHV), and by Middle East respiratory syndrome-associated coronavirus antagonize the o

Protective Association of Endemic HCoV-NL63 With Symptomatic COVID-19 in a Pediatric Cohort.

BACKGROUND: Children experience lower rates of symptomatic and severe coronavirus disease 2019 (COVID-19) than adults, potentially due to cross-reactive immunity from prior endemic human coronavirus (HCoV) infections. However, epidemiologic longitudinal evidence supporting this mechanism in children remains limited. In 2021, outbreaks of HCoV-NL63 and HCoV-OC43 occurred in Furano, Japan. METHODS: This prospective longitudinal cohort study was conducted at a single hospital in Furano. The primary outcome was time to onset of symptomatic COVID-19. Children with HCoV-NL63 (weeks 1-21) or HCoV-OC

Human Coronavirus 229E Uses ORF4/4a to Antagonize the Host Restriction Factor SERINC5.

Serine incorporator 5 (SERINC5) restricts the infectivity of various enveloped viruses, including HIV-1 and severe acute respiratory syndrome coronavirus 2. However, these pandemic viral pathogens have evolved mechanisms to counteract this restriction. Here, we examined the impact of all five human SERINC family members on the seasonal human coronaviruses (hCoVs) 229E and OC43, which account for up to 15% of global mild respiratory infections and can cause severe disease in vulnerable individuals. Our data show that both exogenous and endogenous SERINC1, SERINC3, and SERINC5 significantly redu

A Mouse-Adapted Model of HCoV-OC43 and Its Usage to the Evaluation of Antiviral Drugs.

The human coronavirus OC43 (HCoV-OC43) is one of the most common causes of common cold but can lead to fatal pneumonia in children and elderly. However, the available animal models of HCoV-OC43 did not show respiratory symptoms that are insufficient to assist in screening antiviral agents for respiratory diseases. In this study, we adapted the HCoV-OC43 VR-1558 strain by serial passage in suckling C57BL/6 mice and the resulting mouse-adapted virus at passage 9 (P9) contained 8 coding mutations in polyprotein 1ab, spike (S) protein, and nucleocapsid (N) protein. Pups infected with the P9 virus

Nanochiral graphene quantum dots preferentially target virus-organized membrane states for host-sparing antiviral activity.

Selectively perturbing pathogenic membranes while preserving host-cell integrity is a fundamental challenge in the design of membrane-active biomaterials. This challenge is particularly acute for enveloped viruses because their lipid membranes are derived from host cells and therefore share many of the same molecular constituents. Here, we demonstrate that engineered nanochirality provides a structural parameter for distinguishing virus-organized membrane nanodomains from host-cell membranes. Histidine-functionalized graphene quantum dots (His-GQDs) form enantiomeric nanoscale structures with

Investigating theobromine as a potential anti-human coronaviral agent.

Coronaviruses (CoVs) have long been known to infect humans, mainly alpha-CoV and beta-CoV. The vaccines developed for SARS-CoV-2 are likely not effective against other coronavirus species, whereas the risk of the emergence of new strains that may cause the next epidemic/pandemic is high. The development of antiviral drugs that are effective across different CoVs represents a viable strategy for improving pandemic preparedness. In this study, we aim to identify pan-coronaviral agents by targeting the conserved main protease (Mpro). For drug screening, the catalytic dyad of four human CoVs (HCoV

Coronavirus hemagglutinin-esterase and spike proteins coevolve for functional balance and optimal virion avidity.

Human coronaviruses OC43 and HKU1 are respiratory pathogens of zoonotic origin that have gained worldwide distribution. OC43 apparently emerged from a bovine coronavirus (BCoV) spillover. All three viruses attach to 9-O-acetylated sialoglycans via spike protein S with hemagglutinin-esterase (HE) acting as a receptor-destroying enzyme. In BCoV, an HE lectin domain promotes esterase activity toward clustered substrates. OC43 and HKU1, however, lost HE lectin function as an adaptation to humans. Replaying OC43 evolution, we knocked out BCoV HE lectin function and performed forced evolution-popula

SNX10 enhances HCoV-OC43 infection by facilitating viral entry and inhibiting virus-triggered autophagy.

The ongoing coronavirus epidemic, including the novel coronavirus (SARS-CoV-2), continues to pose a significant threat to global public health. Host targets address multiple stages of the viral life cycle and provide diverse opportunities for therapeutic interventions. This study identified sorting nexin 10 (SNX10) as a facilitator of replication of human coronavirus OC43 (HCoV-OC43), underscoring its potential as a novel antiviral target. The knockout of SNX10 significantly suppressed HCoV-OC43 replication both in vivo and in vitro. Immunoprecipitation-mass spectrometry (‌IP-MS) analysis iden

Multifactorial Analysis Identifies Conditions for Robust HCoV-OC43 Replication in Primary Human Bronchial Epithelial Cells Cultured at Air-Liquid Interface.

Air-liquid interface (ALI) cultures recapitulate key features of the airway epithelium by driving basal cell differentiation into ciliated, club, and goblet cells and by generating a functional mucus barrier, thereby representing a highly relevant model of the human respiratory tract. Using a reduced factorial Design of Experiments (DoE) methodology, we simultaneously investigated the effects of seven variables on human coronavirus OC43 (HCoV-OC43) replication in air-liquid interface (ALI)-cultured primary human bronchial epithelial cells (pHBECs) to identify robust conditions that support inf

Discontinuous template switching generates coronavirus subgenomic RNAs from the 3' viral genome end by 5' to 3' transcription.

In coronavirus (CoV)-infected cells, several structural and accessory proteins are synthesized from subgenome RNAs (sgRNA) containing a common genomic 5'-leader followed by a given open reading frame (ORF). We report that the abundance of these sgRNAs varies with distance from the 3'-end of the genome. Thus, there are more sgRNAs encoding nucleocapsid (N) than spike (S), presumably the results from discontinuous 5'-3' transcription template switch mediated by the viral replication and transcription complex (RTC). We optimized the circular polymerase extension reaction (CPER) methodology to gen

High-Throughput Screening and Identification of Potent Broad-Spectrum Inhibitors of Coronaviruses.

Coronaviruses (CoVs) act as cross-species viruses and have the potential to spread rapidly into new host species and cause epidemic diseases. Despite the severe public health threat of severe acute respiratory syndrome coronavirus and Middle East respiratory syndrome CoV (MERS-CoV), there are currently no drugs available for their treatment; therefore, broad-spectrum inhibitors of emerging and endemic CoVs are urgently needed. To search for effective inhibitory agents, we performed high-throughput screening (HTS) of a 2,000-compound library of approved drugs and pharmacologically active compou

Zein-based nanostructured coatings: A green approach to enhance virucidal efficacy of protective face masks.

Face masks represent a valuable tool to prevent the spreading of airborne viruses; however, they show poor comfort and scarce antiviral efficacy. Zein-based coatings are herein exploited to enhance antiviral performance. Zein functionalization is done through acidifying agents (lactic acid, LA). Coatings are characterized in terms of morphological, mechanical, breathability, and cytotoxicity analyses. The antiviral efficacy is tested in vitro against four viruses (Human Coronavirus OC43, Herpes Simplex Virus type 1, Human Adenovirus type 5, and MPox Virus) according to a biological assay on ce

Epidemiological and clinical features of human coronavirus infections among different subsets of patients.

BACKGROUND: Epidemiological and clinical data of human coronaviruses (HCoVs) infections are restricted to span 1-3 years at most. We conducted a comprehensive 9-year study on HCoVs by analyzing 1137 respiratory samples from four subsets of patients (asymptomatic, general community, with comorbidities, and hospitalized) in São Paulo, Brazil. METHODS: A pan-coronavirus RT-PCR screening assay was performed, followed by species-specific real-time RT-PCR monoplex assays. RESULTS: Human coronaviruses were detected in 88 of 1137 (7.7%) of the samples. The most frequently detected HCoV species were

Identification of a new human coronavirus.

Three human coronaviruses are known to exist: human coronavirus 229E (HCoV-229E), HCoV-OC43 and severe acute respiratory syndrome (SARS)-associated coronavirus (SARS-CoV). Here we report the identification of a fourth human coronavirus, HCoV-NL63, using a new method of virus discovery. The virus was isolated from a 7-month-old child suffering from bronchiolitis and conjunctivitis. The complete genome sequence indicates that this virus is not a recombinant, but rather a new group 1 coronavirus. The in vitro host cell range of HCoV-NL63 is notable because it replicates on tertiary monkey kidney

Favorable antibody responses to human coronaviruses in children and adolescents with autoimmune rheumatic diseases.

BACKGROUND: Differences in humoral immunity to coronaviruses, including severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), between children and adults remain unexplained, and the effect of underlying immune dysfunction or suppression is unknown. Here, we sought to examine the antibody immune competence of children and adolescents with prevalent inflammatory rheumatic diseases, juvenile idiopathic arthritis (JIA), juvenile dermatomyositis (JDM), and juvenile systemic lupus erythematosus (JSLE) against the seasonal human coronavirus (HCoV)-OC43 that frequently infects this age group.

Clinical manifestations of human coronavirus (HCoV)-related diseases are mostly related to the respiratory system, although secondary complications such as headache, anosmia, ageusia, and myalgia have been reported. HCoV infection and replication in chemosensory cells associated with ageusia and anosmia is poorly understood. Here, we characterized HCoV-OC43 and SARS-CoV-2 infection in two types of chemosensory cells, olfactory and taste cells, with their unique molecular and histological characteristics. We first assessed HCoV-OC43 infection in in vitro cultured human olfactory epithelial cell
