Membrane Proteins and Proteomics of Cronobacter sakazakii Cells: Reliable Method for Identification and Subcellular Localization.

Members of the genus Cronobacter are responsible for severe infections in infants and immunosuppressed individuals. Although several virulence factors have been described, many proteins involved in the pathogenesis of such infections have not yet been mapped. This study is the first to fractionate Cronobacter sakazakii cells into outer membrane, inner membrane, periplasmic, and cytosolic fractions as the basis for improved proteome mapping. A novel method was designed to prepare the fractionated samples for protein identification. The identification was performed via one-dimensional electropho

Exogenous α-linolenic acid acts as a negative regulator of Cronobacter sakazakii structural flagellar proteins.

AIM: This work aims to report on the mechanism of action by which Cronobacter sakazakii virulence is impacted by α-linolenic acid (ALA), a C18:3 fatty acid. METHODS AND RESULTS: To elucidate this, two concentrations of ALA (250 and 1000 µmol L-1) were added exogenously to C. sakazakii 29 544 in tryptic soy broth. Quantitative proteomic analysis using label-free mass spectrometry showed significantly different proteomic profiles of treated and control C. sakazakii samples. Across both ALA treatments, a total of 11 flagellar proteins were identified as reduced in abundance and showed a dose-dep

Bacteroides fragilis ameliorates Cronobacter malonaticus lipopolysaccharide-induced pathological injury through modulation of the intestinal microbiota.

Cronobacter has attracted considerable attention due to its association with meningitis and necrotizing enterocolitis (NEC) in newborns. Generally, lipopolysaccharide (LPS) facilitates bacterial translocation along with inflammatory responses as an endotoxin; however, the pathogenicity of Cronobacter LPS and the strategies to alleviate the toxicity were largely unknown. In this study, inflammatory responses were stimulated by intraperitoneal injection of Cronobacter malonaticus LPS into Sprague-Dawley young rats. Simultaneously, Bacteroides fragilis NCTC9343 were continuously fed through gavag

Crystal structures of YeiE from Cronobacter sakazakii and the role of sulfite tolerance in gram-negative bacteria.

SignificanceYeiE has been identified as a master virulence factor of Cronobacter sakazakii. In this study, we determined the crystal structures of the regulatory domain of YeiE in complex with its physiological ligand sulfite ion (SO32-). The structure provides the basis for the molecular mechanisms for sulfite sensing and the ligand-dependent conformational changes of the regulatory domain. The genes under the control of YeiE in response to sulfite were investigated to reveal the functional roles of YeiE in the sulfite tolerance of the bacteria. We propose the molecular mechanism underlying t

Enteric-colonized Cronobacter spp. can disrupt the central nervous system without translocation across the host barriers.

Cronobacter sakazakii is an opportunistic foodborne pathogen known to cause severe neonatal infections. However, its subclinical effects on human health remain largely unexplored. Here, we report that intestinal colonization by Cronobacter is potentially correlated with human neurobehavioral alterations in the absence of bacterial translocation across host barriers. Analysis of fecal microbiota from the children in GUSTO cohort revealed that Cronobacter abundance was significantly and positively correlated with their emotional reactivity and affective problem scores, suggesting an association

Highly efficient production of 2,3-butanediol from xylose and glucose by newly isolated thermotolerant Cronobacter sakazakii.

BACKGROUND: 2,3-Butanediol (2,3-BD), a valuable compound used for chemicals, cosmetics, pesticides and pharmaceuticals, has been produced by various microbes. However, no high-temperature fermentation of the compound at high productivity has been reported. METHODS: Thermotolerant xylose-utilizing microbes were isolated from 6 different districts in Laos and screened for a low accumulation of xylitol in a xylose medium at 37 ˚C. One isolate was found to produce 2,3-BD and identified by 16S rDNA sequencing. The 2,3-BD fermentation capacity was investigated at different temperatures using xylose

Detection of Cronobacter species in powdered infant formula using immunoliposome-based immunomagnetic concentration and separation assay.

Cronobacter species are foodborne pathogens that can affect the human central nervous system. Survivors of Cronobacter infections often suffer from severe neurological impairments, including hydrocephalus, quadriplegia, and developmental delays in all ages, especially in infants and the immunocompromised. Moreover, Cronobacter species pose a high risk in powdered infant formula (PIF) because PIF is a major source of nutrition for infants worldwide. To develop a rapid and sensitive detection method for Cronobacter species in PIF, immunoliposomes and immunomagnetic nanoparticles were synthesized

Opposing regulation of Gig2 by RecA and GlrK controls redox-linked desiccation tolerance in Cronobacter sakazakii.

Cronobacter sakazakii is a desiccation-tolerant foodborne pathogen that poses a serious threat to neonatal health through contamination of powdered infant formula. Despite its clinical relevance and exceptional persistence in dry environments, the molecular basis of its desiccation tolerance remains poorly defined. Here, we systematically investigated the genetic and physiological determinants of C. sakazakii survival under desiccation stress. A functional screen of gene knockout strains revealed intricate relationships between surface hydrophilicity and desiccation tolerance, suggesting the i

Uncovering the pathogenic mechanisms of Cronobacter turicensis: A dual transcriptomics study using a zebrafish larvae model.

PURPOSE: Cronobacter (C.) is an emerging opportunistic pathogen representing a significant cause of mortality in neonatal patients with bacteremia and meningitis. The pathobiology of Cronobacter mediated meningitis has primarily been investigated using in vitro models. In this study, we used zebrafish to investigate in vivo the infection strategy of the sepsis/meningitis-causing strain C. turicensis z3032 (LMG 23827T) and the immune response of zebrafish larvae after central nervous system (CNS) invasion. Global gene expression profiles of both organisms were analyzed using RNA-Seq. METHODS:
