Bioremediation of copper in sediments from a constructed wetland ex situ with the novel bacterium Cupriavidus basilensis SRS.

The H-02 constructed wetland was designed to remove metals (primarily copper and zinc) to treat building process water and storm water runoff from multiple sources associated with the Tritium Facility at the DOE-Savannah River Site, Aiken, SC. The concentration of Cu and Zn in the sediments has increased over the lifetime of the wetland and is a concern. A bioremediation option was investigated at the laboratory scale utilizing a newly isolated bacterium of the copper metabolizing genus Cupriavidus isolated from Tim's Branch Creek, a second-order stream that eventually serves as a tributary to

Biosynthesis of polyhydroxyalkanoate (PHA) by Cupriavidus necator KCTC 2649 using coconut (Cocos nucifera L.) water as carbon source.

Polyhydroxyalkanoates (PHA) are biodegradable polymers synthesized by microorganisms under nutrient-deficient conditions. Coconut water, an abundant yet often discarded agro-industrial by-product, is a cost-effective and environmentally friendly substrate rich in carbohydrates such as glucose, fructose, and sucrose. PHAs are typically produced using costly and unsustainable carbon sources. This study investigated the effects of media sterilization methods and carbon-to-nitrogen (C/N) and carbon-to-phosphorus (C/P) ratios on PHA production by Cupriavidus necator KCTC 2649 using coconut water. Isopropanol production from carbon dioxide by Cupriavidus necator using a zero-gap cell with culture broth as catholyte.

Microbial CO2 fixation is a promising route for sustainable chemical production. The facultative chemolithoautotroph Cupriavidus necator can convert CO2 into value-added compounds using H2 and O2. Here, we developed a bioelectrochemical system (BES) for isopropanol production that integrates CO2 fixation with in situ H2 generation via a zero-gap proton exchange membrane electrolyzer. Using the culture broth as catholyte, a pentlandite-based cathode supplied dissolved H2 directly to engineered C. necator, eliminating the need for pressurized gas. Real-time monitoring of dissolved and off-gas co

Potential of a novel facultative anaerobic denitrifying Cupriavidus sp. W12 to remove fluoride and calcium through calcium bioprecipitation.

This study focused on a novel denitrifying Cupriavidus sp. W12, which can perform microbial induced calcium precipitation (MICP) to remove fluoride (F-) under aerobic and anaerobic conditions. Under anaerobic condition, the removal ratios of F-, calcium (Ca2+), and nitrate (NO3--N) reached 87.52%, 65.03%, and 96.06%, respectively, which were higher than that under aerobic condition (50.17%, 88.21%, and 67.33%, respectively). Higher pH of 8.26 was obtained after 120 h of the strain W12 growth under anaerobic condition than that under aerobic condition (7.77). The F- removal ratio of 98.20% was

Interactions of core-shell quantum dots with metal resistant bacterium Cupriavidus metallidurans: consequences for Cu and Pb removal.

In the present study we address the interactions of carboxyl-CdSe/ZnS core/shell quantum dots (QDs), as a model of water dispersible engineered nanoparticles, and metal resistant bacteria Cupriavidus metallidurans, largely used in metal decontamination. The results demonstrate that QDs with average hydrodynamic size of 12.9 nm adhere to C. metallidurans. The percentage of bacterial cells displaying QD-fluorescence increased proportionally with contact time and QD concentration in bacterial medium demonstrating the association of QDs with the metal resistant bacteria. No evidence of QD internal

Value-added utilisation of industrial by-products from bioenergy processes for growth of the PHB synthesising bacterium Cupriavidus necator.

The utilisation of low-value by-products derived from the production of bioenergy as nutrient media for microbial growth was investigated using the polyhydroxybutyrate (PHB) synthesising bacterium Cupriavidus necator as a case study. The potentially suitable by-products crude glycerol (derived from biodiesel production as carbon source) and digestate (resulting from biogas production as source for nitrogen and other nutrients) contain significant amounts of nutrients but also dissolved and solid impurities. In order to minimise negative effects such as clogging in bioreactor infrastructure and

Harnessing the potential of Cupriavidus necator for CO2 capture from alcoholic fermentation and its bioconversion into poly(3-hydroxybutyrate).

The fermentation process in alcoholic beverage production converts sugars into ethanol and CO2, releasing significant amounts of greenhouse gases. Here, Cupriavidus necator DSM 545 was grown autotrophically using gas derived from alcoholic fermentation, using a fed-batch bottle system. Nutrient starvation was applied to induce intracellular accumulation of poly(3-hydroxybutyrate) (PHB), a bioplastic polymer, for bioconversion of CO2-rich waste gas into PHB. Grape marc, another by-product of wine production, was evaluated as a low-cost carbon source for the heterotrophic growth of C. necator, w

Conceptual Approach for Aerobic Autotrophic Gas Cultivation in Shake Flasks: Overcoming the Inhibitory Effects of Oxygen in Cupriavidus necator.

This study conceptualizes the design of a small-scale system (250 mL-1 L) for the autotrophic cultivation of hydrogen-oxidizing bacteria, such as the representative strain Cupriavidus necator. The research aimed to systematically investigate the impact of bottle volume and gas composition, particularly oxygen concentration, on the growth and performance of C. necator during autotrophic cultivations. To this end, customized, pressure-tight, baffled glass bottles of various sizes (250, 500, and 1000 mL) and gas mixtures with varying oxygen concentrations (4%, 8%, and 12% v/v) were tested. Growth

Synthesis of nickel-iron hydrogenase in Cupriavidus metallidurans is controlled by metal-dependent silencing and un-silencing of genomic islands.

Cupriavidus metallidurans CH34 is able to grow autotrophically as a hydrogen-oxidizing bacterium and produces nickel-dependent hydrogenases, even under heterotrophic conditions. Loss of its two native plasmids resulted in inability of the resulting strain AE104 to synthesize the hydrogenases and to grow autotrophically in phosphate-poor, Tris-buffered mineral salts medium (TMM). Three of eleven previously identified catabolic genomic islands (CMGIs; Van Houdt et al., 2009), two of which harbor the genes for the membrane-bound (CMGI-2) and the soluble hydrogenase (CMGI-3), were silenced in stra

Valorization of prosecco wine lees for sustainable polyhydroxyalkanoates production by Cupriavidus necator DSM 545 and Hydrogenophaga pseudoflava DSM 1034.

Nowadays, in the agricultural and agro-industrial sectors, there is increasing attention on the wine industry. The environmental impact of wine production, particularly in terms of residues generation, is a growing concern. Among generated residues, wine lees, rich in organic matter, phenols compounds, and with low pH, pose environmental challenges due to their disposal requirements. Despite their usage for biogas production and extraction of compounds, such as antioxidants, tartaric acid and ethanol, their potential in bioplastic production, specifically polyhydroxyalkanoates (PHAs), remains

Comparative metabolomic profiling of Cupriavidus necator B-4383 revealed production of cupriachelin siderophores, one with activity against Cryptococcus neoformans.

Cupriavidus necator H16 is known to be a rich source of linear lipopeptide siderophores when grown under iron-depleted conditions; prior literature termed these compounds cupriachelins. These small molecules bear β-hydroxyaspartate moieties that contribute to a photoreduction of iron when bound as ferric cupriachelin. Here, we present structural assignment of cupriachelins from C. necator B-4383 grown under iron limitation. The characterization of B-4383 cupriachelins is based on MS/MS fragmentation analysis, which was confirmed by 1D- and 2D-NMR for the most abundant analog (1). The cupriache

Effective recovery of poly-β-hydroxybutyrate (PHB) biopolymer from Cupriavidus necator using a novel and environmentally friendly solvent system.

This work demonstrates a significant advance in bioprocessing for a high-melting lipid polymer. A novel and environmental friendly solvent mixture, acetone/ethanol/propylene carbonate (A/E/P, 1:1:1 v/v/v) was identified for extracting poly-hydroxybutyrate (PHB), a high-value biopolymer, from Cupriavidus necator. A set of solubility curves of PHB in various solvents was established. PHB recovery of 85% and purity of 92% were obtained from defatted dry biomass (DDB) using A/E/P. This solvent mixture is compatible with water, and from non-defatted wet biomass, PHB recovery of 83% and purity of 90

Leguminous plants nodulated by selected strains of Cupriavidus necator grow in heavy metal contaminated soils amended with calcium silicate.

Increasing concern regarding mining area environmental contamination with heavy metals has resulted in an emphasis of current research on phytoremediation. The aim of the present study was to assess the efficiency of symbiotic Cupriavidus necator strains on different leguminous plants in soil contaminated with heavy metals following the application of inorganic materials. The application of limestone and calcium silicate induced a significant increase in soil pH, with reductions in zinc and cadmium availability of 99 and 94 %, respectively. In addition, improved nodulation of Mimosa caesalpini

Cupriavidus sp. strain Ni-2 resistant to high concentration of nickel and its genes responsible for the tolerance by genome comparison.

The widespread use of metals influenced many researchers to examine the relationship between heavy metal toxicity and bacterial resistance. In this study, we have inoculated heavy metal-contaminated soil from Janghang region of South Korea in the nickel-containing media (20 mM Ni2+) for the enrichment. Among dozens of the colonies acquired from the several transfers and serial dilutions with the same concentrations of Ni, the strain Ni-2 was chosen for further studies. The isolates were identified for their phylogenetic affiliations using 16S rRNA gene analysis. The strain Ni-2 was close to Cu

Sustainable production of β-caryophyllene from fructose and carbon dioxide in Cupriavidus necator H16 through pathway enhancement and global regulatory optimization.

Cupriavidus necator H16, a hydrogen-oxidizing bacterium, is capable of chemolithoautotrophic growth and biosynthesis using carbon dioxide (CO2) as the sole carbon source. This organism has been engineered to convert CO2 into various value-added compounds; however, its application in autotrophic terpenoid biosynthesis remains limited due to low metabolic efficiency and poorly understood regulatory mechanisms. In this study, we developed an efficient C. necator H16 cell factory to autotrophically produce β-caryophyllene, a sesquiterpene with significant potential in the food, cosmetic, and biofu
