Palladium-Catalyzed Defluorinative Cyanation of gem-Difluorinated Cyclopropanes for the Synthesis of 2-Fluoroallylic Nitriles.

We report a palladium-catalyzed ring-opening defluorinative cyanation of gem-difluorinated cyclopropanes (gem-DFCPs) with Zn(CN)2 as the cyanide source. The reaction proceeds smoothly via a sequence of regioselective C-C activation, C-F cleavage, and C-CN formation, providing precise access to synthetically appealing 2-fluoroallylic nitriles with features of a broad substrate scope, extraordinary functional group compatibility, and excellent Z-selectivity. The late-stage modification of gem-DFCPs derived from bioactive complex molecules and medicinally relevant scaffolds enhanced the practical

Enantioselective cyanation of propargylic C-H bonds via cooperative photoredox and copper catalysis.

Herein, we report an enantioselective cyanation of propargylic C-H bonds by combining photoredox catalysis with a copper-catalyzed radical relay in which the propargylic radical was generated by an intramolecular 1,5-HAT process. This reaction provides easy access to optically pure propargyl nitrile compounds under mild conditions.

Controllable cyanation of carbon-hydrogen bonds by zeolite crystals over manganese oxide catalyst.

The synthesis of organic nitriles without using toxic cyanides is in great demand but challenging to make. Here we report an environmentally benign and cost-efficient synthesis of nitriles from the direct oxidative cyanation of primary carbon-hydrogen bonds with easily available molecular oxygen and urea. The key to this success is to design and synthesize manganese oxide catalysts fixed inside zeolite crystals, forming a manganese oxide catalyst with zeolite sheath (MnOx@S-1), which exhibits high selectivity for producing nitriles by efficiently facilitating the oxidative cyanation reaction a

Chelation-assisted palladium-catalyzed direct cyanation of 2-arylpyridine C-H bonds.

A chelation-assisted palladium-catalyzed ortho-cyanation of the sp2 C-H bond by CuCN provided aromatic nitriles in moderate to good yields. Notably, the reaction could be conducted on a 10 mmol scale. The key intermediate of the natural product of Menispermum dauricum DC was concisely synthesized by the procedure. This new approach represents an exceedingly practical method for the synthesis of aromatic nitriles and offers an attractive alternative to the traditional Sandmeyer reaction.

Reduction of ferrate(VI) and oxidation of cyanate in a Fe(VI)-TiO2-UV-NCO- system.

The aqueous photocatalytic degradation of cyanate (NCO(-)), which is a long-lived neurotoxin formed during the remediation of cyanide in industrial waste streams, was studied in the ferrate(VI)-UV-TiO2-NCO(-) system. Kinetics measurements of the photocatalytic reduction of ferrate(VI) were carried out as a function of [NCO(-)], [ferrate(VI)], [O(2)], light intensity (I(o)), and amount of TiO2 in suspensions at pH 9.0. The photocatalytic reduction rate of ferrate(VI) in the studied system can be expressed as -d[Fe(VI)]/dt=kI(o)(0.5) [NCO(-)] [TiO2]. The rate of photocatalytic oxidation of cyana

The effects of cyanate in vitro on red blood cell metabolism and function in sickle cell anemia.

Cyanate, which is in equilibrium with urea, combines with the alpha-amino group of the aminoterminal valine of hemoglobin in an irreversible, specific carbamylation reaction. Partial carbamylation (0.72 residues/hemoglobin tetramer) as determined by cyanate-(14)C incorporation or hydantoin analysis diminishes the in vitro sickling phenomenon. Since cyanate may react not only with hemoglobin but also with functional groups of other red blood cell proteins, the in vitro effect of cyanate was studied on sickle cells. Cells were incubated with 10 mM KCl (control) or 10 mM KNCO (carbamylated) for 1

Decoupling Radical Generation and Copper Catalysis for the Remote C-H Cyanation of Unfunctionalized Sulfonamides.

A decoupled photoredox/copper dual catalytic system for the remote C(sp3)-H cyanation of aliphatic sulfonamides is described. The strategy circumvents the need for substrate prefunctionalization, a limitation of previous "coupled" systems. An acridinium photocatalyst generates a nitrogen radical from a free N-H bond, while an independent copper cycle, sustained by an external oxidant, forges the C-CN bond with high δ-selectivity.

Copper-Catalyzed Asymmetric Tertiary Radical Cyanation for the Synthesis of Chiral Tetrasubstituted Monofluoroacyl Nitriles.

The construction of chiral tetrasubstituted α-fluoro-α-cyano carbonyl compounds remains a key challenge in synthetic organic chemistry because of their popularity in multiple disciplines. In this paper, we report the copper-catalyzed asymmetric fluorinated tertiary radical cyanation reaction of cyclic α-iodo-α-fluoroindanones with TMSCN to achieve chiral nitriles with carbon-fluorine quaternary stereogenic centers. Thus, an array of optically active tetrasubstituted monofluoroacyl nitriles were synthesized with high reaction efficiency and excellent enantioselectivities (up to 91% yield, 99% e

Electrochemical Aminoxyl-Mediated α-Cyanation of Secondary Piperidines for Pharmaceutical Building Block Diversification.

Secondary piperidines are ideal pharmaceutical building blocks owing to the prevalence of piperidines in commercial drugs. Here, we report an electrochemical method for cyanation of the heterocycle adjacent to nitrogen without requiring protection or substitution of the N-H bond. The reaction utilizes ABNO (9-azabicyclononane N-oxyl) as a catalytic mediator. Electrochemical oxidation of ABNO generates the corresponding oxoammonium species, which promotes dehydrogenation of the 2° piperidine to the cyclic imine, followed by addition of cyanide. The low-potential, mediated electrolysis process i

Visible-Light-Mediated Alkenylation, Allylation, and Cyanation of Potassium Alkyltrifluoroborates with Organic Photoredox Catalysts.

Iridium- and ruthenium-free approaches to protected allylic amines and alkyl nitriles under photoredox conditions are reported. An inexpensive organic dye, eosin Y, catalyzes coupling of Boc-protected potassium α-aminomethyltrifluoroborates with a variety of substituted alkenyl sulfones through an α-aminomethyl radical addition-elimination pathway. Allylic and homoallylic amines were formed in moderate yields with high E/Z selectivity. The mechanistic approach was extended using tosyl cyanide as a radical trap, enabling the conversion of alkyltrifluoroborates to nitriles via a Fukuzumi acridin

Electrophilic Cyanate As a Recognition Motif for Reactive Sulfur Species: Selective Fluorescence Detection of H2S.

An ESIPT-based fluorescent dye, 3-hydroxyflavone, is chemically masked with an electrophilic cyanate motif in order to construct a fluorescent probe for cellular sulfur species. This novel probe structure, displays an extremely fast, highly sensitive and selective "turn-on" type fluorescent response toward H2S. We have also documented its utility for imaging of H2S in the living cells.

Reduction of thiocyanate, cyanate, and carbon disulfide by nitrogenase: kinetic characterization and EPR spectroscopic analysis.

Nitrogenase catalyzes the reduction of N2, protons, and a number of alternative substrates that contain C-C, C-N, N-N, and N-O double and triple bonds. Recently it has been shown that nitrogenase also reduces the C==S bond of COS and the C==O bond of CO2. The current work demonstrates that the COS analogs SCN-, CS2, and OCNH are novel substrates for nitrogenase and that the reduction of these substrates produces changes in the electron paramagnetic resonance (EPR) spectrum of nitrogenase, providing insight into the mechanism of substrate reduction by nitrogenase. CH4, HCN, H2S, and NH4+ were d

Transmetallation by Halogen-Cyanide Metathesis in Water: Application to Cyanide Ion-Free Palladium Catalyzed Cyanation.

Aryl nitriles are important synthons in organic chemistry, yet conventional cyanation methods mostly generate free cyanide ions, creating acute safety hazards and wastewater contamination issues. We report a Pd-catalyzed cyanide ion-free cyanation of aryl halides featuring a transmetallation potentially via halogen-cyanide (CN-) metathesis using potassium ferrocyanide as a bound CN- reservoir. Mechanistic studies, CN--sensing strip tests, and IR spectroscopy confirm that no free CN⁻ is released at any stage, eliminating toxicity and downstream CN- liabilities. Reductive cyanation of organic chlorides using CO2 and NH3 via Triphos-Ni(I) species.

Cyano-containing compounds constitute important pharmaceuticals, agrochemicals and organic materials. Traditional cyanation methods often rely on the use of toxic metal cyanides which have serious disposal, storage and transportation issues. Therefore, there is an increasing need to develop general and efficient catalytic methods for cyanide-free production of nitriles. Here we report the reductive cyanation of organic chlorides using CO2/NH3 as the electrophilic CN source. The use of tridentate phosphine ligand Triphos allows for the nickel-catalyzed cyanation of a broad array of aryl and ali

Myeloperoxidase-catalyzed oxidation of cyanide to cyanate: A potential carbamylation route involved in the formation of atherosclerotic plaques?

Protein carbamylation by cyanate is a post-translational modification associated with several (patho)physiological conditions, including cardiovascular disorders. However, the biochemical pathways leading to protein carbamylation are incompletely characterized. This work demonstrates that the heme protein myeloperoxidase (MPO), which is secreted at high concentrations at inflammatory sites from stimulated neutrophils and monocytes, is able to catalyze the two-electron oxidation of cyanide to cyanate and promote the carbamylation of taurine, lysine, and low-density lipoproteins. We probed the r

Cu-catalyzed debrominative cyanation of gem-dibromoolefins: a facile access to α,β-unsaturated nitriles.

An efficient catalytic route for the synthesis of α,β-unsaturated nitriles from easily accessible gem-dibromoolefins has been developed. The method utilized inexpensive reagents such as Cu2O as a catalyst, L-proline as a ligand and NaCN as a cyanide source to afford α,β-unsaturated nitriles in high yields (62-86%). A deuterium exchange study has shown that one of the bromide atoms of gem-dibromoolefins exchanges with cyanide while the other with a deuterium atom.

The influence of oxygen on [NiFe]-hydrogenase cofactor biosynthesis and how ligation of carbon monoxide precedes cyanation.

The class of [NiFe]-hydrogenases is characterized by a bimetallic cofactor comprising low-spin nickel and iron ions, the latter of which is modified with a single carbon monoxide (CO) and two cyanide (CN-) molecules. Generation of these ligands in vivo requires a complex maturation apparatus in which the HypC-HypD complex acts as a 'construction site' for the Fe-(CN)2CO portion of the cofactor. The order of addition of the CO and CN- ligands determines the ultimate structure and catalytic efficiency of the cofactor; however much debate surrounds the succession of events. Here, we present an FT

Simultaneous and independent versus antagonistic inhibition of muscle carbonic anhydrase (CA III) by acetazolamide and cyanate.

The inhibition by cyanate and acetazolamide of pig muscle carbonic anhydrase III (CA III) CO2 hydratase activity was studied in order to explore mechanistic features possibly unique to the muscle isoenzyme. The turnover number for CO2 hydration was found to be 6000 sec-1 with a Km of 83 mM for CO2. Cyanate inhibition (Ki, 3 microM) and acetazolamide inhibition (Ki, 44 microM) were both found to be noncompetitive with respect to CO2. Significantly, acetazolamide and cyanate displayed non-exclusive binding to pig muscle carbonic anhydrase. The similarity of mode and degree of inhibition of muscl
