Engineering an advanced multicomponent pentacyclic triterpenoid nanoplatform for synergistic co-delivery of cyclin-dependent kinase 4/6 inhibitors to enhance cancer chemoimmunotherapy.

Immune checkpoint inhibitors such as monoclonal antibodies anti-PD-L1 hold huge promise for triple-negative breast cancer (TNBC) therapy but face limited immune efficacy due to the cold tumor nature and suppressive tumor immune microenvironment (TIME). To address these challenges, we engineered an innovative multi-component natural triterpenoid (PT)-co-assembled nano-platform that simultaneously serves as immunogenic cell death (ICD)-inducing chemotherapeutics for synergistic delivery of palbociclib (Pal), a cyclin-dependent kinase 4/6 (CDK4/6) inhibitor with distinct immunomodulatory effects

Cyclin-Dependent Kinase 4/6 Inhibitor Outcomes in Patients With Advanced Breast Cancer Carrying Germline Pathogenic Variants in DNA Repair-Related Genes.

PURPOSE: In recent years, unprecedented benefits have been observed with the development of cyclin-dependent kinase (CDK) 4 and 6 inhibitors for the treatment of hormone receptor-positive/human epidermal growth factor receptor 2-negative metastatic breast cancer. However, there is scarce evidence of their value in specific populations, such as patients carrying germline pathogenic variants in DNA repair-related genes. PATIENTS AND METHODS: We retrospectively studied the efficacy of CDK 4/6 inhibitors plus endocrine therapy in patients with hormone receptor-positive/human epidermal growth fact

Cyclin dependent kinase 4/6 inhibitor palbociclib synergizes with BCL2 inhibitor venetoclax in experimental models of mantle cell lymphoma without RB1 deletion.

BACKGROUND: Mantle cell lymphoma (MCL) is a chronically relapsing malignancy with deregulated cell cycle progression. We analyzed efficacy, mode of action, and predictive markers of susceptibility to palbociclib, an approved CDK 4/6 inhibitor, and its combination with venetoclax, a BCL2 inhibitor. METHODS: A panel of nine MCL cell lines were used for in vitro experiments. Four patient derived xenografts (PDX) obtained from patients with chemotherapy and ibrutinib-refractory MCL were used for in vivo proof-of-concept studies. Changes of the mitochondrial membrane potential, energy-metabolic pa

Cyclin-Dependent Kinase 4/6 Inhibitors and Dermatologic Adverse Events: Results from the EADV Task Force "Dermatology for Cancer Patients" International Study.

BACKGROUND: The introduction of cyclin-dependent kinase inhibitors (CDK4/6i) was a great advance in therapeutics for patients with estrogen receptor+/human epidermal growth factor receptor (HER2) locally advanced and metastatic breast cancer. Despite the increasing use of these agents, their adverse drug-related events have not yet been fully characterized. We describe the spectrum of cutaneous adverse reactions occurring in advanced breast cancer patients treated with cyclin-dependent kinase inhibitors, analyzing types, severity, time to onset, and possible treatment outcomes. METHODS: We pe

Deletions and rearrangements of cyclin-dependent kinase 4 inhibitor gene p16 are associated with poor prognosis in B cell non-Hodgkin's lymphomas.

In the present study we examined by Southern blot analysis the presence of deletions and rearrangements of the p16 tumor-suppressor gene in B cell non-Hodgkin's lymphomas (NHLs) in order to determine whether or not these changes can be related to a particular histological subtype and the different clinico-biological and prognostic characteristics of the disease. 103 untreated patients were enrolled in the study. Seven cases displayed alterations in the p16 gene: four cases with homozygous deletions and three with gene rearrangement. The presence of these abnormalities did not correlate with an

Evaluation of cyclin-dependent kinase 4 immunolabelling and a germline p.G201S variant as predictors of clinical outcome in canine mammary tumours: a preliminary study.

Cyclin-dependent kinase 4 (CDK4), a regulator of the G1/S transition, is frequently dysregulated in cancer. Although extensively studied in human breast cancer, its significance in canine mammary tumours remains poorly understood. This preliminary study investigated the prognostic value of CDK4 immunohistochemistry (IHC) and of sequence variation in the CDK4 kinase-domain segment surrounding residue 201. Thirty-three prospectively enrolled female dogs with tumours affecting a single mammary chain underwent unilateral radical mastectomy. An 89-bp CDK4 segment was sequenced and paired with CDK4

An immunohistochemical based assessment of murine double minute 2 (MDM2) and cyclin-dependent kinase 4 (CDK4) in jaw osteosarcoma and benign osseous lesions.

UNLABELLED: The diagnosis of primary bone tumours of the jaw often presents as a diagnostic dilemma. Overlap in clinical, radiological, and histological features between benign lesions like juvenile trabecular ossifying fibroma (JTOF); osteoblastoma (OB) and malignant tumours like low-grade osteosarcoma (LGOS) makes the diagnosis challenging. While osteosarcoma is a malignant tumour with poor prognosis requiring aggressive treatment, benign lesions have generally favourable outcome. This study aimed to assess the immunohistochemical expression of murine double minute 2 (MDM2) and Cyclin-depend

Association between body composition parameters and treatment-related toxicities in patients with metastatic breast cancer receiving cyclin-dependent kinase 4 and 6 inhibitors.

BACKGROUND & AIMS: Nowadays, limited data or no data are available on body composition changes and the development of treatment-related toxicities in metastatic breast cancer (MBC) patients treated with innovative and promising anticancer therapies, including cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitors. Therefore, we evaluated in MBC treated with CDK4/6 inhibitors changes in adiposity and muscularity before and after treatment and whether the changes in these parameters were associated with toxicities, dose reduction or treatment discontinuation. METHODS: We considered ER+/HER2- MBC p

The Effects of Anlotinib Combined with Chemotherapy following Progression on Cyclin-Dependent Kinase 4/6 Inhibitor in Hormone Receptor-Positive Metastatic Breast Cancer.

PURPOSE: Endocrine therapy combined with cyclin-dependent kinase (CDK) 4/6 inhibitors (CDK4/6i) is the preferred treatment for hormone receptor-positive (HR+)/human epidermal growth factor receptor 2-negative (HER2-) metastatic breast cancer (MBC). However, there are currently no recommendations for therapeutic strategies after progression on CDK4/6i-based treatment. This study aimed to examine the efficacy and safety of anlotinib plus chemotherapy in HR+/HER2- MBC after progression on CDK4/6 inhibitors. METHODS: We collected data from 32 patients with HR+/HER2- MBC treated with anlotinib plu

Detection of homozygous deletions of the cyclin-dependent kinase 4 inhibitor (p16) gene in acute lymphoblastic leukemia and association with adverse prognostic features.

A recently described putative tumor suppressor gene, the cyclin-dependent kinase 4 inhibitor (p16), has been shown to be altered by deletions and/or point mutations in various human cancers. To assess the incidence and clinico-biologic correlations of p16 homozygous deletion in hemopoietic tumors, we studied a panel of 244 DNA samples representative of distinct acute (99 cases) and chronic (57 cases) leukemia subtypes, myelodysplastic (22 cases) and myeloproliferative (15 cases) syndromes, and lymphomas (51 cases). A 361-bp probe complementary to the p16 exon 2 gene sequences was generated by

Melatonin Enhances Aquaporin 4 and Alpha-Syntrophin Interaction by Inhibiting Cyclin-Dependent Kinase 5 Activity to Preserve Glymphatic Function in Neonatal Hypoxic-Ischemic Encephalopathy.

The glymphatic system is a critical waste clearance system in the brain, playing an essential role in maintaining homeostasis within the central nervous system. Aquaporin 4 (AQP4), an indispensable component of the glymphatic system, is vital for ensuring the proper function of this system. Melatonin has been proven to be protective in treating hypoxic-ischemic encephalopathy (HIE). The aim of this study was to examine if alterations occur in the glymphatic system function in the brain of HIE model rats, and to determine whether melatonin can enhance the function of the glymphatic system by re

Cyclin-Dependent Kinase 4/6 Inhibitor in Hormone Receptor-Positive, Human Epidermal Growth Factor Receptor 2-Negative Breast Cancer: Global Challenges and Tibremciclib Perspective.

PURPOSE: Hormone receptor-positive (HR+), human epidermal growth factor receptor 2 (HER2)-negative advanced breast cancer represents a substantial proportion of breast cancer cases globally. Cyclin-dependent kinase 4/6 (CDK4/6) inhibitors combined with endocrine therapy are established first-line treatments in high-income settings, improving progression-free survival (PFS) and quality of life. However, access remains limited in low- and middle-income countries (LMICs) because of cost, infrastructure, and policy barriers. METHODS: We critically evaluated global challenges in CDK4/6 inhibitor i

Vitiligo-Like Depigmentation Induced by Cyclin-Dependent Kinase 4/6 Inhibitors in the Treatment of Metastatic Breast Cancer: A Case Report and Literature Review.

Cyclin-dependent kinase (CDK) 4/6 inhibitors are commonly used in the first-line treatment of hormone receptor-positive, human epidermal growth factor receptor 2-negative metastatic breast cancer, and more recently in the adjuvant setting as well. Whilst cutaneous toxicities are well documented, vitiligo-like depigmentation (VLD) is rarely reported in existing literature. Such adverse effects impair tolerability and compromise patients' quality of life. Here we describe a case from our centre of a 72-year-old Chinese woman with recurrent, metastatic hormone-positive breast cancer who developed

Bromodomain-containing-protein-4 and cyclin-dependent-kinase-9 inhibitors interact synergistically in vitro and combined treatment reduces post-traumatic osteoarthritis severity in mice.

OBJECTIVE: Joint injury rapidly induces expression of primary response genes (PRGs), which activate a cascade of secondary genes that destroy joint tissues and initiate post-traumatic osteoarthritis (PTOA). Bromodomain-containing-protein-4 (Brd4) and cyclin-dependent-kinase-9 (CDK9) cooperatively control the rate-limiting step of PRG transactivation, including pro-inflammatory genes. This study investigated whether Brd4 and CDK9 inhibitors suppress inflammation and prevent PTOA development in vitro and in a mouse PTOA model. METHODS: The effects of Brd4 and CDK9 inhibitors (JQ1 and Flavopirid

Array-comparative genomic hybridization of central chondrosarcoma: identification of ribosomal protein S6 and cyclin-dependent kinase 4 as candidate target genes for genomic aberrations.

BACKGROUND: Enchondromas are benign lesions that can occur as solitary tumors or multiple tumors (Ollier disease) and may be precursors of central chondrosarcomas. Recurrent chondrosarcomas can be of a higher grade compared with primary tumors, suggesting possible progression. METHODS: Genome-wide array-comparative genomic hybridization (CGH) was used to investigate copy number changes in enchondromas and central chondrosarcomas to elucidate both primary genetic events and the events related to tumor progression. Analyses of variance, Student t tests, and hierarchical clustering were used for
