Cytarabine intravenous constant rate infusion (IV CRI) toxicity in cats with lymphoma has not been well documented. We aimed to assess toxicities following cytarabine IV CRI as a single-agent or within a multiagent protocol in cats with lymphoma. Medical records of cats with lymphoma treated with cytarabine IV CRI, with documented dose, route, duration and at least one follow-up visit between 1990 and 2024, were retrospectively reviewed. Adverse events (AEs) were retrospectively graded according to VCOG-CTCAE v2. Thirty-eight cats were included, and 70 doses were evaluated for AEs. Median cyta

INTRODUCTION: Cytarabine is one of the main chemotherapy drugs used to treat hematological malignancies. Neurotoxicity is an increasingly recognized adverse effect of high-dose cytarabine therapy and occurs in 10% to 25% of patients. There is very little experience with cytarabine retreatment following neurotoxicity.
CASE REPORT: We report a case of a man who was diagnosed with chemotherapy-induced cerebellar syndrome. He was treated with high doses of cytarabine.
MANAGEMENT AND OUTCOME: The patient was successfully retreated at full dose after experiencing cytarabine-induced cerebellar toxi

Most cases of transient abnormal myelopoiesis (TAM) in neonates with Down syndrome (DS) resolve spontaneously; however, DS-TAM neonates with hydrops fetalis (HF) show poor clinical outcomes. We report three infants with DS-TAM and HF who were treated with exchange transfusion (ET) followed by low-dose cytarabine (LD-CA). All of them survived without developing liver failure, acute leukemia, or other serious adverse events. Our results suggest that this combination treatment with ET and LD-CA would be safe, tolerable and effective as an novel approach for DS-TAM patients with HF.

Conventionally, the diagnosis of neuropathology in a subject requires the identification of a behavioral modification, which provides direction for appropriate histological analyses. However, since the ultimate diagnosis of the pathology largely depends on the initial choice of histological tests, the opportunity exists for inaccurate or insensitive results. An innovative approach using Fourier transform infrared (FT-IR) spectroscopic imaging to diagnose neuropathology should prove useful. This novel method monitors and visualizes the underlying chemistry of the tissue, based on hundreds of vi

We attempted a combination chemotherapy with cytarabine ocfosfate (SPAC) and etoposide for myelodysplastic syndrome (MDS), and acute non-lymphocytic leukemia developing after a prior history of MDS (MDS/ANLL). SPAC and etoposide were administered orally at the dose of 200 mg/day and 25 mg/day for 14 days, as standard regimen. Two cases complete remission (CR), 4 of partial remission (PR) were obtained among 9 patients. The plasma concentration of cytarabine (Ara-C) reached a plateau at around 4.5 ng/ml during the treatment period from the 7th to the 14th day, and it was detectable with a gradu

Two patients with refractory anemia with excess of blasts in transformation having entered remission and in continuing complete remission with the small-dose cytarabine therapy (SDCA) are reported. SDCA was the only chemotherapy used in these two patients. Cytarabine was injected subcutaneously every 12 hours at doses 15 mg and 10 mg, respectively, in cases 1 and 2. Case 1, a 60-year-old woman, achieved remission after a 14-day course of SDCA. Thereafter, she received SDCA for seven consecutive days every month or every other months in the first year and every two or three months in subsequent

A 57-year-old woman with a 2-year history of neurologic symptoms was shown on brain biopsy to have progressive multifocal leukoencephalopathy. Although cell-mediated immune mechanisms were impaired, no underlying disease was found. The administration of transfer factor enhanced cell-mediated immunity, but the neurologic symptoms did not improve. Separate courses of glucocorticoids and cytarabine were associated with rapid neurologic deterioration.

This case study presents the first reported instance of cytarabine-induced hyperbilirubinemia in a pregnant patient undergoing treatment of acute myeloid leukemia. Despite experiencing direct hyperbilirubinemia during induction therapy, the patient exhibited no other signs of liver dysfunction. Lowering the cytarabine dose in subsequent cycles resulted in milder bilirubin elevations without compromising treatment efficacy. The patient gave birth to a healthy baby at term between cycles, underscoring the benign nature of this effect on both maternal and fetal health. This case contributes valua

Neurotoxicity from high-dose cytarabine, a pyrimidine metabolite used in treatment for acute myeloid leukemia, is a known but dose-limiting toxicity which has incidences in up to 14% in patients receiving high doses of the drug. Neurologic symptoms vary but range from somnolence and ataxia to more severe complications such as seizures and even death. There are no validated treatments other than discontinuation of the drug and supportive measures. We present two cases of cytarabine-related neurotoxicity treated with corticosteroids with complete resolution of symptoms.

BACKGROUND: Prolonged cytotoxic concentrations of cytarabine (CA) are required for maximum cytotoxicity. DepoCyt is a human liposomal cytarabine (LC) product that lasts longer in plasma and CSF compared with free CA (FC). The use of LC has not been evaluated in dogs.
OBJECTIVES: To perform a LC pharmacokinetic (PK) study when administered SC in dogs.
ANIMALS: Five healthy female beagles.
METHODS: Three-period, 3-treatment, nonblinded, randomized, and crossover design, including a pilot study. LC was administered at 50 mg/m2 SC and FC was administered at 25 and 50 mg/m2 SC and IV. Plasma CA

Cytarabine (Ara-C) is a nucleoside analogue used in the treatment of cancers and viral infections. It has teratogenic potential and causes a variety of birth defects in fetuses. Alpha-lipoic acid (ALA) is a natural antioxidant offers protection against the developmental toxicity induced by drug- or toxicant-exposure or pathological conditions. This study was aimed at evaluating the protective effect of ALA against Ara-C induced developmental toxicity in rat fetus. Pregnant rats divided into five groups and received normal saline, ALA200 mg/kg, Ara-C12.5 mg/kg, Ara-C25 mg/kg and, Ara-C25 mg/kg

BACKGROUND: Liposomal formulation of anthracyclines provide better systemic and organ-specific tolerance, with potential for less local tissue damage during extravasation. Several small series have reported that most liposomal anthracycline extravasations are consistent with irritant injury without tissue necrosis. There have been no reports published regarding the clinical effects of extravasation of liposomal cytarabine-daunorubicin (CPX-351).
CASE REPORT: The patient received CPX-351 for relapsed acute myelogenous leukemia via a left chest wall port-a-catheter. The catheter became dislodge

BACKGROUND: Relapsed medulloblastoma (MB) is a highly lethal disease, requiring for new effective treatment strategies. Intrathecal (IT) therapy both for de novo or relapsed brain tumors with meningeal metastasis is rarely used in first line and relapse protocols.
PATIENTS AND METHODS: We report on three cases of children with relapsed MB treated with IT liposomal cytarabine administered after mild sedation every 15 days.
RESULTS: The treatment was well-tolerated in all patients, achieving a prolonged progression-free survival (4-11 months) with a good quality of life.
CONCLUSION: This expe

To understand the mechanism of cellular resistance to the nucleoside analogue cytarabine (1-beta-D-arabinofuranosylcytosine, AraC), two resistant derivatives of the human leukemic line CCRF-CEM were obtained by stepwise selection in different concentrations of AraC. CEM/4xAraC cells showed low AraC resistance, whereas CEM/20xAraC cells showed high resistance. Both cell lines showed similar patterns of cross-resistance to multiple cytotoxic nucleoside analogues, with the exception that CEM/20xAraC cells remained sensitive to 5-fluorouridine and 2-deoxy-5-fluorouridine. Both cell lines were sens

We previously reported a high incidence of kerato-conjunctivitis in patients receiving high-dose cytarabine following total body irradiation (TBI) as a conditioning for hematopoietic stem cell transplantation (HSCT) even on prophylaxis with topical corticosteroid. This study aimed to evaluate whether addition of eye rinse, which was designed to remove cytarabine from ocular surface, further reduces the incidence of kerato-conjunctivitis in the same setting. Seventy-six patients receiving cytarabine at a dose of 3 g/m(2) every 12 h for 4 days after receiving TBI (12 Gy) as conditioning for HSCT

Acute myeloid leukemia (AML) patients with high PRDM16 expression frequently experience induction failure and have a poor prognosis. However, the molecular mechanisms underlying these clinical features remain elusive. We found that murine AML cells transformed by MLL::AF9 fusion and oncogenic short-isoform Prdm16 overexpression (hereafter, MF9/sPrdm16) exhibited resistance to cytarabine (AraC), but not to anthracycline, both in vitro and in vivo. Intriguingly, MF9/sPrdm16 cells displayed a gene expression signature of high oxidative phosphorylation (OxPHOS) and increased mitochondrial respirat
