First integrated in vitro regeneration protocol for the endangered aquatic plant Hydrocera triflora (Balsaminaceae) via optimized cytokinin-auxin balance.

Hydrocera triflora, a rare and endangered aquatic species of phylogenetic importance within the family Balsaminaceae, lacks any reported in vitro regeneration system. This study establishes the first complete in vitro propagation protocol for this taxon, integrating optimized sterilization, shoot induction, proliferation, and rooting. Stem explants (~ 3 cm) were disinfected using ethanol, 0.1% mercuric chloride with Tween, and 20% sodium hypochlorite, with the most effective method combining ethanol pre-wash and 0.1% mercuric chloride for 15 min, resulting in 35% contamination and 13% browning

S-nitrosoglutathione Reductase-Mediated Nitric Oxide Affects Axillary Buds Outgrowth of Solanum lycopersicum L. by Regulating Auxin and Cytokinin Signaling.

Auxin and cytokinin are two kinds of important phytohormones that mediate outgrowth of axillary buds in plants. How nitric oxide and its regulator of S-nitrosoglutathione reductase (GSNOR) take part in auxin and cytokinin signaling for controlling axillary buds outgrowth remains elusive. We investigated the roles of GSNOR during tomato axillary bud outgrowth by using physiological, biochemical and genetic approaches. GSNOR negatively regulated NO homeostasis. Suppression of GSNOR promoted axillary bud outgrowth by inhibiting the expression of FZY in both apical and axillary buds. Meanwhile, AU

Dynamic changes in concentrations of auxin, cytokinin, ABA and selected metabolites in multiple genotypes of Douglas-fir (Pseudotsuga menziesii) during a growing season.

Changes in concentrations of several endogenous phytohormones and metabolites were analyzed in the long shoots of nine genotypes of coastal Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco var. menziesii) at five developmental stages: (1) closed buds, (2) flushing buds, (3) rapidly elongating shoots, (4) growing shoots and (5) near full-length shoots during one growing season. When averaged across genotypes, indole-3-acetic acid (IAA) concentration was high at stages 1 and 3. The only pattern that correlated with cone productivity was the one that was unique to IAA, in which high concentratio

Phosphorylation of Arabidopsis response regulator 7 (ARR7) at the putative phospho-accepting site is required for ARR7 to act as a negative regulator of cytokinin signaling.

Cytokinins are plant hormones that regulate diverse aspects of plant growth and development. Arabidopsis cytokinin signal transduction utilizes a multi-step two-component signaling (TCS) system by histidyl-aspartidyl phosphorelays. We here show that phosphorylation of ARR7, an A-type response regulator that acts as a negative regulator of cytokinin signaling, is required for its function in plants. Phosphorylation of ARR7 is inhibited in vitro by mutation in a putative phospho-accepting Asp residue into an Asn residue (ARR7(D85N)). While ectopic expression of ARR7 decreases root-growth inhibit

Exogenous ABA combined with cytokinin enhances drought resistance in Ziziphus jujuba Mill. var. spinosa by regulating xylem development and zeatin signaling pathway.

Drought is a decisive abiotic constraint limiting the growth and yield of jujube, its alleviation is orchestrated by a complex hormonal network that governs the tree's drought-resistance responses. Based on preliminary evidence that abscisic acid (ABA) and cytokinin play coordinated roles in drought adaptation, this study aimed to clarify their individual and combined effects on regulating drought resistance in sour jujube. Phenotypic responses, anatomical alterations, endogenous hormone profiles, and the expression levels of genes involved in the zeatin biosynthesis pathway after exogenous ho

Genome-Wide Identification and Expression Analysis of Cytokinin Response Regulator (RR) Genes in the Woody Plant Jatropha curcas and Functional Analysis of JcRR12 in Arabidopsis.

The cytokinin (CK) response regulator (RR) gene family plays a pivotal role in regulating the developmental and environmental responses of plants. Axillary bud outgrowth in the perennial woody plant Jatropha curcas is regulated by the crosstalk between CK and gibberellins (GA). In this study, we first analyzed the effects of gibberellin A3 (GA3), lovastatin (a CK synthesis inhibitor), decapitation, and their interaction, on the outgrowth of axillary buds. The results indicate that lovastatin completely inhibited GA-promoted axillary bud outgrowth and partially weakened the decapitation-promote

Monoclonal Lym-1 antibody-dependent lysis of B-lymphoblastoid tumor targets by human complement and cytokinine-exposed mononuclear and neutrophilic polymorphonuclear leukocytes.

Lym-1 is a murine IgG2a monoclonal antibody that recognizes a polymorphic variant of HLA-DR antigens on malignant B cells, with minimal cross-reactivity with normal tissues. Because it can be safely administered in vivo, a detailed knowledge of its ability to recruit and trigger the antitumor immune effector systems is required to optimize potential serotherapeutic approaches in B-lymphoma patients. By using Raji cells as a model of B-lymphoma targets, we found that Lym-1 activates complement-mediated lysis efficiently. Moreover, Lym-1 was capable of triggering the antibody-dependent cellular

ABA-cytokinin crosstalk regulate drought tolerance in Ziziphus jujuba var. spinosa through the phenylpropanoid pathway: insights from physiological and multi-omics integration.

Drought stress is a major constraint in jujube production. Although abscisic acid (ABA) and cytokinin exert opposing effects on plant growth, recent findings revealed that their combined application enhanced drought tolerance in jujube; however, the underlying mechanism remains elusive. In this study, three exogenous treatments were applied to sour jujube seedlings under drought stress: 5 mg L-1 ABA, a cytokinin mixture of 50 mg L-1 6-benzylaminopurine (6-BA) and 100 mg L-1 kinetin (KT), and a combined ABA and cytokinins solution. Evolution and roles of cytokinin genes in angiosperms 2: Do ancient CKXs play housekeeping roles while non-ancient CKXs play regulatory roles?

Cytokinin oxidase/dehydrogenase (CKX) is a key enzyme responsible for the degradation of endogenous cytokinins. However, the origins and roles of CKX genes in angiosperm evolution remain unclear. Based on comprehensive bioinformatic and transgenic plant analyses, we demonstrate that the CKXs of land plants most likely originated from an ancient chlamydial endosymbiont during primary endosymbiosis. We refer to the CKXs retaining evolutionarily ancient characteristics as "ancient CKXs" and those that have expanded and functionally diverged in angiosperms as "non-ancient CKXs". We show that the e

ARR1 and ARR12 negatively regulate arsenic stress tolerance by controlling flavonoid metabolism in Arabidopsis.

ARR1/12-mediated cytokinin signaling negatively regulates the accumulation of glycosylated flavonoids, thereby increasing plant susceptibility to As(III) stress. Cytokinins negatively regulate arsenic stress tolerance in plants through cytokinin-signaling type-B Arabidopsis response regulators (B-ARRs), specifically ARR1 and ARR12. However, the mechanism by which cytokinin signaling regulates plant metabolite dynamics, particularly antioxidant flavonoids, in response to arsenic toxicity remains largely unknown. Here, we hypothesized that ARR1/12-mediated cytokinin signaling modulates flavonoid

Auxin synthesis gene tms1 driven by tuber-specific promoter alters hormonal status of transgenic potato plants and their responses to exogenous phytohormones.

Ectopic auxin overproduction in transgenic potato leads to enhanced productivity accompanied with concerted and occasional changes in hormonal status, and causing altered response of transformants to exogenous auxin or cytokinin. Previously, we generated potato transformants expressing Agrobacterium-derived auxin synthesis gene tms1 driven by tuber-specific patatin gene promoter (B33-promoter). Here, we studied the endogenous hormonal status and the response to exogenous phytohormones in tms1 transformants cultured in vitro. Hormonal control of second flushing in Douglas-fir shoots.

Spring-flushing, over-wintered buds of Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) produce new buds that may follow various developmental pathways. These include second flushing in early summer or dormancy before flushing during the following spring. Second flushing usually entails an initial release of apical dominance as some of the current-season upper lateral buds grow out. Four hypotheses concerning control of current bud outgrowth in spring-flushing shoots were tested: (1) apically derived auxin in the terminal spring-flushing shoot suppresses lateral bud outgrowth (second flushin

Topical and nutricosmetic products for healthy hair and dermal antiaging using "dual-acting" (2 for 1) plant-based peptides, hormones, and cannabinoids.

One of the side effects of oral antiaging retinoids is increased hair shedding. Retinoids promote the expression of TGF-β2 from fibroblasts, which stimulate collagen expression but silences keratinocytes. Since keratinocytes normally influence differentiation of dermal papilla cells at the base of the hair follicle, retinoids feasibly inhibit hair growth via the increased expression of TGF-β2, which inhibits Wnt/β-catenin signaling. Fortunately, the plant kingdom provides an array of alternatives as dual-acting nutricosmetics and topicals that work independently of TGF-β2 to confer dermal anti

Vig r 6, the cytokinin-specific binding protein from mung bean (Vigna radiata) sprouts, cross-reacts with Bet v 1-related allergens and binds IgE from birch pollen allergic patients' sera.

SCOPE: Birch pollen associated allergy to mung bean sprouts is caused by cross-reactivity between the birch pollen allergen Bet v 1 and the mung bean allergen Vig r 1. We aimed to determine the allergenicity of the cytokinin-specific binding protein from mung bean (Vig r 6), another allergen related to Bet v 1 with only 31% sequence identity. METHODS AND RESULTS: Bet v 1, Gly m 4, Vig r 1, and Vig r 6 were produced in Escherichia coli. In an ELISA, 73 and 32% of Bet v 1-sensitized birch-allergic patients' sera (n = 60) showed IgE binding to Vig r 1 and Vig r 6, respectively. Of 19 patients wh

Relationships between plant hormones, carbohydrate metabolism and excision-induced adventitious root formation of two contrasting hydrangea cultivars.

Cytokinins and abscisic acid act as auxin antagonists during adventitious root induction, thereby limiting AR development depending on plant genotype and the distance to the wounding site. This study investigated the role of plant hormones in the lower rooting capacity of cuttings of the Hydrangea macrophylla cultivar 'Clarissa' compared to 'Caipirinha', further considering interrelations to carbohydrates. At first, the rooting response to 1 min pulse treatments of the stem base with indole-3-acetic acid (IAA) was analyzed. Without IAA, rooting of 'Clarissa' lagged behind that of 'Caipirinha'.

Functional delineation of rice MADS29 reveals its role in embryo and endosperm development by affecting hormone homeostasis.

Rice MADS29 has recently been reported to cause programmed cell death of maternal tissues, the nucellus, and the nucellar projection during early stages of seed development. However, analyses involving OsMADS29 protein expression domains and characterization of OsMADS29 gain-of-function and knockdown phenotypes revealed novel aspects of its function in maintaining hormone homeostasis, which may have a role in the development of embryo and plastid differentiation and starch filling in endosperm cells. The MADS29 transcripts accumulated to high levels soon after fertilization; however, protein a

The AP2/ERF transcription factor WIND1 controls cell dedifferentiation in Arabidopsis.

Many multicellular organisms have remarkable capability to regenerate new organs after wounding. As a first step of organ regeneration, adult somatic cells often dedifferentiate to reacquire cell proliferation potential, but mechanisms underlying this process remain unknown in plants. Here we show that an AP2/ERF transcription factor, WOUND INDUCED DEDIFFERENTIATION 1 (WIND1), is involved in the control of cell dedifferentiation in Arabidopsis. WIND1 is rapidly induced at the wound site, and it promotes cell dedifferentiation and subsequent cell proliferation to form a mass of pluripotent cell

On the mechanism of rejuvenation of ageing detached bean leaves by low-concentration stressors.

The effect of low concentrations of some stress-inducing compounds of different toxicity and chemical nature, such as Cd and Pb salts or DCMU, was investigated on the senescence of chloroplasts in detached primary leaves of bean (Phaseolus vulgaris L.). After 1 week of senescence followed by root development from the petiole, these agents stimulated chlorophyll accumulation and photosynthetic activity ((14)CO(2) fixation) as compared to the control, thus inducing rejuvenation. Low-concentration stressors increased the level of active cytokinins in roots and leaves during the treatment, as moni

A CLV3-WUS positive feedback loop in individual cells enhances shoot stem cell thermotolerance in Arabidopsis.

CLAVATA3 (CLV3) has long been thought to be a negative regulator of the WUSCHEL (WUS) expression domain, whereas cytokinin signaling increases WUS expression at the transcriptional level. Whether these two pathways in WUS regulation are coordinated in the stem cell niche remains unknown. Here, we show that CLV signaling has a dual-directional effect on WUS by limiting its expression domain but maintaining its expression level. CLV1 phosphorylates the transcription factor BRASSINAZOLE RESISTANT 1 (BZR1) to release the inhibitory effect of BZR1 on cytokinin biosynthesis, which relays the CLV sig

High-throughput quantitative analysis of phytohormones in sorghum leaf and root tissue by ultra-performance liquid chromatography-mass spectrometry.

Plant development, growth, and adaptation to stress are regulated by phytohormones, which can influence physiology even at low concentrations. Phytohormones are chemically grouped according to both structure and function as auxins, cytokinins, abscisic acid, jasmonates, salicylates, gibberellins, and brassinosteroids, among others. This chemical diversity and requirement for highly sensitive detection in complex matrices create unique challenges for comprehensive phytohormone analysis. Here, we present a robust and efficient quantitative UPLC-MS/MS assay for 17 phytohormones, including jasmona
