Immunocytochemical and electron microscopic study of hepatitis B virus antigen and complete particle production in hepatitis B virus DNA transfected HepG2 cells.

The relationship between the presence of hepatitis B virus antigens, their localization and hepatitis B virus replication was studied in different clones of cultured HepG2 hepatoblastoma cells transfected with cloned hepatitis B virus DNA. Intracellular hepatitis B virus antigens were detected by immunofluorescence. The production of these antigens was evaluated in the culture media by enzyme-linked immunoassay. Hepatitis B virus DNA was detected using dot-blot hybridization. Three types of HBcAg staining were observed in transfected HepG2 cells: (a) cells with nuclear HBcAg, (b) cells with cy

Anatomical locus of the common enterobacterial antigen.

Cell walls, membrane vesicles and the soluble cytoplasmic components have been isolated from Escherichia coli 0:14 and Salmonella typhimurium and examined by electron microscopy as well as hemagglutination inhibition with heterologous antisera for enterobacterial common antigen (ECA). The outer and cytoplasmic membranes also were isolated and examined for common antigen. Although common antigen was present in the isolated surface structures it was not localized exclusively in these fractions. Additional experiments revealed that, whatever the nature of the integration of ECA of Kunin into the

Coordinated movement of cytoplasmic and transmembrane domains of RyR1 upon gating.

Ryanodine receptor type 1 (RyR1) produces spatially and temporally defined Ca2+ signals in several cell types. How signals received in the cytoplasmic domain are transmitted to the ion gate and how the channel gates are unknown. We used EGTA or neuroactive PCB 95 to stabilize the full closed or open states of RyR1. Single-channel measurements in the presence of FKBP12 indicate that PCB 95 inverts the thermodynamic stability of RyR1 and locks it in a long-lived open state whose unitary current is indistinguishable from the native open state. We analyzed two datasets of 15,625 and 18,527 frozen-

Development of the First Low Nanomolar Liver Receptor Homolog-1 Agonist through Structure-guided Design.

As a key regulator of metabolism and inflammation, the orphan nuclear hormone receptor, liver receptor homolog-1 (LRH-1), has potential as a therapeutic target for diabetes, nonalcoholic fatty liver disease, and inflammatory bowel diseases (IBD). Discovery of LRH-1 modulators has been difficult, in part due to the tendency for synthetic compounds to bind unpredictably within the lipophilic binding pocket. Using a structure-guided approach, we exploited a newly discovered polar interaction to lock agonists in a consistent orientation. This enabled the discovery of the first low nanomolar LRH-1

Synthesis and biological evaluation of halogenated curcumin analogs as potential nuclear receptor selective agonists.

This report describes the synthesis of analogs of curcumin, and their analysis in acting as nuclear receptor specific agonists. Curcumin (CM), a turmeric-derived bioactive polyphenol found in curry, has recently been identified as a ligand for the vitamin D receptor (VDR), and it is possible that CM exerts some of its bioeffects via direct binding to VDR and/or other proteins in the nuclear receptor superfamily. Using mammalian-two-hybrid (M2H) and vitamin D responsive element (VDRE) biological assay systems, we tested CM and 11 CM synthetic analogs for their ability to activate VDR signaling.

"Neuritic conglomerates" in the cerebral cortex of a patient with Creutzfeld-Jakob disease.

In the cerebral cortex of a patient with Creutzfeld-Jakob disease (CJD), we found peculiar glomeruloid or skein-like structures which have not previously been described. The patient was a 67-year-old man, whose clinical features and neuropathological findings were consistent with CJD. The glomeruloid or skein-like structures were distributed in the deep layers of the cortex and consisted of intricately entangled masses of thick argyrophilic fibers. These structures were immunostained with anti-neurofilament antibodies and were considered to have originated from neuronal cytoplasmic processes,

The cytoplasmic extension of the integrin β6 subunit regulates epithelial-to-mesenchymal transition.

Prognosis for oral cancer patients has not improved in over 60 years due to invasion and recurrence. To understand the invasive behavior of this tumor, we evaluated the role of the αvβ6 integrin. Invasive oral SCC cells express the αvβ6 integrin, which contains an 11-amino-acid extension on its β-subunit unique to the integrin family. We determined that this β6 cytoplasmic extension regulates the composition of the intermediate filament network and the organization of signaling structures called focal contacts. The auto-phosphorylation of FAK, which is localized to focal contacts, was also reg

Reversible suppression of skeletal myotube formation in vitro obtained by varying [CO2].

In primary cultures of chicken skeletal muscle, decreasing the [CO2] of the gaseous phase below 10(-3)% resulted in inhibition of cell proliferation and cytolysis. With 10(-3)% CO2-air, cell proliferation was slightly retarded and myotube formation was inhibited approximately 90% compared to cultures receiving 5% CO2-air. Changes in pH were not effective. Culture in low [CO2] resulted in the accumulation of lipoidal inclusions and unique cytoplasmic structures. Increasing time in culture with low [CO2] resulted in an increase in the length of G1 of the cell cycle. The inhibition was reversed b

Ultrastructural and Immunohistochemical Study of a Gastric Bizarre Leiomyoma: Bizarre Nuclei and Prominent Cytoplasmic Processes Extending Into and Displacing the Stroma.

This study presents the ultrastructural and immunohistochemical findings of a gastric bizarre leiomyoma arising in the vestibule of a 79-year-old male. Histologically, loosely proliferating tumor cells consist of large, multinucleated, bizarre nuclei with intranuclear inclusions and abundant cytoplasm-containing vacuoles. A murky line was apparent between the tumor cells and eosinophilic and heterogenous stroma-like areas. Immunohistochemically, tumor cells exhibited positively stained dot patterns of α-smooth muscle actin and caldesmon, which were distributed in the cytoplasm of tumor cells a

Choroid plexus papilloma: light and electron microscopic study of three cases.

Three cases of choroid plexus papilloma were studied by light and electron microscopy. All had the typical papillary pattern, and, in addition, two exhibited solid areas of tumor growth which predominated in one case. PAS positive (diastase resistant) and argyrophilic cytoplasmic inclusions were present in all three tumors but were particularly abundant in the predominantly solid one. Ultrastructurally, the inclusions appeared as irregularly shaped structures containing lipid droplets, filamentous material and microtubules, and resembled the "silver bodies" of Biondi found in normal choroid pl

Morphometric and fine-structural studies of rat pancreatic acinar cells during early postnatal life.

Pancreatic acinar cells of rats obtained at 1, 2, 3, 5, 7 and 14 days of age were examined using fine structural and morphometric techniques. From 5 days of age onwards, the acinar cells were analysed twice per day, at 20.00 h and 08.00 h. The present study demonstrates changes in the average volume of the cell, nucleus and cytoplasm, and volume densities of various cytoplasmic organelles during the first two weeks after birth. During early postnatal life, the volume density of rER increases, whereas that of zymogen granules decreases. From 5 days of age onwards, the volume densities of these

Proliferative parathyroid lesions in captive-bred American bullfrogs (Lithobates catesbeianus) with metabolic bone disease.

Parathyroid gland lesions in anurans are infrequently reported and most often occur secondary to experimental interventions. Husbandry-related parathyroid changes have not been documented in this order of Amphibia. Three American bullfrogs (Lithobates catesbeianus) living in a captive colony were euthanized due to clinical concern for metabolic bone disease secondary to lack of consistent dietary supplementation with vitamin D3. Necropsy revealed cystic dilation and variable proliferation of unidentified structures within the cranial coelom corresponding to the anatomical location of anuran pa

FXR-targeted drug discovery: Recent advances and therapeutic perspectives.

Farnesoid X Receptor (FXR), a pivotal member of the nuclear receptor superfamily, plays a central role in the regulation of crucial physiological processes. These processes encompass bile acid homeostasis, inflammatory responses, fibrosis, as well as lipid and glucose metabolism. Given its extensive physiological functions, FXR has become an extremely promising therapeutic target for bile acid metabolism disorders. In recent years, the progress in medicinal chemistry has significantly expedited the development of FXR agonists. In 2016, obeticholic acid (OCA) received approval from FDA for mark

Defects in intron recycling suppress the antiviral response via a mechanism of intronic endogenous dsRNA.

Loss of the lariat debranching enzyme DBR1 causes cytoplasmic accumulation of intron lariats, but why this reduces cell-intrinsic immunity is unclear. Here, we show that intronic inverted repeats Alu (IR Alus), normally degraded after splicing, form long double-stranded RNA (dsRNA) structures when lariats escape recycling. Viral introns evolve under pressure to avoid dsRNA, whereas human introns are enriched for them. Using computational, immunostaining, and genomic approaches, we demonstrate that DBR1 deficiency elevates cytoplasmic dsRNA and attenuates RNase L and PKR signaling. Our data sug

Mucolipidosis type III α/β: the first characterization of this rare disease by autopsy.

We report findings from an autopsy of a 45-year-old woman with the rare lysosomal storage disease mucolipidosis type III α/β. Her disease manifested most notably as multiple bone and cartilage problems with tracheal and bronchial malacia. Principal autopsy findings included gross abnormalities in bone and cartilage with corresponding microscopic cytoplasmic lysosomal granules. These cytoplasmic granules were also seen in histologic preparations of the brain, myocardium, heart valves, and fibroblasts of the liver and skin by light and electron microscopy. By electron microscopy there were scatt

The E3 ubiquitin ligase MID1/TRIM18 promotes atypical ubiquitination of the BRCA2-associated factor 35, BRAF35.

MID1/TRIM18 is a member of the TRIM family of ubiquitin E3 ligases characterized by the presence of a conserved RING-containing N-terminal tripartite motif. Mutations in the MID1 gene have been associated with the X-linked form of Opitz Syndrome, a developmental disorder characterized by midline defects and intellectual disability. The effect of MID1 E3 ligase activity within the cell and the role in the pathogenesis of the disease is still not completely unraveled. Here, we report BRAF35, a non-canonical HMG nuclear factor, as a novel MID1 substrate. MID1 is implicated in BRAF35 ubiquitinatio

Cytoplasmic localization and the choice of ligand determine aggregate formation by androgen receptor with amplified polyglutamine stretch.

Polyglutamine tract expansion in androgen receptor is a recognized cause of spinal and bulbar muscular atrophy (SBMA), an X-linked motor neuronopathy. Similar mutations have been identified in proteins associated with other neurodegenerative diseases. Recent studies have shown that amplified polyglutamine repeat stretches form cellular aggregates that may be markers for these neurodegenerative diseases. Here we describe conditions that lead to aggregate formation by androgen receptor with polyglutamine stretch amplification. In transfection experiments, the mutant, compared with the wild-type
