Subtype Distribution of Blastocystis in Communities along the Chao Phraya River, Thailand.

Blastocystis is a common zoonotic enteric protozoan that has been classified into 17 distinct subtypes (STs). A cross-sectional study was conducted to determine the prevalence and subtype distributions of Blastocystis in villagers living along the Chao Phraya River, Ayutthaya Province, Thailand, and to assess the risk of zoonotic infection. In total, 220 stool samples were collected, and DNA was extracted. PCR and sequencing were performed with primers targeting the small-subunit ribosomal RNA (SSU rRNA) genes. Blastocystis was present in 5.9% (13/220) of samples, and ST3 (5.0%; 11/220) was th

Unravelling the Secret of Phantom Phenotypes in Macrolichens: Insights From Cladonia bellidiflora Complex.

Lichens are unique among symbiotic organisms because their distinctive features develop only through interactions between the partners and differ from the appearance of each partner when grown separately in culture. Traditionally, lichen phenotype has been assumed to be determined by the mycobiont; however, exceptions exist that challenge the universal validity of this rule. One example is the phenomenon of 'phantom phenotypes', where lichens with genetically indistinguishable mycobionts exhibit distinct morphologies, sometimes better explained by differences among other symbiotic partners. Sedimentary Ancient DNA Tracks Multi-Kingdom Ecosystem Reorganizations Following Sequential Human Land Use at Crawford Lake.

Crawford Lake has an exceptional stratigraphic record that began recording biannual (varved) sedimentation in the lake basin ~750 years ago, preserving evidence of shifting cultural zones and agricultural practices, from Late Woodland Period Indigenous agriculturalists to the impacts of industrialization during the late 19th century. It was selected as the candidate site for the proposed 'Anthropocene' epoch in 2023-a proposal ultimately rejected in 2024-but the lake's significance extends beyond a formal stratigraphic boundary. Its sediments preserve a long record of human-ecosystem entanglem

Physiological-Biochemical Characteristics and Omics Landscape of Drought Tolerance in Chlorella sp. from the Taklamakan Desert of China.

Microalgae endemic to desert ecosystems possess exceptional stress-resistance traits; however, the physiological and molecular drought-adaptation mechanisms of desert-dwelling Chlorella remain largely uncharacterized. Herein, we combined physiological-biochemical assays, ultrastructural observation, transcriptomics and metabolomics to dissect drought responses in Taklamakan-desert-origin Chlorella sp. under polyethylene glycol 6000 (PEG-6000)-simulated mild (10%), moderate (20%), and severe (30%) drought stress, with mechanistic focus on moderate-drought conditions. Moderate drought repressed

Phycosphere microbiome remodeling and Brevundimonas partnership establishment enhance microalgal nonylphenol tolerance.

INTRODUCTION: Microalgae are vital primary producers in aquatic ecosystems yet increasingly threatened by nonylphenol (NP), a common endocrine disruptor. Current ecotoxicological assessments, however, largely rely on single-species models, overlooking the role of the phycosphere microbiome in host stress adaptation. METHODS: Here, using the NP-tolerant microalgae Dictyosphaerium sp. as a model holobiont, we integrated multi-omics, axenic algae-bacteria co-culture, physiological, and metabolic analyses to unravel how associated microbes contribute to microalgal NP fitness. RESULTS: Under envi

System-level multi-omics uncover proteomic, peptidomic, and metabolic remodeling in Microcystin-LR exacerbated chronic kidney disease.

Chronic kidney disease (CKD) is increasingly linked to environmental toxins, including the cyanobacterial toxin microcystin-LR (MC-LR), particularly in populations with pre-existing renal injury. We have previously demonstrated that MC-LR promotes CKD, however the full underlying mechanism remains poorly understood. This study investigated how MC-LR exacerbates adenine-induced kidney disease in animal models using an integrative multi-omics approach. Male Sprague-Dawley rats were assigned to four groups: control, MC-LR, adenine, and adenine + MC-LR. Histopathology showed that MC-LR alone cause

A giant virus in a small lake: characterization of a new tropical algal virus expands the geographical and genetic diversity of the Brazilian virosphere.

Chloroviruses are large double-stranded DNA (dsDNA) viruses that infect Chlorella-like green algae in freshwater environments. Despite being extensively studied in the Northern Hemisphere, little is known about their diversity and biology in tropical regions. In this study, we report the isolation and characterization of a novel chlorovirus, named Chlorovirus BR-MG-N01, infecting Chlorella variabilis NC64A. The virus was recovered from a freshwater lake in Guanhães, Minas Gerais, Brazil, located in a transition zone between the Atlantic Forest and Cerrado biomes. Electron microscopy revealed i

Copper-dependent iron assimilation pathway in the model photosynthetic eukaryote Chlamydomonas reinhardtii.

The unicellular green alga Chlamydomonas reinhardtii is a valuable model for studying metal metabolism in a photosynthetic background. A search of the Chlamydomonas expressed sequence tag database led to the identification of several components that form a copper-dependent iron assimilation pathway related to the high-affinity iron uptake pathway defined originally for Saccharomyces cerevisiae. They include a multicopper ferroxidase (encoded by Fox1), an iron permease (encoded by Ftr1), a copper chaperone (encoded byAtx1), and a copper-transporting ATPase. A cDNA, Fer1, encoding ferritin for i

Breaking-Down Barriers: Proposal of Using Cellulose Biosynthesis Inhibitors and Cellulase Enzyme as a Novel Treatment Modality for Vision Threatening Pythium Insidiosum Keratitis.

Pythium insidiosum, an Oomycete, causes severe keratitis that endangers vision. Its clinical, morphological, and microbiological characteristics are often indistinguishable from those of fungal keratitis, earning it the moniker "parafungus". Distinctive clinical hallmarks that set it apart from other forms of keratitis include radial keratoneuritis, tentacles, marginal infiltration, and a propensity for rapid limbal spread. The therapeutic approach to Pythium keratitis (PK) has long been a subject of debate, and topical and systemic antifungals and antibacterials have been tried with limited s

Production of vitamins, coenzymes and related biochemicals by biotechnological processes.

Vitamins and related biofactors belong to those few chemicals with a direct positive appeal to people. There is indeed a large need for extra vitamins, other than those derived from plant and animal food sources, due to unbalanced food habits or processing, food shortage or disease. Added vitamins are now either prepared chemically or biotechnologically via fermentation or bioconversion processes. Several vitamins and related biofactors are now only or mainly produced chemically (vitamin A, cholecalciferol (D3), tocopherol (E), vitamin K2, thiamine (B1), niacin (PP or B3), pantothenic acid (B5

Reduction and liquid-solid partitioning of SARS-CoV-2 and adenovirus throughout the different stages of a pilot-scale wastewater treatment plant.

Investigating waterborne viruses is of great importance to minimizing risks to public health. Viruses tend to adsorb to sludge particles from wastewater processes by electrostatic and hydrophobic interactions between virus, aquatic matrix, and particle surface. Sludge is often re-used in agriculture; therefore, its evaluation is also of great interest to public health. In the present study, a pilot scale system treating real domestic wastewater from a large city in Brazil was used to evaluate the removal, the overall reduction, and liquid-solid partitioning of human adenovirus (HAdV), the nove

Effect of Nitrosomonas europaea on Chlorella vulgaris in bio-hydrogels for startup of microalgal-bacterial granular sludge: Performance and microscopic mechanism.

Algal-bacterial granular sludge technology holds significant promise for treating municipal wastewater with carbon emission reduction. However, its practical application has been constrained by the long startup period. In this study, bio-hydrogels co-encapsulating autotrophic ammonia-oxidizing bacteria (Nitrosomonas europaea) and photomixotrophic microalgae (Chlorella vulgaris) were used in a microalgal-bacterial granules system (MBGS) for the treatment of low C/N wastewater. The operating conditions of the MBGS system included inoculation volume ratio of 1:6 (bio-hydrogel granules: wastewater

A chirality change in XPC- and Sfi1-derived peptides affects their affinity for centrin.

The Ca(2+)-binding protein centrin binds to a hydrophobic motif (W(1)xxL(4)xxxL(8)) included in the sequence of several cellular targets: XPC (xeroderma pigmentosum group C protein), Sfi1 (suppressor of fermentation-induced loss of stress resistance protein1), and Sac3 [the central component of the transcription and mRNA export (TREX-2) complex]. However, centrin binding occurs in a reversed orientation (L(8)xxxL(4)xxW(1)) for Sfi1 and Sac3 compared with XPC. Because D-peptides have been investigated for future therapeutic use, we analyzed their centrin-binding properties. Their affinity for c

Genome-based approaches to understanding phosphorus deprivation responses and PSR1 control in Chlamydomonas reinhardtii.

The Chlamydomonas reinhardtii transcription factor PSR1 is required for the control of activities involved in scavenging phosphate from the environment during periods of phosphorus limitation. Increased scavenging activity reflects the development of high-affinity phosphate transport and the expression of extracellular phosphatases that can cleave phosphate from organic compounds in the environment. A comparison of gene expression patterns using microarray analyses and quantitative PCRs with wild-type and psr1 mutant cells deprived of phosphorus has revealed that PSR1 also controls genes encod

Occurrence and environmental stress responses of two plastid terminal oxidases in Haematococcus pluvialis (Chlorophyceae).

The plastid terminal oxidase (PTOX) is a plastoquinol oxidase involved in carotenoid biosynthesis in higher plants, and may also represent the elusive oxidase in chlororespiration. Haematococcus pluvialis is a green alga that has the ability to synthesize and accumulate large amounts of the red carotenoid astaxanthin (ca. 2% of dry weight) under various stress conditions. However, the occurrence and function of PTOX in astaxanthin synthesis and the stress response in this organism is unknown. In this study, two ptox cDNAs were cloned and sequenced from H. pluvialis and were designated as ptox1

cDNA cloning and characterization of a human sperm antigen (SPAG6) with homology to the product of the Chlamydomonas PF16 locus.

Serum from an infertile male with high-titer anti-sperm antibodies was used to identify a novel human sperm antigen by screening of a testis expression library. The clone, initially designated Repro-SA-1 (HUGO-approved symbol SPAG6), was found to encode a sequence highly enriched in testis. The deduced amino acid sequence of the full-length cDNA revealed striking homology to the product of the Chlamydomonas reinhardtii PF16 locus, which encodes a protein localized to the central pair of the flagellar axoneme. The human gene encodes 1.8- and 2.8-kb mRNAs highly expressed in testis but not in pr

Regulation of sesquiterpene cyclase gene expression. Characterization of an elicitor- and pathogen-inducible promoter.

The promoter for a tobacco (Nicotiana tabacum) sesquiterpene cyclase gene, a key regulatory step in sesquiterpene phytoalexin biosynthesis, has been analyzed. The EAS4 promoter was fused to the beta-glucuronidase (GUS) reporter gene, and the temporal and spatial expression patterns of GUS activity were examined in stably transformed plants and in transient expression assays using electroporated protoplasts of tobacco. No GUS activity was observed in any tissues under normal growth conditions. A low level of GUS activity was detected in wounded leaf, root, and stem tissues, whereas a much highe
