Metagonimus yokogawai Ancient DNA Recovered from 16th- to 17th-Century Korean Mummy Feces of the Joseon Dynasty.

Metagonimiasis is foodborne intestinal parasitism occurring by the definitive hosts' ingestion of raw or undercooked fish, mostly commonly sweetfish. Most Metagonimus infection is caused by Metagonimus yokogawai but also rarely by Metagonimus takahashii as well as Metagonimus miyatai. Despite recent molecular work on Metagonimus spp., there are still insufficient data to reveal the genetic characteristics of ancient M. yokogawai in a wide geo-historical scope. In this study, we were successful in the analysis of M. yokogawai ancient DNA (aDNA) using coprolite samples retrieved from 16th- to 17

Ancient DNA Reveals Limited Hunter-Gatherer Ancestry in the Cerny Culture (Middle Neolithic, France) Despite Hunting-Associated Practices.

The arrival of the Neolithic way of life in Europe is associated with the migration of Anatolian farmers and their admixture with local hunter-gatherers. In the Paris Basin (Northern France), the Middle Neolithic Cerny culture (4700-4300 bce) combines agricultural practices with funerary items linked to hunting, suggesting a particular connection with the local descendants of Mesolithic hunter-gatherers. The monumental burials gathering only a handful of deceased indicate the development of an elite. To better understand Cerny social structures, we generated genome-wide data for 28 individuals

Innovative DNA tetrahedron inspired by ancient mortise-and-tenon technique offers new immunotherapy strategy for metastatic breast cancer.

Framework nucleic acids effectively meet the demands for precise size control and accurate targeting in the design of drug delivery systems, while developing a controllable drug delivery system with low immunogenicity and high efficiency for delivering nucleic acid drugs to the tumor immune microenvironment (TIME) remains significant challenge. Inspired by ancient Chinese mortise and tenon joint structures, this study develops an intelligent self-assembling DNA tetrahedron (TDN@siCSF-1R), which consists of a gapped DNA tetrahedron (TDN) and a therapeutic siRNA against Colony-Stimulating Factor

Age estimation of single nucleotide polymorphisms associated with autoinflammatory diseases in anatolia: insights from ancient and modern DNA.

Autoinflammatory diseases are immune system disorders that arise from abnormalities in the innate immune system. With the advent of genome sequencing, several distinct genetic loci associated with different types of autoinflammatory diseases have been identified. However, the evolutionary history of these variants, such as the age of the alleles, has not been fully explored in human populations. In this study, we focus on Anatolia, a region with a rich and dynamic prehistory, serving as a hub for human migration and genetic interactions. We explore the age of mutations associated with autoinfl

Ancient DNA analyses of remains of the Medici family (16th century) provide insights into the genetic variation of Plasmodium falciparum.

We evaluated the presence of six Plasmodium species in osteological remains from Grand Duke Francesco I de' Medici (GDFIDM; 1541-1587) and Cardinal Giovanni de' Medici (CGDM; 1543-1562). Targeted enrichment sequencing for mitochondrial genomes recovered 1,865 and 185 base pairs (bp) of P. falciparum from CGDM and GDFIDM, respectively, and 43 bp of P. malariae from GDFIDM. The CGDM P. falciparum sequence represents a previously uncharacterized haplotype with two unique mutations in intergenic and cytochrome c oxidase subunit 1 loci. Network analyses using nine additional ancient samples (Iron A

Detection of 400-year-old Yersinia pestis DNA in human dental pulp: an approach to the diagnosis of ancient septicemia.

Ancient septicemic plague epidemics were reported to have killed millions of people for 2 millenniums. However, confident diagnosis of ancient septicemia solely on the basis of historical clinical observations is not possible. The lack of suitable infected material has prevented direct demonstration of ancient septicemia; thus, the history of most infections such as plague remains hypothetical. The durability of dental pulp, together with its natural sterility, makes it a suitable material on which to base such research. We hypothesized that it would be a lasting refuge for Yersinia pestis, th

Integrating Osteology and Ancient DNA: Cranial Variation, Hemoglobin S, and Paternal Lineage in a Roman-Period Individual from Anatolia.

(1) Background: Integrated bioarchaeological approaches combining osteological and ancient DNA analyses provide powerful insights into health, disease, and population history in past societies. However, the relationship between rare skeletal variations, genetic disorders, and ancestry remains insufficiently explored within single individuals. This study aimed to investigate the combined osteological, paleopathological, and genetic characteristics of a Roman-period individual from southwestern Anatolia. (2) Methods: A multidisciplinary approach was applied to the skeletal remains of an adult ma

Ancient DNA of Metagonimus yokogawai Recovered from Joseon Period Human Remains Newly Discovered at Goryeong County in South Korea.

PURPOSE: Metagonimiasis, commonly seen in East Asian countries, is a parasitic disorder caused by definitive hosts' ingestion of undercooked freshwater fishes. Recently, genetic analysis has proved 28S rRNA and cytochrome c oxidase subunit I (COI) mtDNA gene to be a successful marker differentiating species of the genus Metagonimus. In the present study, using specimens from the newly discovered Joseon Dynasty human remains of Goryeong, we obtained updated genetic data on genus Metagonimus, which was also prevalent during the Joseon period. METHODS: The ancient DNA (aDNA) was retrieved from t

DNA typing of ancient parasite eggs from environmental samples identifies human and animal worm infections in Viking-age settlement.

Ancient parasite eggs were recovered from environmental samples collected at a Viking-age settlement in Viborg, Denmark, dated 1018-1030 A.D. Morphological examination identified Ascaris sp., Trichuris sp., and Fasciola sp. eggs, but size and shape did not allow species identification. By carefully selecting genetic markers, PCR amplification and sequencing of ancient DNA (aDNA) isolates resulted in identification of: the human whipworm, Trichuris trichiura , using SSUrRNA sequence homology; Ascaris sp. with 100% homology to cox1 haplotype 07; and Fasciola hepatica using ITS1 sequence homology

Embryophyte-wide detection of natural Agrobacterium-mediated horizontal gene transfer reveals an ancient role for mini T-DNAs.

Agrobacterium transfers DNA into plant cells, leading to tumors, hairy roots (HR), and natural genetically modified organisms (nGMOs). Transferred DNAs (T-DNAs) from agrobacteria and T-DNA-derived cellular T-DNAs (cT-DNAs) from nGMOs vary considerably and may carry up to 15 different genes. Among these, opine synthase (ops) genes encode the synthesis of opines used as nutrients by the agrobacteria. Earlier studies predicted large numbers of naturally transformed plant species, but only few have been identified and studied so far. We therefore developed a general method to detect cT-DNAs in all

Sedimentary ancient DNA as part of a multimethod paleoparasitology approach reveals temporal trends in human parasitic burden in the Roman period.

The detection of parasite infections in past populations has classically relied on microscopic analysis of sediment samples and coprolites. In recent years, additional methods have been integrated into paleoparasitology such as enzyme-linked immunosorbent assay (ELISA) and ancient DNA (aDNA). The aim of this study was to evaluate a multimethod approach for paleoparasitology using microscopy, ELISA, and sedimentary ancient DNA (sedaDNA) with a parasite-specific targeted capture approach and high-throughput sequencing. Using 26 samples dating from c. 6400 BCE to 1500 CE that were previously anal

Analysis of the three Yersinia pestis CRISPR loci provides new tools for phylogenetic studies and possibly for the investigation of ancient DNA.

The precise nature of the pathogen having caused early plague pandemics is uncertain. Although Yersinia pestis is a likely candidate for all three plague pandemics, the very rare direct evidence that can be deduced from ancient DNA (aDNA) analysis is controversial. Moreover, which of the three biovars, Antiqua, Medievalis or Orientalis, was associated with these pandemics is still debated. There is a need for phylogenetic analysis performed on Y. pestis strains isolated from countries from which plague probably arose and is still endemic. In addition there exist technical difficulties inherent

Comparison of Illumina NovaSeq 6000, GeneMind SURFSeq 5000, Salus Evo, and MGI DNBSEQ-G400 for Ancient DNA Whole-Genome Sequencing.

Background: Ancient DNA (aDNA) research is one of the biological fields that has been transformed by the development of next-generation sequencing (NGS). To date, Illumina sequencing has dominated aDNA research. However, its high costs are driving the adoption of alternative sequencing platforms. Combining data generated on different platforms may introduce artifacts arising from platform-specific errors. This study systematically compared the performance of four sequencing platforms (Illumina NovaSeq 6000, GeneMind SURFSeq 5000, Salus Evo, and MGI DNBSEQ-G400) for whole-genome aDNA sequencing

Molecular Diagnosis of Syphilis in Brazilian Ambulatory Patients: Detection of Treponema pallidum subsp. pallidum in Serum Using Ancient DNA Protocols.

The rising incidence of syphilis in recent decades underscores the need to improve diagnostic and control strategies. The infection caused by Treponema pallidum subsp. pallidum is commonly diagnosed using serological tests. However, these methods exhibit limitations in the early or late stages of disease, when antibody responses and/or bacterial loads are low. Molecular biology detection using serum samples is also hampered by low circulating bacterial loads during asymptomatic periods. Ancient DNA (aDNA) studies apply methods adapted to recovering low concentrations and degraded DNA. In this

Ancient DNA Investigation of a Medieval German Cemetery Confirms Long-Term Stability of CCR5-Δ32 Allele Frequencies in Central Europe.

The CCR5-Δ32 mutation present in European populations is among the most prominently debated cases of recent positive selection in humans. This allele, a 32-bp deletion that renders the T-cell CCR5 receptor nonfunctional, has important epidemiological and public health significance, as homozygous carriers are resistant to several HIV strains. However, although the function of this allele in preventing HIV infection is now well described, its human evolutionary origin is poorly understood. Initial attempts to determine the emergence of the CCR5-Δ32 allele pointed to selection during the 14th-cen

Leprosy in a Lombard-Avar cemetery in central Italy (Campochiaro, Molise, 6th-8th century AD): ancient DNA evidence and demography.

BACKGROUND: The study of past infectious diseases increases knowledge of the presence, impact and spread of pathogens within ancient populations. AIM: Polymerase chain reaction (PCR) was used to examine bones for the presence of Mycobacterium leprae ancient DNA (aDNA) as, even when leprosy is present, bony changes are not always pathognomonic of the disease. This study also examined the demographic profile of this population and compared it with two other populations to investigate any changes in mortality trends between different infectious diseases and between the pre-antibiotic and antibio

Molecular characterization of mitochondrial Amerindian haplogroups and the amelogenin gene in human ancient DNA from three archaeological sites in Lambayeque - Peru.

Important pre-Inca civilizations, known by their great political and religious structures, inhabited the northern coast of Peru. Archeological and anthropological studies have shown that people from these villages have hierarchical strata, but the genetic structure has been poorly studied. Here, we aimed to perform a molecular characterization of the Amerindian maternal lineages and the amelogenin gene in skeletons collected from three archeological sites in Lambayeque. Ancient DNA (aDNA) samples were analyzed with conventional PCR to assess the nine-base pair (9 bp) deletion corresponding to

Genetic characterization and meta-analysis of the population of the Northern Black Sea region in the 1st millennium CE based on ancient DNA data.

Over the past two decades, the introduction of whole-genome sequencing analysis of ancient DNA has led to a breakthrough in archaeogenetic research, significantly expanding our understanding of human genetic history. In this context the Northern Black Sea region during first millennium CE (1-1,000 CE) is of particular relevance, as it remained a hub of intense cultural exchange and migration. Despite its historical importance, ancient genomic data from this period remains scarce, and a comprehensive synthesis of existing findings is lacking. This study presents a systematic review and meta-ana

Ancient DNA confirmation of lepromatous leprosy in a skeleton with concurrent osteosarcoma, excavated from the leprosarium of St. Mary Magdalen in Winchester, Hants., UK.

To establish a biological profile and disease aetiologies for one of four burials recovered during a Time Team dig at the St. Mary Magdalen leprosarium, Winchester, UK in AD 2000. Osteological techniques were applied to estimate age at death, biological sex, stature and pathology. Visual assessment of the material was supplemented by radiographic examination. Evidence for leprosy DNA was sought using ancient DNA (aDNA) analysis. The remains are those of a male individual excavated from a west-east aligned grave. The skeleton shows signs of two pathologies. Remodelling of the rhino-maxillary ar

The plague of Athens (430-427 BC): Integration of historical, clinical, and ancient DNA data supports an invasive non-typhoidal Salmonella enterica etiology.

The Plague of Athens represents one of the earliest and most detailed descriptions of a large-scale epidemic in recorded history. Emerging in 430 BC during the second year of the Peloponnesian War, it caused extensive mortality, including the death of Pericles, the leader of Athenian democracy at the time. Thucydides, the famous historian, provides an eyewitness account with a rich clinical narrative encompassing symptomatology, complications, mortality rates, epidemic dynamics, post-infection immunity, and animal involvement. In this narrative review, we integrate Thucydides' account with mod
