[Study on molecular phylogeny of Schistosoma sinensium based on nuclear ribosomal DNA].

OBJECTIVE: To determine the phylogenetic relationships between Schistosoma sinensium and other Schistosomatid species using DNA sequence data. Two segments of the nuclear rDNA repeat, the second internal spacer (ITS2) and large subunit (LSU/12S) were selected for sequencing. METHODS: Adult worms stored in 100% methanol were washed 3 times with 0.1 x TE (pH8.0) and the genomic DNA was extracted by the GNT-K method. The target regions were amplified by PCR using specific primers. The PCR products were purified before ligation into the plasmid pT-adv (Clontech). Recombinant plasmids were amplifi

Phenotypic and molecular characterization of Madurella pseudomycetomatis sp. nov., a novel opportunistic fungus possibly causing black-grain mycetoma.

A case of black-grain mycetoma occurring on the lower jaw with an odontogenic origin, which to our knowledge is the first case reported in China, is presented here. The clinical manifestation, histopathological morphology, and microbiological features are described. The new species, Madurella pseudomycetomatis, isolated from the black grains discharged by this patient, was analyzed using sequence data of the multiloci of ribosomal DNA (rDNA) and its ability to ferment carbohydrate as well as morphology. The analyses of the internal transcribed spacer (ITS) region and the D1/D2 hypervariable re

Morphological and molecular characterisation of Ditylenchoides morocciensis sp. nov. (Nematoda: Anguinoidea) from Morocco.

The present study describes a new Ditylenchoides species, isolated from Meknès, Morocco, during nematode surveys conducted to investigate the biodiversity of plant-parasitic nematodes in Mediterranean olive groves and adjacent patches of natural vegetation. Application of integrative taxonomical approaches clearly verified that it is a new species designated herein as Ditylenchoides morocciensis sp. nov., also representing the first report of the genus in Morocco. The new species is parthenogenetic, characterised by a short body 460 (373-528 μm); stylet delicate, relatively short, 8.7 (8.0-9.0

Comparison of the bacterial community structure within the equine hindgut and faeces using Automated Ribosomal Intergenic Spacer Analysis (ARISA).

The horse's hindgut bacterial ecosystem has often been studied using faecal samples. However few studies compared both bacterial ecosystems and the validity of using faecal samples may be questionable. Hence, the present study aimed to compare the structure of the equine bacterial community in the hindgut (caecum, right ventral colon) and faeces using a fingerprint technique known as Automated Ribosomal Intergenic Spacer Analysis (ARISA). Two DNA extraction methods were also assessed. Intestinal contents and faeces were sampled 3 h after the morning meal on four adult fistulated horses fed mea

Multi-Marker Approach for the Identification of Different Heterodera Species (Nematoda: Heteroderidae).

Cyst nematodes of the genus Heterodera are important plant-parasitic nematodes that cause significant crop losses worldwide but are often overlooked due to their non-specific symptoms and complex biology. This study assessed Heterodera diversity in Portugal using an integrative molecular approach based on four genetic markers (mtCOI, 18S rDNA, ITS, and 28S rDNA). Five valid species were identified: Heterodera cruciferae, H. mani, H. schachtii, H. trifolii, and H. zeae, with H. mani reported for the first time in the country. A distinct taxon from Coimbra (central Portugal) may represent a new

Morphological and phylogenetic analyses of Gonatobotryum (Dothideaceae, Dothideales), and the description of a new species from a mangrove sediment in Brazil.

Gonatobotryum is an anamorphic fungus reported mainly as a saprophyte, parasite, or endophyte. The genus has traditionally been identified based on morphological features, with its phylogenetic placement remaining uncertain due to the lack of molecular data. During the analysis of fungi obtained from mangrove sediments in Brazil, nine isolates were morphologically identified as Gonatobotryum and subsequently analysed using the internal transcribed spacer (ITS) region, including the intervening 5.8S nRNA gene and the nuclear ribosomal RNA large subunit (LSU) gene, leading to the description of

Description and phylogenetic position of a new Serpentirhabdias (Nematoda: Rhabdiasidae) from Hydrodynastes gigas (Serpentes: Colubroidea) in Pantanal, Brazil.

Serpentirhabdias Tkach, Kuzmin and Snyder, 2014 comprises about 23 species of lung-dwelling nematodes parasites of snakes distributed worldwide. Among them, eight species were found in Brazil. We describe and examine the phylogenetic position of Serpentirhabdias kuzmini n. sp., a parasite of Hydrodynastes gigas from the Pantanal, Brazil. The new species is characterized by a triangular oral opening, absence of a buccal capsule, presence of onchia in the oesophastome, excretory glands longer than the esophagus, and a gradually tapering tail. Moreover, comparative analysis of partial sequences o

Genetic markers for strongylid nematodes of livestock defined by PCR-based restriction analysis of spacer rDNA.

Twenty-four species of parasitic nematode (order Strongylida) from sheep, goats, cattle or pigs were characterised using a polymerase chain reaction-linked restriction fragment length polymorphism technique (PCR-RFLP). The ribosomal (r)DNA region spanning the first internal transcribed spacer (ITS-1), 5.8S rRNA gene and the second internal transcribed spacer (ITS-2) (designated ITS) was amplified from genomic DNA by polymerase chain reaction (PCR), digested separately with four restriction endonucleases (RsaI, HinfI, DraI or NlaIII) and the fragments separated by agarose gel electrophoresis. Borrelia burgdorferi genotype predicts the capacity for hematogenous dissemination during early Lyme disease.

BACKGROUND: Lyme disease, the most common tickborne disease in the United States, is caused exclusively by Borrelia burgdorferi sensu stricto in North America. The present study evaluated the genotypes of >400 clinical isolates of B. burgdorferi recovered from patients from suburban New York City with early Lyme disease associated with erythema migrans; it is the largest number of borrelial strains from North America ever to be investigated. METHODS: Genotyping was performed by restriction fragment-length polymorphism polymerase chain reaction analysis of the 16S-23S ribosomal RNA spacer and

Central European parasitic flatworms of the family Renicolidae Dollfus, 1939 (Trematoda: Plagiorchiida): molecular and comparative morphological analysis rejects the synonymization of Renicola pinguis complex suggested by Odening.

The Renicolidae are digenean parasites of piscivorous and molluscivorous birds. Although they exhibit few morphological autapomorphies and are highly variable, the numerous suggested re-classifications within the family have never been supported by any molecular analyses. We address the possible synonymization of species within the Renicola pinguis complex suggested previously by Odening. We provide and analyse sequences of two nuclear (ITS2, 28S rDNA) and two mitochondrial (CO1, ND1) DNA loci of central European species of the Renicolidae, namely Renicola lari, Renicola pinguis and Renicola s

First Description of the Causal Agent of Fairy Ring of Cranberry and Its Novel Dispersal Method.

Fairy ring is an economically important disease of cranberry in the northeastern United States, yet its causal agent has remained unresolved for more than a century. Field sampling, targeted isolation, morphological and molecular characterization, pathogenicity tests, and population analyses consistently associated the disease with the genus Helicobasidium (Pucciniomycetes), a known teleomorph of the violet root rot fungus. Internal transcribed spacer and large subunit rDNA sequences placed all isolates from New Jersey and Massachusetts in a distinct lineage within H. longisporum, designated h

SARCOCYSTIS INFECTIONS IN TONGUES OF COUGARS (PUMA CONCOLOR COUGUAR) IN COLORADO.

Sarcocystis species are intracellular protozoan parasites that infect a wide range of intermediate and definitive hosts, including felids. This study reports Sarcocystis species in the tongues of cougars (Puma concolor couguar) in Colorado for the first time. By light microscope, sarcocysts were found in 4 of 30 cougars; they appeared to represent more than 2 morphologic types by light microscopy. One morphologic type had serrations on the sarcocyst wall and resembled Sarcocystis felis reported from domestic and wild felids. In the second type, serrations were not visible on the sarcocyst wall

Cryptic Aspergillus from clinical samples in the USA and description of a new species in section Flavipedes.

BACKGROUND: In the last few decades there has been an emergence of cryptic Aspergillus as agents of human infections due to the increase in immunocompromised population and to the improvement of identification tools. METHODS: Continuing our study on Aspergillus isolates from clinical origin deposited in a mycological reference centre in the United States, we selected 37 isolates belonging to less common sections of the genus, to study their species diversity and detect cryptic species by using a polyphasic approach. RESULTS: From this set of isolates, a total of 16 species were identified; t

Malassezia Species Isolated from Avian Carcasses in a Japanese Zoo, and the Description of Malassezia phoenicopteridarum sp. nov.

Studies on the diversity of lipid-dependent Malassezia yeast that colonize avian hosts remain limited. In this study, we investigated Malassezia species diversity by sampling cerumen from 12 carcasses (representing 10 species across eight genera and four families) housed in a Japanese zoo. Using culture-based and molecular phylogenetic methods, four Malassezia strains were isolated from a lesser flamingo and a toco toucan. Molecular phylogenetic analyses based on the nucleotide sequences of the 26S rRNA D1/D2 domain, rRNA internal transcribed spacer region, and β-tubulin gene identified JCM 39

Morphological and molecular data on Pseudozoogonoides ugui Shimazu, 1974 (Digenea: Microphalloidea: Zoogonidae) ex Pseudaspius hakonensis (Günther, 1877) and taxonomic problems in Zoogoninae genera.

New morphological and molecular data were generated for trematodes recovered from the intestines of the fish Pseudaspius hakonensis from two locations in the south of the Russian Far East. Morphologically, these trematodes are identical to Pseudozoogonoides ugui (Microphalloidea: Zoogonidae) from Japan. According to results of phylogenetic analysis based on 28S rDNA sequence data, P. ugui was closely related to Zoogonoides viviparus, and P. subaequiporus appears as a sister taxon to these two species. Genetic distance values, calculated based on both 28S rDNA and ITS2 rDNA, between P. ugui and

Genetic diversity among clinical isolates of Acremonium strictum determined during an investigation of a fatal mycosis.

Primarily saprophytic in nature, fungi of the genus Acremonium are a well-documented cause of mycetoma and other focal diseases. More recently, a number of Acremonium spp. have been implicated in invasive infections in the setting of severe immunosuppression. During the course of routine microbiological studies involving a case of fatal mycosis in a nonmyeloablative hematopoietic stem cell transplant patient, we identified a greater-than-expected variation among strains previously identified as Acremonium strictum by clinical microbiologists. Using DNA sequence analysis of the ribosomal DNA in
