Injection of a GABA antagonist into the mesopontine reticular formation abolishes haloperidol-induced catalepsy in rats.

THE pedunculopontine nucleus and its adjacent structure of the mesopontine reticular formation are known as a mesencephalic locomotor region, since either electrical or chemical stimulation of these regions induces locomotion in decerebrate animals. In parkinsonism, it is presumed that the pedunculopontine nucleus is under GABAergic overinhibition from the basal ganglia. To reveal the behavioural effects of GABAergic disinhibition of the mesopontine reticular formation in parkinsonism, picrotoxin, a GABAA antagonist, (5 or 10 ng/ 0.25 microliter) or vehicle was injected unilaterally into the m

[The effects of hyperventilation upon the spinal pain modulating system (second report)].

The purpose of this study is to investigate the effect of hyperventilation on the spinal pain modulating system by using naloxone. Under enflurane anaesthesia, cats were prepared with midcollicular decerebration and lumbar laminectomy. The spinal cord was transected at T12-L1. WDR cells, responding primarily to noxious peripheral stimuli, were sampled with a microelectrode at the depth of 2,000 microns from the cord dorsum. Following the control period, ventilation was adjusted to produce hypocapnia of PCO2 20-25 mmHg. After activities of WDR cells were well suppressed, naloxone 0.1 mg.kg-1 wa

Primary brainstem injury without persistent coma and decerebrate rigidity. Report of 2 cases.

Two patients with blunt head injury were examined by CT scanning. It was found that there was brainstem hemorrhage with unremarkable changes in the other parts of the brain. Physical examination showed signs of upper brainstem lesion such as discoordination, bilateral Babinski signs, and internuclear ophthalmic palsy without persistent coma and decerebrate rigidity. The diagnosis of primary brainstem injury, well healed, was established. Basing on the results of these 2 cases, we believe that primary brainstem injury does exist, but it is not necessarily associated with persistent coma and dec

Cardiovascular and respiratory effects of an opioid kappa agonist ethylketazocine and sigma agonist N-allylnormetazocine in acutely decerebrated dogs.

Effects of opioid kappa agonist ethylketazocine (EKC), sigma agonist (+-)-N-allylnormetazocine (NANM), and naloxone alone and in combination on mean blood pressure (MBP), heart rate (HR), respiratory rate (RR) and minute volume (MV) were studied in acutely decerebrated dogs. EKC (0.5 mg/kg) decreased HR, MBP, RR and MV. Post-EKC NLX increased RR and MV and reversed the bradycardia and hypotension produced by EKC. NANM (1 mg/kg) produced respiratory depression and tachycardia without changing MBP. Post-NANM NLX antagonized tachycardia, increased MBP, however did not significantly change RR and

Hypoxia inhibits abdominal expiratory nerve activity.

Our purpose was to examine the influence of steady-state changes in chemical stimuli, as well as discrete peripheral chemoreceptor stimulation, on abdominal expiratory motor activity. In decerebrate, paralyzed, vagotomized, and ventilated cats that had bilateral pneumothoraces, we recorded efferent activity from a phrenic nerve and from an abdominal nerve (cranial iliohypogastric nerve, L1). All cats showed phasic expiratory abdominal nerve discharge at normocapnia [end-tidal PCO2 38 +/- 2 Torr], but small doses (2-6 mg/kg) of pentobarbital sodium markedly depressed this activity. Hyperoxic hy

Responses of neurons in the lateral cervical nucleus of the cat to noxious cutaneous stimulation.

The majority of neurons at the origin of the spinocervical tract are driven by noxious stimulation of their receptive fields. Surprisingly, previous studies have encountered only a small percentage of nociceptive neurons within the terminus of the spinocervical tract, the lateral cervical nucleus (LCN). To determine if previous reports have underestimated the proportion of nociceptive LCN neurons, 129 neurons within the nucleus were physiologically identified and examined in cats prepared using three different methods. Fifty-nine percent of the neurons studied in unanesthetized cats that were

Single unit analysis of the posteroventral cochlear nucleus of the decerebrate cat.

Single unit recordings were obtained in the cochlear nuclear complex of the unanesthetized, decerebrate cat. Sixty-six of 282 units were localized to the posteroventral cochlear nucleus, 17 from the multipolar cell area and 49 from the octopus cell area. Spontaneous rates ranged from less than 1 to 75 spikes per second in the multipolar cell area and from less than 1 to 135 spikes per second in the octopus cell area. Poststimulus time histograms revealed four response types, at the best frequency, in the posteroventral cochlear nucleus. These responses were: (1) primary-like (maximum response

Modulation of hypoxic depressions of ventilatory activity in the newborn piglet by mesencephalic mechanisms.

In neonates, ventilatory responses to hypoxia are 'biphasic,' with an augmentation followed by a decline. The hypoxia-induced augmentations in ventilation are attenuated and the depressions are accentuated following denervation of the peripheral chemoreceptors. Piglets that were decerebrated at a rostral mesencephalic level exhibited these hypoxia-induced depressions. These depressions were lessened following transection through the caudal mesencephalon. Mesencephalic mechanisms play a fundamental role in the brainstem regulation of ventilatory responses to hypoxia.

Reversal of fusimotor reflex responses during locomotion in the decerebrate cat.

The effect of brief trains of electrical stimulation, at 2, 3 and 20 x threshold (T), of cutaneous afferents in the medial plantar nerve on the discharges of single medial gastrocnemius static and dynamic gamma-efferents has been investigated at rest and during locomotion in a decerebrate cat preparation. The units were classified as dynamic (10 units) or static (10 units) indirectly on the basis of their resting and locomotor discharge characteristics. Responses were assessed by calculating the change in mean gamma-rate during the 100 ms after stimulus onset compared with a control period. At

The different changes of phrenic nerve activity and frequency elicited by microinjection of L-glutamic acid into ventrolateral nucleus of the tractus solitarius in cats.

There is only limited information of the neurotransmitters in central respiratory control. L-glutamic acid has been proposed as the primary neurotransmitter in the nucleus of the tractus solitarius (NTS) in cats. To test whether there is a respiratory effect of glutamic acid in the respiratory center or not, we microinjected L-glutamic acid (1 M, 0.1 microliter) via 1 microliter Hamilton microsyringe into the ventrolateral nucleus of NTS, namely dorsal respiratory group (DRG) over two-second intervals with continuous monitoring of phrenic nerve activity (PNA), frequency (f), end tidal CO2, blo

Bulbar respiratory activity during defensive airways reflexes in cats.

Experiments in anaesthetized nonparalyzed cats indicated that mechanical stimulation of the airways and a longitudinal split of the brainstem cause marked alterations of the regulatory function of the bulbar respiratory neurones. The resulting changes in breathing and in defensive airways reflexes depend on intactness of the relevant structures of both halves of the "respiratory centre" and their interconnections.

Effects of microinjection of a cholinergic agonist into the locus coeruleus on the gain of vestibulospinal reflexes in decerebrate cats.

1. Experiments were performed in precollicular decerebrate cats to determine whether activation of locus coeruleus (LC) neurons elicited by local injection of the cholinergic agonist carbachol modifies the dynamic characteristics of responses of forelimb extensors to selective stimulation of labyrinth receptors resulting from roll tilt of the animal. 2. Injection of 0.1-0.4 microliter (usually 0.25 microliter) of carbachol at a concentration of 0.02-0.1 micrograms/microliter of sterile saline into the LC of one side, which slightly increased the tonic contraction of limb extensors ipsilateral

Responses in the posterior lobe of the rat cerebellum to electrical stimulation of cutaneous afferents to the snout.

1. Responses in the cortex of the posterior lobe of the cerebellum to electrical stimulation of afferent fibres from the skin of the snout have been analysed in decerebrate and pentobarbitone-anaesthetized rats by means of surface and microelectrode records. Single shocks were applied either to the exposed follicles of the mystachial vibrissae or to the infraorbital branch of the trigeminal nerve. 2. In decerebrate rats responses were mediated only via mossy fibre afferents. Stimulation of one side of the snout yielded responses with mean latency 2.4 ms throughout the uvula (largest ipsilatera

Respiratory patterns during progressive asphyxia in newborn rabbits.

During acute progressive asphyxia an abrupt transition occurs from regular respiratory efforts to primary apnea and gasping. To study this transition we measured inspiratory duration (TI), expiratory duration (TE), maximal inspiratory tracheal pressure (Pmax), electromyographic activity (EMG) of different muscle groups, and the electrocorticogram (ECoG) of 3- to 5-day-old rabbits following airway occlusion at functional residual capacity (FRC). The onset of primary apnea was associated with other central nervous system events as indicated by decerebrate posture and maximal EMG activity which w

The control of the diuresis following a blood meal in females of the yellow fever mosquito Aedes aegypti (L).

Control of post-feeding diuresis in females of the mosquito Aëdes aegypti has been studied by means of a weighing technique and simple surgical procedures. The primary controlling factor is (as in the larva) a nervous feedback mechanism and not an increased production of diuretic hormone. As the mosquito ingests blood, sensory information from the distending abdomen reaches the mid gut via the nerve cord, brain and stomatogastric system. This information probably inhibits retroperistaltic movements of the mid gut allowing fluid from the Malpighian tubules (which seems to be produced continuall
