Pulse Electron Double Resonance Detected Multinuclear NMR Spectra of Distant and Low Sensitivity Nuclei and Its Application to the Structure of Mn(II) Centers in Organisms.

The ability to characterize the structure of metal centers beyond their primary ligands is important to understanding their chemistry. High-magnetic-field pulsed electron double resonance detected NMR (ELDOR-NMR) is shown to be a very sensitive approach to measuring the multinuclear NMR spectra of the nuclei surrounding Mn(II) ions. Resolved spectra of intact organisms with resonances arising from (55)Mn, (31)P, (1)H, (39)K, (35)Cl, (23)Na, and (14)N nuclei surrounding Mn(2+) centers were obtained. Naturally abundant cellular (13)C could be routinely measured as well. The amplitudes of the (14

Genomic signatures of radiation stress adaptations in Kocuria rhizophila: insights from strain 301 of the Jáchymov radon springs.

Several strains of Kocuria rhizophila have been reported to tolerate ionizing radiation and other environmental stresses; however, this phenotype is unevenly distributed across the species. Here, we characterize K. rhizophila strain 301, isolated from chronically radioactive radon springs in Jáchymov (Czech Republic). Strain 301 displayed exceptional stress resilience, retaining approximately 10% viability after exposure to 1.0 kGy of γ-irradiation, and maintaining ~20% survival following 30 days of desiccation. These responses sharply contrasted with the pronounced sensitivity of the type str

The Effects of Milking and Cleaning Procedures on the Quality and Microbiome of Raw Goat Milk.

The rising popularity of raw goat milk has heightened concerns about its safety. This study examined how differences in milking and cleaning practices influence the quality and microbiota of goat milk from small-scale Oregon farms during July and August. Milk quality was assessed through somatic cell counts (SCCs) and components, while microbiota was evaluated using viable counts and 16S rRNA sequencing. Sequencing revealed a diverse microbial community, dominated by genera such as Staphylococcus, Escherichia-Shigella, and Pseudomonas, with pathogenic taxa like Salmonella and Campylobacter lar

Survival of thermophilic and hyperthermophilic microorganisms after exposure to UV-C, ionizing radiation and desiccation.

In this study, we investigated the ability of several (hyper-) thermophilic Archaea and phylogenetically deep-branching thermophilic Bacteria to survive high fluences of monochromatic UV-C (254 nm) and high doses of ionizing radiation, respectively. Nine out of fourteen tested microorganisms showed a surprisingly high tolerance against ionizing radiation, and two species (Aquifex pyrophilus and Ignicoccus hospitalis) were even able to survive 20 kGy. Therefore, these species had a comparable survivability after exposure to ionizing radiation such as Deinococcus radiodurans. In contrast, there

Producing alcohol and salt stress tolerant strain of Saccharomyces cerevisiae by heterologous expression of pprI gene.

INTRODUCTION: Ethanol is considered a comparatively clean biofuel, and its large scale production has been a long time concern. Saccharomyces cerevisiae has proven to be the suitable microorganism for large scale ethanol production, but production of other alcohols like butanol and using lignocellulosic substrates is restricted due to lacking tolerance toward toxicity of alcohols, and compounds released from substrates. This study aimed to produce a tolerant strain by using pprI gene of Deinococcus radiodurans. MATERIAL AND METHOD: pprI gene was introduced into Saccharomyces cerevisiae. To ev

Therapeutic potency and the related mechanism of deinoxanthin in experimental animal and cell models of periodontitis.

Periodontitis is a chronic inflammatory disease that leads to the destruction of periodontal tissue, ultimately resulting in tooth loss. Studies have aimed to develop biomaterials that effectively prevent inflammatory responses and oxidative stress in the progression of periodontitis, without adverse side effects. Here, we explored whether Deinococcus radiodurans-derived deinoxanthin (DEIX) protects against alveolar bone loss and connective tissue degradation in an experimental rat model of periodontitis and investigated the related mechanisms using human-derived periodontal ligament cells (hP

Plant endophytes: unveiling hidden applications toward agro-environment sustainability.

Endophytic microbes are plant-associated microorganisms that reside in the interior tissue of plants without causing damage to the host plant. Endophytic microbes can boost the availability of nutrient for plant by using a variety of mechanisms such as fixing nitrogen, solubilizing phosphorus, potassium, and zinc, and producing siderophores, ammonia, hydrogen cyanide, and phytohormones that help plant for growth and protection against various abiotic and biotic stresses. The microbial endophytes have attained the mechanism of producing various hydrolytic enzymes such as cellulase, pectinase, x

Alteration in microbial community and antibiotic resistance genes mediated by microplastics during wastewater ultraviolet disinfection.

Microplastics (MPs) could be as a vector to colonize microorganisms and antibiotic resistance gene (ARGs) in surface water. However, little information is known regarding their changes by the presence of MPs in wastewater treatment. Here, the effects of different concentrations and sizes of polystyrene microplastics (PSMPs) on the distribution and removal of microbial communities and ARGs under ultraviolet disinfection of urban sewage have been systematically studied. Results showed that the presence of MPs altered abundance and functions of microorganisms in wastewater, despite different effe

Deep subsurface rock-hosted chemolithotrophic bacterial communities exhibited differential CO2 assimilation and bioconversion potential under varying oxygen level.

Deep continental subsurface hosts diverse microbial ecosystems that are primarily driven by chemolithoautotrophy. In this study, we investigated deep continental igneous rock-hosted bacterial populations enriched under microoxic (ME) and anoxic (AE) conditions. Metataxonomic, metagenomics and metabolomics approaches, along with physiological analyses, were performed to elucidate community composition, CO2 utilization and possible bioconversion potential of subsurface rock enrichment cultures under chemolithoautotrophic conditions. Following prolonged incubation, ME enrichments resulted in high

Integrative Identification of Anti-Photoaging Peptides From Stress-Tolerant Microorganisms via Machine Learning and KEAP1-NRF2 Docking.

Excess reactive oxygen species generated by ultraviolet exposure cause photoaging by degrading collagen and inhibiting its synthesis. This study presents a comprehensive strategy connecting the biological stress responses of γ-irradiated microorganisms to the discovery of novel anti-photoaging peptides. We profiled the radiation-regulated transcriptomes of Deinococcus radiodurans and Cryptococcus neoformans, focusing on DNA repair and oxidative stress responses. From these datasets, peptide libraries were generated in silico, filtered for biochemical properties, and prioritized using a seven-c

Pleural fluid microbiota as a biomarker for malignancy and prognosis.

Malignant pleural effusions (MPE) complicate malignancies and portend worse outcomes. MPE is comprised of various components, including immune cells, cancer cells, and cell-free DNA/RNA. There have been investigations into using these components to diagnose and prognosticate MPE. We hypothesize that the microbiome of MPE is unique and may be associated with diagnosis and prognosis. We compared the microbiota of MPE against microbiota of pleural effusions from non-malignant and paramalignant states. We collected a total of 165 pleural fluid samples from 165 subjects; Benign (n = 16), Paramalign

Biotransformation-guided purification of a novel glycoside derived from the extracts of Chinese herb Baizhi.

Our pursuit of new compounds with enhanced bioavailability and bioactivity prompted us to employ the biotransformation-guided purification (BGP) approach which leverages proficient in vitro biotransformation techniques. Angelica dahurica roots, also called Baizhi in Chinese traditional medicine, are famous for their anti-inflammatory and analgesic properties. Herein, we applied the BGP methodology to Baizhi extracts, employing Deinococcus geothermalis amylosucrase (DgAS), an enzyme demonstrating catalytic competence across diverse substrates, for biotransformation. Microbe-mediated synthesis of defect-rich CeO2 nanoparticles with oxidase-like activity for colorimetric detection of L-penicillamine and glutathione.

To enhance production efficiency, curtail costs, and minimize environmental impact, developing simple and sustainable nanozyme synthesis methods has been the focus of relevant research. In this report, graphite-coated CeO2 nanoparticles (CeO2 NPs) with multiple defects (Ce3+ defects, oxygen vacancies and carbon defects) were synthesized via the culture filtrate of the extremely radioresistant bacterium Deinococcus wulumuqiensis R12 (D. wulumuqiensis R12). The as-prepared CeO2 NPs exhibit remarkable oxidase (OXD)-like activity, efficiently catalyzing the oxidation of the chromogenic substrate 3

Temporal and spatial variation in the mechanisms used by microorganisms to form methylmercury in the water column of Changshou Lake.

The microbiome in artificial lake water and its impact on mercury (Hg) methylation remain largely unknown. We selected the largest artificial lake in southeastern china, Changshou Lake (CSL), which has high background levels of Hg, for our investigation of Hg transformation microorganisms. Five different sections of the water column of CSL were sampled during four seasons. The water samples were subjected to analysis of geochemical parameters, various Hg species and microbiome information. High concentrations of total mercury (THg) were detected in CSL in comparison with those found in natural

In Vivo Analysis of the Viable Microbiota and Helicobacter pylori Transcriptome in Gastric Infection and Early Stages of Carcinogenesis.

Emerging evidence shows that the human microbiota plays a larger role in disease progression and health than previously anticipated. Helicobacter pylori, the causative agent of gastric cancer and duodenal and gastric ulcers, was early associated with gastric disease, but it has also been proposed that the accompanying microbiota in Helicobacter pylori-infected individuals might affect disease progression and gastric cancer development. In this study, the composition of the transcriptionally active microbial community and H. pylori gene expression were determined using metatranscriptomic RNA se

Detoxification of toxins by bacillithiol in Staphylococcus aureus.

Bacillithiol (BSH), an α-anomeric glycoside of l-cysteinyl-d-glucosaminyl-l-malate, is a major low-molecular-mass thiol found in bacteria such as Bacillus sp., Staphylococcus aureus and Deinococcus radiodurans. Like other low-molecular-mass thiols such as glutathione and mycothiol, BSH is likely to be involved in protection against environmental toxins including thiol-reactive antibiotics. We report here a BSH-dependent detoxification mechanism in S. aureus. When S. aureus Newman strain was treated with monobromobimane and monochlorobimane, the cellular BSH was converted to the fluorescent S-c

A lupus-like syndrome develops in mice lacking the Ro 60-kDa protein, a major lupus autoantigen.

Antibodies against a conserved RNA-binding protein, the Ro 60-kDa autoantigen, occur in 24-60% of all patients with systemic lupus erythematosus. Anti-Ro antibodies are correlated with photosensitivity and cutaneous lesions in these patients and with neonatal lupus, a syndrome in which mothers with anti-Ro antibodies give birth to children with complete congenital heart block and photosensitive skin lesions. In higher eukaryotes, the Ro protein binds small RNAs of unknown function known as Y RNAs. Because the Ro protein also binds misfolded 5S rRNA precursors, it is proposed to function in a q

Case studies on potential G-quadruplex-forming sequences from the bacterial orders Deinococcales and Thermales derived from a survey of published genomes.

Genomes provide a platform for storage of chemical information that must be stable under the context in which an organism thrives. The 2'-deoxyguanosine (G) nucleotide has the potential to provide additional chemical information beyond its Watson-Crick base-pairing capacity. Sequences with four or more runs of three G nucleotides each are potential G-quadruplex forming sequences (PQSs) that can adopt G-quadruplex folds. Herein, we analyzed sequenced genomes from the NCBI database to determine the PQS densities of the genome sequences. First, we found organisms with large genomes, including hum

The ephemeral microbiota: Ecological context and environmental variability drive the body surface microbiota composition of Magellanic penguins across subantarctic breeding colonies.

Environmental microbes routinely colonize wildlife body surface microbiota. However, animals experience dynamic environmental shifts throughout their daily routine. Yet, the effect of ecological shifts in wildlife body surface microbiota has been poorly explored. Here, we sequenced the hypervariable region V3-V4 of the 16S rRNA gene to characterize the body surface microbiota of wild Magellanic penguins (Spheniscus magellanicus) under two ecological contexts: (1) Penguins walking along the coast and (2) Penguins sheltered underground in their nest, across three subantarctic breeding colonies i

Characterization of the duodenal bacterial microbiota in patients with pancreatic head cancer vs. healthy controls.

An increasing number of reports have demonstrated that there is an association between the presence of pathogenic microorganisms and pancreatic cancer. However, the role of the duodenal microbiota in pancreatic carcinogenesis remains unknown. In this study, duodenal mucosal microbiota was analyzed in 14 patients with pancreatic head cancer and 14 healthy controls using 16S rRNA gene pyrosequencing methods. Plasma endotoxin activity and the concentrations of the proinflammatory cytokine IL-6 and C-reactive protein (CRP) were measured in blood samples. The urea breath test was used to detect Hel
