Aqueous-phase hydroformylation of 1-octene using hydrophilic sulfonate salicylaldimine dendrimers.

Water-soluble dendritic ligands based on tris-2-(5-sulfonato salicylaldimine ethyl)amine (5) and DAB-(5-sulfonato salicylaldimine) (6) (DAB = diaminobutane) were synthesized by means of Schiff base condensation and sulfonation reactions. These dendritic ligands were fully characterized by (1)H NMR, (13)C NMR and FT-IR spectroscopy, elemental analysis and mass spectrometry. Dendritic ligands (5 and 6) in combination with [RhCl(COD)](2) (COD = 1,5-cyclooctadiene) were evaluated in aqueous biphasic hydroformylation of 1-octene. New water-soluble mononuclear 5-sulfonato propylsalicylaldimine Rh(i)

Dendrimers ended by non-symmetrical azadiphosphonate groups: synthesis and immunological properties.

The synthesis and characterization of new series of phosphorus-containing dendrimers ended by non-symmetrical azamonophosphonates, or azadiphosphonates, or azadiphosphonic acid salts are reported. The sodium salts of the non-symmetrical azadiphosphonic dendrimers are soluble in water. Their influence towards human immune blood cells is assayed ex vivo.

Exploring peptide dendrimers for intestinal lymphatic targeting: formulation and evaluation of peptide dendrimer conjugated liposomes for enhancing the oral bioavailability of Asenapine maleate.

Asenapine maleate (ASPM) is a second-generation atypical antipsychotic that is approved for treating acute schizophrenia and bipolar disorder in adults by the US FDA. The major downside of ASPM therapy is rapid, extensive first-pass hepatic metabolism following its oral administration with a very low oral bioavailability of < 2%. In this work, we developed ASPM nanoformulations conjugated with ligands such as arginine-glycine-aspartic acid (RGD) and peptide dendrimers (PDs) with the intention of improving the oral bioavailability of the drug by targeting it to the intestinal lymphatic system (

Azobenzene-Cored Amphiphilic Dendrimers: Molecular Photoswitching Controls Dendriplex Topology and Gene Delivery.

Azobenzene-cored ionizable amphiphilic Janus dendrimers (Azo-IAJDs) are introduced as molecularly defined, single-component vectors in which reversible photoswitching controls plasmid DNA (pDNA) condensation pathways, dendriplex topology, and transfection behavior. The amphiphiles combine asymmetrically substituted azobenzene cores with ionizable and lipophilic dendrons and undergo efficient, reversible E/Z photoisomerization under biologically relevant conditions. In the presence of pDNA, they form nanocomplexes displaying markedly different supramolecular organizations depending on the archi

Unexpected Temperature Behavior of Polyethylene Glycol Spacers in Copolymer Dendrimers in Chloroform.

We have studied copolymer dendrimer structure: carbosilane dendrimers with terminal phenylbenzoate mesogenic groups attached by poly(ethylene) glycol (PEG) spacers. In this system PEG spacers are additional tuning to usual copolymer structure: dendrimer with terminal mesogenic groups. The dendrimer macromolecules were investigated in a dilute chloroform solution by (1)H NMR methods (spectra and relaxations). It was found that the PEG layer in G = 5 generations dendrimer is "frozen" at high temperatures (above 260 K), but it unexpectedly becomes "unfrozen" at temperatures below 250 K (i.e., mel

Transdermal delivery of nonsteroidal anti-inflammatory drugs mediated by polyamidoamine (PAMAM) dendrimers.

Nonsteroidal anti-inflammatory drugs (NSAIDs) are among the most frequently used drugs in the world, primarily for symptoms associated with osteoarthritis and other chronic musculoskeletal conditions. However, adverse effects caused by oral administration, such as local or systemic disturbance in the gastrointestinal tract, have limited the clinical applications of these drugs. In the present study we have assessed the ability of polyamidoamine (PAMAM) dendrimers to facilitate transdermal delivery of NSAIDs, using Ketoprofen and Diflunisal as model drugs. Applying dendrimer nanoprobes to the diagnosis and evaluation of kidney fibrosis.

Renal fibrosis is the central pathological process driving the progression of chronic kidney disease (CKD), representing a major global health burden. Current diagnostics, like biopsy, are invasive and lack the sensitivity for early, repeated monitoring. This creates an urgent need for noninvasive, molecularly specific platforms. While conventional nanoparticles have been explored for imaging, they face critical structural and physiological hurdles, particularly navigating the glomerular filtration barrier (GFB), leading to unpredictable clearance and off-target accumulation. This review highl

Dendrimer-conjugated isotretinoin for controlled transdermal drug delivery.

BACKGROUND: In the present study, we aimed to develop a novel isotretinoin delivery model for treating skin diseases, revealing its potential advantages in drug delivery and targeted therapy. Using a self-assembly strategy, we grafted a dendrimer, based on a well-defined branched structure for nanomedical devices, with a well-defined nanoarchitecture, yielding spherical, highly homogeneous molecules with multiple surface functionalities. Accordingly, a self-assembled dendrimer-conjugated system was developed to achieve the transdermal delivery of isotretinoin (13cRA-D). RESULTS: Herein, 13cRA

Synthesis and Antimicrobial Properties of a Ciprofloxacin and PAMAM-dendrimer Conjugate.

Infections caused by bacteria resistant to antibiotics are an increasing problem. Multivalent antibiotics could be a solution. In the present study, a covalent conjugate between Ciprofloxacin and a G0-PAMAM dendrimer has been synthesized and tested against clinically relevant Gram-positive and Gram-negative bacteria. The conjugate has antimicrobial activity and there is a positive dendritic effect compared to Ciprofloxacin itself.

Dendrimer nanoarchitectures for cancer diagnosis and anticancer drug delivery.

Dendrimers are novel nanoarchitectures with unique properties including a globular 3D shape, a monodispersed unimicellar nature and a nanometric size range. The availability of multiple peripheral functional groups and tunable surface engineering enable the facile modification of the dendrimer surface with different therapeutic drugs, diagnostic agents and targeting ligands. Drug encapsulation, and solubilizing and passive targeting also equally contribute to the therapeutic use of dendrimers. In this review, we highlight recent advances in the delivery of anticancer drugs using dendrimers, as

Modification of regenerated cellulose membrane based on thiol dendrimer.

This work describes the modification of a highly swollen commercial regenerated cellulose (RC) membrane with a dendritic molecule (thiol DAB dendrimer of generation 3) by the dip coating method, and its possible use in electrochemical systems with saline solutions fluxes. Moreover, cellulosic membrane modification with dendrimer covered-CdSe quantum dot was also performed, since it allows dendrimer assortment by fluorescence. Changes in electrical, mechanical and diffusive membrane parameters were determined by impedance spectroscopy, elasticity curves and diffusional permeability measurements

Distinctive Fluorescence Response of Eosins Confined in PAMAM Dendrimers. A Spectroscopic Study.

Binding of Eos-B to PAMAM dendrimers was evaluated in buffered aqueous solutions, by means of absorption and fluorescence spectroscopies. Progressive changes in the spectra of the dye were recorded with increasing amounts of PAMAM. The corresponding binding constants (Kbind) were estimated by using a 1:1 equilibrium model and different fitting approximations were discussed. After analyzing six generations of PAMAM (G0-G5), a clear correlation of the Kbind values with the size of the dendrimer was observed. In addition, the effect of pH was also tested, recording a significant increase in the c

PEGylated Heterofunctional Dendrimers Enable Multivalent Diclofenac Delivery for ROS-Driven Anticancer Activity.

Cancer drug development faces escalating costs and limited success, driving interest toward drug repurposing strategies. Diclofenac, a widely used nonsteroidal anti-inflammatory drug (NSAID) with emerging anticancer potential, exhibits poor aqueous solubility and rapid systemic clearance, limiting its chemotherapeutic suitability. Here, we engineered PEGylated heterofunctional polyester dendrimers (HFDs) as modular nanocarriers that enable controlled multivalent presentation of diclofenac through orthogonal chemistry. Diclofenac was conjugated within the dendritic interior using copper(I)-cata

Cell internalization kinetics and surface charge accessibility of surface-modified PAMAM dendrimers.

Surface-modified PAMAM dendrimers have important applications in drug delivery, yet a gap remains about the role that surface functionalization plays on their cell internalization capacity. We examined the cell internalization kinetics of PAMAM dendrimers that were surface-modified with acetyl, folate and poly(ethylene glycol), as model functional groups differing in size, charge, and chemical functionality. Dendrimers with 25% functionalization were internalized by HEK cells, but with slower rates and lower maximum uptakes than the native dendrimer between 1-6 h of incubation. Dendrimers with
