A nitroalkene derivative of salicylate, SANA, induces creatine-dependent thermogenesis and promotes weight loss.

The emergence of glucagon-like peptide-1 agonists represents a notable advancement in the pharmacological treatment of obesity, yet complementary approaches are essential. Through phenotypic drug discovery, we developed promising nitroalkene-containing small molecules for obesity-related metabolic dysfunctions. Here, we present SANA, a nitroalkene derivative of salicylate, demonstrating notable efficacy in preclinical models of diet-induced obesity. SANA reduces liver steatosis and insulin resistance by enhancing mitochondrial respiration and increasing creatine-dependent energy expenditure in

The complexity of dementia development and its comorbidities: The collaborative cross-mouse population for multivarious tasks approach.

The rising incidence of dementia and associated neurodegenerative disorders poses a growing public health challenge. These conditions have traditionally been studied as isolated central nervous system disorders; however, emerging evidence suggests that broader systemic factors, including chronic inflammation, immune dysregulation, metabolic dysfunction, and genetic susceptibility, may also play a role. This review examines the interconnection between autoimmune diseases and metabolic syndromes in the pathogenesis and exacerbation of neurodegeneration. Conditions such as rheumatoid arthritis, s

[Obesity and type 2 diabetes mellitus (Update 2026)].

Obesity is a chronic disease with a variety of metabolic, mechanical and psychosocial complications, including a high risk of developing prediabetes and subsequently type 2 diabetes mellitus. The choice of antidiabetic treatment as well as concomitant treatment, increasingly takes this into account. Modern glucagon-like peptide 1 (GLP-1) analogues and the combined gastric inhibitory polypeptide (GIP)/GLP‑1 agonist tirzepatide play an important role in the combined treatment of obesity and type 2 diabetes mellitus. Metabolic surgery is currently indicated for people with type 2 diabetes mellitu

Non-alcoholic fatty liver disease and type 2 diabetes: An update.

The global prevalence of non-alcoholic fatty liver disease (NAFLD) is rising, along with the epidemic of diabesity. NAFLD is present in >70% of individuals with type 2 diabetes. Although the mutually detrimental relationship between NAFLD and type 2 diabetes has been well established, a multitude of recent studies have further shown that type 2 diabetes is closely linked to the development of cirrhosis, hepatocellular carcinoma, liver-related morbidity and mortality. In contrast, NAFLD also negatively impacts type 2 diabetes both in terms of its incidence and related adverse clinical outcomes,

Glycemic and lipid metabolic disorders in diabetic and non-diabetic patients bmi < 35 or > 35 before gastric bypass.

INTRODUCTION: Obesity and diabetes are diseases with high prevalence worldwide. There is currently no effective medical treatment for combat the weight gain. It is precursor of diseases such as diabetes or metabolic syndrome. It is necessary to know if weight gain has cumulative effects on the glycemic and lipid metabolism as precursors of complications or comorbidities. PATIENTS AND METHODS: We studied 131 patients (78 nondiabetic and 53 diabetic), 37 BMI ≤35 (3 BMI < 25, 18 BMI 25-29.9, 16 BMI 30-34.9) and 94 BMI ≥35 (81 BMI 35-49.9 and 13 BMI ≥50).We analyzed BMI, gender, diabetes and t

Metabolic disorders of liver and iron in diabetic and non-diabetic patients BMI < 35 or > 35 before gastric bypass.

INTRODUCTION: The presence of abnormalities in the metabolic pathways of iron and liver functioning can produce insulin resistance or metabolic syndrome. Therefore, it is important to examine those alterations that may lead to the development of diseases. Nutritional status is another important factor that is intimately linked to diabetes and obesity. PATIENTS AND METHODS: We studied 131 patients (78 nondiabetic patients and 53 diabetic), 37 patients BMI ≤35 (3 BMI < 25, 18 BMI 25-29.9, 16 BMI 30-34.9) and 94 patients BMI ≥35 (81 BMI 35-49.9 and 13 BMI ≥50). Semaglutide Improves Liver Steatosis and De Novo Lipogenesis Markers in Obese and Type-2-Diabetic Mice with Metabolic-Dysfunction-Associated Steatotic Liver Disease.

Metabolic-dysfunction-associated steatotic liver disease (MASLD) is a prevalent clinical condition associated with elevated morbidity and mortality rates. Patients with MASLD treated with semaglutide, a glucagon-like peptide-1 receptor agonist, demonstrate improvement in terms of liver damage. However, the mechanisms underlaying this beneficial effect are not yet fully elucidated. We investigated the efficacy of semaglutide in halting MASLD progression using a genetic mouse model of diabesity. Leptin-receptor-deficient mice with obesity and diabetes (BKS db/db) were either untreated or adminis

Improved metabolic regulation is associated with retinoblastoma protein gene haploinsufficiency in mice.

Retinoblastoma protein (pRb) is involved in the control of energy metabolism, and its inactivation protects mice against high-fat diet-induced diabesity. Here, we tested the hypothesis that partial deficiency in the Rb gene could confer metabolic advantages in front of acute challenges to metabolism and as mice age on a regular diet. Rb haploinsufficient (Rb(+/-)) mice and wild-type (WT) littermates were studied from weaning and characterized at 1.5-2.5 mo of age (young adults) and 6-7.5 mo of age (mature adults). Whereas no differences in body weight or composition were observed at young age,

Early life nutrient restriction impairs blood-brain metabolic profile and neurobehavior predisposing to Alzheimer's disease with aging.

Prenatal nutrient restriction (NR) culminating in intra-uterine growth restriction (IUGR) with postnatal catch up growth leads to diabesity. In contrast, postnatal NR with growth restriction (PNGR) superimposed on IUGR (IPGR) protects young and aging adults from this phenotype. We hypothesized that PNGR/IPGR will compromise the blood-brain metabolic profile impairing neurobehavior and predisposing to Alzheimer's disease (AD). NR (50%) in late gestation followed by cross-fostering of rat pups to either ad lib fed (CON) or NR (50%) lactating mothers generated CON, IUGR, PNGR and IPGR male (M) an

Type 2 diabetes subphenotypes are associated with differential outcomes after metabolic and bariatric surgery: An international multicentre retrospective cohort study.

AIMS: Type 2 diabetes (T2D) is characterized by its clinical heterogeneity. Newly described T2D subphenotypes, each with distinct metabolic and co-morbidity risk profiles, may enable a more personalized care. This study examined whether these subphenotypes can predict outcomes in patients undergoing metabolic and bariatric surgery (MBS). MATERIALS AND METHODS: A total of 233 people with T2D from four clinical centres undergoing MBS were retrospectively assigned to T2D subphenotypes based on the Ahlqvist methodology. The primary outcome was T2D remission at 2 years; secondary outcomes included

Metabolic and biochemical profiling reveals phenotypic heterogeneity in Zucker diabetic fatty rats.

BACKGROUND: Zucker diabetic fatty (ZDF) rats exhibit significant phenotypic variability despite genetic uniformity, yet a comprehensive characterization of these divergent phenotypes remains limited. METHODS: Male ZDF (fa/fa) rats (4 months, n = 22) and ZDF (fa/+) lean controls (n = 10) were maintained on standard chow for 6 months. Based on metabolic trajectory, ZDF (fa/fa) rats were stratified into three phenotypes: obese normoglycemic (O, n = 8), diabetic with cachexia (D, n = 8), and diabetic without cachexia (D-C, n = 6). Comprehensive assessments included body weight, glycemic control,

High-fiber diet altered gut microbiota, improved glycemic control, and reduced body weight in newly diagnosed type 2 diabetes and obesity.

Type 2 diabetes and obesity commonly coexist, making glycemia and body weight both central treatment goals. Few trials have directly compared a high-fiber dietary intervention with metformin. The factors associated with individual responses to high-fiber diet also remain unclear. In this single-center, open-label, head-to-head clinical trial, 48 adults with newly diagnosed type 2 diabetes and overweight or obesity were randomized to a high-fiber dietary intervention without glucose-lowering medication (n = 36) or metformin (n = 12) monotherapy for 84 days; 46 completed the trial. Gut Bacteria Provide Genetic and Molecular Reporter Systems to Identify Specific Diseases.

With genetic information gained from next-generation sequencing (NGS) and genome-wide association studies (GWAS), it is now possible to select for genes that encode reporter molecules that may be used to detect abnormalities such as alcohol-related liver disease (ARLD), cancer, cognitive impairment, multiple sclerosis (MS), diabesity, and ischemic stroke (IS). This, however, requires a thorough understanding of the gut-brain axis (GBA), the effect diets have on the selection of gut microbiota, conditions that influence the expression of microbial genes, and human physiology. Differential expression of genes influencing mitotic processes in cord blood mononuclear cells after a pre-conceptional micronutrient-based randomised controlled trial: Pune Rural Intervention in Young Adolescents (PRIYA).

In The Pune Maternal Nutrition Study, vitamin B12 deficiency was seen in 65% of pregnant women, folate deficiency was rare. Maternal total homocysteine concentrations were inversely associated with offspring birthweight, and low vitamin B12 and high folate concentrations predicted higher offspring adiposity and insulin resistance. These findings guided a nested pre-conceptional randomised controlled trial 'Pune Rural Intervention in Young Adolescents'. The interventions included: (1) vitamin B12+multi-micronutrients as per the United Nations International Multiple Micronutrient Antenatal Prepa

Suppression of FoxO1 activity by long-chain fatty acyl analogs.

OBJECTIVE: Overactivity of the Forkhead transcription factor FoxO1 promotes diabetic hyperglycemia, dyslipidemia, and acute-phase response, whereas suppression of FoxO1 activity by insulin may alleviate diabetes. The reported efficacy of long-chain fatty acyl (LCFA) analogs of the MEDICA series in activating AMP-activated protein kinase (AMPK) and in treating animal models of diabesity may indicate suppression of FoxO1 activity. RESEARCH DESIGN AND METHODS: The insulin-sensitizing and anti-inflammatory efficacy of a MEDICA analog has been verified in guinea pig and in human C-reactive protein

Cardiac myocyte contractility, calcium dynamics, and morphology in Zucker diabetic fatty female and male rats - lacking data in this model of obesity and diabetes: A review.

Diabetes mellitus (DM) represents an increasing global health burden with type 2 diabetes mellitus (T2DM) accounting for most cases. T2DM is associated with microvascular and macrovascular complications including myocardial ischemia and contributes to the development of a distinct form of cardiac dysfunction referred to as diabetic cardiomyopathy (DCM). Importantly, DCM contributes to 50-80% of mortality in patients with diabetes independent of other vascular comorbidities and is characterized by structural and functional alterations of the myocardium. Notably, these outcomes exhibit sex-speci

Reduced circulating levels of CXCL14 are associated with cardiovascular damage in people living with HIV.

Combined antiretroviral therapy (cART) has improved quality of life of people living with HIV (PLWH). However, alterations in adiposity with concomitant cardiovascular disease (CVD) and circulating systemic disturbances reminiscent of the metabolic syndrome still occur. We recently identified chemokine C-X-C motif ligand 14 (CXCL14) as a brown adipokine involved in M2 macrophage migration to adipose tissues to promote energy expenditure and insulin sensitivity, also exerting cardioprotective actions. Here, we aimed to study CXCL14 regulation and potential association with CVD in PLWH. Serum le

Relationship between hyperuricemia with deposition and sexual dysfunction in males and females.

PURPOSE: The association between gout, the most common crystal arthropathy, and sexual dysfunctions has often been investigated by studies in the last decades. Despite the presence of shared risk factors and comorbidities and the possible effects on sexual health of long-term gout complications, awareness of this association is severely lacking and the pathogenetic mechanisms have only partially been identified. In the present review, we aimed to investigate the current evidence regarding the potential mechanisms linking sexual dysfunctions and gout. METHODS: A comprehensive literature search
