We sought to delineate the retinal features associated with the high-fat diet (HFD) mouse, a widely used model of obesity. C57BL/6 mice were fed either a high-fat (60% fat; HFD) or low-fat (10% fat; LFD) diet for up to 12 months. The effect of HFD on body weight and insulin resistance were measured. The retina was assessed by electroretinogram (ERG), fundus photography, permeability studies, and trypsin digests for enumeration of acellular capillaries. The HFD cohort experienced hypercholesterolemia when compared to the LFD cohort, but not hyperglycemia. HFD mice developed a higher body weight

Opioids, prescribed for pain relief or as maintenance therapies for opioid use disorder, induce constipation, which can lead to medication compliance issues. However, it is not known how dietary manipulation might impact opioid-induced constipation. To test the hypothesis that eating a high fat diet exacerbates opioid-induced constipation, male and female Sprague-Dawley rats ( n = 8/diet/sex) were fed either a low fat (standard) diet, a high fat/high carbohydrate diet, or a ketogenic diet. Rats were injected with either saline or morphine (1-10 mg/kg) and constipation was measured by counting

An increase of visceral fat affects human bone health causing fragility, mechanical strength reduction, and increased propensity to fractures because of impaired bone matrix microstructure and aberrant bone cell function. Adult Danio rerio (zebrafish) represents a powerful model to study both metabolic diseases and bone metabolism. The aim of this study was to generate an obese adult zebrafish by high-fat diet and evaluate metabolic and bone tissue effects. Fish blood glucose and insulin levels were found to be altered in high-fat diet fish revealing a failure in β-cells insulin production. Bl

The effects of a high fat diet (30% (w/w) corn oil) on chronic streptozotocin-diabetic rats were investigated at the whole body level and at the enzyme level. The diet caused significant decreases in the extent of polydipsia (66% decrease), polyphagia (49%), polyuria (67%) and glycosuria (70%). The activities of selected hepatic enzymes from the glycolytic, gluconeogenic, ureogenic and lipogenic clusters were determined. The fat diet caused significant decreases (range: 47 to 54%) in the activity of the ureogenic enzymes carbamyl phosphate synthetase, ornithine transcarbamylase and arginase; h

OBJECTIVE: Non-alcoholic fatty liver disease (NAFLD) is the most common chronic liver disorder worldwide, affecting 30-40% of adults. High-fat diets contribute significantly to NAFLD by promoting hepatic lipid accumulation, oxidative stress, and inflammation. In this context, cinnamaldehyde (CNMA) has emerged as a promising hepatoprotective agent due to its antioxidant, anti-inflammatory, and lipid-regulatory properties.
MATERIALS AND METHODS: Male Wistar rats were randomly assigned to four groups (n=6 per group): (A) Control; (B) HFD; (C) Control+CNMA; and (D) HFD+CNMA. CNMA was administered

Adrenomedullin (AM) is highly expressed in cardiovascular tissues and has been extensively studied as a vasoactive peptide. Subsequent studies have also demonstrated substantial AM expression in adipose tissue; however, the pathophysiological role of adipocyte-derived AM in obesity remains unclear. We generated adipocyte-specific AM knockout mice (Adiponectin-Cre; Adm^fl/fl, cKO) and fed cKO and littermate controls normal chow (NC) or HFD from 8 to 36 weeks of age. HFD increased systolic blood pressure (SBP) in both genotypes, but SBP was higher in cKO mice under chronic HFD conditions, despit

A high-fat diet (HFD) is a major contributor to metabolic diseases, causing cardiac injury through inflammation, oxidative stress, and apoptosis mechanisms. Atraric acid (AA) is a natural compound with anti-inflammatory and metabolic regulatory activities; however, its protective effects against HFD-induced cardiac injury and the underlying mechanisms remain unclear. This study aimed to investigate whether AA alleviates HFD-induced cardiac injury by regulating eukaryotic elongation factor 2 kinase (eEF2K). Using an HFD-induced mouse model and a primary cardiomyocytes (PCMs) model established w

PURPOSE: Obesity contributes to immune system dysregulation, thereby increasing the risk of developing numerous diseases. Dietary capsaicin (CAP) has been demonstrated to mitigate obesity, while its impact on B cell immunity during obesity remains unexplored. This study aimed to investigate the restorative effect of CAP on B cells deficiency in obese mice, and explore the underlying mechanisms.
METHODS: C57BL/6J mice were fed a standard chow diet or a high-fat diet (HFD), and HFD-fed mice were orally administered CAP. Flow cytometry was employed to evaluate the effect of CAP on B cell functio

At present, non-alcoholic fatty liver disease (NAFLD) is the most prevalent liver disease worldwide, with obesity recognized as a global epidemic and type 2 diabetes a worldwide disease. In this study, 10 C57BL/6 mice were divided into two groups: the control group (SC) and the high-fat diet (HF) group. Both groups were fed their respective diets for 8 weeks. The animals were analyzed for body weight, glucose/insulin resistance, hepatic steatosis, and fibrosis to diagnose NAFLD. Results showed that the HF group animals had significantly higher body weight (P<0.0001), glucose resistance (P=0.00

BACKGROUND: Hepatic glycosphingolipid biosynthesis is implicated in insulin resistance and metabolic dysfunction-associated steatotic liver disease (MASLD). While UDP-glucose ceramide glucosyltransferase (UGCG) serves as the rate-limiting enzyme in glycosphingolipid synthesis, its cell-specific roles in MASLD pathogenesis remain undefined. Our study investigates the mechanistic contribution of LSEC-expressed UGCG to high-fat diet (HFD)-induced insulin resistance and MASLD progression.
METHODS: Primary cell sorting was used to analyze LSEC-specific enrichment of UGCG in wild-type mice under no

Unbalanced lipid metabolism contributes to inflammation in several conditions. Gluten, that triggers celiac disease, may also play a role in diseases associated with a high-fat diet (HFD). Our aim was to investigate the interplay between gluten and HFD in HLA-DQ8 (DQ8) transgenic mice, a model of gluten sensitivity. DQ8 mice were fed the gluten-free diet, the HFD, or the HFD containing 8% gluten (HFD + G) for 12 or 23 weeks. Clinical parameters, liver and intestinal histology, immune parameters were assessed. Twelve weeks of HFD increased body and white adipose tissue weight and reduced glucos

Lipid droplet accumulation in microglia, microglia-mediated neuroinflammation, and subsequent neuronal damage are hallmark features of high-fat diet (HFD)-induced cognitive impairment. In this analysis, this is proposed that a new molecule feimin (B230219D22Rik in mice) is a key negative regulator of LD accumulation and the inflammatory response in HFD-induced cognitive impairment. To test this hypothesis, BV2 microglia is exposed to palmitic acid (PA) in vitro, mimicking the effects of an HFD. This is found that feimin expression is significantly increased following high-lipid stimulation. Fe

Background: Overweight and obesity are currently a worldwide problem, with undesirable health consequences, such as type 2 diabetes. Therefore, much attention has been paid to preventing obesity through diet. Free fatty acids (FFAs) serve as signaling molecules in many biological processes, leading to increased energy expenditure and insulin secretion. Short-chain fatty acids (SCFAs) such as acetic, propionic and butyric acid are bioactive metabolites produced by gut microbes, and their beneficial effects on host metabolism are well studied. However, the effects of hexanoic acid on metabolism

This study investigated the effects of semaglutide (Sema) on the gut microbiota of obese mice induced with high-fat diet (HFD). Male C57BL/6 J mice aged 6 weeks were enrolled and randomly distributed to four groups, which were provided with a normal control diet (NCD,NCD + Sema) and a 60% proportion of a high-fat diet (HFD,HFD + Sema), respectively. HFD was given for 10 weeks to develop an obesity model and the intervention was lasted for 18 days. The results showed semaglutide significantly reduced body weight gain, areas under the curve (AUC) of glucose tolerance test and insulin resistance

Prolonged intake of a high-fat diet (HFD) disturbs the composition of gut microbiota, contributing to the development of metabolic diseases, notably obesity and increased intestinal permeability. Thyme (Thymus vulgaris L.), an aromatic plant, is known for its several therapeutic properties. In this study, we explored the potential of thyme extract (TLE) to mitigate HFD-induced metabolic derangements and improve the gut environment. Eight-week-old C57BL/6 mice were administered 50 or 100 mg/kg TLE for eight weeks. Administration of 100 mg/kg TLE resulted in decreased weight gain and body fat pe

Abstract Traumatic brain injury (TBI) is one of the leading causes of death for children in the United States. Juveniles are more likely to sustain TBIs than most other age groups, and TBI has been shown to result in increased anxiety and stress behaviors. In addition, the hypothalamic-pituitary-adrenal (HPA) axis has previously been shown to become dysregulated after a TBI. Further, many children consume diets high in saturated fats and refined sugars, which are also connected to alterations in HPA axis function and behavior disorders. The goal of the current study was to identify a potential

Non-alcoholic fatty liver disease (NAFLD) comprises a spectrum of liver damage directly related to diabetes, obesity, and metabolic syndrome. The (pro)renin receptor (PRR) has recently been demonstrated to play a role in glucose and lipid metabolism. Here, we test the hypothesis that the PRR regulates the development of diet-induced hepatic steatosis and fibrosis. C57Bl/6J mice were fed a high-fat diet (HFD) or normal-fat diet (NFD) with matching calories for 6 weeks. An 8-week methionine choline-deficient (MCD) diet was used to induce fibrosis. Two weeks following diet treatment, mice were im

Certain monoamine oxidase (MAO) inhibitors exhibit beneficial effects, such as reducing adiposity and metabolic disorders; however, their effects on hepatic lipid metabolism have not been revealed. This study aimed to investigate the effects of a selective MAO-B inhibitor, selegiline, on dyslipidemia and hepatic steatosis in mice induced by a high-fat diet (HFD). Administration of selegiline (0.6 mg/kg body weight) by intraperitoneal injection was found to reduce HFD-induced body weight gain and increases in liver and adiposity coefficients, blood lipids and fatty acid levels. Furthermore, sel
