Di-2-ethylhexyl Phthalate Induces Neurotoxicity of Transforming Microglia into M1 Subtype through Targeting α-Syn Dimerization.

Di-2-ethylhexyl phthalate (DEHP) accumulates in the food chain, threatening human and animal health. However, how DEHP induces neurotoxicity by interfering with microglia has not been fully revealed. Our research aims to explore the effects of DEHP on microglia and identify relevant targets that cause neurotoxicity. The results indicated that DEHP damaged neurons, leading to swelling, demyelination, and abnormal autophagy. Inhibition of autophagy resulted in α-Syn misfolding, characterized by an increase in dimers with increasing additive concentrations (P = 0.0096). Dimer formation was promot

Analysis of toxicity and mechanisms of DEHP in prostate cancer with network toxicology and molecular docking strategy.

Recent decades have seen a significant increase in the use of industrial chemicals, introducing numerous environmental pollutants with uncertain health impacts. Among these, di(2-ethylhexyl) phthalate (DEHP), a key plastic industry component, has raised concerns due to its carcinogenic and endocrine-disrupting potential, particularly its association with tumorigenesis. The current study focused on DEHP's implications for prostate cancer (PCa), a leading health issue for men worldwide. DEHP's endocrine-disrupting properties, which interfere with hormone functions crucial for PCa development and

Endometriosis-like lesions induced by phthalates: new phytotherapic applications to complement traditional cares: a PNRR 2023 project.

BACKGROUND: Endometriosis (E) is an oestrogen-dependent, multifactorial, inflammatory disease causing pelvic pain and infertility. Several concerns have been raised about the role of food contaminants, in particular Bis(2-ethylhexyl) phthalate (DEHP), potentially involved in the onset and propagation of E. Conventional therapies- which have considerable side effects - focus on reducing levels of oestrogens and counteracting inflammation. The potential preventive/protective role of plant extracts (PEs) on phthalate (PH) -induced E is studied by a stepwise approach. METHODS: (i) raw material id

Extracellular vesicles as a potential source of biomarkers for endocrine disruptors in MASLD: A short review on the case of DEHP.

Metabolic dysfunction-Associated Steatotic Liver Disease (MASLD) is a chronic disease with increasing prevalence and for which non-invasive biomarkers are needed. Environmental endocrine disruptors (EDs) are known to be involved in the onset and progression of MASLD and assays to monitor their impact on the liver are being developed. Extracellular vesicles (EVs) mediate cell communication and their content reflects the pathophysiological state of the cells from which they are released. They can thus serve as biomarkers of the pathological state of the liver and of exposure to EDs. In this revi

Nanoparticles for Augmenting Therapeutic Potential and Alleviating the Effect of Di(2-ethylhexyl) Phthalate on Gastric Cancer.

Changes in diet culture and modern lifestyle contributed to a higher incidence of gastrointestinal-related diseases, including gastritis, implicated in the pathogenesis of gastric cancer. This observation raised concerns regarding exposure to di(2-ethylhexyl) phthalate (DEHP), which is linked to adverse health effects, including reproductive and developmental problems, inflammatory response, and invasive adenocarcinoma. Research on the direct link between DEHP and gastric cancer is ongoing, and further studies are required to establish a conclusive association. In our study, extremely low conc

Exposure to di-isononyl phthalate during early pregnancy disrupts decidual angiogenesis and placental development in mice.

Di-isononyl phthalate (DiNP), an endocrine-disrupting chemical, is found in numerous consumer products and human exposure to this phthalate is becoming inevitable. The impact of DiNP exposure on the establishment and maintenance of pregnancy remains largely unknown. Thus, we conducted studies in which pregnant mice were exposed to an environmentally relevant dose (20 µg/kg BW/day) of DiNP on days 1-7 of gestation, then analyzed the effects of this exposure on pregnancy outcome. Our studies revealed that exposure to DiNP during this window led to fetal loss towards the end of gestation. Further

Secondary oxidized di-2-ethylhexyl phthalate metabolites may be associated with progression from isolated premature thelarche to central precocious or early puberty.

Phthalate esters (PAEs) may act as estrogen receptor agonists, and their relationship with precocious puberty is a global health concern. However, their role in isolated premature thelarche (IPT) progression remains unclear. We conducted a cohort study investigating the relationship between IPT progression and urinary PAE metabolites. Girls with IPT aged 6-8 years were regularly followed up every three months for one year. Clinical data and urine PAE metabolite levels were collected. Participants who progressed to central precocious puberty (CPP) or early puberty (EP) had significantly higher

Promising hepatoprotective effects of lycopene in different liver diseases.

The incidence of liver diseases such as hepatitis, fibrosis, cirrhosis, and liver cancer is one of the world's most significant health challenges. Liver diseases can be caused by a variety of circumstances, including viral infection, exposure to xenobiotics, environmental pollutants, metabolic disorders, and others. Lycopene (LYC) is a potent antioxidant of the carotenoid family in red fruits and vegetables. LYC has been found to have multiple biological activities, including antioxidative, anti-inflammatory, and antiapoptotic effects. Notably, LYC has promised hepatoprotective properties agai

Long-Term Exposure to Low-Dose Di-(2-ethylhexyl) Phthalate Impairs Cholesterol Metabolism in Hepatic Stellate Cells and Exacerbates Liver Librosis.

Phthalates are often added to plastic products to increase their flexibility. Di-(2-ethylhexyl) phthalate (DEHP) is one of the most common plasticizers. Previously, a major incident involving phthalate-contaminated foodstuffs occurred, where phthalates were deliberately added to foodstuffs as a substitute for emulsifiers, resulting in a threat to public health. DEHP exposure can cause liver damage and further lead to cancer; however, the effects of long-term exposure to low-dose DEHP on hepatic stellate cells (HSCs) and on liver fibrosis are still unclear. In this study, we showed that chronic

The present lack of evidence for unique rodent germ-cell mutagens.

Six chemicals, diethylhexyl phthalate (DEHP), ethanol, cyclohexylamine (CHA), sodium saccharin (NaS), cadmium chloride (CdCl2) and triflupromazine (TFP), were suggested to be unique germ-cell mutagens (Auletta and Ashby, 1988) by the GeneTox Workgroups of the U.S. Environmental Protection Agency (EPA). If this is a correct classification it would have major consequences when screening for mutagenicity and when labelling genotoxic substances. However, our re-evaluation of the GeneTox literature, including some more recent publications, has failed to find substantive evidence that any of these c

Repeated exposure of bumblebee workers to field-realistic doses of di-2-ethylhexyl and di-n-butyl phthalates lowers worker survival and induces obesity-related phenotypes in workers and brood.

Wild bees, like many other insects, are declining. Multiple drivers have been identified, but the potential effects of endocrine disruptors such as phthalates, remain poorly studied. Yet, di-2-ethylhexyl phthalate (DEHP) and di-n-butyl phthalate (DnBP), two of the most common phthalates that pervade the environment, are known to affect key hormonal and metabolic pathways in invertebrates even at field realistic doses. These ubiquitous pollutants are semi-volatile compounds used to increase the flexibility of plastic materials, or as solvents for certain detergents, pesticides, and other produc

Transcriptomic analysis of the mechanism of male reproductive injuries caused by the mixture of three phthalates in rats.

Phthalates (PEs) are widespread in environment, and human beings are unavoidably exposing to the mixture of PEs, which may induce male reproductive health risks. In order to investigate the mechanism of male reproductive injuries caused by the mixture of di-2-ethylhexyl phthalate, dibutyl phthalate and butyl benzyl phthalate (MPEs), male rats were orally exposed to 16 mg/kg/d MPEs (L-MPEs) and 450 mg/kg/d MPEs (H-MPEs) for 90 days, and the results showed that MPEs decreased the weights of testes, epididymis and periepididymis fat, decreased serum levels of male hormones, increased abnormal spe

Hydrolysis of dibutyl phthalate and di(2-ethylhexyl) phthalate in human liver, small intestine, kidney, and lung: An in vitro analysis using organ subcellular fractions and recombinant carboxylesterases.

Phthalates are widely used plasticizers that are primarily and rapidly metabolized to monoester phthalates in mammals. In the present study, the hydrolysis of dibutyl phthalate (DBP) and di(2-ethylhexyl) phthalate (DEHP) in the human liver, small intestine, kidney, and lung was examined by the catalytic, kinetic, and inhibition analyses using organ microsomal and cytosolic fractions and recombinant carboxylesterases (CESs). The Vmax (y-intercept) values based on the Eadie-Hofstee plots of DBP hydrolysis were liver > small intestine > kidney > lung in microsomes, and liver > small intestine > l

Estimation and potential ecological risk assessment of multiphase PAEs in mangrove wetlands in Dongzhai Harbor, Hainan.

With the application of plastic products, phthalates now widely occur in various environmental media. A large number of ecological risk assessment experiments have only been carried out on a single medium such as water or sediment. There are few reports of ecological risk assessments based on the phase states of phthalic acid esters (PAEs) such as the free dissolved state and the dissolved organic carbon (DOC) adsorption state. In this study, the concentrations of the free dissolved state, the DOC adsorption state, and the easily released PAEs in the sediments, as well as the dissolved organic

Network toxicology integrated with machine learning and SHAP analysis identifies overlapping immune signatures between Di(2-ethylhexyl) phthalate (DEHP) and Sjögren's syndrome.

BACKGROUND: As a prevalent plasticizer in industrial manufacturing and daily-use products, di(2-ethylhexyl) phthalate (DEHP) has raised growing concerns regarding its safety and potential role in disease pathogenesis. Although direct epidemiological evidence linking DEHP exposure to Sjögren’s syndrome (SS) remains limited, DEHP has been reported to exert immunomodulatory and endocrine-disrupting effects that are highly relevant to the core pathological mechanisms of SS. Therefore, this study aimed to systematically explore the potential links between DEHP and SS using an integrated strategy co

MEHP impairs male reproductive function through the lipid metabolism pathway: Evidence from in vitro human immature testis culture.

Di-(2-ethylhexyl) phthalate (DEHP) is a widely used industrial plasticizer whose main metabolite in the body is mono-(2-ethylhexyl) phthalate (MEHP). The reproductive toxicity of DEHP and MEHP has been documented in some studies. However, precise mechanisms remain unclear, and most research has focused on animal models. In this study, an in vitro culture system of immature human testicular tissue was employed to explore the etiological link between MEHP and developmental toxicity in immature human testes. The examination of micro-morphology, subcellular structure and testosterone secretion dem

Developmental neurotoxicity evaluation of di(2-ethylhexyl) phthalate (DEHP) and three alternative plasticizers in human neurospheres.

Plasticizers like di-(2-ethylhexyl) phthalate (DEHP) are commonly used in medical devices to enhance plastic flexibility. DEHP is classified as a CMR1b substance due to its adverse effects on reproduction and fertility, and it has been linked to neurodevelopmental disorders such as ADHD, autism spectrum disorder, and learning disabilities. While DEHP is scheduled for phase-out by 2030, data on the developmental neurotoxicity (DNT) of alternative plasticizers remain scarce. We evaluated the DNT potential of DEHP and three alternative plasticizers: di-(2-ethylhexyl) terephthalate (DEHT), di-(2-e
