Computational Study on a Copper-Catalyzed Atroposelective Dehydro-Diels-Alder Reaction of Ynamide via Vinyl Cation: Mechanistic Investigations and Chiral Induction Model.

Dehydro-Diels-Alder (DDA) reaction serves as one of the most prominent methods for the synthesis of aromatic compounds. Herein, our study elucidates the mechanism and origin of atroposelectivity of Cu(I)-catalyzed DDA reactions of aryl-substituted 1,6-diyne substrate via density functional theory calculations. The intramolecular nucleophilic cyclization is identified as the rate- and stereo-determining step, with a 2.3 kcal/mol free energy difference between the two atropo-determining transition states. Chiral induction arises from a global-local synergy: the chiral ligand's bulky side arms sh

Enzyme inhibitory activities of acetylene and sesquiterpene compounds in atractylodes rhizome.

In an attempt to elucidate the physiological activities of (6E,12E)-tetradecadiene-8,10-diyne-1,3-diol diacetate (TDEYA), which was detected as a hydrolysis form (TDEY) in the plasma after oral administration of a decoction of Atractylodes rhizome in rats, we examined the inhibitory effects of various enzymes which are considered to participate in the regulation of body fluid levels and inflammatory reactions. TDEY and TDEYA did not show inhibitory effects on carbonic anhydrase (CA) or angiotensin converting enzyme (ACE) at concentrations less than 1.0 x 10(-3) M. However, both acetylene compo

Antitrypanosomal properties of Panax ginseng C. A. Meyer: new possibilities for a remarkable traditional drug.

African trypanosomiasis is still a major health problem in many sub-Saharan countries in Africa. We investigated the effects of three preparations of Panax ginseng, Panax notoginseng, isolated ginsenosides, and the polyacetylene panaxynol on Trypanosoma brucei brucei and the human cancer cell line HeLa. Hexane extracts and the pure panaxynol were toxic and at the same time highly selective against T. b. brucei, whereas methanol extracts and 12 isolated ginsenosides were significantly less toxic and showed only weak selectivity. Panaxynol was cytotoxic against T. b. brucei at the concentration

Cross-linkable liposomes stabilize a magnetic resonance contrast-enhancing polymeric fastener.

Liposomes are commonly used to deliver drugs and contrast agents to their target site in a controlled manner. One of the greatest obstacles in the performance of such delivery vehicles is their stability in the presence of serum. Here, we demonstrate a method to stabilize a class of liposomes that load gadolinium, a magnetic resonance (MR) contrast agent, as a model cargo on their surfaces. We hypothesized that the sequential adsorption of a gadolinium-binding chitosan fastener on the liposome surface followed by covalent cross-linking of the lipid bilayer would provide enhanced stability and

Falcarindiol and dichloromethane fraction are bioactive components in Oplopanax elatus: Colorectal cancer chemoprevention via induction of apoptosis and G2/M cell cycle arrest mediated by cyclin A upregulation.

Oplopanax elatus (Nakai) Nakai has a long history of use as an ethnomedicine by the people living in eastern Asia. However, its bioactive constituents and cancer chemopreventive mechanisms are largely unknown. The aim of this study was to prepare O. elatus extracts, fractions, and single compounds and to investigate the herb's antiproliferative effects on colon cancer cells and the involved mechanisms of action. Two polyyne compounds were isolated from O. elatus, falcarindiol and oplopandiol. Based on our HPLC analysis, falcarindiol and oplopandiol are major constituents in the dichloromethane

Falcarinol is a covalent cannabinoid CB1 receptor antagonist and induces pro-allergic effects in skin.

The skin irritant polyyne falcarinol (panaxynol, carotatoxin) is found in carrots, parsley, celery, and in the medicinal plant Panax ginseng. In our ongoing search for new cannabinoid (CB) receptor ligands we have isolated falcarinol from the endemic Sardinian plant Seseli praecox. We show that falcarinol exhibits binding affinity to both human CB receptors but selectively alkylates the anandamide binding site in the CB(1) receptor (K(i)=594nM), acting as covalent inverse agonist in CB(1) receptor-transfected CHO cells. Given the inherent instability of purified falcarinol we repeatedly isolat

Bifunctional ligand-enabled gold-catalyzed cycloisomerization of 2'-ethynylbiaryl-2-yl carbonyl compounds.

Gold(I) vinylidenes are powerful yet typically fleeting electrophiles whose synthetic exploitation has, until now, relied largely on two strategies: 1,2-migration of a halide or silyl group from a gold-activated alkyne, or dual gold catalysis of diyne substrates. Here, we introduce a ligand-enabled catalytic entry to these reactive intermediates based on a prior stoichiometric approach reported by Fürstner. A bifunctional biaryl-2-ylphosphine bearing a remote N,N-dialkylamide group promotes in situ formation of a gold acetylide without an external base; the proton thereby relayed to the amide

Ring-Closing Metathesis Approaches towards the Total Synthesis of Rhizoxins.

Efforts are described towards the total synthesis of the bacterial macrolide rhizoxin F, which is a potent tubulin assembly and cancer cell growth inhibitor. A significant amount of work was expanded on the construction of the rhizoxin core macrocycle by ring-closing olefin metathesis (RCM) between C(9) and C(10), either directly or by using relay substrates, but in no case was ring-closure achieved. Macrocycle formation was possible by ring-closing alkyne metathesis (RCAM) at the C(9)/C(10) site. The requisite diyne was obtained from advanced intermediates that had been prepared as part of th

Antimycobacterial polyacetylenes from Levisticum officinale.

No conflicts of interest concerning financial matters or personal relationships exist between the authors and those who might bias this work. The present work is in part included the PhD thesis of A. Schinkovitz (University of Graz) but has not been published elsewhere previously. The dichloromethane extract of the roots of Levisticum officinale L. (Apiaceae) exhibited significant antimycobacterial activity against Mycobacterium fortuitum and Mycobacterium aurum in a microtiter plate dilution assay and was further analysed following a bioassay-guided fractionation strategy. 3(R)-Falcarinol (3(

Suppressed excitonic effects enable high mobility and high-yield photoconductivity in a pyridine-coordinated two-dimensional polymer crystal.

Two-dimensional polymers (2DPs) and their layer-stacked covalent organic frameworks (2D COFs) offer modular, atomically precise platforms for organic optoelectronics, yet their photoconductive responses remain fundamentally constrained by strong excitonic effects and localized charge transport. Here, we demonstrate that a diyne-linked 2DP crystal with axial pyridine coordination overcomes this limitation, enabling simultaneous efficient free-carrier generation and band-like transport. Introducing pyridine ligands that axially coordinate to Cu-porphyrin nodes transforms weak van der Waals stack

Functional Poly(dihalopentadiene)s: Stereoselective Synthesis, Aggregation-Enhanced Emission and Sensitive Detection of Explosives.

The development of polymeric materials with novel structures and unique properties and functionalities is of both academic and industrial significance. In this work, functional poly(dihalopentadiene)s were synthesized by boron trihalide-mediated multicomponent polymerization routes in a stereoselective manner. The polymerizations of tetraphenylethylene-containing diyne, BX₃ (X = Cl, Br) and p-tolualdehyde proceed smoothly in dichloromethane under mild conditions to afford high molecular weight poly(dihalopentadiene)s with a predominant (Z,Z)-configuration in moderate to good yields. Panaxynol mitigates chemotherapy-induced intestinal mucositis by improving the colonic microenvironment in murine models.

Chemotherapy-induced mucositis (CIM) is a debilitating side effect impacting as many as 90% of patients with cancer undergoing treatment. Patients receiving 5-fluorouracil (5FU), a first-line chemotherapeutic in colorectal cancer, experience significant gastrointestinal distress that perpetuates poor patient quality of life and reduces treatment tolerance, efficacy, and survival. Natural compounds have shown promise in improving CIM through their pleiotropic actions, including immune and mucosal regulation. We examined whether panaxynol, a bioactive compound isolated from American ginseng, can

Synthesis and Antiviral Evaluation of 5-(4-Aryl-1,3-butadiyn-1-yl)-uridines and Their Phosphoramidate Pronucleotides.

The emergence of RNA viruses driven by global population growth and international trade highlights the urgent need for effective antiviral agents that can inhibit viral replication. Nucleoside analogs, which mimic natural nucleotides, have shown promise in targeting RNA-dependent RNA polymerases (RdRps). Starting from protected 5-iodouridine, we report the synthesis of hitherto unknown C5-substituted-(1,3-diyne)-uridines nucleosides and their phosphoramidate prodrugs. The modifications at C5 include 4-(trifluoromethyl)benzene (a), 4-pentyl-benzene (b), 3,5-dimethoxy-benzene (c), 4-(trifluorome

Mechanistic Analysis of Bioorthogonal Double Strain-Promoted Alkyne-Nitrone Cycloadditions Involving Dibenzocyclooctadiyne.

The reactions of nitrones with cyclooctadiynes were studied to establish the relative rates of sequential reactions and to determine the limits and scope of this bioorthogonal chemistry. We have established the second-order rate constants for the consecutive additions of a variety of nitrones onto diyne and studied the structure-activity relationships via Hammett plots. Results show that the addition of the second nitrone to the monointermediate occurs significantly faster than the first, with both reactions being faster than analogous reactions with azides. Computational chemistry supports th

Deciphering the anti-Mycobacterium tuberculosis activity of bioinspired lipidic alkynylcarbinols.

This article reports the first in-depth structure-activity relationship investigation of the anti-Mycobacterium tuberculosis (Mtb) potential of falcarindiol, a representative natural acetylenic lipid found in medicinal herbs of various traditional pharmacopeias. Prompted by our previous work on bioinspired anticancer lipidic alkynylcarbinols, we implemented a chemistry-driven deconstruction-reconstruction approach. This led to a detailed analysis of the key structural elements required for an optimized balance between potent anti-mycobacterial activity and low cytotoxicity against mammalian ce

Inhibition of 15-hydroxyprostaglandin dehydrogenase activity in rabbit gastric antral mucosa by panaxynol isolated from oriental medicines.

Panaxynol is a polyacetylene compound with anti-inflammatory and anti-platelet-aggregatory effects isolated from commonly used oriental medicines. The effects of panaxynol on the activity of prostaglandin-synthesizing and catabolizing enzymes in the rabbit gastric antral mucosa have been examined. At concentrations ranging from 25 to 200 microM panaxynol had no effect on the synthesis of prostaglandins E2, F2alpha and D2 from exogenous arachidonic acid in the microsomal fraction of the gastric mucosa whereas at 25-200 microM it dose-dependently inhibited the activity of 15-hydroxyprostaglandin
