Assaying Mechanonociceptive Behavior in Drosophila Larvae.

Drosophila melanogaster larvae have been extensively used as a model to study the molecular and cellular basis of nociception. The larval nociceptors, class IV dendritic arborization (C4da) neurons, line the body wall of the animal and respond to various stimuli including noxious heat and touch. Activation of C4da neurons results in a stereotyped escape behavior, characterized by a 360° rolling response along the body axis followed by locomotion speedup. The genetic accessibility of Drosophila has allowed the identification of mechanosensory channels and circuit elements required for nocicepti

Alzheimer's Disease, Drosophila melanogaster and Polyphenols.

Alzheimer's disease (AD) is an insidious neurological disorder that affects memory, one of the human brain's main cognitive functions. Around 5.2 million Americans currently have AD, and the number threatens to climb to 7 million by 2020. Our native country, Colombia, is no exception with an estimated 260,000 individuals to be affected by AD in 2020. A large, genetically-isolated community in Antioquia, Colombia, with early-onset familial Alzheimer's disease due to a presenilin-1 mutation is ideally suited for the study of molecular mechanisms of AD, and hence accelerate the discovery of new o

Sleep deprivation suppresses aggression in Drosophila.

Sleep disturbances negatively impact numerous functions and have been linked to aggression and violence. However, a clear effect of sleep deprivation on aggressive behaviors remains unclear. We find that acute sleep deprivation profoundly suppresses aggressive behaviors in the fruit fly, while other social behaviors are unaffected. This suppression is recovered following post-deprivation sleep rebound, and occurs regardless of the approach to achieve sleep loss. Genetic and pharmacologic approaches suggest octopamine signaling transmits changes in aggression upon sleep deprivation, and reduced

A Drosophila ABC transporter regulates lifespan.

MRP4 (multidrug resistance-associated protein 4) is a member of the MRP/ABCC subfamily of ATP-binding cassette (ABC) transporters that are essential for many cellular processes requiring the transport of substrates across cell membranes. Although MRP4 has been implicated as a detoxification protein by transport of structurally diverse endogenous and xenobiotic compounds, including antivirus and anticancer drugs, that usually induce oxidative stress in cells, its in vivo biological function remains unknown. In this study, we investigate the biological functions of a Drosophila homolog of human

Endogenously imprinted genes in Drosophila melanogaster.

Genomic imprinting is an epigenetic state that results from differential processing of chromosomes during gametogenesis and which can cause differential expression of genes depending on the sex of the parent transmitting that gene. In Drosophila, many examples of imprinted marker genes have been documented and imprinting of these genes involves highly conserved epigenetic regulators. However, no endogenously imprinted genes have yet been identified. Here we present a phenotypic and transcriptional analysis of parthenogenetic (gynogenetic) and genotypically identical but sexually produced adult

Drosophila Answers to TDP-43 Proteinopathies.

Initially implicated in the pathogenesis of CFTR and HIV-1 transcription, nuclear factor TDP-43 was subsequently found to be involved in the origin and development of several neurodegenerative diseases. In 2006, in fact, it was reported for the first time the cytoplasmic accumulation of TDP-43 in ubiquitin-positive inclusions of ALS and FTLD patients, suggesting the presence of a shared underlying mechanism for these diseases. Today, different animal models of TDP-43 proteinopathies are available in rodents, nematodes, fishes, and flies. Although these models recapitulate several of the pathol

Modeling spinal muscular atrophy in Drosophila.

Spinal Muscular Atrophy (SMA), a recessive hereditary neurodegenerative disease in humans, has been linked to mutations in the survival motor neuron (SMN) gene. SMA patients display early onset lethality coupled with motor neuron loss and skeletal muscle atrophy. We used Drosophila, which encodes a single SMN ortholog, survival motor neuron (Smn), to model SMA, since reduction of Smn function leads to defects that mimic the SMA pathology in humans. Here we show that a normal neuromuscular junction (NMJ) structure depends on SMN expression and that SMN concentrates in the post-synaptic NMJ regi

Drosophila spermatogenesis: insights into testicular cancer.

Geneticists have a long history of studying reproduction in the fruitfly, Drosophila melanogaster, and in recent years it has become apparent that many of the genes that regulate invertebrate reproduction have been conserved through vertebrate evolution. As with other higher eukaryotes, spermatogenesis in Drosophila is characterized by a regenerative germline stem cell population that divides asymmetrically to produce mitotic spermatogonia which will eventually differentiate into spermatocytes. Germline tumours consisting of undifferentiated germ cells have been associated with both loss-of-fu

Drosophila TGIF proteins are transcriptional activators.

The information carried by transforming growth factor beta (TGF-beta) signaling molecules induces profound responses in target cells. To restrict this information to appropriate cells, TGF-beta signaling pathways are tightly regulated by dynamic interactions with transcriptional activators and repressors. Numerous cross-species experiments have shown that TGF-beta family members and their signal transduction machinery (receptors and Smad signal transducers) are functionally conserved between vertebrates and invertebrates. TG-interacting factor (TGIF) is a homeodomain-containing transcriptional

Induced RNA Interference Can Suppress Persistent Drosophila A Virus (DAV) Infection in Cultured Drosophila Melanogaster S2 Cells.

Background: The RNA interference (RNAi) pathway is a highly conserved antiviral mechanism in eukaryotes, including insects such as fruit flies. Many viruses have therefore evolved mechanisms that protect their transcripts and/or genomes; how effectively RNAi can act is thus a question that must be answered case-by-case. Methods: We demonstrate that the RNAi machinery can successfully combat Drosophila A virus (DAV) in persistently infected Drosophila melanogaster S2 cell lines, but only when supported with exogenous triggers. Since such an infection poses a challenge to the fitness of cells an

Hippo, Drosophila MST, is a novel modifier of motor neuron degeneration induced by knockdown of Caz, Drosophila FUS.

Mutations in the Fused in Sarcoma (FUS) gene have been identified in familial ALS in human. Drosophila contains a single ortholog of human FUS called Cabeza (Caz). We previously established Drosophila models of ALS targeted to Caz, which developed the locomotive dysfunction and caused anatomical defects in presynaptic terminals of motoneurons. Accumulating evidence suggests that ALS and cancer share defects in many cellular processes. The Hippo pathway was originally discovered in Drosophila and plays a role as a tumor suppressor in mammals. We aimed to determine whether Hippo pathway genes mo

Wrinkling of the eye in hybrids between Drosophila virilis and Drosophila lummei is caused by interaction of maternal and zygotic genes.

Hybrids of Drosophila virilis females and Drosophila lummei males have visible developmental anomalies in external adult structures. Reciprocal hybrids are normal, and the anomalies are not found in other interspecific F1 hybrids of the virilis subgroup. Antibiotic treatments with ampicillin, streptomycin and tetracycline did not cure the syndrome. The genetic basis of the wrinkling of the compound eye was analysed in detail. Both male and female F1 progeny of D. virilis mothers were affected, indicating that wrinkling of eyes is maternally determined. F1 hybrid females produced healthy progen

Optimizing irradiation dose for Drosophila melanogaster males to enhance heterospecific Sterile Insect Technique (h-SIT) against Drosophila suzukii.

The spotted-wing drosophila (Drosophila suzukii), a highly invasive agricultural pest, poses significant challenges to fruit production worldwide. Traditional chemical control methods are costly and raise concerns about resistance and environmental sustainability. The Heterospecific Sterile Insect Technique (h-SIT) has emerged as a promising alternative. Sterile heterospecific males (Drosophila melanogaster) can be used to suppress D. suzukii populations through reproductive interference, primarily mediated by post-zygotic isolation mechanisms. Although this approach ensures the absence of via

Genetic analysis of female mating recognition between Drosophila ananassae and Drosophila pallidosa: application of interspecific mosaic genome lines.

Drosophila ananassae and Drosophila pallidosa are closely related species that can produce viable and fertile hybrids of both sexes, although strong sexual isolation exists between the two species. Females are thought to discriminate conspecific from heterospecific males based on their courtship songs. The genetic basis of female discrimination behavior was analyzed using isogenic females from interspecific mosaic genome lines that carry homozygous recombinant chromosomes. Multiple regression analysis indicated a highly significant effect of the left arm of chromosome 2 (2L) on the willingness

Daily variation of nociceptive responses in Drosophila.

The circadian clock regulates a wide range of physiological and behavioral rhythms. Daily variation in nociceptive responsiveness to harmful stimuli has been demonstrated in mammalian models; however, whether such variation is present in non-mammalian species including insects has remained unclear. Here, we found that behavioral responses to noxious heat stimuli in the fruit fly Drosophila melanogaster exhibit significant variation throughout the day. In contrast, null mutants for the clock genes per and tim failed to exhibit daily variation in nociceptive responses. Consistently, we found dai

Slmap Is Required For Spermiogenesis in Drosophila melanogaster.

Spermatogenesis is a conserved process across animals, involving the maintenance and differentiation of germ stem cells, haploid spermatid production via meiosis, and morphologically distinct sperm formation during spermiogenesis. We previously demonstrated that Slmap (Sarcolemma-associated protein) knockdown causes male sterility with no sperm in the seminal vesicle. Here, we show that Slmap knockdown in the early Drosophila melanogaster germline led to severe spermiogenesis defects, including aberrant paracrystalline material deposition in both mitochondrial derivatives, fusion of the two de

Determinants of dicentric chromosome breakage in Drosophila.

Eukaryotic genomes often have fragile sites where chromosomes are particularly prone to break. In Drosophila, when dicentric ring chromosomes try to segregate, they break at nonrandom hotspots. Here, we precisely map breakage hotspots produced by dicentric ring chromosomes in Drosophila. Our study provides three key results about the nature of dicentric chromosome breakage. First, duplications produced by dicentric ring chromosome breakage are surprisingly complex and involve many structural rearrangements, indicating that healing of these breaks is not a simple process. Second, characterizati

The Use of Drosophila to Understand Psychostimulant Responses.

The addictive properties of psychostimulants such as cocaine, amphetamine, methamphetamine, and methylphenidate are based on their ability to increase dopaminergic neurotransmission in the reward system. While cocaine and methamphetamine are predominately used recreationally, amphetamine and methylphenidate also work as effective therapeutics to treat symptoms of disorders including attention deficit and hyperactivity disorder (ADHD) and autism spectrum disorder (ASD). Although both the addictive properties of psychostimulant drugs and their therapeutic efficacy are influenced by genetic varia

Drosophila Trachea as a Novel Model of COPD.

COPD, a chronic obstructive pulmonary disease, is one of the leading causes of death worldwide. Clinical studies and research in rodent models demonstrated that failure of repair mechanisms to cope with increased ROS and inflammation in the lung leads to COPD. Despite this progress, the molecular mechanisms underlying the development of COPD remain poorly understood, resulting in a lack of effective treatments. Thus, an informative, simple model is highly valued and desired. Recently, the cigarette smoke-induced Drosophila COPD model showed a complex set of pathological phenotypes that resembl

Regulation of Nucleolar Dominance in Drosophila melanogaster.

In eukaryotic genomes, ribosomal RNA (rRNA) genes exist as tandemly repeated clusters, forming ribosomal DNA (rDNA) loci. Each rDNA locus typically contains hundreds of rRNA genes to meet the high demand of ribosome biogenesis. Nucleolar dominance is a phenomenon whereby individual rDNA loci are entirely silenced or transcribed, and is believed to be a mechanism to control rRNA dosage. Nucleolar dominance was originally noted to occur in interspecies hybrids, and has been shown to occur within a species (i.e, nonhybrid context). However, studying nucleolar dominance within a species has been c
