Acetylcholine facilitation, atropine block of synaptic excitation of cortical neurons.

Sinigle neurons in cat visual cortex were driven by afferent stimuli to determine the effects of drugs liberated from multibarreled micropipettes on synaptic transmission. Acetylcholine increased, and atropine decreased, the number of neuron discharges fired in response to stimulation of the mesencephalic reticular formation without necessarily affecting responses to other stimuli or affecting spontaneous firing.

Nitrogen Monoxide Releasing Nitric Ester Derivatives of Ibuprofen and Naproxen as COX Inhibitors, Anti-Inflammatory and Hypolipidemic Compounds.

Nitric esters are among the compounds that can liberate nitrogen monoxide (NO) in the organism. Due to the vasodilatation caused by nitrogen monoxide, NO-donors have been shown to protect endothelial function, acting as vasodilators, promoting efficient oxygen supply to tissues, to lower blood pressure, and to inhibit platelet aggregation. Incorporation of a NO-liberating moiety in the structure of non-steroidal anti-inflammatory drugs results in anti-inflammatory agents that are safer for the gastrointestinal system. In this research, ibuprofen and naproxen, two commonly applied non-steroidal

The direct gas chromatographic analysis of barbiturates as their sodium salts.

The addition of formic acid to the carrier gas will permit the on-column liberation of the free acids following the direct injection of barbiturates as their sodium salts. These free acids may then be resolved upon an Apiezon L column: the presence of formic acid also minimizes the adsorption effects which may be associated with the gas chromatographic analysis of these relatively polar drugs.

Dual temperature-sensitive liposome-in-liposome structure for advanced drug delivery systems.

Hierarchical liposome-in-liposome (LIL) structures enable sophisticated multi-stage drug delivery, their application is however often limited by complex, time-consuming fabrication and the use of organic solvents. Here we report a simple two-step hydration (TSH) method for fabricating stable LIL architectures in aqueous media within 2 h. This process employs ultrasonication for the formation of structurally robust inner liposomes (IL) and gentle hydration for their subsequent encapsulation within outer liposomes (OL), achieving high-quality dual-spacing configurations. Direct drug milling in organic PLGA solution facilitates the encapsulation of nanosized drug into PLGA microparticles.

The objective of this study was to prepare poly(lactide-co-glycolide) (PLGA) microparticles loaded with nanosized drug by combining non-aqueous wet bead milling and microencapsulation. 200-300 nm dexamethasone, hydrocortisone and dexamethasone sodium phosphate nanosuspensions were successfully prepared by wet bead milling the drug in dichloromethane using PLGA as a stabilizer. PLGA microparticles loaded with nanosized drugs were then prepared by a solid-in-oil-in-water (S/O/W) solvent evaporation method or solid-in-oil-in-oil (S/O/O) organic phase separation method. The microparticles were cha

Smart stimuli-responsive hydrogels for drug delivery in periodontitis treatment.

Periodontitis is a chronic inflammatory disease initiated by pathogenic biofilms and host immunity that damages tooth-supporting tissues, including the gingiva, periodontal ligament and alveolar bone. The physiological functions of the oral cavity, such as saliva secretion and chewing, greatly reduce the residence of therapeutic drugs in the area of a periodontal lesion. In addition, complex and diverse pathogenic mechanisms make effectively treating periodontitis difficult. Therefore, designing advanced local drug delivery systems and rational therapeutic strategies are the basis for successf

Biocompatibility and Physiological Thiolytic Degradability of Radically Made Thioester-Functional Copolymers: Opportunities for Drug Release.

Being nondegradable, vinyl polymers have limited biomedical applicability. Unfortunately, backbone esters incorporated through conventional radical ring-opening methods do not undergo appreciable abiotic hydrolysis under physiologically relevant conditions. Here, PEG acrylate and di(ethylene glycol) acrylamide-based copolymers containing backbone thioesters were prepared through the radical ring-opening copolymerization of the thionolactone dibenzo[c,e]oxepin-5(7H)-thione. The thioesters degraded fully in the presence of 10 mM cysteine at pH 7.4, with the mechanism presumed to involve an irrev

Carboxymethyl starch encapsulated 5-FU and DOX co-loaded layered double hydroxide for evaluation of its in vitro performance as a drug delivery agent.

Achieving a new oral drug delivery system with controlled drug release behavior is valuable in cancer therapy. Therefore, for the first time, doxorubicin (DOX) and 5-fluorouracil (5-Fu) were simultaneously co-loaded on the as-synthesized layered double hydroxides LDH(MgAl). The resulted system was encapsulated with carboxymethyl starch to improve its efficiency for colon cancer therapy. Several characterization techniques were used to evaluate the successful synthesis of the CMS@LDH(MgAl)@DOX,5-Fu microspheres. The scanning electron microscopy result showed that the size of prepared microspher

Fabrication and Evaluation of Transdermal Delivery of Carbamazepine Dissolving Microneedles.

This project aims to prepare hydrogel microneedle patches (MNs) as a painless method to deliver carbamazepine transdermally. This can be used as a sustained release system that offers the advantages of lower gastrointestinal side effects and avoids the first-pass metabolism of the drug. MNs were composed of two medicated layers, a microneedle layer and a base layer. MNs were fabricated using polyvinyl alcohol with or without polyvinylpyrrolidone Kollidon 30 as a matrix polymer and in the presence of selected solubilizing agent (polyethylene glycol 400, Tween 80, or α-tocopherol polyethylene gl

Controlled drug delivery with nanoassemblies of redox-responsive prodrug and polyprodrug amphiphiles.

Self-assembled nanostructures are highly promising for controlled delivery of therapeutic and diagnostic agents. However, for conventional drug delivery nanosystems, there exist some intrinsic limitations such as low drug-loading contents, premature and burst release, nanocarrier matrix-associated toxicity and immunogenicity, and poor shelf stability. To address these issues, the covalent integration of active drug molecules into prodrug and polyprodrug amphiphiles and fabrication of self-delivery nanomedicines via controlled molecular self-assembly have emerged as a new paradigm. Moreover, it

Poly(styrenesulfonate)-Modified Ni-Ti Layered Double Hydroxide Film: A Smart Drug-Eluting Platform.

Drug-eluting stents (DESs) are widely used in the palliative treatment of many kinds of cancers. However, the covered polymers used in DESs are usually associated with stent migration and acute cholecystitis. Therefore, developing noncovered drug-loading layers on metal stents is of great importance. In this work, Ni-Ti layered double hydroxide (Ni-Ti LDH) films were prepared on the surface of nitinol via hydrothermal treatment, and the LDH films were further modified by poly(styrenesulfonate) (PSS). The anticancer drug doxorubicin could be effectively loaded onto the modified films, and drug

Valproic acid induced liver injury: An insight into molecular toxicological mechanism.

Valproic acid (VPA) is an anti-seizure drug that causes idiosyncratic liver injury. 2-propyl-4-pentenoic acid (Δ4VPA), a metabolite of VPA, has been implicated in VPA-induced hepatotoxicity. This review summarizes the pathogenesis involved in VPA-induced liver injury. The VPA induce liver injury mainly by i) liberation of Δ4VPA metabolites; ii) decrease in glutathione stores and antioxidants, resulting in oxidative stress; iii) inhibition of fatty acid β-oxidation, inducing mitochondrial DNA depletion and hypermethylation; a decrease in proton leak; oxidative phosphorylation impairment and ATP

Cell-Penetrating Drug Carrier by Molecular Recognition of Sphingomyelin on Plasma Membranes.

Plasma membranes not only maintain the intracellular microenvironment through their phospholipid bilayer but also eliminate exogenous compounds outside the cell membranes. Most drugs especially with high polarity are prevented from entering into cells to exert their effects. Therefore, it is of great significance to design effective drug carriers with a penetrating ability toward plasma membranes. In this study, a dual-templated MIP (dt-MIPs) carrier with controllable microstructure and high drug loading capacity was prepared using highly expressed sphingomyelin on the plasma membrane and teno

Hydrogels for localized chemotherapy of liver cancer: a possible strategy for improved and safe liver cancer treatment.

The systemic drug has historically been preferred for the treatment of the majority of pathological conditions, particularly liver cancer. Indeed, this mode of treatment is associated with adverse reactions, toxicity, off-target accumulation, and rapid hepatic and renal clearance. Numerous efforts have been made to design systemic therapeutic carriers to improve retention while decreasing side effects and clearance. Following systemic medication, local administration of therapeutic agents allows for higher 'effective' doses with fewer side effects, kidney accumulation, and clearance. Hydrogels

Balancing Solid-State Stability and Dissolution Performance of Lumefantrine Amorphous Solid Dispersions: The Role of Polymer Choice and Drug-Polymer Interactions.

Amorphous solid dispersions (ASDs) are of great interest due to their ability to enhance the delivery of poorly soluble drugs. Recent studies have shown that, in addition to acting as a crystallization inhibitor, the polymer in an ASD plays a role in controlling the rate of drug release, notably in congruently releasing formulations, where both the drug and polymer have similar normalized release rates. The aim of this study was to compare the solid-state stability and release performance of ASDs when formulated with neutral and enteric polymers. One neutral (polyvinylpyrrolidone-vinyl acetate
