Tumor-Specific T Cell Dysfunction Is a Dynamic Antigen-Driven Differentiation Program Initiated Early during Tumorigenesis.

CD8(+) T cells recognizing tumor-specific antigens are detected in cancer patients but are dysfunctional. Here we developed a tamoxifen-inducible liver cancer mouse model with a defined oncogenic driver antigen (SV40 large T-antigen) to follow the activation and differentiation of naive tumor-specific CD8(+) T (TST) cells after tumor initiation. Early during the pre-malignant phase of tumorigenesis, TST cells became dysfunctional, exhibiting phenotypic, functional, and transcriptional features similar to dysfunctional T cells isolated from late-stage human tumors. Thus, T cell dysfunction seen

Coupled information diffusion--pest dynamics models predict delayed benefits of farmer cooperation in pest management programs.

Worldwide, the theory and practice of agricultural extension system have been dominated for almost half a century by Rogers' "diffusion of innovation theory". In particular, the success of integrated pest management (IPM) extension programs depends on the effectiveness of IPM information diffusion from trained farmers to other farmers, an important assumption which underpins funding from development organizations. Here we developed an innovative approach through an agent-based model (ABM) combining social (diffusion theory) and biological (pest population dynamics) models to study the role of

Fertility programs for lactating dairy cows: A novel presynch + timed artificial insemination program (Double E-Synch) produces similar ovarian dynamics, synchronization, and fertility as Double-Ovsynch.

Fertility programs were implemented for the first postpartum timed artificial insemination (TAI) in 800 (primiparous and multiparous) lactating dairy cows, evaluating 2 presynchronization (presynch) strategies and 2 TAI protocols, in a 2 × 2 factorial arrangement. Weekly, cows were enrolled into 1 of 4 groups (Ovs+Ovs [Double-Ovsynch], Ovs+OvsP4/E2, PreP4/E2+Ovs, and PreP4/E2+OvsP4/E2 [Double E-Synch]). On d -17 (34 ± 3 DIM), the Ovsynch [Ovs] presynch was initiated with 10 µg of buserelin acetate (GnRH), and cows received 0.5 mg of cloprostenol sodium PGF2α analog (PGF) on d -10, and 10 µg of

EXPRESS: Programmed Oscillatory Oxygen Therapy: Reframing Oxygen as a Dynamic Signal for Restoring Cerebrovascular Regulation After Ischemic Stroke.

Successful recanalization after ischemic stroke does not always restore adequate microvascular perfusion within previously ischemic tissue, partly because persistent microcirculatory dysfunction, impaired neurovascular coupling, and perfusion-metabolism mismatch continue to disrupt cerebral blood flow regulation. Conventional continuous oxygen therapy primarily increases systemic oxygen availability, but may be insufficient to restore impaired cerebrovascular responsiveness, while excessive or prolonged oxygen exposure may increase oxidative stress and attenuate adaptive oxygen-sensitive pathw

Dual-dynamic carboxymethyl chitosan hydrogel with photothermal properties and programmed radical/NO release properties for enhanced healing of infected wounds.

Bacterial-infected wound healing still faces substantial challenges, including multidrug-resistance, uncontrollable bleeding, long-term inflammation, and impaired angiogenesis. However, current treatment strategies cannot meet the complex demands of skin wound healing. We here developed a dual dynamic CFP hydrogel based on Schiff base and borate ester bonds between carboxymethyl chitosan (CMCS), polyvinyl alcohol (PVA), and 2-formylphenylboronic acid (2-FPBA). Meanwhile, customized L-arginine/epigallocatechin-3-gallate/ferric ion (EAF) nanoparticles and the thermal-labile 2,2'-azobis[2-(2-imid

The multiplex crosstalk between non-coding RNAs, programmed cell death and related mechanisms: a dynamic duo in hematological malignancies.

Hematological malignancies, including leukemia, lymphoma, and multiple myeloma (MM), are cancers originating in the hematopoietic system, characterized by diverse pathogenesis and clinical features. Non-coding RNAs (ncRNAs) such as microRNAs (miRNAs) and long non-coding RNAs, play critical roles in regulating gene expression and influencing cell fate. Accumulating evidence indicates that ncRNAs are key modulators of programmed cell death (PCD) pathways, affecting tumor development, progression, and drug resistance in these malignancies. PCD, a precisely programmed and regulated cell death proc

Multimodal Exercise Program of Balance and Strength Improves Dynamic Balance, Strength and Functionality and Decreases the Risk of Falls in Adults With Haemophilia.

INTRODUCTION: Haemophilia is a hereditary bleeding disorder that leads to joint damage and musculoskeletal impairments, affects balance and increases the risk of falls. Although exercise is beneficial, little is known about the clinical effects of programmes that focus on balance and proprioception in people with haemophilia (PwH). The aim of this study is to evaluate the effectiveness of a 12-week multimodal balance and strength training programme (MBSTP) on balance, fall risk, strength, functionality, kinesiophobia and quality of life (QoL) in adult PwH. METHODS: A single-blind non-randomiz

Disrupting microtubule dynamics to rewire apoptosis, differentiation, and EMT programs in oral squamous cell carcinoma through indenamine derivatives.

Oral squamous cell carcinoma (OSCC) remains a major global health burden with limited treatment options and high rates of recurrence. In this study, we screened 22 synthesized indenamine derivatives designed to target microtubules and identified three lead compounds-SSB2, SSB21, and SSB22 showing potent cytotoxicity against OSCC cells while sparing normal epithelial cells. These compounds induced G2/M cell cycle arrest, disrupted microtubule organization, and triggered apoptotic cell death. Transcriptomic analysis revealed significant downregulation of oncogenic and cell cycle-related genes an

Effects of a warm-up program on jump-landing pattern and lumbopelvic function in female basketball players with dynamic knee valgus.

Dynamic knee valgus (DKV), commonly observed during functional movements, is recognized as a key biomechanical factor contributing to the risk of anterior cruciate ligament (ACL) injury. Given its importance in injury prevention, this study aimed to investigate the effects of a warm-up program on jump-landing pattern and lumbopelvic function in female basketball players exhibiting dynamic knee valgus. The present study employed a quasi-experimental design. Thirty female basketball players with DKV were screened using the single-leg landing (SLL) test and subsequently randomized into control (n

Validating the Predictions of a Dynamic Transmission Model Using Real-World Data from a Universal Varicella Vaccination Program in Germany.

Dynamic transmission models (DTMs) have been used to estimate various aspects of the public health impact of varicella vaccination programs. The aim of this study was to validate the predictions of a DTM-developed using the typical approach to varicella modeling-using real-world data from a country with a long-term universal varicella vaccination (UVV) program and to assess the sensitivity of the predictions to changes in model input parameters. A compartmental, age-stratified DTM was developed using the settings corresponding to the existing UVV program in Germany. The model-predicted total n

Differential expression of paralog RNA binding proteins establishes a dynamic splicing program required for normal cerebral cortex development.

Sam68 and SLM2 are paralog RNA binding proteins (RBPs) expressed in the cerebral cortex and display similar splicing activities. However, their relative functions during cortical development are unknown. We found that these RBPs exhibit an opposite expression pattern during development. Sam68 expression declines postnatally while SLM2 increases after birth, and this developmental pattern is reinforced by hierarchical control of Sam68 expression by SLM2. Analysis of Sam68:Slm2 double knockout (Sam68:Slm2dko) mice revealed hundreds of exons that respond to joint depletion of these proteins. More

The impact of social network dynamics on engagement in drug use reduction programs among men and women who use drugs.

BACKGROUND: Cross-sectional studies have shown strong relationships between social network characteristics and substance use disorder (SUD) treatment engagement. The current study examined associations between longitudinal changes in egocentric social networks of male and female people who use drugs (PWUD) and engagement in drug use reduction programs, broadly defined as either formal SUD treatment or self-help groups. METHOD: Using data from an HIV prevention and care study in Baltimore, MD, this study categorized PWUD into those who engaged and did not engage in any drug use reduction progr

Dynamic single-cell landscape reveals a T-cell-suppressed microenvironment and a conserved neutrophil program in the lung metastatic niche.

While the immune landscape of primary tumors is increasingly understood, the dynamic immune remodeling within the evolving lung metastatic microenvironment remains poorly characterized. Here, by systematically comparing the microenvironments of primary and metastatic tumors using single-cell transcriptomics across diverse cancer models, we reveal that lung metastases consistently establish a profoundly T-cell-suppressed microenvironment. Concurrently, we identified a strictly metastasis-specific neutrophil gene program. Crucially, this program is highly concurrent with the metastatic microenvi

Exhausted CD8+ T cell fate is programmed by dynamic CTCF-mediated enhancer activation and invariant CTCF-imposed barriers.

Exhausted CD8+ T (TEX) cells undergo extensive genome reorganization during differentiation, yet the drivers of this process remain elusive. Here we show that CTCF programmed CD8+ TEX cell fates through two distinct modes of action. CTCF acquired de novo binding sites and concordantly induced open chromatin in early CD8+ TEX cells responding to chronic viral infection. The dynamic CTCF binding activated enhancers and promoted chromatin looping. Consequently, genetic ablation of CTCF diminished chromatin accessibility and interaction strength, impairing CD8+ TEX cell proliferation, effector fun

Dynamic programmed death 1 expression by virus-specific CD8 T cells correlates with the outcome of acute hepatitis B.

BACKGROUND & AIMS: Recent studies have shown that programmed death 1 (PD-1) expression can impair virus-specific CD8 T-cell responses during chronic viral infection, including hepatitis B virus (HBV). This study aimed to characterize the PD-1 expression during acute hepatitis B (AHB) and further address whether and how the PD-1-mediated pathway balances antiviral immunity versus immunopathology, possibly contributing to disease progression. METHODS: Peripheral and intrahepatic PD-1 expression was investigated longitudinally in 23 human HLA-A2-positive patients with acute hepatitis B. Four pat

Effects of long-term treatment with bovine somatotropin on follicular dynamics and subsequent oocyte and blastocyst yield in an OPU-IVF program.

Fourteen Holstein-Friesian heifers between 15 and 22 months of age with normal reproductive tracts were used in an experimental set up to investigate the effect of a long term rBST-treatment on the follicular population prior to transvaginal ovum pick-up (OPU). The estrous cycle of the animals was synchronized by means of a double injection of 2 ml cloprostenol 11 days apart. The heifers were divided in 2 equal groups (n = 7) in which the animals had the same average body weight, one group receiving a weekly subcutaneous injection of 640 mg recombinantly derived bovine somatotropin (rBST) on M
