Selective modulation of P-glycoprotein activity by steroidal saponines from Paris polyphylla.

Bio-guided fractionation of the roots of Paris polyphylla (Trilliaceae), based on inhibition of P-glycoprotein-mediated daunorubicin efflux in K562/R7 cell line, led to isolation and identification of the three saponins 3-O-Rha(1-->2)[Ara(1-->4)]Glc-pennogenine, gracillin and polyphyllin D, and the two ecdysteroids 20-hydroxyecdysone and pinnatasterone. These compounds were tested for multidrug reversion on P-glycoprotein (ABCB1) with both drug-selected and transfected cell lines, and also on Breast Cancer Resistance Protein (BCRP/ABCG2). By contrast to a weak efficiency on BCRP, the three sap

Juvenile hormone-induced histone deacetylase 3 suppresses apoptosis to maintain larval midgut in the yellow fever mosquito.

SignificanceJuvenile hormone (JH), a sesquiterpenoid, regulates many aspects of insect development, including maintenance of the larval stage by preventing metamorphosis. In contrast, ecdysteroids promote metamorphosis by inducing the E93 transcription factor, which triggers apoptosis of larval cells and remodeling of the larval midgut. We discovered that JH suppresses precocious larval midgut-remodeling by inducing an epigenetic modifier, histone deacetylase 3 (HDAC3). JH-induced HDAC3 deacetylates the histone H4 localized at the promoters of proapoptotic genes, resulting in the suppression o

Tick pheromones and their use in tick control.

Tick pheromones that regulate assembly, attraction/aggregation/attachment, and mating behavior have been described. Most of the compounds regulating these behaviors are purines, substituted phenols, or cholesteryl esters. Other pheromonal compounds include organic acids, hematin, or ecdysteroids. Novel devices have been developed that combine the specific compounds comprising these pheromones with an acaricide. When applied to tick-infested vegetation or directly to the body surfaces of livestock or companion animals, these devices are effective for tick control. This review summarizes the cur

Restriction of nutrient intake results in the increase of a specific Manduca sexta allatotropin (Manse-AT) mRNA in the larval nerve cord.

The Manduca allatotropin (Manse-AT) gene is expressed as three mRNAs that differ from each other by alternative splicing. The level of one of these mRNAs (RNA-3) is specifically increased in the nerve cord of last instar larvae that were starved, parasitized, or fed the ecdysteroid agonist RH-5992. Each of these treatments results in reduction of feeding and increased levels of juvenile hormone (JH). The normal decline in JH biosynthesis by the corpora allata does not occur in starved or RH-5992-fed larvae. The increase in RNA-3 levels has the capacity to increase the production of Manse-AT an

Exogenous 20-hydroxyecdysone induces epidermal carbonic anhydrase but inhibits exoskeletal calcification in the post-ecdysial blue crab, Callinectes sapidus.

The crustacean exoskeleton is composed primarily of chitin, proteins and various inorganic compounds. It is the inorganic compounds, such as calcium and magnesium, that underlie the exoskeletal mineralization process following ecdysis. Little is known about the hormonal mechanism for this process in crustaceans. Carbonic anhydrase (CA) in the epidermis has been suggested to aid in deposition of calcium and magnesium carbonates to the exoskeleton in crustaceans by generating bicarbonate ions, resulting in mineralization. Given a similar pattern of fluctuation in prevalence between epidermal CA

Ovary ecdysteroidogenic hormone functions independently of the insulin receptor in the yellow fever mosquito, Aedes aegypti.

Most mosquito species must feed on the blood of a vertebrate host to produce eggs. In the yellow fever mosquito, Aedes aegypti, blood feeding triggers medial neurosecretory cells in the brain to release insulin-like peptides (ILPs) and ovary ecdysteroidogenic hormone (OEH). Theses hormones thereafter directly induce the ovaries to produce ecdysteroid hormone (ECD), which activates the synthesis of yolk proteins in the fat body for uptake by oocytes. ILP3 stimulates ECD production by binding to the mosquito insulin receptor (MIR). In contrast, little is known about the mode of action of OEH, wh

[Role of Schistosoma mansoni bilharziasis in male hypogonadism].

In tropical and subtropical areas, schistosomiasis may cause anatomic anomalies of the genital organs responsible for permanent or reversible infertility. Furthermore, it has been suggested that this parasitic infection may have adverse consequences on the endocrine system. To specify the effects of schistosomiasis on endocrine function, production of pituitary gonadotropins and testosterone in rats and hamsters experimentally infected with Schistosoma mansoni was studied. Prior to infection, hormone levels were within the normal range for our laboratory in all animals studied. Hormone levels

Demonstration of protein kinase C activity in crustacean Y-organs, and partial definition of its role in regulation of ecdysteroidogenesis.

Ecdysteroid-producing Y-organs from the crab Cancer antennarius were shown to possess enzyme activity that was stimulated in vitro by addition of Ca2+, phosphatidylserine, or the protein kinase C activator, phorbol 12-myristate 13-acetate (PMA; ED50, 4 nM). In the presence of calcium and phosphatidylserine, PMA increased protein kinase C activity dose-dependently to a maximum 4-fold increase at 100 nM PMA. Stimulated protein kinase C activity was unaffected by calmodulin (100 nM) but was inhibited by 100 nM trifluoperazine. Pretreatment of cultured Y-organ segments with PMA elevated basal prot

Ecdysone Elicits Chronic Renal Impairment via Mineralocorticoid-Like Pathogenic Activities.

BACKGROUND/AIMS: Ecdysteroids are steroidal insect molting hormones that also exist in herbs. Ecdysteroid-containing adaptogens have been popularly used to improve well-being and by bodybuilders for muscle growth. However, the use of ecdysone in mammals is also associated with kidney growth and enlargement, indications of disturbed kidney homeostasis. The underlying pathogenic mechanism remains to be clarified. METHODS: Virtual screening tools were employed to identify compounds that are homologous to ecdysone and to predict putative ecdysone-interacting proteins. The kidney effect of ecdyson

A cell surface protein controls endocrine ring gland morphogenesis and steroid production.

Identification of signals for systemic adaption of hormonal regulation would help to understand the crosstalk between cells and environmental cues contributing to growth, metabolic homeostasis and development. Physiological states are controlled by precise pulsatile hormonal release, including endocrine steroids in human and ecdysteroids in insects. We show in Drosophila that regulation of genes that control biosynthesis and signaling of the steroid hormone ecdysone, a central regulator of developmental progress, depends on the extracellular matrix protein Obstructor-A (Obst-A). Ecdysone is pr

[Phytotherapeutic aspects of diseases of the circulatory system. 6. Leuzea carthamoides (WILLD.) DC: the status of research and possible use of the taxon].

Leuzea carthamoides (WILLD.) DC., Asteraceae, has become an important taxon usable in human practice and animal production in recent years. Originally an endemic plant of southern Siberia, now it is widely grown not only in the Union of Independent States, once the original region of introduction, but also in central and eastern Europe. The principal constituents of the whole plant are ecdysteroids (5 beta-cholest-6-on-7-ene derivatives) and flavonoids. In addition, the aerial part contains sesquiterpenic lactones of the guajanolide type, and the roots contain polyines (on the thiophene base).

Functional Characterization of Eccyp307a1 in Early Ovary Development of Exopalaemon carinicauda.

According to a previous study, in insects, cyp307A1 plays a central role in ecdysteroid synthesis, which is a member of the cytochrome P450 family. However, the function of cyp307A1 in crustaceans remains unclear. In this study, we explored the function of Eccyp307a1 in Exopalaemon carinicauda through a series of experiments. The sequence of Eccyp307a1 encoded 529 amino acids, and the protein was found to possess typical P450 domains and heme-binding sites. The mRNA of Eccyp307a1 was expressed at a higher level during the early stages of ovary development, but was expressed less during the mat

Supreme glutathione-dependent ketosteroid isomerase in the yellow-fever transmitting mosquito Aedes aegypti.

The steroid hormone ecdysone is essential for the reproduction and survival of insects. The hormone is synthesized from dietary sterols such as cholesterol, yielding ecdysone in a series of consecutive enzymatic reactions. In the insect orders Lepidoptera and Diptera a glutathione transferase called Noppera-bo (Nobo) plays an essential, but biochemically uncharacterized, role in ecdysteroid biosynthesis. The Nobo enzyme is consequently a possible target in harmful dipterans, such as disease-carrying mosquitoes. Flavonoid compounds inhibit Nobo and have larvicidal effects in the yellow-fever tr

Endocrine strategies for the control of ectoparasites and insect pests.

The increasing knowledge about endocrine mechanisms in arthropods facilitates the biorational search for drugs against insect pests and parasites that interfere with arthropod hormone action. Juvenile hormone mimics have been successfully applied for about 20 years; however, resistance to juvenile hormone analogues has developed. The introduction of moulting hormone agonists, which compete for binding to the ecdysteroid receptor, is expected in the near future. Despite the considerable progress that has been achieved in peptide hormone research during the last few years, no successful insectic

Croquemort elicits activation of the immune deficiency pathway in ticks.

The immune deficiency (IMD) pathway directs host defense in arthropods upon bacterial infection. In Pancrustacea, peptidoglycan recognition proteins sense microbial moieties and initiate nuclear factor-κB-driven immune responses. Proteins that elicit the IMD pathway in non-insect arthropods remain elusive. Here, we show that an Ixodes scapularis homolog of croquemort (Crq), a CD36-like protein, promotes activation of the tick IMD pathway. Crq exhibits plasma membrane localization and binds the lipid agonist 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphoglycerol. Crq regulates the IMD and jun N-term

Expression of the early-late gene encoding the nuclear receptor HR3 suggests its involvement in regulating the vitellogenic response to ecdysone in the adult mosquito.

The insect steroid hormone, 20-hydroxyecdysone (20E), is a key factor controlling critical developmental events of embryogenesis, larval molting, metamorphosis, and, in some insects, reproduction. We are interested in understanding the molecular basis of the steroid hormone ecdysone action in insect egg development. The yellow fever mosquito, Aedes aegypti, in addition to being an important vector of human diseases, represents an outstanding model for studying molecular mechanisms underlying egg maturation due to stringently controlled, blood meal-activated reproductive events in this insect.

Ovary ecdysteroidogenic hormone activates egg maturation in the mosquito Georgecraigius atropalpus after adult eclosion or a blood meal.

The rockpool mosquito, Georgecraigius atropalpus, is a facultatively autogenous species that produces its first egg clutch without a blood meal shortly after emergence. Several days after depositing this clutch, females must take a blood meal to produce a second egg clutch. Decapitation of females shortly after emergence or blood ingestion prevents egg maturation. Here, we report that a single injected dose of the neuropeptide ovary ecdysteroidogenic hormone (OEH) fully restored egg maturation in decapitated females in both circumstances. This neuropeptide and two insulin-like peptides (ILPs)

Pre-molt cholesterol regulation of ecdysone receptor (EcR) signaling in the Y-organ contributes to dietary control of molting in Scylla paramamosain.

Molting death syndrome (MDS), characterized by incomplete ecdysis and failed shell hardening, severely constrains mud crab (Scylla paramamosain) aquaculture. Cholesterol is an essential precursor for ecdysteroid synthesis, but its stage-specific roles and endocrine targets during the molt cycle remain unclear. Here we examined how dietary cholesterol and ecdysone receptor (EcR) signaling interact to regulate molting physiology and MDS risk. Juvenile crabs were fed a basal diet (0% cholesterol added) or a diet supplemented with 0.75% cholesterol (air-dry basis) and followed through a complete m

Metabolomic profiling reveals deep chemical divergence between two morphotypes of the zoanthid Parazoanthus axinellae.

Metabolomics has recently proven its usefulness as complementary tool to traditional morphological and genetic analyses for the classification of marine invertebrates. Among the metabolite-rich cnidarian order Zoantharia, Parazoanthus is a polyphyletic genus whose systematics and phylogeny remain controversial. Within this genus, one of the most studied species, Parazoanthus axinellae is prominent in rocky shallow waters of the Mediterranean Sea and the NE Atlantic Ocean. Although different morphotypes can easily be distinguished, only one species is recognized to date. Here, a metabolomic pro
