In Vivo Assessment of Alveolar Macrophage Efferocytosis Following Ozone Exposure.

Ozone (O3) is a criteria air pollutant that exacerbates and increases the incidence of chronic pulmonary diseases. O3 exposure is known to induce pulmonary inflammation, but little is known regarding how exposure alters processes important to the resolution of inflammation. Efferocytosis is a resolution process, whereby macrophages phagocytize apoptotic cells. The purpose of this protocol is to measure alveolar macrophage efferocytosis following O3-induced lung injury and inflammation. Several methods have been described for measuring efferocytosis; however, most require ex vivo manipulations.

Mer-mediated eosinophil efferocytosis regulates resolution of allergic airway inflammation.

BACKGROUND: Eosinophils play a central role in propagation of allergic diseases, including asthma. Both recruitment and retention of eosinophils regulate pulmonary eosinophilia, but the question of whether alterations in apoptotic cell clearance by phagocytes contributes directly to resolution of allergic airway inflammation remains unexplored. OBJECTIVES: In this study we investigated the role of the receptor tyrosine kinase Mer in mediating apoptotic eosinophil clearance and allergic airway inflammation resolution in vivo to establish whether apoptotic cell clearance directly affects the re

Lysosomal Cholesterol Hydrolysis Couples Efferocytosis to Anti-Inflammatory Oxysterol Production.

RATIONALE: Macrophages face a substantial amount of cholesterol after the ingestion of apoptotic cells, and the LIPA (lysosomal acid lipase) has a major role in hydrolyzing cholesteryl esters in the endocytic compartment. OBJECTIVE: Here, we directly investigated the role of LIPA-mediated clearance of apoptotic cells both in vitro and in vivo. METHODS AND RESULTS: We show that LIPA inhibition causes a defective efferocytic response because of impaired generation of 25-hydroxycholesterol and 27-hydroxycholesterol. Reduced synthesis of 25-hydroxycholesterol after LIPA inhibition contributed to

Restoring cigarette smoke-induced impairment of efferocytosis in alveolar macrophages.

Cigarette smoke has been associated with susceptibility to different pulmonary and airway diseases. Impaired alveolar macrophages (AMs) that are major phagocytes in the lung have been associated with patients with airway diseases and active smokers. In the current report, we show that exposure to second-hand cigarette smoke (SHS) significantly reduced efferocytosis in vivo. More importantly, delivery of recombinant granulocyte-macrophage colony-stimulating factor (GM-CSF) to the alveolar space restored and refurbished the efferocytosis capability of AMs. Exposure to SHS significantly reduced e

Microglia efferocytosis: an emerging mechanism for the resolution of neuroinflammation in Alzheimer's disease.

Alzheimer's disease (AD) is a complex neurodegenerative disorder characterized by significant neuroinflammatory responses. Microglia, the immune cells of the central nervous system, play a crucial role in the pathophysiology of AD. Recent studies have indicated that microglial efferocytosis is an important mechanism for clearing apoptotic cells and cellular debris, facilitating the resolution of neuroinflammation. This review summarizes the biological characteristics of microglia and the mechanisms underlying microglial efferocytosis, including the factors and signaling pathways that regulate

Efferocytosis: Current status and future prospects in the treatment of autoimmune diseases.

Billions of apoptotic cells are swiftly removed from the human body daily. This clearance process is regulated by efferocytosis, an active anti-inflammatory process during which phagocytes engulf and remove apoptotic cells. However, impaired clearance of apoptotic cells is associated with the development of various autoimmune diseases, such as rheumatoid arthritis, systemic lupus erythematosus, and inflammatory bowel disease. In this review, we conducted a comprehensive search of relevant studies published from January 1, 2000, to the present, focusing on efferocytosis, autoimmune disease path

The Efferocytosis-Driven Macrophage Polarization: A Concerto in the Tumor Immunosuppressive Microenvironment.

Efferocytosis plays a critical role in maintaining tissue homeostasis and controlling inflammation. However, in the tumor microenvironment, this process is hijacked to promote immune suppression. Treatments like radiotherapy and chemotherapy lead to widespread tumor cell death, which triggers efferocytosis and drives macrophages into an immunosuppressive state, allowing the tumor to evade the immune system and resist treatment. Nonetheless, the way efferocytic signals alter macrophage function and promote protumor effects during the recognition, engulfment, and digestion of apoptotic cells rem

Efferocytosis related KCTD12 is a clinico-immune target in lung adenocarcinoma.

BACKGROUND: Efferocytosis, the clearance of apoptotic cells by phagocytes, contributes to immune homeostasis but may also promote tumor immune tolerance. However, its transcriptional landscape and clinical relevance in lung adenocarcinoma (LUAD) remain incompletely understood. METHODS: We systematically analyzed efferocytosis-associated genes across TCGA and multiple GEO datasets to classify LUAD subtypes and construct a prognostic risk model. The prognostic and immunological relevance of this model was validated in four independent cohorts and further assessed through immune infiltration, ge

Efferocytosis: the art of cellular clearance and novel perspectives in disease therapy.

Efferocytosis, the process of apoptotic cell clearance, is a fundamental biological mechanism for maintaining tissue homeostasis. However, its role in disease pathogenesis is often oversimplified, neglecting a critical knowledge gap: how the single process could drive opposing pathological outcomes. This review provides a comprehensive analysis centered on the functional duality of efferocytosis. By synthesizing evidence across a spectrum of human pathologies-from atherosclerosis and neurodegeneration to cancer-we establish a core paradigm: impaired efferocytosis is a central pathogenic driver

Identification and analysis of key genes related to efferocytosis in colorectal cancer.

UNLABELLED: The impact of efferocytosis-related genes (ERGs) on the diagnosis of colorectal cancer (CRC) remains unclear. In this study, efferocytosis-associated biomarkers for the diagnosis of CRC were identified by integrating data from transcriptome sequencing and public databases. Finally, the expression of biomarkers was validated by real-time quantitative polymerase chain reaction (RT-qPCR). Our study may provide a reference for CRC diagnosis. BACKGROUND: It has been shown that some efferocytosis related genes (ERGs) are associated with the development of cancer. However, it is still un

A multifunctional nanoparticle for efferocytosis and pro-resolving-mediated endometriosis therapy.

Endometriosis is an inflammation-dependent disorder characterized by the abnormal growth of endometrium-like lesions. In recent years, there is a great deal of interest in the development of anti-inflammatory therapy. Herein, an acid-sensitive calcium carbonate nanoparticle (CaNP) incorporated BML-111 (BML@CaNP) was prepared. BML@CaNP acted as a Ca2+ nanomodulator for efferocytosis (macrophages engulf apoptotic cells). Specifically, BML@CaNP induced the apoptosis of endometriotic stromal cells and enhanced the efferocytosis of macrophages. In addition, the particle can also deliver BML to the

Circulating cochlin LCCL domain binds to dying cells and enhances efferocytosis.

Clearance of dead cells by efferocytosis is a critical process for homeostasis, notably by limiting inflammation. Defective efferocytosis has been associated with autoimmune, neurodegenerative, and cardiovascular diseases, as well as chronic infections, making its regulation a potential therapeutic target. Here, we identify circulating cochlin LCCL (Limulus factor C, Cochlin, and Lgl1) domain as a regulator of efferocytosis. Using cell binding assay for recombinant cochlin LCCL domain, we establish its tropism for dead or dying cells of both immune and non-immune lineages from murine and human

Rosa26-INDIA Identifies Early Apoptotic B Cells Before Macrophage-Mediated Efferocytosis.

Apoptotic cells are removed by efferocytosis, raising the possibility that in vivo apoptotic cell frequencies underestimate cell death. Rosa26-INDIA is an apoptosis reporter that detects activated caspase-3 (Casp3) via Förster resonance energy transfer (FRET) loss, but the relationship between Casp3 activation, phosphatidylserine exposure, membrane permeabilization, and efferocytic clearance remains unclear. We combined live-cell imaging and macrophage depletion to evaluate apoptotic B cell detection by Rosa26-INDIA. Live-cell imaging revealed that FRET loss preceded loss of detectable INDIA,

Targeting efferocytosis for tissue regeneration: From microenvironment reprogramming to clinical translation.

Efferocytosis, phagocytic clearance of apoptotic cells (ACs), is an essential biological process that resolves inflammation and regulates tissue regeneration in various organ systems. Through removal of apoptotic cell debris, efferocytosis attenuates secondary necrosis and dampens the release of damage-associated molecular patterns (DAMPs). More importantly, it can reprogram phagocytes towards a pro-reparative phenotype via the secretion of anti-inflammatory mediators, metabolic rewiring, and the production of growth factors. There are four closely regulated stages in the entire process: "find

Baoganning decoction mitigates liver fibrosis via regulating IDO1-mediated macrophage efferocytosis.

BACKGROUND: Liver fibrosis poses a significant global health challenge with limited treatments. Baoganning (BGN) decoction, a traditional Chinese medicine formula administered for liver fibrosis, has shown promise in preliminary studies by targeting indoleamine 2,3-dioxygenase 1 (IDO1), a key immunoregulator in the liver microenvironment. However, the precise underlying mechanisms remain unclear. PURPOSE: This study aimed to elucidate the mechanism by which BGN alleviates liver fibrosis via IDO1-mediated regulation of macrophage function. METHODS: The anti-fibrotic effects of BGN were evalua

Irgm1 Improves Postinfarction Cardiac Repair by Promoting Neutrophil Clearance and Efferocytosis.

Delayed neutrophil clearance after myocardial infarction (MI) significantly disrupts the myocardial microenvironment, but the underlying mechanisms remain unclear. Macrophage-mediated efferocytosis of infiltrating neutrophils is crucial for resolving inflammation and restoring homeostasis post-MI. However, the specific regulatory mechanisms governing neutrophil clearance and efferocytosis remain undefined. This study demonstrates a significant correlation between increased IRGM expression in peripheral blood neutrophils of patients with MI and improved prognostic outcomes. Neutrophil-specific

FASN-dependent lipogenesis controls macrophage efferocytosis in alcohol-associated liver disease.

BACKGROUND & AIMS: Defective clearance of apoptotic hepatocytes contributes to inflammation of the liver and progression of alcohol-associated liver disease (ALD), but mechanisms regulating macrophage efferocytosis during alcohol exposure remain unclear. We investigated whether fatty acid synthase (FASN)-dependent lipid metabolism controls hepatic macrophage efferocytosis in ALD. METHODS: Human liver tissues from patients with alcohol-related cirrhosis (AC) and controls (n = 18 per group), together with experimental ALD mouse models (six per group), were analyzed for hepatocyte apoptosis and

NLRP3 inflammasome constrains liver regeneration through impairing MerTK-mediated macrophage efferocytosis.

The NOD-like receptor protein 3 (NLRP3) inflammasome plays a crucial role in human acute and chronic liver diseases. However, the role and cell-specific contribution of NLRP3 in liver regeneration remains unclear. Here, we found that NLRP3 was highly activated during the early stage of liver regeneration via 70% partial hepatectomy (PHx) mice model and clinical data. Global NLRP3 depletion or pharmacologically blocking NLRP3 significantly enhanced liver regeneration, while NLRP3 overexpression impaired it after PHx. Furthermore, mice with myeloid-specific knockout of Nlrp3 (Nlrp3Δmye), rather
