Iminocoumarin-based low affinity fluorescent Ca2+ indicators excited with visible light.

A series of iminocoumarin-based fluorescent Ca2+ indicators were synthesized and the spectral profiles of their free and Ca2+ bound forms were studied. The newly-synthesized compounds incorporate the Ca2+ chelating structure of BAPTA. The chromophore moieties are iminocoumarins substituted at the 3-position with benzothiazolyl, benzoxazolyl and benzimidazolyl groups. These compounds are excited with visible light and their Ca2+ dissociation constants range from 5.4 to 27.5 microM. Fluorescence spectra studies of these probes indicated a clear shift in their excitation wavelength maxima upon Ca

Two actions of calcium regulate the supply of releasable vesicles at the ribbon synapse of retinal bipolar cells.

Ribbon synapses of sensory neurons are able to sustain high rates of exocytosis in response to maintained depolarization, but it is not known how this is achieved. Using the capacitance technique, we have found that Ca(2+) regulates the supply of releasable vesicles at the ribbon synapse of depolarizing bipolar cells from the retina of goldfish. Ca(2+) had two actions that could be differentiated by introduction of the Ca(2+) chelator EGTA; one action stimulated refilling of the rapidly releasable pool of vesicles from a reserve pool, and a second action stimulated recruitment of vesicles to t

Cell cycle dependent variation of calmodulin in Tetrahymena.

The level of calmodulin (CaM), a ubiquitous calcium-binding protein of eukaryotic cells was determined at different phases of the cell cycle in a synchronized Tetrahymena population. It was found that the concentration of CaM at G1 was approximately half of the concentration of S and this 2 x G1 level of CaM was maintained through the G2 and M stages of the cell cycle. To ascertain the role of CaM in the initiation of DNA synthesis, the cells were treated with trifluoperazine (TFP), a CaM antagonist, and EGTA (Ca2+-chelator) at the G1/S boundary. It was found that DNA synthesis was inhibited i

Intracellular hydrolysis of EGTA-esters.

It has been established that the hydrolysis of EGTA-acetoxymethylester (AME) by red blood cells is about the tenth of the hydrolysis of acetylthiocholine (Ac-S-Ch). This splitting of AME could be inhibited by about 50% by prostigmine at a concentration of 0.75 X 10(-5) mol/l, while the splitting of Ac-S-Ch was totally inhibited by the same prostigmine concentration. The hydrolysis of AME by the so-called white-ghost preparation was considerably inhibited by prostigmine (KI = 5 X 10(-8) mol/l), and this inhibition proved to be a competitive one. The splitting of AME by membrane-free cytosol fra

Effects of tityustoxin (TsTX) from scorpion venom on the release and synthesis of acetylcholine in brain slices.

Tityustoxin (TsTX) increased the release and synthesis of acetylcholine (ACh) in slices of rat brain hippocampus, hypothalamus, thalamus, cortex and striatum. The effect was highest in slices of hippocampus and cortex and smallest in hypothalamus. These effects of TsTX were dependent on the presence of Na+ and Ca2+ in the incubation medium. EGTA, 0.1 mM, blocked the effect of TsTX in all areas, except for the hippocampus and frontal striatum, where a concentration of 10 mM was required. Tetrodotoxin blocked the increase in the release and synthesis of ACh induced by tityustoxin.

Chemistry and biological applications of photo-labile organic molecules.

Photo-labile molecules have been widely used not only in organic synthesis but also in biological study. The chemistry of the typical photo-labile organic molecules, including their structure, mechanism underlying their photo-lability and strategies to integrate them with biomolecules, is reviewed to illustrate the structural basis for photo-activable caged compounds. Biological applications of representative photo-labile caged molecules were also illustrated for a general understanding on the important roles of caged compounds in dynamic biological studies. This tutorial review would provide

Ionomycin, a carboxylic acid ionophore, transports Pb(2+) with high selectivity.

Studies utilizing phospholipid vesicle loaded with chelator/indicators for polyvalent cations show that ionomycin transports divalent cations with the selectivity sequence Pb(2+) > Cd(2+) > Zn(2+) > Mn(2+) > Ca(2+) > Cu(2+) > Co(2+) > Ni(2+) > Sr(2+). The selectivity of this ionophore for Pb(2+) is in contrast to that observed for A23178 and 4-BrA23187, which transport Pb(2+) at efficiencies that are intermediate between those of other cations. When the selectivity difference of ionomycin for Pb(2+) versus Ca(2+) was calculated from relative rates of transport, with either cation present indiv

BAPTA-AM decreases cellular pH, inhibits acidocalcisome acidification and autophagy in amino acid-starved T. brucei.

To investigate the role of Ca2+ signaling in starvation-induced autophagy in Trypanosoma brucei, the causative agent of human African trypanosomiasis, we used cell-permeant Ca2+ chelator BAPTA-AM and cell impermeant chelator EGTA, and examined the potential involvement of several intracellular Ca2+ signaling pathways in T. brucei autophagy. The results showed an unexpected effect of BAPTA-AM in decreasing cellular pH and inhibiting acidocalcisome acidification in starved cells. The implication of these results in the role of Ca2+ signaling and cellular/organellar pH in T. brucei autophagy is d

Enhancement in corneal permeability of riboflavin using calcium sequestering compounds.

Ethylenediaminetetraacetic acid, ethylenediamine-N,N'-disuccinic acid and ethylene glycol-bis(2-aminoethylether)-N,N,N',N'-tetraacetic acid are polyaminocarboxylic acids that are able to sequester metal ions. Calcium is implicated in maintenance of intercellular matrix, zonula occludens (tight junctions) and zonula adherens of epithelium and endothelium cells. Corneal epithelium is impervious to many aqueous formulations due to it being lipophilic, whereby transcellular drug transit is resisted, whilst tight junctions restrict access via the paracellular route. Research has shown that integrit

Spontaneous haemolytic activity of Atlantic halibut (Hippoglossus hippoglossus L.) and sea bass (Dicentrarchus labrax) serum.

The spontaneous haemolytic (SH) activity of sera was compared in groups of cultured halibut and sea bass. The optimum assay temperature was determined for each species and different red blood cell donors were tested. The effects of heat inactivation, storage temperature and of different agents like EDTA, EGTA, yeast cell components and bacterial LPS were compared. Halibut sera gave optimum lysis with sheep red blood cells (RBC) at 16 degrees C whereas sea bass sera showed optimum lysis with rabbit RBC at 37 degrees C. The haemolytic activity of halibut sera was inactivated at 45 degrees C whil

Direct modulation of volume-regulated anion channels by Ca(2+) chelating agents.

Ca(2+) chelating agents are widely used in biological research for Ca(2+) buffering. Here we report that BAPTA, EDTA and HEDTA produce fast, reversible, voltage-dependent inhibition of swelling-activated Cl(-) current (I(Cl,swell)) in LNCaP prostate cancer epithelial cells that is unrelated to their Ca(2+) binding. BAPTA was the most effective (maximal blockade 67%, IC(50)=70 microM, at +100 mV) followed by EDTA and HEDTA. I(Cl,swell) blockade by EDTA was pH-dependent. BAPTA blocked I(Cl,swell) also in other cell types. We conclude that Ca(2+) chelating agents block I(Cl,swell) by acting direc

In vitro and in vivo protein phosphorylation in Avena sativa L. coleoptiles: effects of Ca2+, calmodulin antagonists, and auxin.

In vitro and in vivo protein phosphorylations in oat (Avena sativa L.) coleoptile segments were analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis and by two-dimensional gel electrophoresis. In vitro phosphorylation of several polypeptides was distinctly promoted at 1 to 15 micromolar free Ca2+ concentrations. Ca2(+)-stimulated phosphorylation was markedly reduced by trifluoperazine, chlorpromazine, and naphthalene sulfonamide (W7). Two polypeptides were phosphorylated both under in vitro and in vivo conditions, but the patterns of phosphorylation of several other polypeptid

The role of divalent cations in epidermolysis.

In experiments designed to elucidate the role of divalent cations in maintaining the integrity of the epidermis, newborn mouse skin was incubated in ethylenediamine tetraacetic acid (EDTA) or ethylene glycol tetraacetic acid (EGTA). In EDTA, epidermolysis occurred and was confirmed by rubbing the specimen to demonstrate that a sheet of epidermis could be split off. After 30 min incubation in 0-01 mol/EDTA the split occurred in the lower granular-upper spinous layer; after 45 min, it was in a spinous-suprabasilar location and at 60 min and later at the dermal-epidermal junction. The physiological effects of induced prepartum subclinical hypocalcemia in multiparous Holstein cows.

Subclinical hypocalcemia (SCH) is prevalent during the periparturient period in dairy cows due to increased calcium (Ca) demand for milk and colostrum synthesis. Transient SCH has been shown to activate a negative feedback loop, thereby improving Ca homeostasis. Here, we investigate the impact of induced SCH during the prepartum period on Ca homeostasis pre- and postpartum, and milk production. Thirty multiparous Holstein cows were enrolled 21 d before the expected due date, in a randomized complete block design, and were assigned to receive either a 6-h continuous intravenous infusion of sali

Origins of Ca2+ Imaging with Fluorescent Indicators.

In 1980, Roger Tsien published a paper, in this journal [Tsien, R. Y. (1980) Biochemistry, 19 (11), 2396], titled "New calcium indicators and buffers with high selectivity against magnesium and protons: design, synthesis, and properties of prototype structures". These new buffers included 1,2-bis(o-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid, or BAPTA, which is still widely used today. And so, the world was set alight with new ways in which to visualize Ca2+. The ability to watch fluctuations in intracellular Ca2+ revolutionized the life sciences, although the fluorescent indicators used to

Adsorption of Cd(II) and Pb(II) by a novel EGTA-modified chitosan material: kinetics and isotherms.

In this study, a novel adsorbent was synthesized by functionalizing chitosan with ethylene glycol-bis(2-aminoethylether)-N,N,N',N'-tetraacetic acid (EGTA) ligands. The adsorption capability of EGTA-modified chitosan was investigated by the removal of Cd(II) and Pb(II) from aqueous solutions. The adsorption and regeneration studies were performed by batch techniques. The effects of pH, contact time, and initial metal concentration were studied. Metal uptake by EGTA-chitosan was 0.74 mmol g(-1) for Cd(II) and 0.50 mmol g(-1) for Pb(II). The adsorption mechanism, that the adsorbent formed octahed

Calcium regulates independently ciliary beat and cell contraction in Paramecium cells.

Intracellular Ca(2+) concentration is a well-known signal regulator for various physiological activities. In many cases, Ca(2+) simultaneously regulates individual functions in single cells. How can Ca(2+) regulate these functions independently? In Paramecium cells, the contractile cytoskeletal network and cilia are located close to each other near the cell surface. Cell body contraction, ciliary reversal, and rises in ciliary beat frequency are regulated by intracellular Ca(2+) concentration. However, they are not always triggered simultaneously. We injected caged calcium into Paramecium caud

Modulation of phosphoenolpyruvate carboxylase in vivo by Ca2+ in Amaranthus hypochondriacus, a NAD-ME type C4 plant: possible involvement of Ca2+ in up-regulation of PEPC-protein kinase in vivo.

The properties of phosphoenolpyruvate carboxylase (PEPC) were studied, with respect to calcium (Ca2+), in leaves of Amaranthus hypochondriacus, a C4 plant. Experiments were conducted in vitro (by adding Ca2+ during enzyme assay) or in vivo (by feeding Ca2+ to intact leaves through petiole). Inclusion of 10 microM Ca2+ during assay marginally increased (<30%) malate sensitivity of PEPC in extracts from dark-adapted leaves. The effect of Ca2+ was marginal on PEPC in extracts from illuminated leaves. Upon applying a low concentration of Ca2+ to leaves, the PEPC activity in leaves increased by 1.5
