Circulating saturated fatty acids and incident chronic kidney disease: a meta-analysis of de-novo prospective cohort investigations.

BACKGROUND: Chronic kidney disease (CKD) is a global health problem which is associated with poor outcomes, and its prevalence is expected to increase. Identifying novel risk factors for CKD may lead to improved outcomes. Circulating saturated fatty acids (SFAs) have been posited as contributors to CKD risk. OBJECTIVES: We aimed to evaluate associations between circulating SFAs (measured in phospholipids in 7 cohorts, serum or plasma total in 5 cohorts, and cholesterol esters in 1 cohort) and incident CKD in 13 cohorts, and to pool results by meta-analysis across the studies. METHODS: SFAs w

Modulation of the metabiome by rifaximin in patients with cirrhosis and minimal hepatic encephalopathy.

UNLABELLED: Hepatic encephalopathy (HE) represents a dysfunctional gut-liver-brain axis in cirrhosis which can negatively impact outcomes. This altered gut-brain relationship has been treated using gut-selective antibiotics such as rifaximin, that improve cognitive function in HE, especially its subclinical form, minimal HE (MHE). However, the precise mechanism of the action of rifaximin in MHE is unclear. We hypothesized that modulation of gut microbiota and their end-products by rifaximin would affect the gut-brain axis and improve cognitive performance in cirrhosis. Aim To perform a systems

Essential fatty acid deficiency in parenterally fed preterm infants.

To determine the incidence of essential fatty acid (EFA) deficiency during short term fat-free parenteral nutrition, the authors investigated prospectively the EFA status of nine low birthweight (1145 +/- 343 g) preterm (28.2 +/- 1.9 weeks) infants, in whom delivery of dietary fat was delayed postnatally for 2-9 days. Serial determinations of plasma fatty acids showed that during fat-free alimentation, the major EFA, linoleic acid (LA), decreased rapidly (-0.75% total fatty acids per day), accompanied by a rise in endogenously produced non-essential fatty acid, eicosatrienoic acid (Mead acid).

[Changes in fatty acids composition in cerebrospinal fluid of patients with myelopathy].

The composition of fatty acids in the cerebrospinal fluid of patients with cervical myelopathy, thoracic myelopathy, cauda equina syndrome, and spinal cord injury were measured by gas chromatography. The relationship between the change in fatty acids composition and the improvement of symptoms and pathology were studied. The percent distribution of eicosadienoic acid (C20: 2) in fatty acids composition was found to be significantly increased in the cerebrospinal fluid of patients with cervical and thoracic myelopathies (p less than 0.05), and with incomplete spinal cord injury (p less than 0.0

Peroxisomes in infantile phytanic acid storage disease: a cytochemical study of skin fibroblasts.

Cultured skin fibroblasts from six patients demonstrating clinical signs and biochemical characteristics of infantile phytanic acid storage disease (IPSD) were examined by electron microscopy, using cytochemical procedures for the demonstration of peroxisomal catalase activity. In four of the six fibroblast cell lines peroxisomes strongly reactive for catalase were present in numbers similar to those found in normal fibroblasts. Liver biopsy tissue from one of these patients showed no typical hepatic peroxisomes, but did show small, marginally reactive bodies. In two other IPSD fibroblast cell

Phytanic acid oxidase deficiency in childhood.

Tables 2 and 5 summarize the major clinical and biochemical findings in these patients. Cases 1 and 2 resemble clinically the previous cases of children reported as suffering from infantile phytanic acid storage disease, Zellweger's disease, or neonatal adrenoleucodystrophy. Cases 3 and 4 differ strikingly from these and from one another. Numerous questions remain unanswered, but it seems likely that these patients have in common defects in peroxisomal function which are related but not identical. Why some patients with phytanic acid oxidase deficiency do not have significant elevation of seru

Activated neutrophils release mediators that may contribute to myocardial injury and dysfunction associated with ischemia and reperfusion.

Neutrophils accumulate in the ischemic myocardium and exacerbate postischemic cardiac dysfunction and injury. The formation of lipoxygenase metabolites of AA, derived either directly from the neutrophils or by interactions with other blood elements or cells, may promote neutrophil-mediated injury. Recognition of the roles played by neutrophils and AA metabolites in reperfusion injury may lead to the development of new therapies that can be used in conjunction with thrombolytic drugs to reduce the complications associated with restoring blood flow to the ischemic heart.

The cerebro-hepato-renal (Zellweger) syndrome: lamellar lipid profiles in adrenocortical, hepatic mesenchymal, astrocyte cells and increased levels of very long chain fatty acids and phytanic acid in the plasma.

Clinical, radiological, histological and biochemical aspects of two cases of cerebro-hepato-renal syndrome (CHRS) are reported. CT scan disclosed a demyelinating process and gyral abnormalities reflecting the observed neuropathological findings. Trilamellar and lamellar inclusions were found in brain astrocytes, hepatic mesenchymal and adrenal cells. The morphologic features of these inclusions are similar to those observed in childhood adrenoleukodystrophy, neonatal adrenoleukodystrophy and infantile Refsum's disease. In the two CHRS patients, increased plasma levels of very long chain fatty

Inhibitors of the arachidonic acid cascade in the management of ocular inflammation.

At the present time, corticosteroids are still the most effective class of drugs for the treatment of ocular inflammation. However, since their prolonged use may result in severe ocular side effects, it would be therapeutically beneficial to develop nonsteroidal anti-inflammatory drugs that have similar or greater efficacy than steroids, but do not share their ocular side effects. Several currently available non-steroidal drugs have been used clinically as prophylactic or therapeutic agents for the following: 1. Prevention of pupillary constriction during intraocular surgery (cataract extracti

Phytanic acid oxidase activity in cultured skin fibroblasts. Diagnostic usefulness and limitations.

Patients with Refsum's disease lack the ability to degrade phytanic acid to pristanic acid and CO2. This defect is expressed in fibroblasts from the patients. An assay system for the degradation of phytanic acid in cultured skin fibroblasts is described. The assay makes it possible to single out patients with Refsum's disease from the cob-web of clinically related conditions. The sensitivity is, however, not good enough to diagnose the heterozygous state. A defect of the same pronounced degree as in Refsum's disease is also found in fibroblasts from patients with Zellweger's syndrome, neonatal

Transcellular metabolism of eicosanoids.

In this chapter, current concepts of eicosanoid biochemistry and function have been summarized. Emphasis has been placed on the importance of ascertaining a complete profile of eicosanoids synthesized by a given tissue under varying conditions of stimulation. The concept of transient intermediates, which are involved in each of the known pathways, was reviewed. Such intermediates can also represent substrate for cell-cell interactions, which have been classified and discussed. Knowledge of the synthetic mechanisms, metabolism, and catabolism of eicosanoids has surpassed our comprehension of th

Pharmacology and significance of nonsteroidal anti-inflammatory drugs in the treatment of skin diseases.

Intensive studies of the molecular pathways involved in common inflammatory skin disorders, coupled with detailed pharmacologic evaluation of the responses of skin to the end products of these pathways, have resulted in a much clearer understanding of the mode of action of nonsteroidal anti-inflammatory drugs. In particular the development of lipoxygenase inhibitors is prompting intense interest in their possible role as anti-inflammatory agents in psoriasis and other dermatoses. Because of the potency of these and other classes of new anti-inflammatory drugs, careful monitoring of their pharm

Viral-bacterial pneumonia in calves: effects on plasma eicosanoids and long chain fatty acids.

In the pathogenesis of bovine pneumonic pasteurellosis, immunodepression induced by stress or respiratory viral infection permits superinfection of the lungs with Pasteurella hemolytica, which results in exudative fibrinous pneumonia. Therefore, bovine pneumonic pasteurellosis was induced by sequential inoculations of calves with bovine herpes virus-1 (BHV-1, 3 X 10(7) tissue culture infectious dose 50 (TCID50)/nostril), followed 3 days later by challenge with P. hemolytica (15 X 10(9) colony-forming units (cfu) intratracheally). To study the pathogenic mechanisms of the disease, we examined t

Eicosanoid synthesis by rabbit hydronephrotic cortical interstitial cells in culture.

Rabbit hydronephrotic cortical interstitial cells in primary culture were labeled with [1-14C]arachidonic acid and the eicosanoids released after stimulation with bradykinin or A23187 were studied by reverse-phase high performance liquid chromatography. The major arachidonic acid metabolite formed was prostaglandin (PG)E2, comprising more than 30% of the total radioactivity released. 12-Hydroxyheptadecatrienoic acid, probably representing spontaneous breakdown of the cyclic endoperoxides PGG2 and/or PGH2, made up 10 to 15% of the radioactivity released. Other cyclooxygenase products that were

Essential fatty acid deficiency depletes rat glomeruli of resident macrophages and inhibits angiotensin II-induced eicosanoid synthesis.

Essential fatty acid (EFA) deficiency exerts a beneficial effect on immune-mediated glomerulonephritis, preventing both the tissue injury and consequent mortality. Because both macrophages and eicosanoids are thought to play pathogenic roles in glomerulonephritis, and because macrophages play an important role in modulating arachidonate metabolism at sites of renal injury, the effects of EFA deficiency on the population of resident glomerular macrophages and on glomerular eicosanoid generation were examined. EFA deficiency led to a striking reduction in the number of resident glomerular macrop

Chemotactic peptide stimulation of leukotrienes from healthy and inflamed rabbit colons.

Prostaglandins, thromboxanes and leukotrienes are increased in human and experimental colitis. To evaluate the biosynthesis of these eicosanoids, colon inflammation was induced in rabbits by formalin enema followed by i.v. immune complexes, and the distal colon was perfused ex vivo. Bradykinin increased synthesis of prostaglandin E2 and thromboxane B2 more from colitis than from control colons (both P less than .001) but had no effect on leukotriene synthesis. The inflammatory cell agonist N-formylmethionyl-leucyl-phenylalanine (30 ng) also induced greater synthesis of prostaglandin E2 (70 +/-

Biological effects of Avicennia marina (Forssk.) vierh. extracts on physiological, biochemical, and antimicrobial activities against three challenging mosquito vectors and microbial pathogens.

Mosquitoes are principal vector of several vector-borne diseases affecting human beings leading to thousands of deaths per year and responsible for transmitting diseases like malaria, dengue, chikungunya, yellow fever, Zika virus, Japanese encephalitis, and lymphatic filariasis. In the present study, we evaluated the different solvent extracts of mangrove Avicennia marina for their toxicity against larvae of three major mosquito vectors, as well as selected microbial pathogens. The larvicidal mortality of third instars was observed after 24 h. Highest larval mortality was found for the acetone

The ocular effects of eicosanoids and other autacoids: historic background and the need for a broader perspective.

Great progress has been made in describing the synthesis of most known eicosanoids by ocular tissues. This progress has included identification of the spectrum of eicosanoids synthesized by different ocular tissues of different species, as well as comparisons of the relative proportion of individual eicosanoids produced by ocular tissues of different species. In contrast to this wealth of information, the lack of antagonists that are specific to individual eicosanoids has greatly hindered efforts to determine the specific ocular functions of each member of this family of autacoids. Furthermore
